Effect of Sinapis alba L. on expression of interferon-gamma and interleukin-4 production in anti-CD3/anti-CD28-stimulated CD4(+) T cells (CD4+ T cells에서 백개자가 IFN-$\gamma$와 IL-4 생성에 미치는 영향)

  • Objective : Sinapis alba L. (SA) is a korean traditional herbal medicine that is usually used to prevent or treat inflammatory diseases, such as respiratory infection and rheumatoid arthritis. However, the effects of SA supplementation in vitro on serum antibody levels, splenocyte and peritoneal macrophage immune responses have not yet been determined. In this study, we examined the effect of SA on the production of Th1/Th2 cytokines. Methods : Splenocytes were isolated from naive C57BL/6 mice. Cells were enriched for CD4+ cell populations by first staining the cells with anti-CD4 (BD PharMingen, Calif, USA). CD4+ T cells were selected on a (CS) column, and the flow-through was collected as CD4+ T cells. Isolated cells were activated by overnight incubation on 24-well plates coated with $1{\mu}g/mL$ anti-CD3, $1{\mu}g/mL$ anti-CD28 and with SA ($100{\mu}g/mL$). Primary macrophages were collected from the peritoneal cavities of mice (8-week-old female C57BL/6). The peritoneal macrophages were washed and plated with RPMI-1640 overnight for the experiments. After 48-hours cultures, samples were centrifuged at 2000 rpm for 10 minutes, and the supernatants were stored at $-80^{\circ}C$. Mouse IL-4, IFN-$\gamma$ and TNF-$\alpha$ were quantified using ELISA kits (BioSource International, Camarillo, Calif, USA) according to the manufacturer's protocols. Results : SA at 100ug/ml decreased the generation of Th1 cytokine (IFN-$\gamma$) by 0.5-fold. However, SA has no effect on Th2 (IL-4) production. Conclusions : These results suggest that SA may play an important role in the control of T-cell-mediated autoimmunity by down-regulation of Th1 cytokine (especially IFN-$\gamma$, TNF-$\alpha$). These data may contribute to the design of new immunomodulating treatments for a group of autoimmune diseases.

IL-17A Secreted by Th17 Cells Is Essential for the Host against Streptococcus agalactiae Infections

  • Streptococcus agalactiae is an important bacterial pathogen and causative agent of diseases including neonatal sepsis and meningitis, as well as infections in healthy adults and pregnant women. Although antibiotic treatments effectively relieve symptoms, the emergence and transmission of multidrug-resistant strains indicate the need for an effective immunotherapy. Effector T helper (Th) 17 cells are a relatively newly discovered subpopulation of helper CD4+ T lymphocytes, and which, by expressing interleukin (IL)-17A, play crucial roles in host defenses against a variety of pathogens, including bacteria and viruses. However, whether S. agalactiae infection can induce the differentiation of CD4+ T cells into Th17 cells, and whether IL-17A can play an effective role against S. agalactiae infections, are still unclear. In this study, we analyzed the responses of CD4+ T cells and their defensive effects after S. agalactiae infection. The results showed that S. agalactiae infection induces not only the formation of Th1 cells expressing interferon (IFN)-γ, but also the differentiation of mouse splenic CD4+ T cells into Th17 cells, which highly express IL-17A. In addition, the bacterial load of S. agalactiae was significantly increased and decreased in organs as determined by antibody neutralization and IL-17A addition experiments, respectively. The results confirmed that IL-17A is required by the host to defend against S. agalactiae and that it plays an important role in effectively eliminating S. agalactiae. Our findings therefore prompt us to adopt effective methods to regulate the expression of IL-17A as a potent strategy for the prevention and treatment of S. agalactiae infection.

