국내 젖소에서 Theileria buffeli 주요 표면 단백질 유전자의 다양성 분석 (The polymorphism of Theileria buffeli major surface protein associate with their clinical signs in holstein in Korea)

  • Theileria (T.) buffeli (formerly T. sergenti/T. orientalis) is the major hemo-protozoan distributed in the Far East Asian countries such as Korea, China and Japan. It is responsible for the clinical symptoms of anorexia, ateliosis, anemia, fever and icterus. It also causes abortion and sudden death under severe cases, resulting in economic losses for many livestock farms. The objective of this study was to analyze the genetic diversity of the major surface protein (Msp) gene in T. buffeli in Holstein in Korea, and we characterized the association of the diversification of the Msp gene and its relationship with the pathogenicity of Theileria. For this, complete blood counts and Theileria PCR sequence analysis were performed from 57 Holstein in Jeju Island. A total of 26 PCR positive Holstein (16 anemic and 10 non-anemic) were then randomly selected based on 18s rRNA sequence typing of the Theileria Msp gene. The DNA sequence of the T. buffeli Msp gene in Holstein showed 99.0%, 99.2%, 99.9%, 99.5%, 98.7%, 98.4% and 98.4% homology with T. sergenti, Theileria spp., T. sergenti, Theileria spp., Theileria spp., Theileria spp. and Theileria spp., respectively. The result showed a genetic variation of 57.7% (type I), 3.8% (type II), 15.4% (type III), 7.7% (type IV), 13.5% (type V) and 1.9% (type VI). Type I is the most frequent type in both anemic and non-anemic Holstein while type II was found in only non-anemic Holstein. This results of our study help confirm the diversity of Msp gene types and demonstrate that the gene type distribution of Msp genes varies among Theileria-infected Holstein in Jeju Island.

국내 한우의 타일레리아 주요항원단백질 유전자의 다양성 (Genetic Diversity in the Major Surface Protein Gene of Theileria Buffeli in Korean Indigenous Cattle)

  • 본 연구는 국내 타일레리아에서 주요항원단백질(major surface protein) 유전자의 다양성을 분석해 보고자 수행되었다. 나아가 Msp 유전자의 다양성과 타일레리아의 병원성과의 관계도 분석하였다. 제주에 있는 목장으로부터 총 177마리의 한우 혈액을 공시재료로 사용하여 혈액검사와 18S rRNA를 표적으로 하는 PCR을 실행하였다. 그 후, 타일레리아 18S rRNA에 양성인 28마리 (16마리 빈혈군과 12마리의 정상군)를 무작위로 선발하여 Msp유전자의 염기 서열을 반복하여 분석하였다. 총 56개의 염기서열 결과는 다변성 부위(517-571 bp)에 따라 크게 type I에서 type V까지 5가지 형태로 나눌 수 있었는데, 이는 유전자은행(GenBank)에 등록되어 있는 다음의 유전자와 98.9% 이상 일치하였다 (Theileria spp. from China-EU584237; T. sergenti from China-DQ078264; Theileria spp. from Thailand-AB081329; Theileria spp. from Japan-AB218442; T. sergenti from Japan-AB016280). 그 분포는 22, 15, 9, 8, 2개가 각각 type I에서 V까지 분포하였고 빈혈과 관계없이 type I이 가장 많이 나타나는 것으로 밝혀졌다(37.5%의 빈혈군과 41.7%의 정상군). 나머지 type중에서는 type II가 빈혈군에서 가장 많이(37.5%) 나타났으며, 반면 type IV는 정상군에서 많이 (25%) 나타났다. 본 연구는 국내 타일레리아 Msp유전자의 다양성을 밝히는데 좋은 자료로 활용될 수 있을 것이다.

