한우(韓牛)의 선천성용혈감수성검사(先天性溶血感受性檢査)에 대한 methylene blue 청색소실시험법(靑色消失試驗法)의 적용(適用) (Application of methylene blue color test for the detection of inherited susceptibility to hemolysis of Korean native cattle)

  • Blood samples were obtained from Korean native cattle and dairy cattle of Holstein species in the slaughter house and methylene blue color tests were performed for the detection of the inherited susceptibitity to hemolysis. Glucose-6-phosphate dehydrogenase activities expressed as the optical density obtained by methylene blue color test were the highest as 0.54 in male Korean cattle, 0.62 in female Korean cattle and 0.72 in dairy cattle of Holstein species. Percent hemolysis, packed cell volume and plasma protein contents were measured and compaired with relation to the results of methylene blue color test and no correlation were observed in each.

돼지 brucellosis에 대한 항체가 조사 (Serosurvey for antibodies against brucellosis in pigs)

  • In order to investigate serum antibodies for detection of brucellosis in pigs, a total of 1208 sera were tested by Rose Bengal test (RBT), the standard tube agglutination test (STAT) and competitive ELISA (cELISA). The sera were collected from pigs of Gyeonggi, Chungnam, Chungbuk, Jeonnam and Jeonbuk, provinces during the period 2002 to 2004. All the sera were screened by RBT, and were confirmed by STAT and cELISA. Among 1208 sera, 26 sera (2.2%) were positive in screening test. All the 26 positive sera were positive by STAT, while all the sera were negative by cELISA. On the basis of this study, farmed pigs may be exposed to Brucella species. Furthermore, these results suggest that establishment of diagnoses for detection of porcine brucellosis is necessary.

천연활엽수림(天然闊葉樹林)에서의 수종간(樹種間) 상관관계(相關關係)와 공변이관계(共變異關係)의 분석(分析) (Analysis of Inter-Species Association and Covariation in a Natural Deciduous Forest)

  • 신갈나무와 굴참나무가 우점종(優占種)인 활엽수림(闊葉樹林)에서 출현하는 37가지 목본식물(木本植物)에 대한 수종간(樹種間) 상관(相關)(association) 관계(關係)와 공변이(共變異)(covariation) 관계(關係)를 $X^2$ 검정(檢定)을 이용하여 분석(分析)하였다. 50개의 $20m{\times}20m$ 정방형(正方形) 표본구내(標本區內)에서 각 수종(樹種)의 출현(出現) 비출현(非出現)이 매개변수(媒介變數)가 되는 상관관계(相關關係)를 분석(分析)한 결과, 단풍나무는 고로쇠나무 및 음나무와 고도(高度)(${\alpha}$=0.01)의 정(正)의 상관관계(相關關係)를 갖고 있으나, 소나무와는 고도(高度)의 부(負)의 상관관계(相關關係)를 갖고 있었다. 졸참나무와 음나무, 참회나무와 노린재나무, 노린재나무와 다릅나무, 다릅나무와 산벚나무, 그리고 생강나무와 느릅나무는 상호 정(正)의 상관관계(相關關係)가 있는 것으로 나타났다. 각 수종(樹種)의 밀도(密度)와 흉고단면적(胸高斷面積)으로 비교된 공변이(共變異)의 분석(分析) 결과는 상관관계(相關關係)와 매우 부합되는 바가 많았다. 더우기 표본구(標本區)에 생존(生存)하는 수종(樹種)의 개체수(個體數)와 흉고단면적(胸高斷面積)이 매개변수(媒介變數)가 됨으로써 상관관계(相關關係)보다는 공변이관계(共變異關係)를 맺고있는 수종(樹種)들의 쌍(雙)이 많은 것으로 나타났다. 특히 천이(遷移)의 초기(初期) 혹은 중반단계(中盤段階) 수종(樹種)인 소나무는 대부분의 천이(遷移) 후기(後期) 수종(樹種) 혹은 극성상(極盛相) 수종(樹種)들과 부(負)의 공변이관계(共變異關係)가 있는 것으로 판단되었다. 이러한 생태학적(生態學的)인 정보(情報)들은 생태계(生態界) 분류(分類), 활엽수(闊葉樹) 혼효림(混淆林) 조성(造成), 그리고 미래목 선정등의 보육(保育)작업에 응용할 수 있을 것이다.

