Callophyllis japonica extract improves high-fat diet-induced obesity and inhibits adipogenesis in 3T3-L1 cells

  • The anti-obesity potential of an ethanolic extract of the edible red alga Callophyllis japonica extract (CJE) was investigated in mice fed a high-fat diet (HFD). CJE administration into HFD mice revealed suppression of body weight, adipose tissue weight, serum total cholesterol, triglyceride, and glucose levels in a dose-dependent manner. Also, it reduced serum levels of glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, and lactate dehydrogenase, as well as the accumulation of fatty droplets in liver tissue. CJE and its ethyl acetate fraction inhibited adipogenesis in 3T3-L1 adipocytes by down-regulating the adipocyte-specific transcriptional regulators. Taken together, these results suggest that CJE reduces obesity in mice fed an HFD by inhibiting lipid accumulation and adipogenesis in the adipose tissues.

A Laboratory Study of the Effect of Phytoplankton Concentration, Water Flow and Their Interaction on the Growth of the Sandy Shore Suspension Feeding Clam Gafrarium tumidum

  • The effects of water flow rate and phytoplankton concentration on the growth of the sandy shore clam Gafrarium tumidum was investigated in a laboratory flume study using a $3{\times}3$ factorial design. After 60 days, shell length, shell weight and tissue dry weight increased significantly with phytoplankton concentration. For the effect of flow rate, growth was faster when flow rate increased from low to medium level; further increases in flow rate, however, either did not sustain faster growth or resulted in a reduction in growth. The condition index (CI) of a standard-sized clam was significantly higher at low flow rate than at medium and high flow rates and was negatively correlated with phytoplankton concentration. The uncoupled growth of shell and tissue in response to flow rate and phytoplankton concentration may be adaptations to low food environments, so that energy can either be stored to sustain life or reserved for gametogenesis during the reproductive period.

갈근이 비만 랫드 간조직의 비만관련 유전자 발현에 미치는 영향 (The effects of Pueraria lobata extract on gene expression in liver tissue of rat with estrogen-deficient obesity)

  • Objective : It is known that Pueriaria lobata has an anti-osteoporetic effect, anti-cancer effect, anti-pyretic effect, and anti-diabetic effect. The aim of this study was to evaluate anti-obesity effect of Pueriaria lobata extract (PLE), and elucidate the effect of it on gene expression related to lipid metabolism. Method : The experiments were performed with the use of ovariectomized rats as estrogen-deficient obesity model. They were grouped NC (normal control), OC (estrogen-deficient control), PLH (100mg/kg of PLE), PLL (20mg/kg). PLE was orally administered for 6 weeks. Body weights and serum lipid level were estimated, and real-time PCR was performed to investigate the effect of PLE on gene expression in liver. Results : PLE decreased the body weight and serum cholesterol and triglyceride, but increased HDL-cholesterol. And PLE increased leptin, CYP27, CPT1, CYP8B1, ACAT2, LDLR, and SCD1, but reduced $PPAR{\gamma}$, PGC1A, HMG-CoA-R, ACAT1, SCD1, and APoB gene expression in liver tissue of rat with estrogen-deficient obesity. Conclusion : It is concluded that Pueriaria lobata reduced body weight, and its effect was expressed by regulation of gene expression related to lipid metabolism in rats with estrogen-deficient obesity.

가토 기흉에서 본 $^{86}Rb$의 분포 ($^{86}Rb$ Distribution in the Lung of the Rabbit with Pneumothorax)

