Changes of Frozen-Thawed Semen Characteristics in Miniature Pig and Duroc (Miniature Pig와 Duroc 종간의 동결-융해 후 정액 성상 비교)

  • The purpose of this study was undertaken to compare ability of frozen-thawed sperm characteristics between two strains (miniature pig and Duroc). The semen was collected by gloved-hand method into a pre-warmed ($37^{\circ}C$) thermos bottle. The semen was diluted with same volume extender and added to LEY solution for freezing. The diluted semen was placed in 0.5 ml straws, and freezing was initiated by exposing the straws to liquid nitrogen ($LN_2$) vapours for 10 min before placing them into $LN_2$ for cryopreservation. The frozen-semen straw were thawed at 20, 37 and $50^{\circ}C$ for 1 min, 45 sec and 10 sec within water-bath. The semen sample were evaluated at 0, 3, 6, 9, and 12 h after incubation at $37^{\circ}C$ for analysis of sperm ability. Abnormality of spermatozoa in miniature pig was significantly (p<0.05) higher than that in Duroc at 0, 9 and 12 h of post-thawing incubation after frozen-thawing. The percentage of F-patterned spermatozoa in miniature pig was significantly (p<0.05) lower, while the percentage of AR (acrosome reacted spermatozoa) pattern was higher in the miniature than in the Duroc. On the other hand, there was no significant difference in the viability of spermatozoa thawed at different temperature ($20^{\circ}C\;and\;37^{\circ}C$) between two species, but the viability in miniature pig was higher (p<0.05) than in Duroc when sperm was thawed at $50^{\circ}C$. In conclusion, this study suggest that suitable freezing method for miniature pig semen is required for increasing post-thawing viability and fertilization capacity.

Numerical Analysis of Pile Foundation Considering the Thawing and Freezing Effects (융해-동결작용을 고려한 말뚝 기초에 관한 수치해석 연구)

  • Numerical analysis was conducted to determine the effect of soil behavior by thawing and freezing of seasonal frozen soil on pile foundations. The analysis was performed using the finite element method (FEM) to simulate soil-pile interaction based on the atmosphere temperature change. Thermomechanical coupled modeling using FEM was applied with the temperature-dependent nonlinear properties of the frozen soil. The analysis model cases were applied to the MCR and HDP models to simulate the elastoplastic behavior of soil. The numerical analysis results were analyzed and compared with various conditions having different length and width sizes of the pile. The results of the numerical analysis showed t hat t he HDP model was relat ively passive, and t he aspect and magnit ude of t he bearing capacit y and displacement of the pile head were similar depending on the length and width of the pile conditions. The vertical displacement of the pile head by thawing and freezing of the ground showed a large variation in displacement for shorter length conditions. In the MCR model, the vertical displacement appeared in the maximum thaw settlement and frost heaving of 0.0387 and 0.0277 m, respectively. In the HDP model, the vertical displacement appeared in the maximum thaw settlement and frost heaving of 0.0367 and 0.0264 m, respectively. The results of the pile bearing capacity for the two elastoplastic models showed a larger difference in the width condition than the length condition of the pile, with a maximum of about 14.7% for the width L condition, a maximum of about 5.4% for M condition, and a maximum of about 5.3% for S condition. The significance of the effect on the displacement of the pile head and the bearing capacity depended on the pile-soil contact area, and the difference depended on the presence or absence of an active layer in the soil and its thickness.

An Experimental Study for the Effective Use Scheme of Snow Removal Materials on Road (도로 제설재의 효과적 사용방안에 관한 실험적 연구)

  • The amount of snow due to global warming and climate change has recently increased. The effective management of snow removal materials will be required. First, on the basis of domestic winter temperature, it is not necessary to get a baseline to less than the freezing point of $-52^{\circ}C$ for the calcium chloride($CaCl_2$) of 30%.. In terms of cost comparison between calcium chloride($CaCl_2$) and sodium chloride($NaCl$), the calcium chloride($CaCl_2$) is 2 to 3 times more expensive, and the supply of the calcium chloride($CaCl_2$) is not produced in domestic country and is in the conditions that have to imports all needed. Accordingly, the effective use scheme of snow removal materials should be considered to multifaceted ways. Thus, the objective of this study is to develop effective method and to replace from calcium choride($CaCl_2$) to sodium chloride($NaCl$) solution in the current snow removal operating system that uses a pre-wetted salt spreading method. The effective method that equals to the quality of the existing snow removal materials was developed in this study through performance tests for deicing chemicals, corrosion test of steel and freezing and thawing tests of concrete.

