Lipids in microalgae are energy-rich compounds and considered as an attractive feedstock for biodiesel production. To redirect carbon flux from competing pathways to the fatty acid synthesis pathway of Tetraselmis sp., we used three types of chemical inhibitors that can block the starch synthesis pathway or photorespiration, under nitrogen-sufficient and nitrogen-deficient conditions. The starch synthesis pathway in chloroplasts and the cytosol can be inhibited by 3-(3,4-dichlorophenyl)-1,1-dimethylurea and 1,2-cyclohexane diamine tetraacetic acid (CDTA), respectively. Degradation of glycine into ammonia during photorespiration was blocked by aminooxyacetate (AOA) to maintain biomass concentration. Inhibition of starch synthesis pathways in the cytosol by CDTA increased fatty acid productivity by 27% under nitrogen deficiency, whereas the blocking of photorespiration in mitochondria by AOA was increased by 35% under nitrogen-sufficient conditions. The results of this study indicate that blocking starch or photorespiration pathways may redirect the carbon flux to fatty acid synthesis.
As a part of the investigation on useful compounds from microalgae, and its recently that marine planktonic algae have been recognized as potial sources of antibacterial and antifungal substances, we searched for antimicrobial active compounds from the extracts of six microalgae-Lyngbya sp., Tetraselmis sp., Microcystis sp., Chlorella sp., Navicula sp. and Rhalassiosira sp.-treated with several solvents. There were two active species-Lyngbya ap., Teraselmis sp.- in the antimicrobial activity test to bacteria, yeast and molds, especially the activity existed in the extracts by ethyl acetate of supernatants to the microalgae incubatio. and there won’t any activity in two diatoms to the test microorganism.
It is important to design photobioreactor by cheap material for economical microalgal biomass production. In this study, two types of marine photobioreactors (MPBR), made by either polyvinyl chloride (MPBR-PVC) or high density poly ethylene (MPBR-HDPE), are used and performance of these were compared. Tetraselmis sp. KCTC 12236BP is a green marine alga that isolated from Ganghwa Island, Korea, and the strain was used for marine cultivations using MPBR-PVC and MPBR-HDPE. The cultivations were performed three times in the spring season of 2012 using MPBR-PVC and of 2013 using MPBR-HDPE in the coastal area of Young Heung Island. As the results, MPBR-PVC shows higher biomass productivities than MPBR-HDPE, due to its high light transmittance. In the cultivations using MPBR-PVC, the average sea water temperature was $11.5^{\circ}C$ during the first experiment and $16.5^{\circ}C$ during the second and third experiments. Average light intensities during three times for experiments were 407.5, 268.1 and $273.0{\mu}{\cdot}E{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. The maximum fresh cell weight and average biomass productivity were $1.2g{\cdot}L^{-1}$ and $0.12g{\cdot}L^{-1}{\cdot}day^{-1}$. These results showed that Tetraselmis sp. KCTC12236BP were adapted well with the environmental conditions from ocean, and grow in the MPBR-PVC and MPBR-HDPE.
As a possible feedstock for biodiesel, the marine green alga Tetraselmis sp. was cultivated under different conditions of phototrophic, mixotrophic and heterotrophic cultures. Glycerol, a byproduct from biodiesel production process, was used as the carbon source of mixotrophic and heterotrophic culture. The effects of glycerol supply and nitrate-repletion were compared for different trophic conditions. Mixotrophic cultivation exhibited higher biomass productivity than that of phototrophic and heterotrophic cultivation. Maximum lipid productivity of 55.5 mg L-1 d-1 was obtained in the mixotrophic culture with 5 g L-1 of glycerol and 8.8 mM of nitrate due to the enhancement of both biomass and lipid accumulation. The major fatty acid methyl esters (FAME) in the produced biodiesel were palmitic acid (C16:0), oleic acid (C18:1), linoleic acid (C18:2), and linolenic acid (C18:3). The degree of unsaturation was affected by different culture conditions. The biodiesel properties predicted by correlation equations based on the FAME profiles mostly complied with the specifications from the US, Europe and Korea, with the exception of the cold-filter plugging point (CFPP) criterion of Korea.

