Solution Dynamics Studies for the Lck SH2 Domain Complexed with Peptide and Peptide-Free Forms

  • It is well known that Src Homology 2(SH2) domain in many intracellular signal transduction proteins is very important. The domain has about 100 amino acid residues and bind phosphotyrosine-containing peptide with high affinity and specificity. Lck SH2 domain is a Src-like, lymphocyte-specific tyrosine kinase. An 11-residue phosphopeptide derived from the hamster polvoma middle-T antigen, EPQp YEEIPIYL, binds with an 1 nM dissociation constant to Lck SH2 domain. And it is known that the phosphotyrosine and isoleucine residues of the peptide are tightly bound by two well-defined pockets on Lck SH2 domain's surface. To investigate the conformational changes during complexation of SH2 domain with phosphopeptide we have performed the molecular dynamics simulation for Lck SH2 domain with peptide and peptide-free form at look in aqueous solution. More than 3000 water molecules were incorporated to solvate Lck SH2 domain and peptide. Periodic boundary condition has been applied in molecular dynamics simulation. Data analysis with the results of that simulation shows that the phosphopeptide makes primary interaction with the Lck SH2 domain at six central residues, The comparison of the complexed and uncomplexed SH2 domain structures in solution has revealed only relatively small change. But the hydrophilic and hydrophobic pockets in the protein surface show the conformational changes in spite of the small structural difference between the complex and peptide-free forms.

Phase Evolution and Electrical Properties of PZT Films by Aerosol-Deposition Method (에어로졸 증착법에 의해 제조된 PZT 막의 상변화와 전기적 특성)

  • $Pb(Zr_{0.52}Ti_{0.48})O_3$ (PZT) films with a thickness of $5{\sim}10{\mu}m$ at the morphotropic phase boundary were fabricated by aerosol-deposition (AD), and their phase evolution and electrical properties were investigated. The microstructure of the AD PZT films revealed nanosized grains with a low crystallinity and a dense structure at room temperature. The AD PZT films showed a mixture of tetragonal and rhombohedral phases. The post-annealing temperature was varied to study the phase transition behavior. The crystallinity of the AD PZT films was enhanced by annealing at 450, 550, and $650^{\circ}C$ for 2 h. At $650^{\circ}C$, the tetragonal and rhombohedral phases reacted to form a bridge phase between the two phases. The polarization-electric field hysteresis loops of the AD PZT film annealed at $650^{\circ}C$ exhibited a smaller cohesive field and a lower slim hysteresis than the films annealed at 450 and $550^{\circ}C$.

Wake Comparison between Model and Full Scale Ships Using CFD (CFD를 이용한 모형선과 실선 스케일의 반류 비교)

  • Assessment of hydrodynamic performance of a ship hull has been focused on a model ship rather than a full-scale ship. In order to design the propeller of a ship, model-scale wake is often extended to full-scale based upon an empirical method or designer's experience, since wake measurement data for a full-scale ship is very rare. Recently modern CFD tools made some success in reproducing wake field of a model ship, which implicates that there are some possibilities of the accurate prediction of full-scale wakes. In this paper firstly the evaluation of model-scale wake obtained by Fluent package was performed. It was found that CFD calculation with the Reynolds-stress model (RSM) provided much better agreement with wake measurement in the towing tank than with the realizable k-$\varepsilon$ model (RKE). In the next full-scale wake was calculated using the same package to find out the difference between model and full-scale wakes. Three hull forms of KLNG, KCS, KVLCC2 having measurement data open for the public, were chosen for the comparison of resistance, form factor, and propeller plane wake between model ships and full-scale ships.

  • The structural layout of mechanical and civil structures is commonly obtained using conventional methods. For example, the shape of structures such as electric transmission towers and offshore substructures can be generated systematically. However, with rapid advancements in computer graphic technology, advanced structural analyses and optimum design technologies have been implemented. In this study, the structural shape of a jacket substructure for an offshore wind turbine is investigated using a topology optimization technique. The structure is subjected to multiple loads that are intended to simulate the loading conditions during actual operation. The optimization objective function is defined as one that ensures compliance of the structure under the given boundary conditions. Optimization is carried out with constraints on the natural frequency in addition to the volume constraint. The result of a first step model provides quick insights into the optimum layout for the second step structure. Subsequently, a 3D model in the form of the frustum of a quadrilateral pyramid is developed through topology optimization.

  • Man has an unconscious desire to free oneself from everyday selves and a strong intellectual desire to see and analyze things in a different perspective. This provided a momentum for architectural design that has long been confined to its limits of modernism of being rational and objective. The ever-changing society that we live in filled with various attempts and pluralistic characteristics shows a complex set of attributes that can hardly be defined by a single term. As society moves towards an uncertain and volatile state, the boundary between literature, philosophy and art has dimmed out and converged. Such tendencies have been observable in recent establishments in architecture in the form of introducing external elements and create an alien and farfetched architectural space. Using the 'depaysement' defamiliarization method expressing irrationality and randomness, it is applied in various aspects, sharing the counter-rational structure and features of such architectural space. Hence, it is the purpose of this study to delve into the unique characteristics of depaysement through Magritte's work which reflects the unique pictorial world built under the influence of surrealism, and discover the $D\acute{e}paysement$ tendencies in modern architectural space. It is apparent in Modern architecture that, owing to the development in digital technology, it is showing process changes in design and an interfacial $D\acute{e}paysement$ effect in cyberspace, an interlace of reality and the virtual world. In this study, the focus of analysis was architectural properties such as the program, figure/spatial, material and the objective side, but due to mutual text properties, it is bound to be an overlapping analysis.