An Influence of Unit-Water Content Distribution in Ready-Mixed Concrete on Strength and Durability of Concrete (레미콘 단위수량 산포가 콘크리트 강도 및 내구성에 미치는 영향)

  • Various problems such as durability degradation may happen when extra water is added to concrete. Because of these reasons, the change of water content is managed by using rapid evaluation method of unit water content such as electric capacity method, heat drying method making use of micro wave, unit capacity mass method among various methods. Especially, in Japan, guidance for the change of water content ($\pm$ 10, 15, 20 kg/$m^3$ etc.) were regulated and used. However, it is the real situation that the guidance which were regulated in South Korea evaluate suitability only considering production and measurement error under the circumstances which are not considering the degree of durability degradation. Therefore, this study tries to investigate the influence of addition of extra water in the concrete on the durability degradation of concrete when it was added by artificial manipulation or by management error. From the test results, a guideline of the contents of extra water for the quality control is suggested with the consideration of the degree of durability degradation and the probable error resulted from the addition of extra water. The contents of extra water for tests are set as 0, 15, 25, 35 kg/$m^3$. To examine the durability degradation of concrete, freezing and thawing, carbonation, chloride penetration and compressive strength are tested.

Study of Vitrification of Immatured Pig Oocytes: Compared with Open Pulled Straw(OPS), Electron Microscopic Grid(EMG) and Nylon Loop System(NLS) (미성숙 돼지 난자의 유리화 동결에 관한 연구: Open Pulled Straw(OPS), Electron Microscopic Grid(EMG) 및 Nylon Loop System(NLS)의 비교)

  • This study evaluated the efficiency and compared with different materials of loading vessels for vitrification-plastic/glass, copper grid and nylon. The loading method, vitrification, cryop-reservation and warming method of the oocytes were examined. The loading samples prepared in manual or company-made and sterilized, loaded the COCs selected on each samples and cultured for maturation during 40 hours, and then exposed sequentially to ethylene glycol solution. Thawing method was reversely treated and exposed for warmed oocytes. After oocytes were thawed, fertilized and cultured in vitro for 3-4 hours, rates of development and morphological appearance were examined. The results were as summarized: ㆍOPS from company-made or hand-made of the hematocrit micropipettes, NLS from fishing line and EMG from company-made for EM were used for loading oocytes, respectively. ㆍThe efficiency of freezing method and loading convenience were orderly higher in OPS, NLS and EMG. The optimal capacity per vessel was orderly lowered in NLS, EMG and OPS, respectively. ㆍAfter oocytes were warmed, the recovery rate, morphology and rate of development were orderly higher in OPS, NLS and EMG, respectively. ㆍIn conclusion, OPS has the advantages of achieving a little more survival and preserving results than other two loading methods.

Development of manufacturing method for the honey coating semi-dried mulberry fruit using hot air dryer

  • Semi-dried mulberry fruit can be distributed at room temperature and maintained its chewy texture and shape. It can be used for its own itself or food materials. We develop the honey coating semi-dried mulberry fruit manufacturing method through hot air dryer. After extracting the moisture from the thawing process, honey was coated and dried. Drying time for semidried mulberry fruit was shortened by manufacturing on the day of work without going through aging process. The first the mulberry fruit juice was separated from the frozen mulberry fruit at room temperature of the laboratory or in the dryer of $60{\sim}90^{\circ}C$, then the first dried by a hot air drier at $60^{\circ}C$ for 3 to 4 h. Next, it is coated with honey, which is equivalent to 20% weight of the first dried mulberry fruit, and then the second dried at a temperature of $38^{\circ}C$ for 20 ~ 30 h. The honey coating semi-dried mulberry fruit by above method was shiny on the surface and retained its shape. The moisture content was about 15% and it was confirmed that it could be stored at room temperature.

