The Effects of Insambackhotang on Mouse with Diabetic Nephropathy Induced by Alloxan (인삼백호탕(人蔘白虎湯)이 Alloxan으로 유발(誘發)된 mouse의 당뇨병성(糖尿病性) 신증(腎症)에 미치는 영향(影響))

  • To investigate recovery effects of Insambackhotang, which have been used clinically in diabetes therapny, on kidney failure of a diabetes-induced mouse by Alloxan administration, body and kidney weight changes of mice, BUN, creatinine, glucose and MDA level in serum, MDA level in kidney tissue. 1. A hyperglycemia(250-400mg/dl) mouse induced by Alloxan(75mg/kg) showed significant decline of kidney function: increase of BUN and creatinine in serum, excretion of glucose, protein, ketone in urine were observed at 4 days after the treatment. 2. Increase of the mouse body and kidney weight and a ratio of the kidney/body weight of Insambackhotang treated group as compared to the control group was significantly inhibited. 3. The BUN, creatinine level in serum of Insambackhotang treated group as compared to the control group were significantly inhibited. 4. The MDA level in serum and kidney tissue of Insambackhotang treated group as compared to the control group were significantly inhibited.

  • This study was attempted to observe the effect of Job's tear diet on change of body weight and tissue in rat. The changes of body weight were higher in Job's tear non-diet group(A) and seemed to lower in Job's tear diet group(c) as expected. The weight of each organ appeared to heavy more C group than A,Bgroup in Brain, Spleen. Lungs and Kidney. Liver was heavier in Job's tear to diet one day among six day group(B) and ovary was lighter in C group, The levels of each organ TBA-value were higher in Brain, Liver of A group and showed to higher Spleen, Lungs, Ovary of C group TBA-value of Kidney was revealed to lower more C group than A,B group.

The Effects of Capsaicin Intake with High-Fat Diet on Tissue Glycogen Contents in Exercise-Trained Rats (캡사이신 첨가 고지방식이가 운동시 조직 글리코겐 농도에 미치는 영향)

  • This study is to investigate the effects of capsaicin with high-fat diet on tissue glycogen contents in exercise-trained rats. Forty male Sprague-Dawley rats were offered a high-fat diet for 2 wks in individual cages and were exercise-trained by a animal treadmill running throughout the experimental period. After 2 wks of the prefeeding with high-fat diet, the rats were divided into two group: high-fat diet group(CON)and high-fat diet + capsaicin(0.014%) group(CAP). The rats were killed by decapitation at 10 hr(rest), 1 hr and 2 hr after treadmill running(27m/min, 6$^{\circ}$). Body weight and epididymal adipose tissure weight were significantly lower in CAP than in CON, but soleus muscle weight was not different between the two groups. Glycogen contents in liver, soleus and gastrocnemius white muscles were significantly lower in CAP than in CON at rest, 1 hr and 2 hr (p<0.05). However, glycogen content in gastrocnemius red muscle was significantly higher in CAP compared with CON at 2 hr after the exercise(p<0.05). These results indicate that capsaicin intake with high-fat diet would decrease glycogen contents in liver and muscle, however, this effect on glycogen metabolism could be changed by muscle type.

Effect of Cadmium and Lead with Single or Mixed Admin stration on Rat Bones (카드뮴 및 납의 수여가 백서골성분에 미치는 영향)

  • This study was carried out in order to clarify the combined effect of cadmium and lead on rat bones when exposed to single metal (1 mg cd/kg body weight) and cambined metals (1 mg cd + 4 mg pb/kg body weight). Seventy five mature rats of Sprague-Dawley species were divided into a control group and a treatment group that were administered by daily peritoneum injection for 7 to 9 weeds, and their body weights were measured every week. The results were summarized as follows 1. Body weight gains of the combined injection group and the single injection groups were lower than that of the control group. 2. In case of the combined injection group, the amount of cadmium accumulated in femur was more than that of the cadmium group. These amounts of cadmium accumulated showed an increasing trend. 3. The lead amount in bone tissue of the combined injection group presented also an increasing trend. In this case, the additive action of cadmium to the lead accumulation in bone tissue was conspicuous. 4. The amounts of calcium and phosphorus in femur showed clear a decreasing trend in the cadmium group and combined administration group. The ratio of calcium and phosphorus (ca/p) in the cadmium group was not different from that of the control group, but in the combined injection group the ratio was a little lower.