  • $^{86}Rb$ uptake of some organs and tissues, ego both lungs, both renal cortices. small intestine, liver and skeletal muscle were studied in the control and the rabbit subjected to pneumothorax. $^{86}Rb$ in the form of chloride mixed with physiological saline was intravenously. injected. The doses were $100{\mu}c$ for a rabbit. The rabbits were sacrificed at intervals of 10, 20, 40, and 60 seconds after the injection of $^{86}Rb$, by the injection of saturated KCI solution. After sacrification, the organ and tissue sample were quickly removed. $^{86}Rb$ uptake in gm of the organs and tissues were measured. On the basis of uptake value, administered doses and body weight, % dose/gm tissues per 200 gm body weight was calculated. Followings were the results; 1. Pneumothorax resulted in a marked elevation in $^{86}Rb$ uptake value of collapsed lung and returned to normal level lately. 2. Contralateral lung of pnemothorax also showed marked elevation in $^{86}Rb$ uptake value and recovered to normal level. 3. Initial $^{86}Rb$ uptake value of liver, small intestine of the rabbit with pneumothorax showed some elevation as compared to control, but that of late stage were similar with control. 4. Local blood flow determination by means of $^{86}Rb$ uptake were inadequate in the collapsed lung of pneumothorax. 5. It was suggested that the mechanism for the initial elevation of $^{86}Rb$ uptake value in each organs and tissue were different from each other.

카페인 첨가가 흰쥐 간의 항산화 활성에 미치는 영향 (The Effect of Caffeine on the Antioxidative Activities of Mouse Liver)

  • The purpose of this study was to examine the antioxidative activities and tissue cell of mouse liver added caffeine. The body weight of all experimental groups increased during experimental periods, but the body weight of caffeine-containing groups were lower than those of any other experimental groups. Superoxide dismutase and catalase activities tended to decrease significantly with caffeine-containing groups, but increased in control and ginseng-containing groups. Hydroperoxide contents were increase significantly with caffeine-containing groups. Lipid peroxidation levels decreased in ginseng-containing groups, but it increased significantly with caffeine-containing group. Protein contents were a tendency of similar between control and ginseng-containing groups, but it showed a increasing tendency in caffeine-containing groups. Microscopic observation of mouse liver cell were similar tissue in ginseng and caffeine-containing groups, but it showed somewhat more injuring only at the liver cell of anhydrous caffeine group, and became the suspicion in liver diseases. This results show that antioxidative activities are slightly higher in non-caffeine and ginseng-containing drinks than caffeine-cotaining drinks. From this standpoint, we suggest that too much drinking of caffeine-containing drinks for a long time is undesirable.

Heat-Killed Lactiplantibacillus plantarum LRCC5314 Mitigates the Effects of Stress-Related Type 2 Diabetes in Mice via Gut Microbiome Modulation

  • The incidence of stress-related type 2 diabetes (stress-T2D), which is aggravated by physiological stress, is increasing annually. The effects of Lactobacillus, a key component of probiotics, have been widely studied in diabetes; however, studies on the effects of postbiotics are still limited. Here, we aimed to examine the mechanism through which heat-killed Lactiplantibacillus plantarum LRCC5314 (HK-LRCC5314) alleviates stress-T2D in a cold-induced stress-T2D C57BL/6 mouse model. HK-LRCC5314 markedly decreased body weight gain, adipose tissue (neck, subcutaneous, and epididymal) weight, and fasting glucose levels. In the adipose tissue, mRNA expression levels of stress-T2D associated factors (NPY, Y2R, GLUT4, adiponectin, and leptin) and pro-inflammatory factors (TNF-α, IL-6, and CCL-2) were also altered. Furthermore, HK-LRCC5314 increased the abundance of Barnesiella, Alistipes, and butyrate-producing bacteria, including Akkermansia, in feces and decreased the abundance of Ruminococcus, Dorea, and Clostridium. Thus, these findings suggest that HK-LRCC5314 exerts protective effects against stress-T2D via gut microbiome modulation, suggesting its potential as a supplement for managing stress-T2D.

고수 에탄올 추출물의 고지방식이 비만 동물모델에서의 항비만효과 (Ant-Obesity Effect of Coriandrum sativum L. Ethanol Extract in High Fat-Induced Obesity Animal Model)