Freezing-Thawing Phenomenon and Durability-Improving Method of Plain Concrete Exposed to Outdoor in Winter Season (동절기 옥외 노출 콘크리트의 동결융해 현상과 내구성 증진방안)

  • 본 고는 동절기 이후 무근 콘크리트의 상부에서 쉽게 발생하는 스케일링 현상에 주목하여 무근 콘크리트의 동해 발생원인을 고찰하고, 이에 대한 내구성을 향상시키기 위한 방안을 기존문헌조사를 통하여 1) 강도향상, 2) 진공배수공법 적용, 3) 흡수방지재 시공의 방법을 선정하였다. 그 후 각 방법의 동결융해저항성 향상 평가를 정량적으로 평가하기위하여 동결융해 시험을 통한 상대동탄성계수를 측정하였다. 그 결과 1), 2)번의 경우 동탄성계수가 약 15% 향상, 3)의 경우 강도에 따라 7~13%향상됨을 실험적으로 확인하였다. 따라서 상기의 방법 모두 무근 콘크리트의 동결융해 저항성 향상에 유효한 방법으로 판단되며, 이를 통해 무근콘크리트의 빈번히 발생하는 품질저하 및 이로인한 유지보수비용 절감을 도모할 수 있을 것으로 사료된다.

A Fundamental Study on Very High Strength and High Flowable Concrete using Industrial By-products (산업부산물을 활용한 고유동화 초고강도 콘크리트의 기초물성 및 동결융해특성)

  • This paper presents the fundamental study on rational manu(acture of Very High Strength(VHS) concrete using industrial by-products as like silica fume, slag and fly ash. In this study, we had tested various mixing cases to manufacture the VHS concrete(target compressive strength : over 1,000 kgf/$cm^{2}$) which is easily workable (target slump flow : 60$\pm$l0cm), The main variables studied are; 1) test variables to find the optimum replacement ratio of mineral admixture, 2) test variables to find a rational water-binder ratio, a proper binder content, 3) test variables to find the method for reduction of slump loss, 4) test variables to know the influence of air entrainment on frost resistance. From the test results, it is concluded that the rational mix design can be made by using 40% slag, 10% silica fume, and water reducing agent(slump loss reduction type). We found that it is unnecessary to entrain air for freeze-thawing resistance.

Effect of Extraction Time on the Rheological Properties of Sericin Solutions and Gels

  • Recently, silk sericin has attracted the attention of researchers owing to its useful properties as a biomaterial including 1) good wound healing and cell activities, 2) fast gelation character, and 3) high water retention property. In the present study, silk sericin was prepared using different extraction times in hot water and the effect of extraction time on the rheological properties of sericin solutions and gels was examined. It was found that the production yield of sericin increased with extraction time. The shear viscosity of sericin solutions and gels decreased with increasing extraction time due to a decrease in sericin molecular weight. When the sericin solution transformed to a gel, the viscosity increased and the shear thinning behavior was more evident. In addition, the shear stress measurements indicated that the slip between the sericin samples and the measuring plate of the rheometer was increased by the gelation of sericin. The compression strength of sericin gel could be increased remarkably (by more than 100 fold) by preparation using the freezing and thawing method.

Encapsulation of Plasmid DNA in Pegylated Liposome

  • The purpose of the study was to prepare the pegylated liposome carrying plasmid DNA with optimal encapsulation efficiency. Plasmid DNA (pCEP4 clone 790, 10.6 kb) was entrapped in the pegylated liposome composed of neutral lipid, POPC (l-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine), cationic lipid, DDAB (dimethyl dioctadecyl ammonium bromide) and anionic lipids, DSPE-PEG 2000 (distearoyl phosphatidyl ethanolamine polyethylene glycol 2000) and DSPE-PEG 2000-maleimide by freezing/thawing method. Free plasmid DNA was separated from the encapsulated one by Sepharose CL-4B column chromatography. The DNA amount encapsulated into the pegylated liposome was increased as cationic lipid concentration, initial amount of plasmid DNA and total lipid amount were increased.
