In general, the blood cell culture is a common method for animal chromosome preparation. However, every animal and its cells have unique physiological characteristics and functions. Hence, it is very difficult to find the suitable method of chromosome preparation using animal lymphocyte culture. This study was carried out to fine the suitable method of chromosome preparation using lymphocytes cultures in mammalians and aves including cattle, rat, mouse and chicken. To seek the optimal method of lymphocyte culture in each animal, $2^3$ factorial experiment was designed. The design evaluated three main effects in culture duration, kinds of mitogen supplements and colcemid exposure time with two levels within each effect. The mitotic index and the score of chromosome morphology were analyzed. In results, the suitable methods of lymphocyte culture for chromosome preparation were 72 hours culture, pokeweed mitogen(PWM) supplement and 90 minutes of colcemid exposure in cattle, 72 hours culture, PWM supplement and 50 minutes of colcemid exposure in chicken, 96 hours culture, concanavalin A supplement and 90 minutes of colcemid exposure in rat, and 72 hours culture, PWM supplement and 50 minutes of colcemid exposure in mouse, respectively. In conclusion, kinds of mitogen, culture duration and colcemid exposure time significantly affected the mitotic index and chromosome morphology, in animal lymphocyte culture. The interaction effects between/among treatment factors were also statistically significant.

This paper examines theoretical trends and policy issues concerning science and public. The dominant perspective about it was changed from “popularization of science” to “public understanding of science (PUS)”. PUS pays attention to active roles of public in the understanding of science based on the contextual model. And recently various concepts are attempted to complement PUS such as “heterogeneous PUS”, “quantitative PUS”, “public understanding of research”, and “public participation in science”. PUS related activities in Korea can be analyzed using the concept of “science and technology culture system”. The characteristics of science and technology culture system in Korea can be summarized as follows: Its purpose is confined to support existing national innovation system; Its constituents have been unevenly and separately developed; Its material and institutional infrastructure is not sufficiently prepared; Its organizing mechanism is strongly influenced by government's execution power.

The productivity of ultra-small rotifer, Synchaeta kitina was investigated at the exchange rate of culture water (10, 20, 30, 40 and 50%) and initial inoculation densities (250, 600 and 900 inds. per mL) in semi-continuous culture. Also, the possibility of mass culture was investigated in a 100 L culture tank. Tetraselmis suecica was used as the feed for S. kitina in all experiments. The production of S. kitina increased with an increase in exchange rate of culture water. The highest production ($82.0{\times}10^5$ inds.) was achieved at 40% exchange rate of culture water. The production of S. kitina increased with an increase of initial inoculation density during the first week and the highest total production ($17.4{\times}10^6$ inds.) was achieved at 900 inds. per mL of initial inoculation density. However, on the second week, all treatments were not significantly different in total production (P>0.05). During the two weeks, total production of S. kitina at 900 inds. per mL of initial inoculation density was higher than at 600 inds. of initial inoculation density, but there was no significant difference (P>0.05). In the 100 L culture tank, density of S. kitina was kept from 516 to 890 inds. per mL and S. kitina was daily harvested $15.5{\times}10^6$ to $26.7{\times}10^6$ during the experimental period. The production cost for 100 million rotifers in semi-continuous culture was 63,656 won. The results from this study indicate that the optimal exchange rate of culture water and initial inoculation density for the semi-continuous culture of ultra-small rotifer, S. kitina are 40% and 600 inds. per mL, respectively.

The creative characteristics of Culture and Arts organization workers who connect consumers and producers of culture and arts(artists, creators, and producers) are very important in the culture and arts field. Where the cultural and arts organization workers are very important as their achievements are directly related to the appreciation of culture and art. The purpose of this study is to investigate the effect of creativity on cultural and artistic organizations workers regarding self-efficacy and work performance. The research was conducted on 208 people in national and public cultural organizations and art organizations in Gwangju Jeollanam-do. As a result of the research, originality and challenge of creative characteristics have a positive effect on self-efficacy. However, openness did not have a statistically significant effect on self-efficacy. In addition, self-efficacy has a positive effect on job performance.

The Yellow River, one of the longest rivers in the world and the second longest river in the north of China, enjoys the reputation of "Mother River". In recent years, more and more researches have pointed to the traditional culture of the Yellow River Basin. The traditional sports of the Yellow River basin belong to the ancient culture of the Yellow River basin. We assume the responsibility in protection and inheritance. This paper from the coupling of tourism and traditional sports to unfold the study by using the methods of literature and data, logical analysis, expert interview methods and so on: 1. The development of tourism leads to the development of Chinese traditional sports. 2. Tourism can make traditional sports get around and promote them as a kind of culture. 3. Let more people know, understand and learn traditional sports culture in a comfortable way, so as to pave the way for the inheritance of traditional sports. The research results of this paper can provide theoretical basis for more research on the development and inheritance of traditional culture in the Yellow River Basin.

The purpose of this study was to delve into the features of paradigm in conjunction with changing convergence culture in the 21st century's knowledge-based society, the influence of convergence paradigm on fashion, and changes in convergence culture. After identifying the features of the paradigm of knowledge-based society in association with changing convergence culture, the characteristics of fashion paradigm were examined in con=junction with the characteristics of the paradigm, and the convergence phenomena of modern fashion were investigated to figure out fashion trends. The findings of the study were as follows: According to the features of ubiquitous technology based on the theories of Mark Weiser and other futurologists, the characteristics of changing convergence culture in knowledge-based society could be classified into invisibility, relationship and materialized virtuality. When relevant theories were reviewed, invisibility, relationship and materialized virtuality are identified as the unique features of the paradigm of changing convergence culture in knowledge-based society. But those characteristics aren't separate concepts. Instead, they are being connected and fused together, thanks to universalized digital technology and ubiquitous technology. Changing convergence culture has brought a change to fashion paradigm as well. There is a type of design that lets all the characteristics stand out, and another design fuses just some of them together conceptually or formally. That is, fashion paradigm is marked by a combination of invisibility, relationship and an embodiment of virtual reality to be in line with changes in convergence culture.

The objective of this study was to establish the optimum protocol for the isolation and culture of porcine muscle satellite cells. Mononuclear muscle satellite cells are a kind of adult stem cell, which is located between the basal lamina and sarcolemma of muscle fibers and is the primary source of myogenic precursor cells in postnatal muscle. Muscle satellite cells are a useful model to investigate the mechanisms of muscle growth and development. Although the isolation and culture protocols of muscle satellite cells in some species (e.g. mouse) have been established successfully, the culture system for porcine muscle satellite cells is very limited. In this study, we optimized the isolation procedure of porcine muscle satellite cells and elaborated the isolation and culture process in detail. Furthermore, we characterized the porcine muscle satellite cells using the immunofluorecence. Our study provides a reference for the isolation of porcine muscle satellite cells and will be useful for studying the molecular mechanisms in these cells.