  • This study investigated the anti-obesity effects of Coriandrum sativum L. ethanol extracts in a high fat diet-induced obesity model (DIO). We confirmed the anti-obesity effects by analysing the expression of the related proteins, weight gain, dietary intake, dietary efficiency, blood biochemistry, histological analysis and western blot analysis. After oral administration of Coriandrum sativumL. ethanol extracts at concentrations of 250 and 500 mg/kg, a significant improvement in dietary efficiency, reduction in weight gain, triglycerides, total cholesterol and LDL-cholesterol in blood lipid was observed for 8 weeks. In addition, improvement in blood glucose and metabolism confirmed through glucose tolerance test was observed. Further, the concentration of alanine transaminase (ALT) in blood was significantly decreased, which improved the fatty liver caused by high-fat diet intake as confirmed by liver tissue analysis. This phenomenon was confirmed to decrease the expression of fat accumulation-related PPARγ and FAS protein in the liver tissue. Especially, it is believed that FAS, a liposynthetic enzyme, has a stronger inhibitory effect than PPARγ. Therefore, Coriandrum sativum L. ethanol extract is thought to improve obesity by reducing blood lipids levels, improving glucose metabolism and inhibiting synthesis of the fat that accumulates in the liver in high-fat diet-induced obesity animal models.

낙엽송 (Larix leptolepis) 배발생조직의 초저온보존 및 식물체 재분화 (Cryopreservation of Embryogenic Tissue and Plant Regeneration in Larix leptolepis)

  • 본 연구에서는 낙엽송 배발생조직의 장기저장을 위한 가능성에 대하여 조사하였다. 0.4M혹은 20% PEG로 24시간 전 처리하여 -0.33$^{\circ}C$/min의 동결온도 하강율로 초저온시켰을 때 높은 조직생중량을 보인 반면 -0.5 혹은 -1.$0^{\circ}C$/min의 경우 초저온 후 조직의 생중량이 저조한 것으로 나타났다. 1, 7, 및 28일간 초저온보존된 조직을 회수하여 재생장시킨 다음 PCR을 이용하여 변이분석을 한 결과 초저온보존 기간에 관계없이 DNA변이는 전혀 나타나지 않았다. 초저온보존된 배발생 조직으로부터 체세포배 유도 및 식물체 재분화를 유도할 수 `있었다.

Dysregulation of Cannabinoid CB1 Receptor Expression in Subcutaneous Adipocytes of Obese Individuals

  • The endocannabinoid system (ECS) plays a key role in the regulation of appetite, body weight and metabolism. We undertook the present study to further clarify the regulation of the cannabinoid CB1 receptor (CB1, CNR1) in human adipose tissue in obesity. CB1 receptor mRNA expression was ~1.6-fold (p<0.004) and 1.9-fold higher (P<0.05) in subcutaneous adipocytes from obese compared to non-obese subjects in microarray and quantitative real-time PCR studies, respectively. Higher CB1 receptor mRNA expression levels in both adipose tissue (~1.2 fold, P<0.05) and adipocytes (~2 fold, P<0.01) were observed in samples from visceral compared to subcutaneous depots collected from 22 obese individuals. Immunofluorescence confocal microscopy demonstrated the presence of CB1 receptor on adipocytes and also adipose tissue macrophages. These data indicate that adipocyte CB1 receptor is up-regulated in human obesity and visceral adipose tissue and also suggest a potential role for the ECS in modulating immune/inflammation as well as fat metabolism in adipose tissue.

가미마행감석탕 및 포황이 비만유도백서의 체중 및 지질대사에 미치는 영향 억제 (Effects of Gamimahaenggamseok-tang and Typhae Pollen on Body Weight and Lipid Metabolism of Rats with Obesity induced by High Fat Diets)

  • The purpose of this study is to examine the effects of Gamimahaenggamseok-tang and Typhae Pollen on Body Weight and Lipid Metabolism of Rats with Obesity induced by high fat diet. Sixty four male Sprague-Dawley rats, around 250g of B.W., were divided into normal, control, sample A and sample B groups. Normal groups were orally administrated with saline and normal animal feed, and control groups with saline and high fat diet, sample A with liquid extract of Gamimahaenggamseok-tang and high fat diet, and sample B with liquid extract of Typhae Pollen and high fat diet. After 4 and 8 weeks administration, body weight, epididymal fat pad weight, weight of liverㆍspleenㆍkidney, abdominal skin fold thickness, feed intake, and lipid level in liver tissue on morphological examination; total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, free fatty acid, and phospholipid on serum examination were measured in all of the groups.
