TFF1 유전자의 C/T 다형성과 위암 민감성과의 연관성 (An Association of C/T Polymorphism in the TFF1 Gene and the Susceptibility to Gastric Cancer)

  • 목적: TFF1 유전자의 -2bp에 위치하고 있는 단일염기다형성의 유전자형 및 대립형질 빈도와 H. pylori 감염 및 위암발생 민감성과의 연관성을 조사하고자 하였다. 대상 및 방법: 2000년 1월부터 2003년 12월까지 위 샘암종으로 근치적 수술을 시행 받은 167명과 정상 건강인 299명의 DNA를 이용하였다. TFF1 유전자의 단일염기다형성의 유전자형은 중합효소연쇄반응 후 제한효소를 이용한 제한 분절길이다형성과 single strand conformation polymorphism의 방법으로 분석하였다. 또한, H. pylori 감염은 Giemsa 염색으로 조사하였다. 결과: 정상 건강인의 TFF1 단일염기다형성에 대한 유전자형 및 대립형질의 빈도를 위암 환자군과 비교한 결과 통계적으로 의의가 없었다(P=0.715 & P=0.595). 위암 환자군을 성별로 나누어 정상 건강인과 비교한 경우에도 통계적 의의를 발견할 수 없었다. 또한, 위암 조직의 조직학적 소견 및 환자 연령과 TFF1 단일염기다형성의 유전자형 및 대립형질 빈도를 조사한 결과, 위암 조직의 조직학적 소견 및 연령은 TFF1 유전자 단일염기다형성과 무관하였다(P=0.088 & P=0.551). 한편, H. pylori 감염은 모두 39예에서 관찰되었는데 TFF1 유전자형과는 통계적으로 연관성이 없었다(P=0.7200). 결론: TFF1 유전자의 -2bp에 존재하는 단일염기다형성은 TFF1 단백의 합성에 큰 영향이 없으며 H. pylori 감염과 위암의 발생과는 무관하다는 것을 의미한다.

  • Trefoil factor 1 (TFF1, also known as pS2) is strongly expressed in the gastrointestinal mucosa and plays a critical role in the differentiation of gastric glands. Since approximately 50% of all human gastric cancers are associated with decreased TFF1 expression, it is considered a tumor suppressor gene. Tff1 deficiency in mice results in histological changes in the antral and pyloric gastric mucosa, with severe hyperplasia and dysplasia of epithelial cells, resulting in the development of antropyloric adenoma. Here, we generated Tff1-knockout (KO) mice, without a neomycin resistant ($Neo^R$) cassette, using the clustered regularly interspaced short palindromic repeats/CRISPR-associated nuclease 9 (CRSIPR/Cas9) system. Though our Tff1-KO mice showed phenotypes very similar to the previous embryonic stem (ES)-cell-based KO mice, they differed from the previous reports in that a reduction in body weight was observed in males. These results demonstrate that these newly established Tff1-KO mice are useful tools for investigating genetic and environmental factors influencing gastric cancer, without the effects of artificial gene insertion. Furthermore, these findings suggest a novel hypothesis that Tff1 expression influences gender differences.

  • We has designed, fabricated, and measured a Single flux quantum (SFQ) toggle flip-flop (TFF). The TFF is widely used in superconductive digital electronics circuits. Many digital devices, such as frequency counter, counting ADC and program counter be used TFF Specially, a program counter may be constructed based on TFF We have designed the newly TFF and obtained high bias margins on test. In this work, we used two circuit simulation tools, WRspice and Julia, as circuit optimization tools. We used XIC for a layout tool. Newly designed TFF had minimum bias margins of +/- $37\%$ and maximum bias margins of +/-$37\%$(enhanced from +/- $37\%$). The designed circuits were fabricated by using Nb technology The test results showed that the re-optimized TFF operated correctly on 100kHz and had a very wide bias margins of +/- $53\%$.

위 선종 및 선암에서 Trefoil Factor Family 1 단백의 발현 양상 (Expression Pattern of the Trefoil Factor Family 1 in Gastric Adenoma and Carcinoma)

  • Purpose: The trefoil factor family 1 (TFF1) has a protective effect against gastric mucosal damage induced by nonsteroidal anti-inflammatory drugs or ethanol. In addition, a TFF1 knockout mouse model has exhibited circumferential adenomas with high-grade dysplasia, of which $30\%$ progressed into frankly invasive carcinomas. We tried to determine whether the expression pattern of the TFF1 could be involved in the development of sporadic gastric carcinomas. Materials and Methods: We examined TFF1 expression in a series of 43 sporadic gastric carcinomas and 18 gastric adenomas by immunohistochemistry. Results: Strong positive TFF1 staining was identified primarily in the normal gastric mucosa, mainly in the cytoplasm of the superficial and foveolar epithelium. We found TFF1 expression in $55.8\%$ (24 out of 43) of the gastric carcinomas and in $16.7\%$ (3 out of 18) of the gastric adenomas. Statistically, TFF1 immunoreactivity was significantly higher in diffuse-type ($82.4\%$) than in intestinal-type ($38.5\%$) carcinomas(p=0.0058, Fisher's exact test). Conclusion: Our findings provide sufficient evidence that the expression of TFF1 in gastric cancer may simply disclose gastric-type differentiation of neoplastic cells and provide further support for the existence of at least two pathways of malignant transformation of the gastric mucosa: one via intestinal metaplasia and adenomatous dysplasia, leading to glandular carcinomas with intestinal-type differentiation, and the other via hyperplastic changes or de novo changes, leading to diffuse carcinomas and to a subset of glandular carcinomas displaying gastric-type differentiation.

