Survey of Beet Red Contents in Foods using TLC, HPLC (TLC, HPLC를 이용한 식품 중 비트레드 함량조사)

  • This study was performed far development of new analytical method of beet red in foods. In this study, analysis of beet red in foods has been carried out by detection of betanine and isobetanine, the main color component of beet red as indicator compounds. The qualitative analysis technique consisted of clean-up of the colors with a $C_{18}$ cartridge, separation of the colors by cellulose TLC plate using acetone:3-methyl-1-butanol:distilled water (7:7:6) as a solvent system. Also, the quantitative analysis was performed using X-terra RP at wavelength 538 nm and $0.1\%$ phosphoric acid : methanol (90:10) as a solvent. The quantitative results of beet .ed were as follows:900.22$\∼$27701.60 $\mu$g/g for 60 item in nutrient supplement food, $21.95\∼713.40{\mu}g/g$ for 30 items and N.D. for 18 items in cindy, and $155.85{\∼}505.37{\mu}g/g$ for 12 items in ice creams, $43.52\∼64.75{\mu}g/g$ for 18 items and N.D. for 54 item in sauce, N.D. for 12 items in retort food.

Comparison of Carotenoid Pigments in Chinese muddy loach, Misgurnus mizolepis, and Muddy loach, Misgurnus anguillicaudatus, in the Subfamily Cobitidae (미꾸리아과에 속하는 미꾸라지와 미꾸리의 Carotenoid 색소성분의 비교)

  • Differences in carotenoid composition in the integuments of wild and cultured chinese muddy loach Misgurnus mizolepis and muddy loach Misgurnus anguillicaudatus were compared. Total carotenoid contents in the integuments of the wild and cultured chinese muddy loach were $4.76mg\%\;and\;3.43mg\%$, respectively. The important carotenoids in the integuments of the wild chinese muddy loach were lutein($30.5\%$), ${\beta}$-cryptoxanthin($24.6\%$), ${\beta}$-carotene($20.6\%$) and cynthiaxanthin($11.7\%$). In addition, zeaxanthin($4.7\%$), tunaxanthin ($4.5\%$), and a-cryptoxanthin($1.0\%$) were present in small amounts. In the integuments of the cultured chinese muddy loach, lutein($35.4\%$), ${\beta}$-cryptoxanthin($17.9\%$), cynthiaxanthin($16.0\%$) and ${\beta}$-carotene($12.7\%$) were present as important carotenoids. In addition, zeaxanthin($8.1\%$), tunaxanthin($5.0\%$), a-cryptoxanthin($0.9\%$) were found in small amounts. Total carotenoid contents in the integuments of the wild and cultured muddy loach were $4.00mg\%\;and\;2.99mg\%$, respectively. The important carotenoids in the integuments of the wild muddy loach were lutein($32.9\%$), ${\beta}$-cryptoxanthin($18.8\%$), cynthiaxanthin($17.0\%$) and ${\beta}$-carotene($15.1\%$). In addition, zeaxanthin($6.5\%$), tunaxanthin($6.0\%$) and a-cryptoxanthin($1.5\%$) were found in small amounts. In the integuments of the cultured muddy loach, lutein($51.8\%$), cynthiaxanthin($19.9\%$) and ${\beta}$-cryptoxanthin($10.8\%$) were observed as important carotenoids. In addition, ${\beta}$-carotene($5.0\%$), zeaxanthin($4.8\%$), tunaxanthin($4.5\%$) and a-cryptoxanthin($0.2\%$) were found in small amounts.