  • Allergic diseases have increased over the past several decade worldwide including developing countries. Allergic inflammatory responses are caused by Th (T helper)2 immune responses, triggered by allergen ingestion by antigen presenting cells such as dendritic cells (DCs). Intestinal microorganisms control the metabolism and physiological functions of the host, contribute to early immune system maturation during the early life, and homeostasis and epithelial integrity during life. Bifidobacteria have strain-specific immunostimulatory properties in the Th1/Th2 balance, inhibit TSLP (thymic stromal lymphopoietin) and IgE expression, and promote Flg (Filaggrin) and FoxP3 (Treg) expression to alleviate allergies. In addition, unmethylated CpG motif ODN (oligodeoxynucleotides) is recognized by TLR (toll-like receptors)9 of B cells and plasmacytoid dendritic cells (pDCs) to induce innate and adaptive immune responses, while the butyrate produced by Clostridium butyricum activates the GPR (G-protein coupled receptors)109a signaling pathway to induce the expression of anti-inflammatory gene of pDCs, and directly stimulates the proliferation of thymically derived regulatory T (tTreg) cells through the activation of GPR43 or inhibits the activity of HADC (histone deacetylase) to differentiate naive $CD4^+$ T cells into pTreg cells through the histone H3 acetylation of Foxp3 gene intronic enhancer.

Suppressive Effects of Cyanidin-3-glucoside on Th2 Cytokines Production in RBL-2H3 Cells (RBL-2H3 세포에서 Cyanidin-3-glucoside의 Th2 사이토카인 발현 억제 효과)

  • Cyanidin-3-glucoside (C3G), an anthocyanin, is one of the most widespread dietary flavonoids. We investigated the effects of C3G in PMA/ionomycin (PI)-induced RBL-2H3 cells. C3G inhibited the production of IL-4 and IL-13 and also decreased the level of mRNA in a dose-dependent manner. Furthermore, western blot analysis implied that C3G down-regulated the protein level of c-Jun, NF-ATc1 and NF-${\kappa}B$ but not c-Fos. Taken together, we suggest that C3G may have suppressive effects on Th2 cytokines and will be studied further to develop as functional foods that help alleviate allergy symptoms.

  • I investigated that tyrosine hydroxylase immunoreactive cells distribution in mesencephalon of korean native goat newborn by immunohistochemical method. The results obtained in this study were summarized as following. 1. It were observed TH-IR cells in substantia nigra pars compacta, ventral tegmental area, substantia nigra pars reticular, central linear nucleus and retrorubral field of Midbrian. 2. TH-IR cells were observed that to mass on several areas in substantia nigra pars compacta and substantia nigra pars reticular. 3. TH-IR cell process observed short or non and it were protruded irregular direction.

A Concise Synthesis of (-)-Cytoxazone via Regioselective and Stereoselective Introduction of Amino Group using Chlorosulfonyl Isocyanate

  • In 1998, Osada and co-workers isolated 140 mg of a novel cytokine modulator from 18 L of the culture broth of Streptomyces RK95- 31 isolated from a soil sample in Hiroshima Prefecture. This new immunosuppressant was named (-)-cytoxazone and its absolute configuration was determined on the basis of its NMR, CD and X -ray analysis. It interferes with cytokine IL4, IL10 and IgG production by selective inhibition of the signaling pathway of Th2 cells, but not Thl cells. Inhibitors of Th2-dependent cytokine production have potential as potent chemotherapeutic agents in the field of immunotherapy. The (-)-cytoxazone is different from known immunomodulators such as FK 506 and rapamycin in respect of structure and biological activity. As such cytoxazone should be a useful tool for understanding signaling pathways in Th2 cells, the synthesis of (-)-cytoxazone is of interest for the development of new cytokine modulators.

  • This study was carried out to elucidate the physiological and ecological mechamism of the branch pattern and electronmicroscopic structure of photosynthetic organs in Zoysia japonica Steud. The angles between the internodes of rhizome and the branches of Z.iaponica were changed from 0 to 52 degrees. The rhizomes tended to branch from the 3th node to the 10th node. When the terminal bud was damaged by various environmental factors, the initial branching formation was appeared immediately to the 4th node. The internode growth of the rhizome started from the beginning of April and continued to the end of Octobor. It became evident that there is a division labor between two different kinds of photosynthetic cells of mesophyll and bundle sheath tissues in Z.japonica The thick-walled bundle sheath cells surrounds the leaf vascular bundles as the Kranz type and separates them from the mesophyll cells.