Harmful algal blooms (HABs) are a serious problem for public health and fisheries industries, thus there exists a need to investigate the possible ways for effective control of HABs. In the present study, we investigated the algicidal effects of a newly developed GreenTD against the HABs (Chattonella marina, Heterosigma akashiwo, Cochlodinium polykriokides, and Heterocapsa circularisquama) and non-HABs (Chaetoceros simplex, Skeletonema sp. and Tetraselmis sp.), which is focused on the different population density and concentration gradients of algicidal substances. The time series viability of target alga was assessed based on the activity of Chl. a photosynthetic efficiency in terms of $F_v/F_m$, and in vivo fluorescence (FSU). Effective control of Raphidophyta, C. marina and H. akashiwo was achieved at a GreenTD concentration of $0.5{\mu}gL^{-1}$ and $0.2{\mu}gL^{-1}$, respectively, and regrowth of both the species was not observed even after 14 days. The inhibitory ratio of the dinoflagellate, C. polykriokides was more than 80% at $0.2{\mu}gL^{-1}$ of GreenTD. H. circularisquama was constantly affected in the presence of $0.2{\mu}gL^{-1}$ of GreenTD in the high- and low-population density experimental groups. On the other hand, diatoms, C. simplex, and Skeletonema sp. were not significantly affected even in the presence of $0.2{\mu}gL^{-1}$ of GreenTD and exhibited re-growth activity with the passage of incubation time. In particular, green alga Tetraselmis sp. remained unaffected even in the presence of the highest concentration of GreenTD ($1.0{\mu}gL^{-1}$), implying that non-HABs were not greatly influenced by the algicidal substances. As a result, the algicidal activity of GreenTD on the harmful and nonharmful algae was as follows: raphidophyte>dinoflagellates>diatoms>green alga. Consequently, our results indicate that inoculation of GreenTD substances into natural blooms at a threshold concentration ($0.2{\mu}gL^{-1}$) can maximize the algicidal activity against HABs species. If we consider the dilution and diffusion rate in the field application, it is hypothesized that GreenTD will demonstrate economic efficiency, thus leading to effective control against the target HABs in the closed bay.

Indigenous species are needed for more accurate toxicity assessments in tropical regions. Thus, we determined not only the optimum culture conditions for stable maintenance of Nitocra sp. isolated from tropical regions but also the availability of copepods for marine ecotoxicological evaluation. Experiments on temperature, salinity, and diet as factors for optimum culture conditions as well as acute and chronic toxicity tests for ecotoxicological assessment were carried out. Data on optimum culture conditions were analyzed for statistically significant observations using one-way analysis of variance (ANOVA). Optimum temperature and salinity for Nitocra sp. were $29^{\circ}C$ and 24~39‰, and Nitocra sp. fed Tetraselmis suecica had relatively faster development and higher survival than other microalga. Under optimum culture conditions, toxicity tests were carried out. The $LC_{50}$ level and NOEC (no observed effect concentration) levels of copper and arsenic were calculated in the acute toxicity test. In the chronic test of Cu and As, developmental time and survival traits were usable endpoints for toxicity assessments. As a result, tropical copepod Nitocra sp. seems to be a potential candidate organism for marine ecotoxicological evaluation.