  • Objectives: Due to the heterogeneous receptor distribution and changes of receptor status over time, the biochemical measurement of estrogen receptor status of biopsy specimens is not sufficient to diagnose breast cancer. As a result, I-123 labeled estradiols have been applied for the diagnosis. The purpose of this study was to develop a suitable radioligand for imaging estrogen receptor-positive human breast tumors. Methods: Among the various estradiol derivatives, $17{\alpha}-[^{123}I]$iodovinyl estradiol ($[^{123}I]$IVE) has been prepared from $17{\alpha}$-ethynyl estradiol. Labeling of $E-17{\alpha}-[^{123}I]$iodovinyl estradiol (E-$[^{123}I]$IVE) was carried out using peracetic acid with $[^{123}I]NaI\;and\;Z-[^{123}I]IVE$ labelling was archived using chloamine-T/HCl solution with $[^{123}I]$NaI. Labeling yield was determined by silica thin-layer chromatography (TLC) and radiochemical purity was measured by high performance liquid chromatography (HPLC). The biodistribution of E-$[^{123}I]$IVE was measured in immature female rats at 60 min, 120 min and 300 min after injection. Results: The labeling yield of two isomers was 92% and 94% ($E-[^{123}I]IVE\;and\;Z-[^{123}I]IVE$, respectively). The radiochemical purity was more than 98% after purification. The highest uptake was observed at 120 min in uterus (3.11% ID/g for E-$[^{123}I]$IVE). Conclusion: These results suggest the possibility of using E-$[^{123}I]$IVE as an imaging agent for the evaluation of the evaluation of the presence of estrogen receptor in patients with breast cancer.

Identification and Quantification of Phytosterols in Maize Kernel and Cob (옥수수 종실 및 속대의 Phytosterol 동정과 함량 변이)

  • Unsaponifiables in the kernel and the cob of 7 maize varieties were analyzed by using thin-layer chromatography (TLC) and gas chromatography (GC) for the identification of phytosterols and their concentrations. The unsaponifiables of the kernel were clearly separated into band I (campesterol, stigmasterol, and ${\beta}$-sitosterol), band II (${\Delta}^5$-avenasterol), band III (${\Delta}^7$- stigmastenol), and band IV (${\Delta}^7$-avenasterol). In the cob, on the other hand, three or more bands were separated in addition to bands. The GC analysis of unsaponifiables showed good separation of campesterol, stigmasterol and ${\beta}$-sitosterol, but the mixture of ${\Delta}^7$-avenasterol (retention time[RT] 22.846), ${\Delta}^7$-stigmastenol (RT 22.852), and ${\Delta}^5$-avenasterol (RT 22.862) showed poor separation. Phytosterol content of the maize kernel was 635.9 mg/100 g, and that of the cob was 273.0 mg/100 g, respectively. The phytosterol content of the kernel was 2.4-fold higher than that of the cob. The phytosterol content of the kernel was higher in the order ${\beta}$sitosterol 80.05% > campesterol 10.5% > stigmasterol 9.46%, but that of the cob was higher in the order ${\beta}$-sitosterol 59.43% > stigmasterol 31.72% > campesterol 10.98%. Based on these results, it appears that the phytosterols of the maize kernel are synthesized in the maize cob and are transferred to the kernel, because the precursors (${\Delta}^7$-avenasterol, ${\Delta}^7$-stigmastenol, and ${\Delta}^5$-avenasterol) of major phytosterols were detected in maize cobs.

Amount and Chemical Characteristics of the Epicuticular Waxes on Leaves at Active Tillering and Heading Stages of Rice Varieties (벼 품종들의 분얼성기 및 출수기의 엽표면Wax의 양 및 화학적 조성)

  • Differences in the amount and chemical characteristics of the epicuticular waxes on rice leaves were studied for the active tillering and heading stages of rice varieties differing widely in gross leaf-surface property and genetics. The amount of waxes on surfaces of rice leaf-blades was determined by extraction with chloroform and chemical composition of the waxes was characterized by thin layer chromatography, gas liquid chromatography and infrared spectrophotometry. The amount of waxes varied by variety and significantly with growth stage. The amount at the heading stage was 1.7 to 3.6 mg/g fresh weight of leaves, which was two to three times as much as that at the tillering stage of 0.8 to 1.8 mg/g fresh weight. The waxes consisted of seven chemical classes, namely diols, fatty acids, fatty alcohols, fatty aldehydes, fatty esters, saturated and unsaturated hydrocarbons. Diols and unsaturated hydrocarbons were identified as new chemical classes of the rice epicuticular waxes. The polar constituents such as dials, fatty acids and fatty alcohols and the non-polars such as fatty aldehydes, fatty esters, and saturated and unsaturated hydrocarbons were identified at the heading stage, but at the tillering stage only the non-polar compounds were identified. In the carbon numbers (C) of the chemical classes, diols were composed entirely of C30 and acids were mainly of C30 and C31. In alcohols, primary alcohols were composed of C13 and C32, and the secondary alcohols were of C14, C16 and / or C30 regardless of the rice varieties. The acid portion of fatty esters, mainly composed of C22 and C23, showed low cabon numbers compared with the aldehydes. The alcohol portion of them showed a wide distribution in carbon numbers from C13 to C26 depending on the rice varieties. Hydrocarbons had odd carbon numbers, consisting mainly of C29 and C31.

