Some sulfur coompounds dissimilation characteristcs of Thermococcus sp. DT1331 were studied. DT1331 had ether-like lipid compounds in addition to esters in the cell membrane. The specific growth rates of DT1331 decreased with increasing head spaces of the cultures. However, when the ratio of head space volume to medium volume was 5.60, the strain showed no growth. DT1331 showed vigo- rous growth with 1% or more elemental sulfur addition. Cystine could substitute elemental sulfur and DT1331 showed moderate growth with 0.1% or more cystine concentration. The specific growth rate and maximum cell concentration of Thermococcus sp. DT1331 in the presence of elemental sulfur were 0.80 hr$^{-1}$ and 2.0 x 10$^{8}$ cells/ml, respectively, while they were 0.67 hr$^{-1}$ and 1.1 x 10$^{8}$ cells/ml, respectively in the presence of cystine.
The hyperthermophilic archaebacterium Thermococcus profundus Isolated from a deep-sea hydrothermal vent system, produced several amylolytic enzymes such as extracellular amylase and pullulanase, intracellular a-1,4-91ucosidase in respone to the presence of complex carbohydrates In the growth medium. This strain showed high activities on 0.5% maltose than on complex carbohydrates One of the amylases was partially purified by ammonium sulfate precipitation, DEAE-Toyopearl chromatography. The amylase exhibited maximal activity at pH 5.5 and 80$^{\circ}C$, and was stable in the range of pH 5.5 to 9.5 and up to 80$^{\circ}C$ for 30 min. The enzyme activity was no dependence on Ca2+ and not inhibited by detergents. The amylase hydrolyzed soluble starch, amylose, amylopectin and glycogen to produce maltose and maltotriose with trace amounts of glucose, but not pullulan and ${\alpha}$-, ${\beta}$-, ${\gamma}$-cyclodextrin. Malto-oligosaccharides ranging from maltotetraose to maltoheptaose were hydrolyzed in an endo fashion.
열수구(Hydrothermal vent)는 빛이 없는 환경에서 생명체의 진화가 일어나는 독특한 환경을 유지하고 있다. 남태평양 Tonga의 Tofua arc의 열수구로부터 퇴적물을 채취하여 산화철[iron(III)], 황(elemental sulfur, $S^0$) 그리고 질산염을 전자수용체로 사용하고, 수소($H_2$), yeast extract를 전자공여체로 사용하여 배양에 의한 미생물의 다양성을 연구하였다. 배양 온도는 각각 $65^{\circ}C$와 $80^{\circ}C$였으며, 연속희석배양법과 16S rRNA 유전자의 PCR-Denaturing Gradient Gel Electrophoresis를 분석하고, 검출된 염기서열의 정보분석을 통하여 고세균을 동정하였다. 16S rRNA 유전자의 계통분류학적 분석 결과 배양된 대부분의 고세균은 Thermococcus 속(T. alcaliphilius, T. litoralis, T. celer, T. barossii, T. thoreducens, T. coalescens)에 속하며 그들과 98-99%의 상동성을 가지고 있었다. Thermococcus 속의 미생물들이 일반적으로 이용할 수 없는 질산염과 산화철을 전자수용체로 첨가한 배양에서 관찰되었으나, 이는 환원제로 첨가한 $Na_2S$의 산화물을 이용하여 성장한 것으로 추정된다. Thermococcus 속에 속하는 고세균 외의 다양한 고세균의 배양을 위해서는 $Na_2S$ 대신 다른 환원제를 사용하는 배양조건의 이용이 요구된다.
초고온 archaeon인 Thermococcus profundus에서 실린드형태의 단백질 복합체를 분리, 생화학적 특성과 구조를 규명하였다. 전자현미경과 영상처리 (image processing)를 이용한 구조 분석으로 8개의 subunit으로 이루어진 링 (ring) 형태가 두 개 겹쳐져 가운데 통로를 가지는 실린드형태로 이루어진 복합체임을 알 수 있었다. 또한 60kDa단백질(P60 complex)로 구성된 homomultimer임을 보여 주고 있다. 이 복합체는 $80^{\circ}C$ 이상에서 도 강한 ATPase의 활성을 보여주는 강한 내열성 단백질임을 알 수 있다. P60 complex는 초고온 archaeon이 높은 온도에서 적응하며 생존하는데 매우 중요한 역할을 할 것으로 추정할 수 있다.
초고온 archaeon인 Thermococcus profundus에서 매우 거대한 단백질 복합체를 분리 및 구조를 규명하였다. 거대 복합체는 93kDa단백질(P93 complex)로 구성된 homomultimer이며, 강한 내열성을 보여주고 있다. 순수 분리한 P93 complex를 SDS(최종 농도 1%)와 $85^{\circ}C$에서 12시간 항온시킨 후, SDS-PAGE와 전자현미경에서 구조적 변화를 관찰할 수 없었다. 음착색된 P93 complex의 전자현미경 사진에서 하나의 형태만을 보여주고 있으며, 구조의 규명을 위해 image processing을 하였다. 이의 구조는 3대칭 중심에 core(혹은 hole)이 뚜렷이 존재하며 이를 중심으로 단백질이 모여 있는 형태를 보여 주고 있다. 또한 P93 complex는 가장자리에서 뚜렷한 형태를 보여주지 않은 부분, 즉 flexible부분을 포함하고 있다. Gel filtration과 2차원 구조를 기초로 P93 complex는 24 homomultimer로 되어 있음을 추정하였다.