Identification of Colletotrichum spp. associated with pepper anthracnose in Korea (oral)

  • Pepper anthracnose is one of the major limiting factors in pepper production. Boring last over 10 years, Colletotrichum gloeosporioides has been known as the most prevalent species among five Colletotrichum spp. involved as anthracnose causing agents. Recently, however, the change of major species with pepper anthracnose has been proposed. Identification study was peformed on 12 test isolates collected from anthracnose disease symptoms on pepper during 2001-2002 and 25 reference isolates obtained from several other host plants. The identification of the isolates with morphological observation and IfS region sequence comparison resulted that 11 ones from 12 test isolates colleted from pepper anthracnose during 2001-2002 were identified as C. acutatum. PCR using species-specific primers designed from ITS region sequence suggested a rapid diagnosis method in identifying C. acutatum from C. gloeosporioides.

식물의 렉틴 성분 스크리닝 (Screening of Plants for Lectins Constituents)

  • The erythrocytes agglutination test was applied to the common korean plants for lectin activity screening by using human blood. During the four years, 108 species from 46 families of floras were collected, identified and subjected to the test after being divided into several different parts. Only 13 species demonstrated strong lectin activities. Meanwhile 66 species did not shown any agglutination. All others were observed as having low activity or as having hemolytic constituents.

  • The objective of this study was to differentiate the beef species between Hanwoo and Holstein from a total of 1,081 beef samples using PCR-RFLP of MC1R gene. When a PCR product of 403 bp specific band amplified from bovine MC1R gene sequence was digested with restriction enzyme MspA1I, Hanwoo type showed 2 bands, 220 bp and 183 bp size bands. Holstein type, however, showed three bands, 220 bp, 138 bp and 45 bp size band, respectively. The results of the differential test for beef species were as following; 7 samples (0.64%) were determined to Holstein type, of which 4 were submitted from administrative authorities, other 3 from self-collection planing, and none from civilian clients including school.

Inability of Mate and Species Recognition by Male Asian Toads, Bufo gargarizans

  • In recent years, we frequently observed missmatched pairs between male Asian toads, Bufo gargarizans, and bullfrogs, Rana catesbeiana, at the toad breeding ponds, where scramble competition for mating occurred among the male toads. Thus, we performed two-choice experiments to investigate recognition ability of mates and species in male toads. The test males did not discriminate sexes, but the clasped stimulus males immediately produced release calls and stopped it while the clasped stimulus female did not. In addition, the test male toads did not discriminate reproductive state of females and even species. However, male toads chose larger individuals. The present results indicate that the main reason of missmatched amplexus by the male toads is due to 1) the lack of recognition cues of conspecifics, 2) the lack of communication tools like release calls, and 3) the larger size of bullfrogs than male toads themselves.

In Vitro Inhibitory Activities of Essential Oils from Two Korean Thymus species against Antibiotic-Resistant Pathogens

  • The in vitro inhibitory activities of essential oils from Thymus magnus and T. quinquecostatus as well as their main constituents were evaluated against susceptible and resistant species of Streptococcus pneumoniae, Staphylococcus aureus, Salmonella enteritidis, and Salmonella typhimurium. Notably, the essential oil fraction of T. magnus and its main components displayed significant inhibitory action against both antibiotic-susceptible and resistant strains of S. pneumoniae, S. aureus, and S. typhimurium with minimum inhibitory concentrations (MICs) ranging from 0.125 to 8 mg/mL. The differential MIC values imply that the oil fraction and its main components exhibit distinct patterns of activity against the tested bacterial species. Moreover, the disk diffusion test revealed that the inhibitory activities of oil fraction and components were dose-dependent. Data from the checkerboard titer test confirmed synergism between the antibiotic, norfloxacin, and T. magnus oil or thymol, particularly against the resistant strains of S. aureus.