Pain-reducing Effect by Transplants of Isolated Xenogeneic Chromaffin Cells in Mouse (추출된 이종 크롬 친화성 세포의 이식에 의한 마우스에서의 통증 완화 효과)

  • Objective : Adrenal medullary chromaffin cells are known to release analgesic substances such as opioides and catecholamines. Transplantation of them is a novel method that challenges current approaches in treating chronic pain. The transplantation of xenogeneic chromaffin cells into the central nervous system(CNS) supply antinociception in animals. In this study, we investigated the analgesic effects of rat adrenal medullary chromaffin cells transplanted into the CNS of the mouse. To study the antinociceptive efficacy of transplanted chromaffin cells, the survival of rat adrenal medullary chromaffin cells transplanted into the CNS of mouse was determined. Methods : The adrenal medullary chromaffin cells isolated from rat were transplanted into the striatum of mouse. These cells were confirmed of the release of Met-enkephalin and Leu-enkephalin by HPLC, and immunoblots for tyrosine hydroxylase(TH). Two weeks after transplantation, we performed immunohistochemistry for TH to determine the survival of implanted cells and assessed pain sensitivity at the same time. Results : The isolated rat adrenal medullary chromaffin cells were positive for anti-TH antibody and released Met-enkephalin and Leu-enkephalin more than rat endothelial cells. Transplanted rat chromaffin cells were stained with anti-TH antibody in striatum of mouse after 2 weeks. Pain sensitivity was reduced on the chromaffin cell-transplanted mouse compared to endothelial cell-transplanted mouse by the hot plate test. Conclusion : These results suggest that the rat chromaffin cells were suitably transplanted into the CNS of mouse. This approach could be used as a therapy for reducing of chronic pain induced by cancer or neuronal injury.

  • Reproductives structures of Pachymeniopsis elliptica (Holmes) Yamade (Rhodophyta, Grateloupiaceae) are investigated. In female gametophyte the carpogonial branch and auxiliary cell are produced in separate accessory branch system, the primary ampullar filament originated from mid-cortical layer. After fertilization, auxiliary cell joined with connecting filament becomes a fusion cell by fusing with several neighboring ampullar cells. The fusion cell produces a gonimoblast initial. It divides into gonimoblast cells, which later convert to carposporangia. In male gametophyte superficial cortical cells of vegetative filament produce two spermatangial mother cells which cut off up to three spermatangia respectively. Tetrasporangial initials are formed from the 6th to 12th cells of the cortical layer in tetrasporophyte, and divided cruciately to form tetrasporangium. Some of the sporangia are, however, divided zonately.

Alleviating Effects of Vitamin C on the Gramoxone Toxicity in the Mucosubstances of Rat Duodenum (흰쥐 십이지장 점액질에 미치는 Gramoxone 독성에 대한 비타민 C의 완화 효과)

  • The effect of vitamin C on th egramoxone toxicity in the duodenal globlet cells of rats were investigated suing histochemical methods. Rats in control, gramoxone and gramoxone+vitmain C (Vt.C) group, aged 6 to 7 weeks, were fed 18% casein diet. In th egramoxone group, neutral and acid mucins of the globlet cells in villi and crypts of duodenum tended to decrease as compared with the control group. And the globle cells secreting nonsuphated mucins tended to increase in number, being usually accompanied by a decrease of the globlet cells secreting suphated mucins which are prominent in the duodenal mucosa of control group. However, the goblet cells secreting nonsuphated mucins tended to increase in the gramoxone + Vt.C group. Morphological changes of the globlet cells in the gramoxone group were noted vacuolation and demolition of globlet cells, while those changes were not significant in the gramoxone +Vt. C group. It seems to be that Vt. C has alleviating effects on the gramoxone toxicity in secretion and production of the duodenal goblet cells.