Carotenoid Pigments in Echiurid, Urechis unicinctus (개불의 carotenoid 색소성분)

  • Carotenoid pigments of echiurid, Urechis unicinctus were investigated during March, April and May as a part of comparative biochemical studies of carotenoid pigments for the marine organisms other than pisces. Total carotenoid contents were found to be 1.19 mg/100 g in March, 0.98 mg/100 g in April and 0.84 mg/100 g in May, indicating that total carotenoid content was negatively affected by the temperature of sea water that echiurid resided. The carotenoid isolated in March composed of $16.3\%$ diatoxanthin monoester, $14.8\%$ $\beta$-carotene and $12.6\%$ cynthiaxanthin monoester, $8.4\%$ cynthiaxanthin diester, $8.2\%$ zeaxanthin monoester, $7.3\%$ diatoxanthin diester $4.2\%$ astaxanthin $2.9\%$ diatoxanthin, $2.4\%$ triol, $2.3\%$ cynthiaxanthin, $1.7\%$ isocrytoxanthin, $1.5\%$ zeaxanthin diester, $0.8\%$ zeaxanthin and $0.5\%$ lutein. The carotenoid isolated in April composed of $21.9\%$ diatoxanthin monoester, $17.2\%$ cynthiaxanthin monoester and $16.6\%$ $\beta$-carotene $10.9\%$ zeaxanthin monoester, $5.6\%$ cynthiaxanthin diester, $4.9\%$ diatoxanthin diester, $3.1\%$ astaxanthin, $2.4\%$ triol, $2.3\%$ diatoxanthin, $1.7\%$ isocrytoxanthin, $1.5\%$ lutein, $1.1\%$ zeaxanthin, $1.0\%$ cynthiaxanthin and $1.0\%$ zeaxanthin diester. Similarly, the carotenoid isolated in May composed of $25.3\%$ diatoxanthin monoester, $19.7\%$ cynthiaxanthin monoester, $13.0\%$ $\beta$-carotene, and $12.6\%$ zeaxanthin monoester, $5.8\%$ cynthiaxanthin diester, $5.1\%$ diatoxanthin, $3.0\%$ astaxanthin, $2.4\%$ triol, $2.2\%$ diatoxanthin, $1.3\%$ isocrytoxanthin, $1.2\%$ zeaxanthin, $1.1\%$ zeaxanthin diester, $1.0\%$ lutein and $0.9\%$ cynthiaxanthin. Based on these data, monoester-type carotenoids ($37.1\~57.6\%$) and diester-type carotenoids ($11.5\~17.2\%$) of total carotenoids in echiurid were the major carotenoids. Meanwhile, when the sea water temperature was elevated and the contents of total carotenoid in echiurids were decreased, the contents of zeaxanthin monoester, diatoxanthin monoester and cynthiaxanthin monoester were increased, but the contents of zeaxanthin diester, diatoxanthin diester and cynthiaxanthin diseter were decreased, indicating that changes in ester-type caroteoids were differently affected by the sea water temperature.