We investigated the effect on dietary EPA (eicosapentaenoic acid) source (EP) into formulated diets for growth of brackish flea, Diaphanosoma celebensis. The highest density and specific growth rate (SGR) of brackish fleas were observed on Tetraselmis suecica (TE) trial, but these of trial were not significantly differed with that of Chlorella (CH) + Crypthecodinium sp. (CR) + EP trial, contained EPA oil (P>0.05). Contrastively, CH trial showed the lowest SGR. And in the RNA/DNA ratio, 0.08 of TE trial was the highest ratio out of whole trials, but the trial not significantly differed with that of CH + CR + EP trial (P>0.05). But, the ratio of CH trial was the lowest ratio at 0.05 out of whole trials (P<0.05). Through out results, the EPA source for flea culture was showed a positive effect through their growth and SGR. Therefore, we suggested that a usage of the source with CH could replace T. suecica as good diet for culture of the brackish flea.
Hydrothermal liquefaction of lipid-extracted Tetraselmis sp. feedstock containing 80 wt.% water was conducted in a batch reactor at different temperatures (300, 325, and $350^{\circ}C$) and reaction times (5, 10, 20, 40, and 60 min). The biocrude yield, elemental composition and higher heating value obtained at various reaction conditions were used to predict the optimum conditions for maximizing energy recovery of biocrude with good quality. A maximum energy recovery of 67.6% was obtained at $325^{\circ}C$ and 40 min with a high energy density of 31.8 MJ/kg and lower contents of nitrogen and oxygen. Results showed that reaction conditions of $325^{\circ}C$, 40 min was most suitable for maximizing energy recovery while at the same time achieving improved quality of biocrude.
Five microalgal species isolated from the Jeju coast and four microalgal stock strains in hatchery were cultured in order to investigate their adapation to extreme changes in environmental factors such as salinity, water temperatue, adn nutrients. In case of salinity variation, Nitzschia sp. of Bacillariophyceae, Isochrysis galbana of Haptophyceae and Tetraselmis gracilis of Prasinophyceae showed optimum growth at the low salinity of 20 and 25 psu. Amphora coffeaeformis and Chetoceros simplex of Bacillariophyceae, and Pavlova lutheri of Haptophyceae adapted well at the relatively high salinities of 30 and 35 psu. However Phaeodactylum tricornutum of Bacillariophyceae and Chlorella sp. of Chlorophyceae showed euryhaline property In case of water temperature variation, most of all the species studied wer inhibited at 10℃. C. simplex, Nitzschia sp., p. tricornutum, Chlorella sp. and T. gracilis grew well at above 20℃. A. coffeaeformis, I. galbana and P. lutheri adapted also at the high temperature of 30℃. Each microalgal strain showed different growth rates and its maximum biomass. Generally microalgal populations from the Jeju coast grow well in relatively high salinity and high water temperature. Their growth were inhibited at low water temperature, but not likely affected at low salinity. This study indicates that the microalgal populations could not be affected by abnormally low salinity phenomena, which have happened occasionally around the west Jeju coast in summer and have led macrobenthic animals to mass mortality.
Although culture techniques for the abalone Haliotis discus hannai are well known, mass culture of the benthic microalgae that are essential live food for the abalone larvae is still not practiced. This study was conducted to identify the microalgal species suitable for the growth of early larvae of H. discus hannai. The growth and attachment rates of 31 microalgal species were examined. Acrylic plates were used as the substrate. Among the 31 microalgal species, nine showing high growth and attachment rates were selected and tested for their dietary values via factors including settlement, metamorphosis, and survival rates of abalone larvae. Tetraselmis hazeni and Rhaphoneis sp. induced the highest settlement rate (65-69%) in abalone larvae. The metamorphosis rate was highest (57%) in larvae fed Rhaphoneis sp. and was also significantly higher in larvae fed Oscillatoria splendida (29%) and T. hazeni (22%) than in those fed other species. The highest survival rate of the larvae during the 15 days after metamorphosis was 67% in those fed Rhaphoneis sp., followed by T. hazeni (42%) and O. splendida (35%). In conclusion, Rhaphoneis sp. is the most suitable diatom for use as a live food for the culture of early larvae of H. discus hannai. In addition, T. hazeni and O. splendida are also potential species to be further developed and utilized in larval culture.
