Bacterial Community Analysis during Composting of Garbage using Denaturing Gradient Gel Electro-phoresis (Denaturing Gradient Gel Electrophoresis를 이용한 음식물 쓰레기 퇴비화 세균 군집 분석)

  • The microbial community during composting of gargage was analyzed using 16S rDNA PCR - DGCE (denaturing gradient gel electrophoresis). Pseudomonas spp. was found throughout the process, and thermophilic Bacillus spp. was dominated at the thermophilic stage. Six thermophilic bacteria were isolated and identified as B. caldoxylolyticus, B. thermoalkalophilus, and B. thermodenitrificans.

A Study on Reduction of Food Waste (음식물 쓰레기 소멸화에 관한 연구)

  • The physical and chemical transformation and reduction degree of food waste were investigated in a food waste reduction machine using thermophilic bacteria. The first operation of the reduction machine for grain, vegetables, fishes and flesh wastes proceeded during three weeks. The first and second reduction percentages of the wastes were 98.3% and 93.2%, respectively. The residue of food waste was composed of fruits, fish, and vegetables. The temperature distribution of the reduction machine ranged between 30 and 6$0^{\circ}C$ appropriate for growth of thermophilic bacteria. At initial stage the pH in the reduction machine decreased with organic acids produced, but increased as the organic acids decomposed by different thermophilic bacteria. In the reduction machine, the moisture content of the food waste was reduced from 80-90% to 10-20% after 24 hours, and the salinity of residue was 0.29% after the second operation. The degree of odor was most high between 2 and 4 hours.

Morphological features of thermophilic activated sludge treating food industry wastewater in MBR

  • Microscopic examination of the activated sludge and morphological characterization of the flocs provides detailed information about the treatment process. The aim of this study is to investigate the morphological parameters of flocs obtained from a thermophilic jet loop membrane bioreactor (JLMBR) in different sludge retention times (SRTs), considering EPS and SMP concentration, hydrophobicity, zeta potential. The results showed that irregularity decreased with the increasing SRT. The compactness value was calculated to be less than 1 for all SRTs. However, the sludge had a more compact structure when the SRT increased. Zeta potential increased whereas hydrophobicity and floc size reduced, with increasing SRT. Furthermore, 2-D porosity calculated using the hole ratio was higher at greater SRTs. Hence, there was a significant correlation between the results obtained using the imaging technique and operation conditions of thermophilic JLMBR.

  • The thermophilic aerobic digestion (TAD) process is recognized as an effective method for rapid waste activated sludge (WAS) degradation and the deactivation of pathogenic microorganisms. Yet, high energy costs due to heating and aeration have limited the commercialization of economical TAD processes. Previous research on autothermal thermophilic aerobic digestion (ATAD) has already reduced the heating cost. However, only a few studies have focused on reducing the aeration cost. Therefore, this study applied a two-step aeration control strategy to a fill-and-draw mode semicontinuous TAD process. The NADH-dependent fluorescence was monitored throughout the TAD experiment, and the aeration rate shifted according to the fluorescence intensity. As a result, the simple two-step aeration control operation achieved a 20.3% reduction in the total aeration, while maintaining an effective and stable operation. It is also expected that more savings can be achieved with a further reduction of the lower aeration rate or multisegmentation of the aeration rate.

  • This study has been conducted to optimum operating conditions for effective acid fermentation according to OLR(organic loading rate) in the mesophilic and thermophilic acid fermentation process. The results are summarized as follows. In order to obtain reasonable acid fermentation efficiency in performing acid fermentation of food wastes in thermophilic condition, organic loading rate was required below 20 gVS/L.d. As $SCOD_{Cr}/TKN,\;SCOD_{Cr}/T-P$ of thermophilic acid fermented food wastes In organic loading rate 20 gVS/L.d were 18.9, 73.4 respectively, it was possible to utilize as external carbon source for denitrification in sewage treatment plant after solid-liquid separation as well as co-digestion of fermented food wastes and sewage sludge.

Inactivation of Pathogenic Bacteria by Addition of Thermophilic Bacteria in the Thermophilic Aerobic Oxidation(TAO) System (고온호기산화장치의 고온미생물 첨가에 의한 병원성 미생물의 불활성화)

  • This study analyzed temperature increase, microorganism changes, and inactivation of pathogenic microorganisms in pig slurry when treated with thermophilic microorganisms in Thermophilic Aerobic Oxidation(TAO) system. An amount of $6 m^3$ of pig slurry was treated in an $18 m^3(3.0\times2.5\times2.4 m)$ reactor for 5 to 7 days in two groups: the control of pig slurry only and the treatment of pig slurry with 6 liters of thermophilic microorganism(Bacillus sp.). To study the microorganism changes in the reactor, the populations of aerobic mesophilic microorganisms, thermophilic microorganisms and general pathogens were analyzed. To study the inactivation of pathogenic microorganisms, the levels of E. coli, Salmonella sp, Crytosporidium parvum and Giardia lamblia were analyzed. The temperature inside the reactor ranged from 18 to $62^{\circ}C$ for the control while far the treatment group it ranged from 18 to $66^{\circ}C$, showing a slightly higher array. With regard to changes in microorganisms, both mesophilic and thermophilic organisms decreased from $3.1\times10^6$ to $1.2\times10^2$ CFU/ml and from $1.0\times10^4$ to $8.0\times10^1$ CFU/ml, respectively, in the control. In the treatment, on the other hand, mesophilic organisms decreased from $3.0\times10^8$ CFU/ml to $8.6\times10^5$ CFU/ml while thermophilic organisms increased sharply from $2.0\times10^6$ to $1.2\times10^8$ CFU/ml. For pathogens, Salmonella and Giardia were not detected either before or after the treatment, while E. coli and C. parvum were found to be $10^5$ CFU/ml each before treatment and negative after it. From this experiment, it was concluded that thermophilic microorganisms could effectively sanitize liquid compost by generating high temperature in the TAO system, which in turn would inhibit the growth of pathogenic organisms.

