Anti-Oxidant Effects of Highly Bioavailable Curcumin Powder in High-Fat Diet Fed- and Streptozotocin-Induced Type 2 Diabetic Rats

  • Curcumin is a hydrophobic polyphenol extracted from turmeric that exhibits a variety of biological functions has albeit with limited efficacy as a functional food material owing to its low absorption when administered orally. The newly developed curcumin powder formulation exhibits improved absorption rate in vivo. This study evaluates the anti-oxidant effects of $Theracurmin^{(R)}$ (TC), which is highly bio-available in curcumin powder. The antioxidant activity of TC was investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, ferrous reducing antioxidant power (FRAP) assays, NO radical, superoxide radical, $H_2O_2$ scavenging activity, and total antioxidant capacity (TAC). Additionally, we evaluated the antioxidant activity of TC in high-fat diet (HFD)-fed streptozotocin (STZ)-induced Type 2 diabetic rats. As a result of oral administration of TC for 13 weeks in type 2 diabetic rats, the group administration of 2,000 mg/kg significantly increased FRAP, superoxide dismutase (SOD), and reduced the level of glutathione (GSH) in liver tissue 1.9, 1.2, and 1.2-times, respectively. Furthermore, serum TAC levels increased by 1.3-fold after the rats were administered with a dose of 500 mg/kg. These results were consistent with the in vitro assay results. In conclusion, TC exhibited its potential as a functional food material through its antioxidant properties.

CT 영상 및 몬테칼로 계산에 기반한 근접 방사선치료계획의 선량분포 평가 방법 연구 (Dose Verification Study of Brachytherapy Plans Using Monte Carlo Methods and CT Images)

  • 대다수의 근접치료용 방사선치료계획장치는 AAPM TG-43의 계산식에 기반을 둔 선량계산 알고리듬을 적용하고 있으나 이는 조직의 비균질성을 적절히 고려하지 못한다. 본 연구에서는 몬테칼로 방법을 이용하여 강내고선량근접치료계획을 검증하는 체계를 구축하고자 하였으며, 특히 환자의 CT 영상을 이용하여 물질정보로 변환한 후 직접 몬테칼로 계산을 수행하는 방법의 타당성에 초점을 맞추었다. 판형 팬텀 및 자궁경부암 환자의 CT 영상을 Plato (Nucletron, Netherlands) 치료계획장치를 이용하여 근접치료계획을 수행한 후 여기서 얻어진 인자들을 이용하여 EGSnrc 기반의 DOSXYZnrc 코드로 몬테칼로 계산을 수행하였으며, EBT 필름측정 결과와 비교하였다. DOSXYZnrc 코드의 선원 모델링 특성 상 후장전 장치의 $^{192}Ir$ 선원들을 직육면체 형태로 근사화하여 모델링하였으며 계산 시 체적소의 크기는 $2{\times}2{\times}2\;mm^3$로 하였다. 균질 매질 내에서는 TG-43 기반의 선량계산 결과와 몬테칼로 선량계산 결과가 잘 일치함을 확인할 수 있었으나 고밀도 물질이 포함된 비균질 매질 내에서는 오차가 커졌다. 환자의 경우 A점 및 B점의 오차는 3% 이내, 평균선량 오차는 5% 정도였다. 그러나 기존 선량계산 알고리듬의 경우 고밀도 물질의 영향을 적절히 고려하지 못하여 표적의 선량을 과대평가하여 실제로는 더 적은 선량이 들어갈 우려가 있다. 본 연구에서 제안된 선량계산 검증체계는 타당하며 선량 계산 결과도 실제와 잘 일치함을 확인할 수 있었다. 또한 기존의 선량계산 알고리듬으로 계산된 치료계획결과를 확인할 경우에는 주의가 필요하며, 몬테칼로 방법과 같은 독립적인 검증 시스템이 유용할 것이다.

개 파보바이러스와 코로나바이러스 진단을 위한 신속진단키트의 임상적 유용성 (Clinical Evaluation of a Rapid Diagnostic Test Kit for Canine Parvovirus and Coronavirus)