Theileria sergenti 분열소체(merozoite)의 분열전체(schizont)로의 복귀 (Reversion of Theileria sergenti merozoite to schizont)

  • Theileria spp.의 생활환에 대해서는 여러종류의 책을 통해서 잘 소개되어 있다. 그중 Theileria spp.의 분열소체 즉, merozoite는 주로 숙주의 적혈구내에 존재하는데 진드기에 의해 흡혈되지 않으면 더이상 발육하지 못하고 생을 마감한다고 여겨 왔다. 그러나 적혈구내 merozoite가 임파구에 다시 들어가 schizont로 복귀하여 분열 증식된다는 가설은 아직까지 증명된 바 없다. 본 실험은 T sergenti merozoite의 schizont로의 복귀를 입증하고자 수행되었다. T sergenti에 감염되지 않은 3개월령의 송아지를 비장적출시킨 후 T sergenti merozoite에 감염된 순수적혈구를 인공감염시켰다. 인공감염후 경시적으로 혈액과 임파액을 채취하여 적혈구내 T sergenti 감염을 조사하고 백혈구 감별혈구를 계산하였으며 임파구내 schizont 출현을 관찰하여 다음과 같은 결과를 얻었다. 1. 혈구내원충 감염율(parasitemia : PE)은 인공감염후 28일째 최고치인 10.5%를 보였으며 그후 5%이내의 수준을 유지하다가 70일째 다시 8.5%의 상승점을 보였다. 2. 백혈구 감별혈구계산에서는 감염초기에는 호중구가 주종을 이루다가 감염후 19일을 기점으로 임파구(60~80%)가 급격히 증가하여 실험종료 때까지 유지되었다. 3. 인공감염후 19~23일, 59~63일 사이에 말초혈액내 임파구에서 분열 증식하고 있는 schizont를 관찰할 수 있었다. 4. 인공감염후 7일부터 림프액내 임파구의 크기가 커지면서 blast-formation이 진행되었으며 실험종료때까지 유지되었다. 이상의 결과로 보아 적혈구내 merozoite가 임파구에 다시 들어가 schizont로 복귀하여 분열 증식함을 입증하여 기존의 T sergenti 생활사는 수정되어야 된다고 사료된다.

전라북도 동부지역 사슴에서 진드기매개성 병원체에 대한 감염 실태 조사 (A survey for tick-borne disease agents from farm deer in the eastern area of Jeonbuk)

  • Ticks cause economic losses to the deer industry by decreasing the growth and production of the farmed animals. The mediation of ticks affects humans and animals by causing contagious disease both directly and indirectly. Blood from farmed deer from the areas near Jangsu branch was collected for screening of infectious protozoa and rickettsial disease. Seventy deer blood samples were collected from 30 different deer farms located in Jinan, Jangsu and Muju. This blood samples were used for blood slide smear examination and hematological analysis. DNA from these samples was extracted and was used for PCR analysis for detection of gene fragments of Theileria spp, Babesia spp, Anaplasma spp and Ehrlichia spp. In the blood slide smear examination and PCR analysis all samples did not show presence of protozoal and rickettsial diseases. Eight blood samples showed anemia, 1 sample showed iron deficiency and 7 samples showed regenerative anemia. Results for PCR analysis showed 2 samples were positive for T orientalis. All DNA samples were negative for Babesia spp, Anaplasma spp, and Ehrlichia spp.

Molecular phylogenetic studies on clinical bovine piroplasmosis caused by benign Theileria in Shaanxi Province, China

  • A group of benign Theileria species, which are often referred to as T. orientalis/T. buffeli/T. sergenti group, has low pathogenicity in cattle. Herein, we report on Theileria spp. in cattle on a farm from China. Based on phylogenetic analysis of the major piroplasm surface protein gene sequences, we detected 6 genotypes that were categorized as Types 1, 2, 3, 4, and 5 as well as an additional Type 9 genotype. The new epidemiological features of the T. orientalis/T. buffeli/T. sergenti parasites in China indicate a greater diversity in the genetics of these species than had been previously thought.

한국과 일본 소에 감염된 Theileria 분리주의 small subunit ribosomal 유전자의 동정 및 분석 (Identification and sequence analysis of small subunit ribosomal RNA gene of bovine Theileria isolates from Korea and Japan)

