  • The crystal protein gene (cp) of Bacillus tizuringienszs subsp. liuvstaki (B.t.k.) HI173 was subcloned into HanzHI site of central region (Tn5-cp) or BglII site of IS50L region (IS50L-cp) in Tn5, and transposed into the chromosomal DNA of five strains of root-colonizing Pseudomonas. The expression of cp gene in Acwiomoncrs transconjugants was demonstrated by immunoblot analysis and bioassay against larvae of the Hyphantria cunea.

  • Enterobacter sp.B54 which shows antagonistic activity to Phytophthora capsici on potato dextrose agar was selected among 112 strains isolated from Korean soil. After Tn5 lac-induced mutants were obtained through Pl :: Tn5 lac mutagenesis, 2 mutants for loss of antibiosis and 1 mutant for increased antibiosis were screened by using in vitro fungal inhibition assay. When the 3 mutants affected in antibiosis were analyzed by southern hybridization with pRZ102 (ColEl :: Tn5) as a probe, its results suggest that Tn5 lac was randomly inserted into different chromosomal sites in these mutants.

  • The bacteriophage Mu and transposon Tn5 containing plasmid pJB4JI-transferred transposon Tn5 to Caulobuter crescentus. When several thousand of transposon Tn5 insertion mutants were examined, we found auxotrophic and motility mutants at frequencies of 2% and 3%, respectively. Transposition of transposon Tn5 was analyzed by the reverse field electrophoresis and Southern hybridization. The results indicated that transposon Tn5 was randomly inserted to Caulobuter crescentus chromosome but the plasmid vector, pJB4JI, was not maintained.

  • This paper presents results from a study of the age and weight at puberty of Thai does. A randomized block $3{\times}3$ factorial design was used. Factors were genotype (Thai Native; TN, 75% TN $\times$ 25% Anglo-Nubian; AN and 50 TN $\times$ 50% AN does), year of kidding (1989, 1990 and 1991) and birth type (single or twin) as a block. It was shown that there were no significant (p > 0.05) difference in age at puberty among genotypes ($167.6{\pm}6.0$, $157.6{\pm}7.0$ and 160.0 = 4.7 days for TN, 75% TN $\times$ 25% AN and 50% TN $\times$ 50% AN does, respectively) and between birth types. However, does kidded in 1991 had significantly (p < 0.01) longer age at puberty ($194.0{\pm}7.4$ days) than did does kidded in 1989 ($143.0{\pm}4.5$ days) or in 1990 ($148.1{\pm}5.7$ days). There was an interaction effect between genotype and year. Fifty % TN $\times$ 50% AN does had significantly (p < 0.05) higher weights at pubery ($17.2{\pm}0.4kg$) than those of TN($14.3{\pm}0.5kg$ and 75% TN $\times$ 25% AN ($14.4{\pm}0.6kg$) does. There was no significant difference in weights at puberty between TN and 75% TN $\times$ 25% AN does. There was no effect of birth type on weight at puberty. There was an interaction effect between genotype and year of kidding on age and weight at puberty and on growth rate from birth to weaning.

  • The spectinomycin resistant strain of slow growing R. japonicum R-168 was selected to be participated in conjugation with E. coli WA803/pGS9. Tn5 was introduced from suicide vector pGS9 into R. japonicum R-168 $spr^{r}$ chromosome at the frequency of $1.0\times 10^{-5}-5.0\times 10^{-7}$ and the transconjugante were selected on the yeast extract-mannitol plate containing kanamycin ($50{\mu}$g/ml) and spectinomycin ($100{\mu}$g/ml) after 8-9 days incubation. All transconjugants we tested were found to contain Tn 5 DNA on their genome, which was confirmed by Southern hybridization experiments. R. japonicum RNa75, which had been selected through plant test, was found to be defective in symbiotic nitrogen fixing ability and the production of leghemoglobin in soybean nodules formed by the inoculation of this mutant. In addition, this mutant strain hardly developed nitrogenase activity asymbiotically in contrast with the wild type strain, indicating that some nitrogen fixing gene might be blocked in this strain and the production of leghemoglobin could be decreased by the interference in nitrogen fixing genes.

Usefulness of Cardiac Troponin I as a Prognostic Marker in Noncardiac Critically Ill Patients (비순환기계 중환자의 예후인자로서 Cardiac Troponin I의 유용성)

  • Background : Cardiac troponin I (cTnI) is a specific marker of myocardial injury. It is known that a higher level of cTnI is associated with a poor clinical outcome in patients with acute coronary syndrome. An elevation in cTnI is also observed in various noncardiac critical illnesses. This study evaluated whether cTnI is useful for predicting the prognosis in noncardiac critically ill patients. Methods : From June 2003 to July 2004 at Gyeongsang National University Hospital, we enrolled 215 patients (male:142, female:73, mean age:$63{\pm}15$ years ) who were admitted for critical illness other than acute coronary syndrome at the medical intensive care unit(ICU). The severity score of critical illness (SAPS II and SOFA) was determined and serum cTnI level was measured within 24 hours after admission to the ICU. The mortality rate was compared between the cTnI-positive (${\geq}0.1{\mu}g/L$) and cTnI-negative ($cTnI<0.1{\mu}g/L$) patients at the $10^{th}$ and $30^{th}$ day after admission to the ICU. The mean cTnI value was compared between the survivors and non-survivors at the $10^{th}$ and $30^{th}$ day after admission to the ICU in the cTnI-positive patients. The correlation between cTnI and the severity of the critical illness score (SAPS II and SOFA) was also analyzed in cTnI-positive patients. Results : 1) The number of cTnI-negative and positive patients were 95(44%) and 120(56%), respectively. 2) The mortality rate at the $10^{th}$ and $30^{th}$ day after admission to the ICU was significantly higher in the cTnI-positive patients (29%, 41%) than in the cTnI-negative patients (12%, 21%)(p<0.01). 3) In the cTnI-positive patients, the mean value of the cTnI at the $10^{th}$ and $30^{th}$ day after admission to the ICU was significantly higher in the non-survivors ($4.5{\pm}9.2{\mu}g/L$, $3.5{\pm}7.9{\mu}g/L$) than in the survivors($1.8{\pm}3.6{\mu}g/L$, $2.0{\pm}3.9{\mu}g/L$) (p < 0.05). 4) In the cTnI-positive patients, the cTnI level was significantly correlated with the SAPS II score (r=0.24, p<0.001) and SOFA score (r=0.30, p<0.001). Conclusion : The cTnI may be a useful prognostic marker in noncardiac critically ill patients.