  • 개 파보바이러스(canine parvovirus type 2, CPV-2)와 코로나바이러스(canine coronavirus, CCoV)는 개에서 위장관염을 일으키는 주요 병원체이다. 두 바이러스는 전염성과 이환율이 높고 특정한 치료법이 없어 신속 정확한 진단이 필요하다. 동물용 신속진단키트 (rapid diagnostic test, RDT)는 빠르고, 간편하여 진료현장에서 널리 활용되고 있다. 이에 본 연구에서는 성능평가를 통해 CPV-2/CCoV RDT의 임상적 유용성을 확인하고자 하였다. 성능평가 항목으로 최소검출한계(limit of detection, LoD), 교차반응, 간섭, 민감도, 특이도, 음성우도비(negative likelihood ratio, NLR), 카파통계량(kappa value, κ) 등을 확인하였다. 성능평가 결과, LoD는 CPV-2 9.7×10 50% tissue culture infections dose (TCID50)/mL, CCoV 2.5×102 TCID50/mL로 나타났다. 병원체 9종에 의한 교차반응과 간섭물질에 대한 간섭은 관찰되지 않았다. RDT는 두 바이러스의 검출에 있어 민감도 90.0%, 특이도 100.0%, NLR=0.1, κ=0.90으로 나타났다. 결론적으로 CPV-2/CCoV RDT는 높은 민감도, 특이도, κ와 낮은 NLR을 보여 선별검사로써 유용할 것으로 생각된다.

흰쥐의 스트레스성 위궤양에 대한 Alove vera의 항궤양작용 (The Antiulcer Effects of Alove vera on the Stomach ulcer Induced by Stress in rats)

  • This was performed to investigate the antiulcer effects of Aloe vera on the stomach ulcer induced by restraint and water-immersion stress in rats. For this experiment, 60 male Sprague-Dawley strain were used. The experimental groups were divided into five: a control(CA) and 4 aloe treatment groups. Each dose of aloe was 50 mg/kg BW(AA), 100mg/kg BW(AB), 200 mg/kg BW(AC), and 400 mg/kg BW(AD). The rats were allocated to each group by 12 and observed for 4 weeks. The results are as following. 1. The stomach surface pH in each group showed no significant difference, byt the values of aloe treatment groups were higher than the value of the control group. 2. The gastric wall mucus was significantly increased in all aloe treatment groups(p<0.05) compared with the control group. Especially in AC and AD group the differences were higher(p<0.01). 3. At shear rate rate 11.25, 45.0, 90.0, 225.0 sec-1, whole blood viscosity and plasma viscosity were measured. Most of the values of aloe treatment groups were significantly low compared with those of the control group(P<0.05). 4. The ulcer index of aloe treatment groups were significantly low campared with control group(p<0.05). Especially in AC and AD group the differences were more significant(p<0.01). 5. Less severe ulcers were observed in AA and AB group than in the control group. Tissues of AC and AD group had only slight ulcers and necrosis of tissue was not observed in these groups. Especially in AD group, there was more mucus than other groups and it seemed that alove vera stimulated the epithelial regeneration. From the results of this study, it can be concluded that the oral administration of Alove vera results in protection of stomach ulcer by stimulating the secretion of gastric mucus and the circulation of blood.

백서를 이용한 포르말린 통증 모형하에서 뇌실 내 Gabapentin의 효과 (The Effect of Intracerebroventricular Gabapentin on the Formalin Test in Rats)

  • Background: Systemic or intrathecal administration of gabapentin has been shown to reverse various pain states. However, until now, the effect of intracerebroventricular (ICV) gabapentin to noxious stimuli has not been reported. The authors' aim of this study was to determine the effect of ICV gabapentin on the inflammatory nociceptive model, formalin test, in rats. Methods: ICV catheters were implanted under halothane anesthesia. For the nociceptive test, $50{\mu}l$ of 5% formalin was subcutaneously injected into the hindpaw. The effect of ICV gabapentin, administered 10 min before formalin injection, were examined on flinching, mean arterial pressure and heart rate evoked by a injection of formalin. Results: Injection of formalin into the paw resulted in a biphasic flinching and cardiovascular response. ICV gabapentin produced a dose-dependent suppression of the flinching and mean arterial pressure response during phase 1. In contrast, in phase 2, ICV gabapentin did not attenuate the pain behavior. ICV gabapentin did not affect on the baseline mean arterial pressure and heart rate. Conclusions: ICV gbapentin was effective for the acute noxious stimulus but it had no effect on the facilitated states induced by tissue injury.