  • Licorice Extract and Erythritol, food additives used in korea, are widely used in foods as sweetener. Its application for use in food is regulated by the standard and specification for food additives but official analytical method far determination of these sweetener in food has not been established. Accordingly, we has been carried out to set up analytical method of the glycyrrhizic acid in several foods by the way of thin layer chromatography and high performance liquid chromatography glycyrrhizic acid is qualitative anaylsis technique consists of clean-up with a sep-pak $C_{18}$ cartridge, separation of the sweeteners by Silica gel 60 F254 TLC plate using 1-butanol:4Nammonia solution:ethanol (50:20:10) as mobile solvent. Also, the quantitative analysis for glycyrrhizic acid, was performed using Capcell prk $C_{18}$ column at wavelength 254nm and DW:Acetonitrile (62:38 (pH2.5)) as mobile phase. and we has been carried out to set up analytical method of the erythritol in several foods by the way of high performance liquid chromatography. erythritol is qualitative anaylsis technique consists of clean-up with a DW and hexane. The quantitative analysis for erythritol, was performed using Asahipak NH2P-50 column, Rl and DW:Acetonitrile (25:75) as mobile phase. The glycyrrhizic acid results determined as glycyrrhizic acid in 105 items were as follows; N.D$\∼$48.7ppm for 18 items in soy sauce, N.D$\∼$5.3ppm for 12 items in sauce, N.D$\∼$988.93ppm for 15 items in health food, N.D$\∼$180.7ppm for 26 items in beverages, N.D$\∼$2.6ppm for 8 items in alcoholic beverages repectively and ND for 63 items in the ethers. The erythritol results determined as erythritol in 52 items were as follows; N.D$\∼$155.6ppm for 13 items in gm, N.D$\∼$398.1ppm for 12 items in health foods repectively and ND for 45 items in the others.

  • Distribution of lipid components in the tissue of meat, skin and viscera from carp(Cyprynus carpio) was analyzed using the techniques of column chromatography, thin layer chromatography and gas liquid chromatography according to the previous report(Choi, et al., 1984). Lipid content was varied by the portion such as $3.88\%$ in meat (free lipid, $2.47\%$ ; bound lipid, $1.41\%$), $8.02\%$ in skin(free lipid, $5.65\%$ ; bound lipid, $2.37\%$) and $6.18\%$ in viscera (free lipid, $3.54\%$ ; bound lipid, $2.64\%$). In the all portions of the body, free lipid was composed of $68\%\;to\;92\%$ in neutral lipid, $3\%\;to\;6\%$ in glycolipid and $4\%\;to\;18\%$ in phospholipid whereas bound lipid was composed of $8\%\;to\;20\%$ in neutral lipid, $2\%\;to\;7\%$ in glycolipid and $47\%\;to\;62\%$ in phospholipid. The free lipids of the tissues on the each portion were mostly represented by triglycerides and some diglycerides, but free lipids in viscera contained considerable amounts of free fatty acids. The bound lipids, on the other hand, commonly comprised appreciable amounts of esterified sterol and hydrocarbon, and triglycerides. The phospholipid was mainly consisted of phosphatidyl choline, phosphatidyl ethanolamine and phosphatidyl serine in the both free and bound lipids, and much more phosphatidyl choline in the bound lipid. The predominant fatty acids of free and bound lipids were $C_{16:0},\;C_{18:0},\;C_{20:4},\;C_{22:6}\;and\;C_{18:2}$ acids in polar lipids, and $C_{16:0},\;C_{16:1},\;C_{18:0},\;C_{18:1}\;and\;C_{18:2}$ acids in non-polar lipids, whereas those of neutral lipids were $C_{14:0}(2.54{\sim}6.98\%),\;C_{16:0}(11.20{\sim}21.13\%)$ and $C_{18:0}(1.58{\sim}12.76\%)$ of saturated acids, $C_{16:1}(7.06{\sim}20.70\%),\;C_{18:1}(21.68{\sim}30.50\%)$ and $C_{20:1}(1.76{\sim}6.27\%)$ of monoenoic acids, and $C_{18:2}(4.50{\sim}6.89\%),\;C_{20:4}(1.52{\sim}4.29\%)$ and $C_{22:6}(0.73{\sim}6.62\%)$, respectively. In conclusion, the fatty acid compositions revealed apparent differences between the free lipid and bound lipids in the tissues of body.