Treatment of Food Waste Leachate using Lab-scale Two-phase Anaerobic Digestion Systems (실험실 규모 2상 혐기성 소화를 이용한 음식물 쓰레기 탈리액의 처리)

  • This study was performed to evaluate the treatability of food waste leachate using lab-scale two-phase anaerobic digestion system. Effects of influent pH, hydraulic retention time (HRT), and recycle of methanogenic reactor effluent to the thermophilic acidogenic reactors were investigated. For methanogenic reactors, effects of internal solids recycle and temperature were studied. Performance of the acidogenic reactors was stable under the conditions of influent pH of 6.0 and HRT of 2 d with the recycle of methanogenic reactor effluent, and acidification and VS removal efficiency were about 30% and 40%, respectively. Up to the organic loading rate (OLR) of 7 g COD/L/d, effluent SCOD values of mesophilic and thermophilic methanogenic reactors either lower or kept the same with the internal solids recycle. Also, decreasing tendency in specific methane production (SMP) due to the organic loading increase became diminished with the internal solids recycle. Mesophilic methanogenic reactors showed higher TCOD removal efficiency and SMP than thermophilic condition under the same OLR as VSS was always higher under mesophilic condition. In sum, thermophilic acidogenesis-mesophilic methanogenesis system was found to be better than thermophilic-thermophilic system in terms of both organic removal and methane production.

Application of a Thermophilic Aerobic Digestion Process to Industrial Waste Activated Sludge Treatment

  • Thermophilic aerobic bacteria were applied in the degradation of industrial waste activated sludge (WAS) on a laboratory scale expreiment. The performance of digestion was estimated by measuring the reduction of total suspended solids (TSS), dissolved organic carbon (DOC), and total organic carbon (TOC). Among three strains of Bacillus stearothermophilus and three strains of Thermus species, B. stearothemophilus ATCC 31197 showed the best overall efficiency level for the degradation of industrial WAS, which was collected from a wastewater treatment plant in an oil refinery factory. Industrial WAS coul be successfully detraded in a batch digestion with ATCC 31197. The stability of the digestion process with ATCC 31197 was successfully verified by semi-continuous (fill-and-draw) digestion experiment. From the results of this study, it was shown that the thermophilic aerobic digestion process with ATCC 31197 could efficiently be applied to the degradation of industrial WAS.

Increase of treatment amount of thermophilic oxic process considering calorie/water (C/W) ratio (칼로리/수분 (C/W)비를 고려한 고온호기 처리법에서의 처리량 증가)

  • The signification of calorie/water (C/W) ratio was investigated in the treatment of highly concentrated organic wastes by thermophilic oxic process (TOP). Swine waste was used in this study. When C/W ratio was 1.6, most of swine waste was decomposed and all water was evaporated in the 24-h injection cycle. To improve treatment efficiency of TOP treating swine waste, the effect of shortening the swine waste injection cycle was examined. The shortening of injection cycle was conducted to stimulate the activity of thermophilic bacteria. A high temperature in the reactor was maintained by shortening of the injection cycle. When the swine waste injection cycle was shortened, the C/W ratio was fixed at 1.6. As a result, by shortening the swine waste injection cycle from 24-h to 12 and 6-h, the maximum loading rate of swine waste per day could be improved 1.9 and 3.5 times, respectively.

Effect of Increasing Amounts of Ammonium Nitrogen Induced by Consecutive Mixture of Poultry Manure and Cattle Slurry on the Microbial Community during Thermophilic Anaerobic Digestion

  • Thermophilic anaerobic digestion (TAD) is characterized by higher biogas production rates as a result of assumedly faster microbial metabolic conversion rates compared to mesophilic AD. It was hypothesized that the thermophilic microbiome with its lower diversity than the mesophilic one is more susceptible to disturbances introduced by alterations in the operating factors, as an example, the supply of nitrogen-rich feedstock such as poultry manure (PM). Laboratory scaled TAD experiments using cattle slurry and increasing amounts of PM were carried out to investigate the (in-) stability of the process performance caused by the accumulation of ammonium and ammonia with special emphasis on the microbial community structure and its dynamic variation. The results revealed that the moderate PM addition, i.e., 25% (vol/vol based on volatile substances) PM, resulted in a reorganization of the microbial community structure which was still working sufficiently. With 50% PM application, the microbial community was further stepwise re-organized and was able to compensate for the high cytotoxic ammonia contents only for a short time resulting in consequent process disturbance and final process failure. This study demonstrated the ability of the acclimated thermophilic microbial community to tolerate a certain amount of nitrogen-rich substrate.
