Antioxidative and Radical Scavenging Properties of the Constituents Isolated from Cosmos caudatus Kunth

  • A phytochemical investigation on the methanolic extract of Cosmos caudatus has led to the isolation of quercetin $3-O-{\beta}-D-arabinofuranoside$ (1), quercetin $3-O-{\beta}-D-rhamnoside$ (2), quercetin $3-O-{\alpha]-D-glucoside$ (3) and quercetin (4). These compounds were shown to be the antioxidative constituents of the plant when evaluated using the ferric thiocyanate (FTC) and thiobarbituric acid (TBA), and radical scavengers based on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assays.

  • This study was designed to develope the antioxidative activity on oxidation of human low density lipoprotein(LDL) from marine microbials. Bacillus KS-96 producign antioxidant have been isolated and identified from seawater, Bacillus sp. KS-96. The optimal medium pH was 7.0 and incubation temperature was 30$^{\circ}C$. The antiosidant of potential substance produced extracellularly in the culture broth by Bacillus sp. KS-96 was obtained by elution of silica gel culumn chromatography with hexane, ethylacetate and water. The ethylacetate faction are shown at highest level of antioxidant activity using thiocyanate method among them. By IR, NMR, and GC/MS, antioxidant purified from ehtylacetate fraction was identified and named as 2,6-dimethoxyphenol. 2,6-dimethoxyphenol inhibited the metal mediated oxidation of human LDL at concentration of 50∼100 ${\mu}$g/mL in the presence of 5uM CuSO4 with macrophage or J774 cells.

  • Solvent-free conversion of various epoxides to their corresponding $\beta$-hydroxy thiocyanates was carried out successfully with $NH_4SCN$/zeolite molecular sieve $4{\AA}$ system at room temperature. The reactions were completed within 2 - 7 min to give thiocyanohydrins with perfect regioselectivity and isolated yields. Moreover, the zeolite can be reused for several times without losing its activity.

  • For the development of new COX-2 inhibitors, 1,5-diarylhydantoins 5a∼5c and 1,5-diaryl-2-thiohydantoin 6a∼6c were synthesized from commercially available phenylacetic acids through esterification, bromination, C-N bond formation and cyclization. Esters 2a∼c were efficiently synthesized from the starting materials 1a∼c by refluxing in absolute methanol for 3 hours with catalytic concentrated sulfuric acid. Bromination of 2a∼c was carried out with use of N-bomosuccinimide at rt in dichloromethane. The bromine of 3a∼c was substituted with aniline in ethanol or N,N-dimethylformamide to provide 4a∼c. Hydantoins and 2-thiohydantoins were synthesized from 4a∼c by treatment of potassium isocyanate or potassium thiocyanate in dil-ethanol with triethylamine.

  • In this study, the method of adsorption by activated carbon in the removal of Hg(II) ion in waste water was treated. The influence of kinds of activated carbon and effect of temperature and the influence of coexistent salt on adsorption rates, the influence of pH in the adsorption, equilibrium and adsorption of mercury from activated carbon were investigated. From the adsorption on activated carbon of mercury(II) ion in the presence of cyanide or thiocyanate ion was found that mercury(II) was easily adsorved onto the activated carbon in the form of complex artion such as Hg(CN)$_4^{2-}$, Hg(SCN)$_4^{2-}$ respectively. ZnCl$_2$ activation method had a higher adsorptive ability than steam activation method in adsorption of Hg on activated carbon. Activated carbon adsorbed iodide ion is very effective on adsorption of Hg.

  • Objectives : The verity extract of the Gagam-Hyungbang-Gihwang-tang (GHG) was assessed to determine the mechanisms of its antioxidant activity. Methods : The fellowing effects were measured : GHG exhibited a concentration-treatment; scavenging ${\alpha},\;{\alpha}-diphenyl-\beta-picrylhydrazyl$ (DPPH) radical, linoleic acid oxidation in a thiocyanate assay system, and superoxide anion, hydroxyl radical-induced DNA nicking. We investigated mRNA levels such as superoxide-dismutase. Results : The GHG extract showed dose-dependent free radical scavenging activity, including DPPH radicals, hydroxyl radicals, and superoxide anion, using different systems. The GHG was also found to be effective in protecting plasmid DNA against the strand breakage induced by hydroxyl radicals in Fenton's reaction mixture. Furthermore, SOD-1 mRNA expression levels increased in tat hepatoma H4IIE cells Conclusions : We expect that GHG will to helpful to the development of antioxidant activity treatments.