Influence of Surface Roughness of Tools on the Friction Stir Welding Process

  • Most publications on friction stir welding describe phenomena or results with given process parameters like feed rate, rotation speed, angle and depth of penetration. But without a complete documentation of tool design, the results under the same process parameters are completely different. For this purpose, the Institute of Cutting and Joining Manufacturing Processes (tff), University of Kassel investigated the influence of tool roughness on the friction stir welding process. Therefore a defined surface finish was produced by turning and die sinking. As basis of comparison the constant parameters were rotation speed, feed rate, tilt angle and a heel plunge depth. Sound butt-welds were produced in aluminium alloy 6082 (AlMgSi1) with 1.5 mm sheet thickness with a turned reference tool with a surface of $Ra=0.575{\mu}m$ in position controlled mode. The surfaces are manufactured from a very fine to a very rough structure, classified by the VDI-classes with differences in the arithmetical mean roughness. It can be demonstrated with the help of temperature measures, that less heat is generated at the surfaces of the shoulder and the pin by the higher roughness due to lower active friction contact surface. This can also be seen in the resulting wormhole defects.

  • We designed a high temperature superconducting rapid single flux quantum(RSFQ) T flip-flop(TFF) circuit using Xic and WRspice. According to the optimized circuit parameters, we fabricated the TFF circuit with $Y_1$$Ba_2$Cu$_3$$O_{7-x}$(YBCO) interface-controlled Josephson junctions. The whole circuit was comprised of five epitaxial layers including YBCO ground plane. The interface-controlled Josephson junction was fabricated with natural junction barrier that was formed by interface-treatment process. In addition, we report second design for a new flip-flop without ground palne.e.

  • We have fabricated a planar lightwave circuit (PLC) hybrid-integrated optical sub-assembly of a triplexer using a thin film filter (TFF)-attached wavelength division multiplexer (WDM) and photodiode (PD) carriers. Two types of TFFs were attached to a diced side of a silica-terraced PLC platform, and the PD carriers with a $45^{\circ}$ mirror on which pin-PDs were bonded were assembled with the platform. A clear transmitter eye-pattern and minimum receiver sensitivity of -24.5 dBm were obtained under 1.25 Gb/s operation for digital applications, and a second-order inter-modulation distortion (IMD2) of -70 dBc was achieved for an analog receiver.

  • TFF(Thin Film Fille.) 방식의 2$\times$2 OADM을 이용하여 1510nm 파장과 1530nm 파장의 광원을 첨가하여 add/drop시킨 4 파장 광 모듈을 구성하였고 이 광모듈을 채택하여 PCS(personal communication system) 및 셀룰러 (cellular) 이동 통신뿐만 아니라 차세대 이동 통신 방식은 IMT-2000 서비스와 다중 링크 시켜 광선로를 재활용하는 방식의 4 파장 광 트랜시버를 구현하였다. OADM을 이용한 4파장 optical signal stream을 구성하여 1510 nm, 1530 nm, 1550 nm, 1570 nm 파장에서 각각 -1.6 dBm, -1.7 dBm, -5.6 dBm, -5.8dBm의 광출력을 얻었다. 제작된 광 트랜시버의 LD 모듈에 12 V의 바이어스 전압을 사용하여 약 1.5dBm의 광 출력 특성을 얻었으며, PD 모듈의 증폭단에는 two-stage 저 잡음 증폭기를 제작하여 1.66dB의 잡음지수와 25.7 dB의 이득을 얻었다. 또한 S$_{11}$과 S$_{22}$ 그리고 입ㆍ출력 정재파비는 각각 -5.47dB, -20 dB 그리고 3.27 : 1, 1.22 : 1의 결과 값을 얻었다.다.

Re-Configurable tow-toss OADM Module Using 2×2 Port Optical Device

  • We describe the optimal design and the fabrication of a 2$\times$2 port optical device based on the thin film filter (TFF), and also propose a 4-channel OADM module using these devices. The optical performance of the proposed OADM module is evaluated theoretically and experimentally, and is compared to that of typical OADM modules using 1$\times$2 port optical devices for 4, 8, 16 and 32 drop channels in optical transmission systems. Since the 2$\times$2 port optical device accomplishes the function of wavelength multiplexing and demultiplexing simultaneously in the proposed OADM module, the insertion loss of through channels can be improved by 1.2 dB compare to that of typical OADM modules using 1$\times$2 port optical devices. In addition, both the size and the price of the module can be reduced to 40~50%.

얼굴 표정 인식을 위한 지역 미세 패턴 기술에 관한 연구 (A Study on Local Micro Pattern for Facial Expression Recognition)

  • 본 논문에서는 얼굴 표정 인식을 위한 지역미세패턴(local micro pattern)의 하나인 LBP(Local Binary Pattern) 코드의 잡음에 대한 단점을 해결하기위하여 새로운 미세패턴 방법인 LDP(Local Directional Pattern)를 제안한다. 제안된 방법은 LBP의 문제점을 해결하기 위해 $m{\times}m$ 마스크를 이용하여 8개의 방향 성분을 구하고, 이를 크기에 따라서 정렬한 후 상위 k개를 선정하여 해당 방향을 나타내는 비트를 1로 설정한다. 그리고 8개의 방향 비트를 순차적으로 연결하여 최종 패턴 코드를 생성한다. 실험결과, 제안된 방법은 기존 방법에 비해 회전에 대한 영향이 적으며, 잡음에 대한 적응력이 현저히 높았다. 또한, 제안된 방법을 기반으로 얼굴의 영구적인 특징과 일시적인 특징을 함께 표현하는 새로운 지역미세패턴의 개발이 가능함을 확인하였다.