Comparative Studies on the Lipid Composition in Varieties of Dent Corn (마치종(馬齒種)옥수수의 품종별(品種別) 지질조성(脂質組成)의 비교(比較))

  • This study dealt with the comparison of the individual lipid component and fatty acid composition in the six varieties of dent corn, Zea mays Indentata. The fatty acid and sterol compositions of the total lipid were analyzed by gas liquid chromatography. The total lipid was also fractionated into three lipid classes namely neutral, glyco and phospolipid by the methods of silicic acid column chromatography. The lipid componets of lipid-classes were estimated by thin layer chromatography and TLC-scanner. The contents of total lipid in six varieties of Cdent corn were $3.7{\sim}5.3%$. Total lipid were mainly Composed of triglyceride$(69.8{\sim}75.7%)$ free fatty acid$(13.0{\sim}17.9%)$, lanosterol$(4.8{\sim}6.0%)$. hydrocarbon & esterified sterol$(3.5{\sim}6.0%)$, and polar lipid & pigment$(2.7{\sim}5.9%)$. The contents of triglycerde in $Chech'{\breve{o}}nok$ and Hwangok No.3 were slightly higher then other varieties. The major fatty acid In total lipid from six varieties of dent corn were chiefly consisted of linoleic$(46.0{\sim}61.4%)$, oleic$(21.9{\sim}29.9%)$ and palmitic acid$(10.9{\sim}16.7%)$. Particularly the content of linoleic acid in $Chech'{\breve{o}}nok$ was higher but oleic and palmitic acid in $Chech'{\breve{o}}nok$ were less than other varieties. The compositions of 4-desmethylsterol were mainly composed of siterol $(44.0{\sim}63.2%)$, campestetel$(11.6{\sim}15.5%)$ and stigmasterol$(5.6{\sim}9.1%)$. The content of sitosterol in Chinjuok was higher than other varieties and isofucosterol was detected only in Chinjuok. The compositions of 4-monomethlysterol were mainly composed of obtusifoliol$(17.7{\sim}37.6%)$, gramisterol$(15.0{\sim}27.0%)$ and citrostadienol$(9.1{\sim}17.3%)$. The contents of obtusifoliol and citrostadienol in Kwangok and Chinjuok were less than other varieties. The contents of fractionated neural lipid in Suwon No.19. Kwangok, $Hoengs{\breve{o}}ngok$ and Chinjuok$(90.3{\sim}97.1%)$ were higher than those of $Chech'{\breve{o}}nok$ and Hwangok No.3$(85.5{\sim}86.1%)$. Neutral lipid were mainly composed of triglyceride$(24.7{\sim}80.0%)$, lanosterol$(6.2{\sim}20.2%)$, Cholesterol$(1.0{\sim}50.6%)$, free fatty acid$(4.4{\sim}8.9%)$ and esterified sterol$(1.5{\sim}15.9%)$. The major fatty acid in neutral lipid from six varieties of dent corn were chiefly consisted of linoleic$(26.2{\sim}55.4%)$ oleic$(22.7{\sim}39.1%)$ and palmitic acid$(11.4{\sim}41.6%)$. Particularly the contents of linoleic acid Suwon No.19 and $Chech'{\breve{o}}nok$ were higher but palmtic acid in Suwon No.19 and $Chech'{\breve{o}}nok$ were less tan other varieties. Glycolipd were mainly composed of nlonoglycosflsterol $(17.5{\sim}56.4%)$, monoglycosylce-ramide $(8.2{\sim}25.9%)$ and monoglycoslydiacylglycerol$(12.4{\sim}22.2%)$. The contents of mono-g1ycosrlceramide and monoglycosrlsterol in Chinjuok. Were higher than other varieties. The major fatty acid in glycolipid from six varieties of dent corn were chiefly consisted$(14.6{\sim}39.3%)$, palmitic$(20.0{\sim}26.1%)$, linoleic$(3.6{\sim}26.9%)$ and heptadecanoic acid $(3.3{\sim}24.7%)$. Particutarly the cantents of oleic acid in Chinjuok and heptadecanoic acid in $Chech'{\breve{o}}nok$ were higher than other varieties. Phospholipid were mainly composed of phosphatidyllnositol$(30.9{\sim}86.4%)$ and phosphatidylcholine$(4.5{\sim}22.0%)$. The contents of phosphatidrlinositol in $Hoengs{\breve{o}}ngok$ and Hwanngok No. 3 were less than other varieties. The major fatty acid in phospolipid from six varieties of dent corn chiefly consisted of patmitic$(37.2{\sim}61.6%)$ heptadecanoic$(9.2{\sim}31.8%)$ and oleic acid$(4.3{\sim}17.2%)$. Particuiarlr the content of oleic acid in $Hoengs{\breve{o}}ngok$ was higher but heptadccanoic acid in $Hoengs{\breve{o}}ngok$ was less than othcr varieties.