고흥 외나로도 연안에서 자망, 통발, 주낙에 어획된 어족생물의 종조성 및 어획량 변동 (Species composition and abundance of fishery resources collected by gill net, trap net, and longline near Oenarodo, Go-heung Peninsula, Korea)

  • The species composition and variation in abundance of fishery resources near Oenarodo, Go-heung Peninsula, Korea, were investigated by gill net, trap net, and longline in May, July, and October 2015 and 2016. During the study period, the total catch included 14 species in the gill net, 11 species in the trap net, and 4 species in the longline. The dominant species were Portunus trituberculatus and Raja pulchrain the gill net, Charybdis japonicaand and Octopus vulgarisin in the trap net, and Muraenesox cinereusin in the longline. The Catch Per Unit Effort (CPUE) per individual and per weight in the gill net were similar in May and July of 2015 and 2016. In October 2015, the CPUE per individual was 2.1 ind./h and the CPUE per weight was 505 g/h higher than the results in 2016, but there was no significant difference in the total CPUE between 2015 and 2016. In the trap net, the CPUE per weight was similar in both 2015 and 2016, but the CPUE per individual was 2.7 ind./h higher in October 2015 than in October 2016 and the total CPUE was not significantly different from 2015 to 2016. The CPUE per individual and weight in the longline were significantly higher in July and October 2015 than in the same months of 2016, but the total CPUE in 2015 and 2016 did not show a significant difference.

폐수처리장의 전 방류수 독성 평가 및 방류수 배출하천의 생지표도 영향분석 (The Whole Effluent Toxicity Tests of Wastewater Discharged from Various Wastewater Treatment Plants and Their Impact Analyses on Biological Component)

  • 전남지역에 존재하는 9개 하수종말처리장(A, B, C, D, E, F, H, I, J)의 방류수가 배출되는 하천에 대한 방류수의 영향을 알아보기 위해 하천에 대한 생 지표도 평가 및 전 방류수 독성 평가(Whole effluent toxicity test)를 실시하였다. WET 평가와 생지표도 평가는 U.S. EPA 시험 방법과 Ohio EPA wading method에 근거하여 수행하였다. 전 방류수 독성 평가를 위해 표준 시험 종인 S. capricornutum과 D. magna를 사용하였다. 전 방류수 독성 평가결과 모든 지점에서 S. capricornutum 독성이 나타났으며, D. magna 독성 평가 결과에서는 A지점과 J지점을 제외한 모든 지점에서 독성 효과를 보였다. 표준 시험 종의 독성 평가 결과에 따르면 D지점에서 가장 높은 독성을 나타내었고, E지점과 F지점에서도 높은 독성 결과를 나타내었다. 이들 지역에 대한 생지표도 분석 결과 C1과 D1, J1 지점에서 낮은 종의 수 분포를 나타내었으며, A1, C1, E1, H1, J1 지점에서는 대조군 지역이나 타 지역보다 낮은 개체수를 나타내어 방류수로부터 악영향을 받고 있는 것으로 판단되었다. A1, B1, C1, D1, J1지점에서는 낮은 종의 풍부도를 보였으며, 전 방류수 독성 평가 결과에서 가장 높은 독성을 나타내었던 D지점(D1)에서는 오염에 내성이 강한 종이 매우 우점 하는 것으로 나타났다. 전 방류수 독성 평가 및 생물지수 분석결과에 따르면 모든 하천지역에서 방류수에 의한 영향이 나타나고 있는 것을 알 수 있었으며, D. magna와 S. capricornutum의 독성이 같이 나타나는 지점에서는 생 지표도 분석 결과에서도 동일하게 생태계에 영향을 주는 것으로 나타났으며, S. capricornutum만의 독성이 나타나는 지점에서도 생태계의 변화가 나타나고 있었다.
