Simplistic Determination of Operation Parameters for Simulated Moving Bed (SMB) Chromatography for the Separation of Ketoprofen Enantiomer

  • Since it is troublesome to estimate adequate flow rates in four sections of SMB chromatography, a systematic determination of the flow rates has been suggested by using ketoprofen as a model chiral enantiomer. S-ketoprofen. less retained species, was separated from raffinate stream and the variation in its purity was dependent on the changes of the flow rate of section 4 ($Q_4$), the raffinate flow rate ($Q_{raf}$), and the feed flow rate ($Q_{feed}$) under a fixed switch­ing time t$^{\ast}$. When one parameter was changed at the given experimental condition, purities of product were changed and these phenomena has be well explained by the triangle theory.

  • Tyrosinase transcript In the blood Is known as the marker of malignant melanoma and it has been often determined by using reverse transcription-polymerase chain reaction (RT-PCA) . However, after the PCR process, the quantification of amplified CDMA by the gel electrophoresis is not reliable and time-consuming. for this reason, we tried to quantify the PCR product using a cuvette-type biosensor, where the oligonucleotide probe was immobilized on the cuvette surface and the single strand CDMA, the denatured PCH product, was then hybridized onto the immobilized probe to give a response signal. The response was Immediate and takes 15 min to obtain a stable signal. The biosensor was much more sensitive comparing to the gel electrophoresis method. The quantification of PCR product using a cuvette-type biosensor was feasible and rapid.

  • Non-destructive determination of soluble solids(Brix) in harvested fruits using near infrared(hereafter, NIR) spectroscopy has been reported by many researchers. We have just reported on non-destructive estimation of Brix in harvested melons using a NIR Systems Model 6500 spectrophotometer(Ito et al., 2000). There is a melon cultivar that is difficult to judge the harvest time from the external appearance. If we can determine Brix in growing fruits non-destructively in the field, immature fruits will not be harvested. A portable m spectrophotometer for field use has been just developed by Kubota Corporation. The spectral data of growing melons were measured by the portable spectrophotometer. A commercial program was used for multiple linear regression analysis. Brix in growing melons could be estimated by a multiple regression equation calibrated with harvested melons. Absorbances of 906 and 874 nm were included as the independent variables in the multiple regression equation, and these wavelengths are key wavelengths for non-destructive Brix determination.

Enzymatic Determination of Glucose Using Soybean Sprouts Peroxidase. (콩나물 Peroxidase를 이용한 포도당의 효소적 분석)

  • Soybean sprouts peroxidase can be used for enzymatic determination of glucose. Peroxidase from soybean sprouts was purified by ammonium sulfate precipitation and DEAE Sephacel column chromatography. The glucose could be quantitatively assayed by using glucose oxidase and soybean sprouts peroxidase. The optimum pH and temperature for glucose assay were of pH 5.5 and $40^{\circ}C$, respectively. The relationship between absorbance and glucose concentration was linear. And also the relationship between absorbance and reaction time was linear. The reducing agents such as L-cysteine, dithiothreitol inhibited the glucose assay by glucose oxidase and soybean sprouts peroxidase.

  • A Diffusion analysis method is described for the determination of alcohol content in tinctures and spirits and then, the comparison test of the official alcohol number test with the diffusion analysis method has hen carried out. Throughout this experiments, a transformed Widmark apparatus is used. It is found that the diffusion analysis method using the transformed Widmark apparatus is very simple and occapy a small space. One of the most convenient fact is many sample can be analyzed at a time with this apparatus. Experimental data indicate that 95% of alcohol content obtained by the official alcohol number test is found in the diffusion analysis.

  • A strategy for geostationary orbit (or geostationary earth orbit [GEO]) surveillance based on optical angular observations is presented in this study. For the dynamic model, precise analytical orbit model developed by Lee et al. (1997) is used to improve computation performance and the unscented Kalman filer (UKF) is applied as a real-time filtering method. The UKF is known to perform well under highly nonlinear conditions such as surveillance in this study. The strategy that combines the analytical orbit propagation model and the UKF is tested for various conditions like different level of initial error and different level of measurement noise. The dependencies on observation interval and number of ground station are also tested. The test results shows that the GEO orbit determination based on the UKF and the analytical orbit model can be applied to GEO orbit tracking and surveillance effectively.

  • Determinations of the isotopic contents of U$^{235}$ and U$^{238}$ in ten uranium samples containing 0.72-89.70 at % U$^{235}$ were carried out in two ways utilizing high resolution Ge (Li) gamma-ray spectrometer. One method is based upon the fact that the intensity of 185.5 kev gamma-ray vary linearly with U$^{235}$ content for a given geometry. Another method applied for the direct determination of the U$^{235}$ / U$^{238}$ isotopic ratios is the precision gamma-ray spectrometric analysis of reactor irradiated uranium samples after allowing a fixed cooling time for one hour. The results obtained by both methods well agree with the values calculated from the isotopic contents of highly enriched original uranium samples measured by mass spectrometer. The precision obtained was well below 5% for most of the isotopic ratios investigated.

  • In this paper we consider the concurrent spare part(CSP) requirements problem of new equipment system. When a part fails, the part is replaced and the repair of failed part is impossible and cannibalization is allowed. We assume that the failure of a part knows a Poisson process. The operational availability concept in CSP is defined, and derive a formula to calculate the operational availability using expected machine operating time during CSP period. A mathematical model is derived for making an CSP requirement determination subject to the constraint of satisfying any given operational availability limitation and an heuristic solution search procedure is derived. An illustrative example is shown to explain the solution procedure.

  • This paper persents an algorithm compensating for the change of tension acting on a belt which is used as a force transfer media of the induction motor that drives a washing machine. The induction motor adopted in a washing machine is operated not only for the power-transmission device, but also for the detection of clothes load. The load of clothes is determined by the duration of inertial rotation which is occurred by the induction motor during a specific time. The tension of belt also affects the determination of clothes load as another load and this change of the tension is a significant disturbance for accurate determination. This paper mentions the algorithm compensating for the amount of change in tension and the application of the algotithm proved to be effectively increasing the washing performance and reducing the noise and the vibration.