갑상선암 환자에서 전절제술 후 I-131 치료에서 미만성 간침착 정도의 분석 (Analysis of Diffuse Hepatic Visualization after Iodine-131 Treatment in Patients with Thyroid Carcinoma)

  • Objectives: Any uptake of I-131 after total thyroidectomy means the remant thyroid tissue or distant metastasis of the thyroid cancer. However diffuse hepatic uptake of I-131 without abnormal uptake was showen in many cases on I-131 whole body scan. The aim of this study was to classify the liver uptake after I-131 scan and to evaluate the analysis of this finding. Materials and Methods: Between 1982 and 1998, 104 patients(l4 males, 90 females) with normal liver function underwent I-131 scan after total thyroidectomy. Prospectively we reviewed the films of the whole body scan and analysed the correlations between results of radioiodine uptake, pathologic diagnosis, prognostic factors, lymphatic metastasis, and thyroid function test. Result: Diffuse hepatic uptake was found in 44 of 104(42%) patients. 10 of 39(26%) patients on I-131 100mCi, and 34 of 63(54%) on I-131 150mCi showed hepatic uptake. 52 of 104(50%) patients was locally invasive thyroid cancer. The rate of the hepatic uptake was no significant differences with the thyroid hormone levels(T3, Free T4) and thyroglobulin between uptake group and non-uptake group. Conclusion: The rate of I-131 uptake was high in high-dose radioiodine treatment group. However, we can not find any correlation among the thyroid functions, the extent of metastasis or the extent of local invasion. We need further study to find out the causes of the hepatic uptake of I-131 after total thyroidectomy, besides liver metabolism of I-131 attached thyroid hormones.

Effect of St. John's Wort (Hypericum perforatum) on obesity, lipid metabolism and uterine epithelial proliferation in ovariectomized rats

  • BACKGROUND/OBJECTIVES: This study was conducted to assess the potential of St. John's Wort (Hypericum perforatum) to prevent obesity and abnormalities in lipid metabolism induced by ovariectomy in a rat model without stimulatory activity on uterus. MATERIALS/METHODS: Ovariectomized (OVX) rats were treated for 6 weeks with 70% ethanol extracts of Hypericum perforatum [HPEs: whole plant (WHPE) and flower and leaves (FLHPE)], ${\beta}$-estradiol-3-benzoate at a dose of $50{\mu}g/kg/day$ (E2) or vehicle (distilled water). RESULTS: As expected, OVX increased body weight gain and adiposity and showed higher food efficacy ratio. OVX also increased the serum cholesterol as well as insulin resistance, while reducing uterus weight and uterine epithelial proliferation rate. HPEs (WHPE and FLHPE) showed estrogen-like effect on body weight gain, adipose tissue weight and food efficacy ratio in OVX rats. HPEs prevented hypercholesterolemia induced by OVX more effectively than E2. E2 increased uterus weight and epithelial proliferation rate in OVX rats, while HPEs maintained them at the level of the sham-operated animals. CONCLUSIONS: Our finding demonstrates that HPEs can be considered as an effective agent to prevent OVX-induced obesity without stimulatory activity on uterus.

Clinical Character of Pediatric Head and Neck Rhabdomysarcomas: A 7-Year Retrospective Study

  • Objective: The rhabdomysarcoma (RMS) is most common soft tissue carcinoma in children, mostly found in the head and neck with high degree of malignancy. The current study aimed to summarize clinical data and evaluate treatment outcome of cases in a single hospital. Methods: Forty-one (24 male, 17 female) children with newly diagnosed RMS in Beijing Tong Ren Hospital were enrolled between November, 2004 and May, 2011. The. Students' t and Chi tests were then performed on retrospectively reviewed clinical data, followed by survival analysis based on the Kaplan Meier method using SPSS 17.0 software. Results: Of all cases, 32 were treated by common chemotherapy, and 3 cases with stage III RMS received high-dose chemotherapy and auto-peripheral blood stem cell transplantation (APBSCT). Side-effects in the former were: I grade for 62.5% (20/32), II grade for 28.1% (9/32), III grade account for 9.275% (3/32). Side-effects of 3 cases with APBSCT: 2 were I grade, 1 was III grade. The median follow-up time of 41 RMS cases was 41 months. Four cases were lost to follow-up, 7 cases recurred, and 5 cases died of cerebral metastasis, witha total survival rate was 86.5% (32/37). CR rate was 67.6% (25/37), PR was 18.9% (7/37). Conclusion: Multidiscipline treatment including chemotherapy, radiotherapy, surgery and auto-PBSCT is highly recommended for pediatric patients with head and neck RMS.