A Political Analysis of Fantasies of Supernatural Beings in Television Drama (대중문화 콘텐츠 속 초자연적 존재 판타지의 정치적 의미: <오 나의 귀신님>과 <싸우자 귀신아> 사례를 중심으로)

  • This study, by analyzing two recent television dramas, attempts to identify the ways how popular cultural texts deal with supernatural beings and to discuss political meanings of the ways in the context of neoliberal Korea. The results are: (1) The narratives make a clear line between the supernatural and the ordinary. (2) The supernatural is effectively used in the narratives to extend the boundary of conflict structure towards structural social problems that the society is now facing. (3) When the text resolving the conflicts, the supernatural is also critical, which makes the whle narrative in line with fantasy rather than reality. These results suggest that the conclusion of the previous studies, arguing the use of the supernatural by popular cultural texts tends to function as a form of resistance against neoliberal discourse structure, needs to be negotiated. It is also reaffirmed that we need to explain political meanings of popular cultural texts dealing with supernatural beings, with its double-sided and ambivalent effects.

  • This study is concerned with the educational program Visual Analysis Tool (VAT) for sound development for foreign intonation using personal computer. The VAT can run on IBM-PC 386 compatible or higher. It shows the spectrogram, waveform, intensity and the pitch contour. The system can work freely on either waveform zoom in-out or the documentation of measured value. In this paper, intensity and pitch contour information were used. Twelve French sentences were recorded from a French conversational tape. And three Korean participated in this study. They spoke out twelve sentences repeatly and trid to make the same pitch contour - by visually matching their pitcgh contour to the native speaker's. A sentences were recorded again when the participants themselves became familiar with intonation, intensity and pauses. The difference of pitch contour(rising or falling), pitch value, energy, total duration of sentences and the boundary of rhythmic group between native speaker's and theirs before and after training were compared. The results were as following: 1) In a declarative sentence: a native speaker's general pitch contour falls at the end of sentences. But the participant's pitch contours were flat before training. 2) In an interrogative: the native speaker made his pitch contours it rise at the end of sentences with the exception of wh-questions (qu'est-ce que) and a pitch value varied a greath. In the interrogative 'S + V' form sentences, we found the pitch contour rose higher in comparison to other sentences and it varied a great deal. 3) In an exclamatory sentence: the pitch contour looked like a shape of a mountain. But the participants could not make it fall before or after training.

  • The plastic arts is used when designers draw inspirations to create fashion design. The author referred to fashion magazines for designing ideas of Mondrian's geometrical abstract painting in practical applications used by designers in Paris, Italy, London and New York during the 10-year period(1991-2000). The collections of data were analyzed as following: ◇ Fashion Designing Idea 1. Matching Idea The art is reproduced in the design as how it is with no transformation. First, the painting's complete figure is reproduced on the entire or parts of clothing. Second, the painting's partial figure is reproduced on the entire or parts of clothing. 2. Contrasting Ideas The composition elements in Mondrian's geometrical abstract painting, for example, structures of shapes, vertical and horizontal lines and different colors are applied in the design. First, one particular shape in painting is transformed into different shape of square, circle or triangle and reproduced in designing. Second, one particular shape in painting is disassembled and then reshaped into different form in reproduction. Third, additional lines are put in to create different look from the original painting. Forth, existing lines are extended over the boundary to create different look from the original painting. Fifth, achromatic colors: black and white, and three basic colors: red, blue and yellow in the original painting are modified into different shades or color scheme is increased in broad range.

Analysis of Stress Concentration Problems Using Moving Least Squares Finite Difference Method(I) : Formulation for Solid Mechanics Problem (이동최소제곱 유한차분법을 이용한 응력집중문제 해석(I) : 고체문제의 정식화)

  • The Taylor expansion expresses a differentiable function and its coefficients provide good approximations for the given function and its derivatives. In this study, m-th order Taylor Polynomial is constructed and the coefficients are computed by the Moving Least Squares method. The coefficients are applied to the governing partial differential equation for solid problems including crack problems. The discrete system of difference equations are set up based on the concept of point collocation. The developed method effectively overcomes the shortcomings of the finite difference method which is dependent of the grid structure and has no approximation function, and the Galerkin-based meshfree method which involves time-consuming integration of weak form and differentiation of the shape function and cumbersome treatment of essential boundary.

  • This thesis aims to improve the analysis accuracy of remotely sensed digital imagery, and the improvement is achieved by considering the weight factors(a priori probabilities) of Bayesian MLC in the classification stage. To be concrete, Bayesian decision theory is studied from remote sensing field of view, and the equations in the n-dimensional form are derived from normal probability density functions. The amount of the misclassified pixels is extracted from probability function data using the thres-holding, and this is a basis of evaluating the classification accuracy. The results indicate that 5.21% of accuracy improvement was carried out. The data used in this study is LANDSAT TM(1985.10.21 ; 116-34), and the study area is within the administrative boundary of Seoul.