Effects of Maternal Hypothyroidism on the Pubertal Development in Female Rat Offspring

  • The present study was performed to investigate the effect of maternal hypothyroidism and puberty onset in female rat pups. To do this, we employed propylthiouracil (PTU) to prepare a hypothyroid rat model. Pregnant rats were treated with PTU (0.025%) in drinking water from gestational day 14 to postnatal day 21 of offspring. Comparison of general indices such as body and tissue weights and puberty indices such as vaginal opening (VO) and tissue histology between control and PTU-treated rats were conducted. There was no significant difference in the date of VO between control and PTU group. The body weights of the PTU group were significantly lower, only 36.8% of the control group (p<0.001). Although the absolute thyroid weight was not changed by PTU treatment, the relative weight increased significantly about 2.8 times (p<0.001), indicating that hypothyroidism was successfully induced. On the other hand, the absolute weights of the ovary and uterus were markedly decreased by PTU administration (p<0.001), and the relative weight was not significantly changed. The ovarian histology of PTU group revealed the advanced state of differentiation (i.e., presence of corpora lutea). Inversely, the uterine histology of PTU group showed underdeveloped structures compared those in control group. Taken together, the present study demonstrates that our maternal hypothyroidism model resulted in minimal effect on pubertal development symbolized by VO despite of huge retardation in somatic growth. More sophisticatedly designed hypothyroidism model will be helpful to achieve a better understanding of pubertal development and related disorders.

Effects of treadmill exercise on the regulatory mechanisms of mitochondrial dynamics and oxidative stress in the brains of high-fat diet fed rats

  • [Purpose] The purpose of this study was to investigate the effects of treadmill exercise on oxidative stress in the hippocampal tissue and mitochondrial dynamic-related proteins in rats fed a long-term high-fat diet (HFD). [Methods] Obesity was induced in experimental animals using high fat feed, and the experimental groups were divided into a normal diet-control (ND-CON; n=12), a high fat diet-control (HFD-CON; n=12) and a high fat diet-treadmill exercise (HFD-TE; n=12) group. The rats were subsequently subjected to treadmill exercise (progressively increasing load intensity) for 8 weeks (5 min at 8 m/min, then 5 min at 11 m/min, and finally 20 min at 14 m/min). We assessed weight, triglyceride (TG) concentration, total cholesterol (TC), area under the curve, homeostatic model assessment of insulin resistance, and AVF/body weight. Western blotting was used to examine expression of proteins related to oxidative stress and mitochondrial dynamics, and immunohistochemistry was performed to examine the immunoreactivity of gp91phox. [Results] Treadmill exercise effectively improved the oxidative stress in the hippocampal tissue, expression of mitochondrial dynamic-related proteins, and activation of NADPH oxidase (gp91phox) and induced weight, blood profile, and abdominal fat loss. [Conclusion] Twenty weeks of high fat diet induced obesity, which was shown to inhibit normal mitochondria fusion and fission functions in hippocampal tissues. However, treadmill exercise was shown to have positive effects on these pathophysiological phenomena. Therefore, treadmill exercise should be considered during prevention and treatment of obesity-induced metabolic diseases.