  • Various kinds of medicinal herbs and prescribed herb medicines which have usually been used for treatment of reducing fever, purging intense heat, and detoxication were secreened for antibacterial activity against Proteus vulgaris, S. aureus, and E. faecalis which causing puerperal infection. Minimal inhibitory concentrations of water and ethanol-soluble extracts of Hwangyon(C. japonica), Omae(P. mume), and Omiza(S. chinensis) were determined and using thin-layer chromatography the extracts of Hwangyon and Omiza were separated. The results obtained were as follow : 1. The water-soluble extracts of Keumeunhwa(L. japonica), Chiza(G. jasminoides), Hwangyon(C. japonica), Dansam(S. miltiorrhiza), Omae(P. mume) and Omiza(S. chinensis) showed antibacterial activities against P. vulgaris ; among them, the extracts of Hwangyon, Omae, and Omiza showed high antibacterial activities. The water-soluble extracts of Keumeunhwa(L. japonica), Hwangyon(C. japonica), Hwangkeum(S. baicalensis), Hwangbaek(P. amurense), Dansam(S. miltiorrhiza), Daehwang(R. coreanum), Omae(P. mume), Omiza(S. chinensis), Gingseng(P. ginseng), and Gamcho(G. uralensis) showed antibacterial activities against S. aureus : among them, the extracts of Hwangyon and Omiza showed high antibacterial activities. The water-soluble extracts of Banggi(C. trilobus), Daehwang(R. coreanum), Omae(P. mume), Omiza(S. chinensis), and Gamcho(G. uralensis) inhibited the cell growth of E. faecalis : among them, Omae and Omiza showed high antibacterial activities. The extract of Hwangyon(C. Japonica) and Omae(P. mume) showed no inhibition against E. coli. However, the extract of Omiza(S. chinensis) inhibited cell growth of both E. coli and B. subtilis. 2. The water- and ethanol-soluble extract of Hwangyon(C. Japonica) showed antibacterial activities against P. vulgaris and S. aureus, those of Omae(P. mume) against P. vulgaris and E. faecalis, and those of Omiza(S. chinensis) against all species tested, P. vulgaris, S. aureus, and E. faecalis. With the exception that ethanol-soluble extract of Hwangyon(C. japonica) showed much higher antibacterial activity against S. aureus than water-soluble one, antibacterial activities of both water-and ethanol-soluble ones were similar to each other of other two medicinal herbs. 3. When the prescribed herb medicines were tested, Sambohwan showed antibacterial activities against P. vulgaris and S. aureus. No prescribed herb medicine inhibitied the cell growth of E. faecalis. 4. Minimal inhibitory concentrations(MICs) of both water- and ethanol-soluble extracts of Hwangyon(C. japonica), Omae(P. mume), and Omiza(S. chinensis) against P. vulgaris were $2.5mg/m{\ell}$, $10mm{\ell}$, and $20mg/m{\ell}$, respectively. Those of both Hwangyon(C. japonica) and Omiza(S. chinensis) against S. aureus were $1.25mg/m{\ell}$ and $10mg/m{\ell}$, respectively. MICs of water-soluble extracts of Omae(P. mume) and Omiza(S. chinensis) against E. faecalis were $2.5m{\ell}$ and $5m{\ell}$, and those of ethanol-soluble extracts were $5m{\ell}$ and $10m{\ell}$, respectively Except for those of E. faecalis the cell growth of P. vulgaris and S. aureus were inhibited by much lower concentration of ethanol-soluble extracts used. As a result, the antibacterial compounds against P. vulgaris S. aureus, and E. aecalis are contained in the extracts of Hwangyon(C. japonica), Omae(P. mume), Omiza(S. chinensis), the prescribed herb medicine, Sambohwan, and might be used for treatment of puerperal infection. Further study should be carried out to identify which compounds affect the cell growth inhibition of P. vulgaris S. aureus, and E. faecalis.