Electronic and Vibrational Spectroscopy of cis-Diisothiocyanato(1,4,8,11-tetraazacyclotetradecane)chromium(Ⅲ) Thiocyanate

  • The emission and excitation spectra of $cis-[Cr(cyclam)(NCS)_2]NCS$ (cyclam = 1,4,8,11-tetraazacyclotetradecane) taken at 77 K are reported. The infrared and visible spectra at room temperature are also measured. The vibrational intervals due to the electronic ground state are extracted from the far-infrared and emission spectra. The ten pure electronic origins due to spin-allowed and spin-forbidden transitions are assigned by analyzing the absorption and excitation spectra. Using the observed transitions, a ligand field analysis has been performed to determine the bonding properties of the coordinated ligands in the title chromium(Ⅲ) complex. According to the results, it is found that nitrogen atoms of the cyclam ligand have a strong σ-donor character, while the NCS ligand has medium σ- and π-donor properties toward chromium(Ⅲ) ion.

  • To develop new COX-2 inhibitors, 1,5-diarylhydantoins and 1,5-diaryl-2-thiohydantoins were synthesized from phenylacetic acids by esterification, bromination, C-N bond formation and cyclization. Esters 1-3 were efficiently synthesized from the starting materials by reflux in absolute methanol for 3 h containing concentrated sulfuric acid as catalyst. Bromination was carried out with N-bromosuccinimide at rt in dichloromethane. Bromides 4-6 were reacted with aniline, p-anisidine, sulfanilamide in ethanol (or N,N-dimethylformamide) to provide the amines 7-15. Hydantoins and 2-thiohydantoins 16-46 were synthesized from amines 7-15 by treating them with potassium isocyanate (or potassium thiocyanate) and triethylamine. The synthetic process from alkyl α-anilinophenylacetate 7-15 to 3-alkylhydantoins was carried out in a one-pot reaction using alkyl isocyanate (alkyl isothiocyanate).

Antioxidant effects of the Root of Vitis labrusca Water and Aqua-acupuncture Solution (포도근 열수 추출액 및 약침액이 항산화에 미치는 효과)

  • This study was purposed to investigated the antioxidant effects of water (VLWS) and aqua-acupuncture solution (VLAS) from root of Vitis labrusca. VLWS and VLAS was were assessed to determine the mechanisms of its antioxidant activities. The VLWS and VLAS exhibitde a concentration-dependent inhibition of DPPH free radicals. This VLWS-mediated antioxidant activity was similar to that L-ascorbic acid (vitamin C). In addition, the VLWS and VLAS showed dose-dependent free radical scavenging activity, including superoxide anions and hydroxyl radicals, using different assay systems. The VLWS and VLAS were also found to be strongly inhibited linoleic acid oxidation in a ferric thiocyanate assay (FTC) system. Finally, the VLWS and VLAS significantly prevented Fenton's reagent-induced DNA nicking.

Avidity of serum immunoglobulin G antibodies to Porphyromonas gingivalis in the elderly with chronic periodontitis

  • The present study aimed at evaluating serum immunoglobulin G (IgG) avidity to Porphyromonas gingivalis in elderly patients with mild and severe chronic periodontitis. The avidity of antibodies against P. gingivalis present in the sera of 18 patients with mild chronic periodontitis and 18 patients with severe chronic periodontitis was evaluated using an ammonium thiocyanate-dissociated enzyme-linked immunosorbent assay (ELISA). The results showed that the mean absorbance value in serum IgG antibody titers was significantly higher in the severe chronic periodontitis group than in the mild chronic periodontitis group ($198{\pm}35ELISA$ unit [EU] vs. $142{\pm}32EU$, p < 0.01). However, there was no significant difference between the two groups in antibody avidity ($65{\pm}57EU$ vs. $54{\pm}27EU$). These findings suggest that humoral immune responses to P. gingivalis between mild and severe chronic periodontitis in elderly patients are characterized by the differences in the quantity rather than the quality of the antibodies.