  • The electrophoretic analysis, variations of protein and nucleic acid contents with sexual maturation, of the liver tissue of male, and of the muscle tissue of male and female B. pectinirostris(Linnaeus) were investigated. As the result of protein analysis of the liver and muscle tissue of male by the SDS-PAGE electrophoretic analysis, the protein of the liver tissue of male was presented 15.8-101.5kd in molecular weight, and appeared 19 bands in all. The protein bands of the muscle tissues of male and female were almost same, their bands were presented 16.9-116.5kd in molecular weight, and appeared 23 bands in all. And in the liver tissue, the protein contents of the liver tissue of male appeared the maximum in June(the mature stage) and afterwards decreased gradually in July(the spawning stage) and August(the degenerative stage). In the muscle tissues of male and female protein contents were shown the maximum in June, and afterwards decreased gradually, the minimum in July, increased a little in August. The total RNA contents of the liver tissue of male, and their contents of the muscle tissue of male and female appeared the maximum in June(the mature stage), and afterwards decreased gradually in July(the spawning stage) and August(the degenerative stage).

The Relationship Between Bone Mineral Density and Adipose Tissue of Postmenopausal Women (폐경 후 여성의 골밀도 수치와 지방조직과의 연관성)

  • Postmenopausal women are at increased risk for osteoporosis and obesity due to changes in hormones. The relationship between osteoporosis and body weight is known, and its relation with body fat mass is discussed. The purpose of this study was to evaluate the bone mineral density(BMD) changes of epicardial adipose tissue(EAT) and abdominal subcutaneous fat. The subjects of this study were 160 postmenopausal women who underwent BMD and echocardiography. The thickness of the epicardial adipose tissue was measured in three sections and the BMD were meassured according to the diagnostic criteria. The results of this study that age increase the risk of osteoporosis increases, and as the weight and BMI decrease, the risk of osteoporosis increases(p<0.05). The relationship between changes in bone mineral density and adipose tissue in postmenopausal women, increased epicardial adipose tissue was negatively correlated with the bone mineral density(p<0.05). conversely, increased abdominal subcutaneous fat thickness was positively correlated with bone mineral density(p<0.05). In other words, the effect of bone mineral density on the location of adipose tissue was different. If Echocardiography is used to periodically examine changes in the thickness of the epicardial adipose tissue, it may be prevented before proceeding to osteoporosis.

Pinus Densiflora Bark Extract (PineXol) Decreases Adiposity in Mice by Down-Regulation of Hepatic De Novo Lipogenesis and Adipogenesis in White Adipose Tissue

  • PineXol, extracted from Korean red pine bark, has beneficial effects, such as antioxidant, antiinflammatory, and antilipogenic activities in vitro. We tested the hypothesis that PineXol supplementation could have anti-obesity effects on mice fed a high-fat diet (HFD). Four-week-old male C57BL/6 mice were fed normal chow (18% kcal from fat) or a HFD (60% kcal from fat). HFD-fed animals were also subjected to PineXol treatment at a dose of 10 or 50 mg/kg body weight (BW) (PX10 or PX50, respectively) body weight. The body weight and body fat mass in the PX50 group were statistically lower than those in the HFD group (p < 0.05 and p < 0.001, respectively). The concentration of hepatic triglycerides, total cholesterol, and low-density lipoprotein cholesterol were reduced in the PX50 group compared with the HFD group (p < 0.01). Acetyl CoA carboxylase (p < 0.01), elongase of very long chain fatty acids 6 (p < 0.01), stearoyl CoA desaturase 1 (p < 0.05), microsomal triglyceride transfer protein (p < 0.01), and sterol regulatory element-binding protein 1 (p < 0.05) were significantly decreased in the PX50 group compared with that in the HFD group. In white adipose tissue, CCAAT-enhancer-binding protein alpha (p < 0.05), peroxisome proliferator-activated receptor gamma (p < 0.001), and perilipin (p < 0.01) were decreased in the PX50 group compared with those in the HFD group. Therefore, the current study implies the potential of PineXol for the prevention and/or amelioration of obesity, in part by inhibition of both hepatic lipid synthesis and adipogenesis in white adipose tissue.