Amplified synthesis and stability of Tn5 polypeptides in escherichia coli (대장균에서의 Tn5 단백질 증폭생합성 및 안정성)

  • Plasmid DNA molecules containing strong promoter upstream from IS50L or IS50R, the two insertion sequences that flank Tn5, were constructed to amplify the synthesis of Tn5-encoded polypeptides. When proteins made by cells that contain these plasmids were analyzed on polyacrylamide gels, enhanced synthesis of IS50R polypeptides could be detected. Synthesis of this polypeptide apparently is initiated within the large open reading frame of this element. In addition, the stability of IS50L-and IS50R-encoded polypeptides was analyzed. It was found that IS50L polypeptides are relatively unstable in vivo. This instability could account for the observed inability of this element to promote transposition.

Isolation and Characterization of Tn5 Insertion Mutants of Pseudomonas fluorescens Antagonistic to Rhizoctonia solani (Rhizoctonia solani 길항세균 Pseudomonas fluorescens의 Tn5 삽입 돌연변이주 분리 및 특성)

  • Pseudomonas fluorescens Biovar III strains S-2 antagonistic to Rhizoctonia solani was subjected to Tn5 mutagenesis by the transposon vector pGS9. Ampicillin and kanamycin resistant (Ampr, Kmr) transconjugants were recovered at a frequency of 1.3$\times$10-7 per initial recipient cell, when recipient cells were washed twice in TE buffer before conjugation. Of the ca. 3000 transconjugants, a frequency of noninhibitory (Inh-), nonfluorescent (Flu-) and auxotorphic (Pro-) mutants were 0.27%, 0.47% and 0.40%, respectively. In these mutants, all Inh- mutants showed the same colony morphology as wild type, whereas all Flu- and Pro- mutants inhibited the growth of R. solani. These mutants were also susceptible to chloramphenicol, indicating only the Tn5 element, except for parts of pGS9, was integrated into the recipient genome. In a Southern blot analysis, the Tn5 element inserted into one site on the chromosome for each of the chosen mutants. However, Tn5 insertion sites of Inh-, and Pro- mutants were differed in each other. These indicate that the genes essential for R. solani inhibition, fluorescent production and auxotrophic are chromosomally located, but not linked to each other.

Quality Control on the BOD, COD, TN, TP of Water Quality Pollutant (수질오염물질 BOD, COD, TN, TP의 정도관리(I))

  • Quality control was carried out the three analytes, BOD, COD, TN, TP of high and low concentration for thirteen laboratories. Evaluation values of Quality control was determined from average value which were made by six times test, repeatedly and periodically. This value had not only error ratio of within 5% from prepared sample value but also high precision of standard deviation, 0.44 (BOD, low concentration), 2.15 (BOD, high concentration), 0.12 (COD, low concentration), 1.63 (COD, high concentration), 0.35 (TN, low concentration), 1.99 (TN, high concentration), 0.05 (TP, low concentration), 0.14 (TP, high concentration). Upper (Lower) Warning Limit (ULWL) and Upper (Lower) Acceptance Limit (ULAL) values made from quality control chart. Three (5.30, 9.70, 5.30 mg/L) and five laboratories were over ULAL value in low and high BOD concentration, respectively. Two (41.00, 30.60 mg/L) and four laboratories were over ULAL value in low and high COD concentration, respectively. while, other eleven laboratories appeared reliable data. One laboratory (0.70 mg/L) was over ULAL at low TN concentration, while other eleven laboratories had reliable values. In case of high TN concentration, eleven laboratories were over ULAL value, it should be focuced for improvement of reliability about measurement and analysis of TN. Four (1.14, 0.45, 0.64, 1.49mg/L) and seven laboratories were over ULAL in low and high TP concentration, respectively.

  • Background: Neurovascular compression (NVC) is a well-known cause of trigeminal neuralgia (TN). However, patients with idiopathic TN (ITN) do not have evidence of NVC on magnetic resonance imaging (MRI), and other patients may remain asymptomatic despite evidence of NVC on MRI. This suggests that there may be additional risk factors for TN development other than NVC. Although epidemiological factors, such as age and sex differences, are useful for understanding the pathophysiology of TN, detailed statistics for each TN subtype are currently unavailable. Therefore, this study aimed to classify patients with TN into the following groups based on data extracted from past medical records: classical TN (CTN), secondary TN, and ITN. Methods: The characteristics of the groups and their differences were explored. Results: CTN was more common in women than in men, as previously reported, whereas ITN was more common in men than in women. The ratio of pain sites located on the right side of the face was high in all groups. Patients with CTN were also prone to NVC on the asymptomatic side. Conclusion: By investigating TN subtype, it may be possible to elucidate the pathophysiology of TN. This would greatly improve treatment outcomes.