Preparation and Characterization of Temperature-Sensitive Poly(N-isopropylacrylamide)-g-Poly(L-lactide-co-$\varepsilon$-caprolactone) Nanofibers

  • Biodegradable and elastic poly(L-lactide-co-$\varepsilon$-caprolactone) (PLCL) was electrospun to prepare nanofibers, and N-isopropylacrylamide (NIPAAm) was then grafted onto their surfaces under aqueous conditions using $^{60}Co-{\gamma}$ irradiation. The graft yield increased with increasing irradiation dose from 5 to 10 kGy and the nanofibers showed a greater graft yield compared with the firms. SEM confirmed that the PLCL nanofibers maintained an interconnected pore structure after grafting with NIPAAm. However, overdoses of irradiation led to the excessive formation of homopolymer gels on the surface of thc PLCL nanofibers. The equilibrium swelling and deswelling ratio of the PNIPAAm-g-PLCL nanofibers (prepared with 10 kGy) was the highest among the samples, which was consistent with the graft yield results. The phase-separation characteristics of PNIPAAm in aqueous conditions conferred a unique temperature-responsive swelling behavior of PNIPAAm-g-PLCL nanofibers, showing the ability to absorb a large amount of water at < $32^{\circ}C$, and abrupt collapse when the temperature was increased to $40^{\circ}C$. In accordance with the temperature-dependent changes in swelling behavior, the release rate of indomethacin and FITC-BSA loaded in PNIPAAm-g-PLCL nanofibers by a diffusion-mediated process was regulated by the change in temperature. Both model drugs demonstrated greater release rate at $40^{\circ}C$ relative to that at $25^{\circ}C$. This approach of the temperature-controlled release of drugs from PNIPAAm-g-PLCL nanofibers using gamma-ray irradiation may be used to design drugs and protein delivery carriers in various biomedical applications.

악성 두피 종양(Scalp) 환자의 M3 Wax Bolus를 이용한 방사선치료 (Radiation Therapy Using M3 Wax Bolus in Patients with Malignant Scalp Tumors)

  • 목 적: 두피 악성종양의 치료에 광자선을 사용할 때 필요한 Bolus 재질들의 단점으로 인하여 3D Printer용 헬멧형 bolus가 제작되고 있다. 하지만 사용되는 재질인 PLA은 조직등가물질에 비해 높은 밀도를 가지고 있으며 환자가 착용할 경우 불편한 점들이 발생한다. 이에 본 연구에서는 3D Printer를 이용한 M3 wax 헬멧을 제작하여 악성 두피종양을 치료하는 방법을 시도해 보고자 한다. 대상 및 방법: 헬멧형 M3 wax의 모델링을 위해 두부인체모형팬텀을 CT로 촬영해 DICOM file로 획득하고, 두피 위에 헬멧이 위치할 부위를 Helmet contour로 제작하였다. M3 Wax 헬멧의 제작은 paraffin wax를 녹이고, 산화마그네슘, 탄산칼슘을 섞어 용해시킨 후 PLA 3D 헬멧의 내부에 넣고 표면의 PLA 3D 헬멧을 제거하였다. 치료계획은 총 10 Portal의 Intensity-Modulated Radiation Therapy(IMRT)로 세웠으며, 치료선량은 200 cGy로 eclipse의 Analytical Anisotropic Algorithm(AAA)를 사용하였다. 그 후 EBT3 film과 Mosfet(Metal Oxide Semiconductor Field Effect Transistor: USA)를 이용해 선량검증을 실시하였으며, CT 모의치료실과 동일한 조건으로 두부인체모형팬텀을 재현해 IMRT Plan을 측정하였다. 결 과: CT상에서 측정된 Bolus의 Hounsfield unit(HU)는 $52{\pm}37.1$으로 나타났다. M3 wax bolus 측정점 A, B, C에서 TPS의 선량은 186.6 cGy, 193.2 cGy, 190.6 cGy으로 확인되었고, Mostet으로 3회 측정한 선량은 $179.66{\pm}2.62cGy$, $184.33{\pm}1.24cGy$, $195.33{\pm}1.69cGy$, 오차율은 -3.71 %, -4.59 %, +2.48 %였다. EBT3 Film으로 측정된 선량은 $182.00{\pm}1.63cGy$, $193.66{\pm}2.05cGy$, $196{\pm}2.16cGy$이었으며, 오차율은 -2.46%, +0.23 %, +2.83 %로 확인되었다. 결 론: M3 wax bolus는 2 cm의 두께로 제작되어 뇌 부분의 선량을 보다 쉽게 낮추어 치료계획을 수립할 수 있었다. 치료선량 검증에서의 EBT3 Film과 Mosfet의 선량계의 A, B, C 측정값에서도 두피의 표면선량 최대 오차율은 5 % 이내로 측정되었으며, 일반적으로 3 % 이내로 정확하게 측정되었다. M3 wax bolus는 제작과정 기간이 3D Printer보다 빠르고 비용이 저렴하며, 재사용 가능하고, 인체조직 등가물질로서 두피 악성종양 치료에 매우 유용한 Bolus이다. 따라서 3D Printer의 대용량 Bolus, Compensator의 제작시간 및 비용이 비싼 단점을 극복하는 주조형 M3 wax bolus의 사용이 추후 확대될 것으로 사료된다.