  • 한국과 일본의 서로 다른 지역으로부터 소의 Theileria 분리주에 있어서 6가지 type(A부터 E 그리고 H)과 subtype(B1)의 small subunit ribosomal RNA(SSUrRNA) 유전자를 밝혔다. 이들 유전자 염기서열을 비교하여 본 바 염기서열의 위치 212~231, 261~270 그리고 632~690으로 3군데의 hypervariable region이 관찰되었다. SSUrRNA 유전자 염기서열 type A는 한국의 전북 분리주(KCB), 충남 분리주(KCN), 제주 분리주(KCJ)그리고 실험실 보관주(KLS)와 일본의 Shintoku 분리주(JHS)인 5개의 분리주에서 나타났으며, 이 염기서열은 Kenya의 Marula 분리주인 Theileria buffeli의 SSUrRNA 유전자(GenBank accession number Z15106)와 일치하였다. 한국의 경북 분리주(KKB)에서는 type B만이 관찰되었으나 그 외의 분리주에서는 2 type 이상의 유전자 염기서열이 관찰되었다. KCB와 JHS 분리주에서는 type A와 B, 강원 분리주(KKW)에서는 type B와 H, KCN 분리 주에서는 type A, C 및 D 그리고 KCJ 분리주에서는 type A, B, E 및 subtype B1이 관찰되었다. 한국과 일본 소의 Theileria 분리주에 있어서 여러 type의 SSUrRNA 유전자 염기서열이 나타나는 것으로 보아 혼함감염이 되어 있는 것으로 판단된다.

Phylogenetic Analysis of Ruminant Theileria spp. from China Based on 28S Ribosomal RNA Gene

  • Species identification using DNA sequences is the basis for DNA taxonomy. In this study, we sequenced the ribosomal large-subunit RNA gene sequences (3,037-3,061 bp) in length of 13 Chinese Theileria stocks that were infective to cattle and sheep. The complete 28S rRNA gene is relatively difficult to amplify and its conserved region is not important for phylogenetic study. Therefore, we selected the D2-D3 region from the complete 28S rRNA sequences for phylogenetic analysis. Our analyses of 28S rRNA gene sequences showed that the 28S rRNA was useful as a phylogenetic marker for analyzing the relationships among Theileria spp. in ruminants. In addition, the D2-D3 region was a short segment that could be used instead of the whole 28S rRNA sequence during the phylogenetic analysis of Theileria, and it may be an ideal DNA barcode.

Molecular and serological surveillance of equine piroplasmosis in the Republic of Korea between 2016 and 2017

  • Equine piroplasmosis (EP) is caused by Babesia caballi and Theileria equi infection. We investigated antigen and antibody of EP in horses in the Republic of Korea during 2016-2017. Antigen and antibody of T. equi was detected 0.06% (1/1,650). Phylogenetic analysis of 18S rRNA revealed that the T. equi was highly homologous with the strains from China, Mongolia, and Spain. Two Theileria spp. were also detected and highly homologous with T. buffeli, T. luwenshuni, and T. orientalis.

서부 경남지역의 도살축우에 대한 주혈기생충의 역학적 조사 (Epizootiological survey of blood parasites in slaughtered cattle of western area of Kyeongnam)

  • This survey was conducted to observe the prevalence of blood parasites in slaughtered cattle included Korean native cattle, Charolias, Hereford, Aberdeenangus and Holstein breeds in the Western area of Kyeongnam. The results obtained are summarized as follows: 1. The prevalence of T sergenti was shown 71.8% as 395 heads of a total of 550 heads examined and from Jaunary to November the monthly prevalence of T sergenti was shown the range of 61.1% to 84% except 38.5% in December. The other blood parasites included Babesia and Anaplasma were not detected from the blood samples except Setaria spp microfilariae. 2. The distribution of parasitaemia levels of T sergenti in positive cattle was shown 93.9% in the range of 1~10/1000 rbc, 4.1% in 11~20, 1.3% in 21~30 and 0.8% above the range of 31. 3. The pervalence of T sergenti by breeds of slaughtered cattle was shown 71.2% in Korean native cattle, 72.7% in Charolias, 78.3% in Hereford and 81. 8% in others (Aberdeen-angus and Holstein) respectively. Also the parasitaemia levels in these cattle were shown higher levels in imported cattle included Charolias, Hereford, Aberdeen-angus and Holstein breeds comparing with Korean native cattle. 4. The prevalence of Setaria spp microfilariae in slaughtered cattle was shown 6.9% and by monthly prevalence of the parasite was shown higher in March, April and May compared with June, July, August and October. But in the winter season included January, February, November and December the parasite was not detected from the blood samples. 5. The distribution of parasitaemia levels of Setaria spp microfilariae per ml of blood was shown 65.8% in the range of 1~50, 13.2% in 51~100 and 10.5% in 101~200 and above the range of 201, respectively.