A novel hyperthermophilic, anaerobic, heterotrophic archaeon, designated strain $NA1^T$, was isolated from a deep-sea hydrothermal vent area (depth, 1,650 m) within the Papua New Guinea-Australia-Canada-Manus (PACMANUS) field. Cells of this strain were motile by means of polar flagella, coccoid-shaped with a diameter of approximately $0.5-1.0{\mu}m$, and occurred as single cells. Optimal temperature, pH, and NaCl concentration for growth were $80^{\circ}C$, 8.5, and 3.5%, respectively. The new isolate was an obligate heterotroph that utilized yeast extract, beef extract, tryptone, peptone, casein, and starch as carbon and energy sources. Elemental sulfur was required for growth and was reduced to hydrogen sulfide. The G+C content of the genomic DNA was 52.0 mol%. Phylogenetic analysis of the 16S rRNA gene indicated that strain $NA1^T$ belongs to the genus Thermococcus, and the organism is most closely related to T. gorgonarius, T. peptonophilus, and T. celer; however, no significant homology was observed among species by DNA-DNA hybridization. Strain $NA1^T$ therefore represents a novel species for which the name Thermococcus onnurineus sp. novo is proposed. The type strain is $NA1^T$ (=KCTC 10859, =JCM 13517).

Genome organization near cyclomaltodextrinases (CDases) was analyzed and compared for four different hyperthermophilic archaea: Thermococcus, Pyrococcus, Staphylothermus, and Thermofilum. A gene (CL1_0884) encoding a putative CDase from Thermococcus sp. CL1 (tccd) was cloned and expressed in Escherichia coli. TcCD was confirmed to be highly thermostable, with optimal activity at $85^{\circ}C$. The melting temperature of TcCD was determined to be $93^{\circ}C$ by both differential scanning calorimetry and differential scanning fluorimetry. A size-exclusion chromatography experiment showed that TcCD exists as a monomer. TcCD preferentially hydrolyzed ${\alpha}$-cyclodextrin (${\alpha}$-CD), and at the initial stage catalyzed a ring-opening reaction by cleaving one ${\alpha}$-1,4-glycosidic linkage of the CD ring to produce the corresponding single maltooligosaccharide. Furthermore, TcCD could hydrolyze branched CDs (G1-${\alpha}$-CD, G1-${\beta}$-CD, and G2-${\beta}$-CD) to yield significant amounts (45%, 40%, and 46%) of isomaltooligosaccharides (panose and $6^2$-${\alpha}$-maltosylmaltose) in addition to glucose and maltose. This enzyme is one of the most thermostable maltogenic amylases reported, and might be of potential value in the production of isomaltooligosaccharides in the food industry.
A hyperthermophilic archaeon, $Thermococcus$$onnurineus$ NA1 was studied to investigate its fermentation characteristics using various carbon sources including formate, maltose and carbon monoxide during the anaerobic batch cultivation at $80^{\circ}C$. Formate was the best carbon source for cell growth and hydrogen production among others. In the batch culture on formate, it was found that the cell concentration increased exponentially by 12 hrs of culture, after which the cell growth and formate consumption was retarded. Hydrogen production was continued more than 24 hrs although the cell growth was ceased at 18 hrs. Hydrogen production rate was directly correlated with the cell growth and formate degradation up to 18 hrs, and the average hydrogen production yield was 1.05 mole-$H_2$/mole-formate. Cell growth and hydrogen production were optimized at the initial pH 6-7, while inhibited at the initial pH lower than 5 and higher than 9.

$OGL-20P^T$-358 is a novel 66 amino acid residue protein from the hyperthermophile Thermococcus thioreducens sp. nov., strain $OGL-20P^T$, which was collected from the wall of the hydrothermal black smoker in the Rainbow Vent along the mid-Atlantic ridge. This protein, which has no detectable sequence homology with proteins or domains of known function, has a calculated pI of 4.76 and a molecular mass of 8.2 kDa. We report here the backbone $^1H$, $^{15}N$, and $^{13}C$ resonance assignments of $OGL-20P^T$-358. Assignments are 97.5% (316/324) complete. Chemical shift index was used to determine the secondary structure of the protein, which appears to consist of primarily ${\alpha}$-helical regions. This work is the foundation for future studies to determine the three-dimensional solution structure of the protein.
초고온성 고세균 Thermococcus onnurineus NA1은 개미산, 일산화탄소, 또는 전분 등을 이용해서 수소를 생산하는 것으로 알려져 있다. 본 연구에서는 T. onnurineus NA1의 고정화 세포를 이용한 수소생산을 고찰하였다. 고정화 실험결과, T. onnurineus NA1은 표면에 아민기가 코팅된 규조토 담체에 정전기적 인력에 의해 효과적으로 고정화되었고, 1 g의 담체에 고정화 될 수 있는 최대 세포의 양은 71.7 mg-dcw로 확인되었다. 고정화 세포를 이용한 세 번의 반복회분식 배양을 통해 개미산으로부터 수소생산 특성을 고찰하였고, 그 결과 배양이 반복됨에 따라 고정화 세포 농도의 증가에 기인하여 초기수소생산속도가 2.3 에서 4.0 mmol l−1 h−1로 상당량 증가됨이 관찰되었다. 따라서, T. onnurineus NA1의 고정화세포 시스템은 수소생산을 위한 좋은 대안이 될 수 있을 것으로 사료된다. 본 연구는 초고온성 고세균의 고정화세포를 수소생산에 적용한 첫 번째 사례이다.
