  • This paper reports the production and characterization of crude xylanase from the newly isolated Humicola sp. Ly01. The highest (41.8 U/ml) production of the crude xylanase was obtained under the optimized conditions (w/v): 0.5% wheat bran, 0.2% $KH_2PO_4$, and 0.5% peptone; initial pH 7.0; incubation time 72 h; $30^{\circ}C$; and 150 rpm. A considerable amount of the crude xylanase was induced using hulless barley bran or soybean meal as the carbon source, but a small amount of the enzyme was produced when supplementary urea was used as the nitrogen source to wheat bran. The crude xylanase showed apparent optimal cellulase-free xylanase activity at $60^{\circ}C$ and pH 6.0, more than 71.8% of the maximum xylanase activity in 3.0-30.0% (v/v) ethanol and more than 82.3% of the initial xylanase activity after incubation in 3.0-30.0% (v/v) ethanol at $30^{\circ}C$ for 2 h. The crude xylanase was moderately resistant to both acid and neutral protease digestion, and released 7.9 and 10.9 ${\mu}mol/ml$ reducing sugar from xylan in the simulated gastric and intestinal fluids, respectively. The xylooligosaccharides were the main products of the hydrolysis of xylan by the crude xylanase. These properties suggested the potential of the crude enzyme for being applied in the animal feed industry, xylooligosaccharides production, and high-alcohol conditions such as ethanol production and brewing.

  • This study has shown that geography teachers need the professionalism in a Teaching and learning aspect. Together with the transition of epistemology and the operation of the Subject Matter-based curriculum, etc the demand for professionalism of teachers is much higher than ever. But the less discussion about what the professionalism of geography teachers based on subject traits and its components are has been mentioned. Especially the previous studies related with geography teachers usually distinguished the contents from the methods in the geography teaching in a dichotomy and tried to apply to a class model in the theoretical aspects, so they $didn't^{\circ}{\varnothing}t$ present the enough measures to improve the professionalism of geography teachers in the synthetic aspects. In this study, after I found the components of professionalism required to geography teachers in a teaching and learning, I tried to present measures to improve the professionalism of geography leathers in the macroscopic aspects. The geography teacher should be a teacher to design a region as a process than a result, and a teacher as a reflective practitioner than a technical practitioner. The improvement of professionalism should be achieved through the process of cooperation with an educational reflection in the individual aspects.

  • Recently, there is a matter of common interests in 'classroom' because many pedagogues and teachers feel urgently the understanding of educational field. This study aims to investigates classroom researches for the last twenty years, asserts the theme 'the spatiality of classroom' which is important to understand diverse point of views in classroom theories and is overlooked at the same time, and considers it in terms of 'classroom space metaphors'. Specially, the author reviews the recent trend in classroom researches, and names it 'classroom theory' in that it deals with classroom not as a means but as an object and has undergone methodological transition. In the mean time, the author lays great emphasis on 'the spatiality of classroom' which is important to expand classroom theory and is overlooked at the same time. To approach this, the author categorizes the classroom space metaphors of existing classroom researches into 'classroom as a work space' and 'classroom as a learning space' by drawing the concept 'metaphor'. And then the author presents new metaphor 'classroom as a lived space' on the basis of the recent classroom theory, and tries to abstract its educational significance in classroom by way of Thirdspace.

  • This study was done to investigate the effect of adolescent smoking on dietary intakes and nutritional status of serum lipids and antioxidant vitamins. Subjects were 82 somkers whose average pack-year was 0.73, and 85 nonsmokers of male technical high school students in Seoul. Anthropometric measurement was performed and % body fat was also analyzed by Bioelectrical Impedance Fatness Analyzer(GIF-891). Dietary intakes and habits were examined through questionnaires and nutrient intakes were analyzed by Computer Aided Nutritional (CAN) analysis programs. Serum TG and total cholesterol levels were measured by Spotchem sp-4410 and serum HDL-cholesterol levels were measured by test kit. serum vitamin C level was measured by 2,4-dinitrophenylhydrazine method and serum levels of vitamin A and vitamin E were measured by HPLC. All data were statistically analyzed by SAS PC package program. There was a significant difference in body fat percentage and systolic blood pressure while other anthropometric measurements were not different between smokers and monsmokers. Caloric intakes(2335㎉) in adolescent smokers tended to be higher than that of nonsmokers (2,175㎉) but the difference was not statistically significant. Intakes of protein(76.67g) and niacin(16.49㎎) in adolescent male smokers were significantly higher(P<0.05) than those of nonsmokers although other nutrient intakes were not significantly different. Analysis of serum lipids showed that serum HDL-cholesterol level of smokers was significantly different. Analysis of serum lipids showed that serum HDL-cholesterol level of smokers was significantly lower(p<0.05) than that of nonsmokers, whereas other lipid levels were not significantly different. Serum vitamin C level was also significantly lower(p<0.05) in adolescent smokers than in nonsmokers. In addition, serum vitamin E level, which was 7.85㎎/1 in smokers, was lower than that of 9.20㎎/1 in nonsmokers(p<0.05) while serum vitamin A level was not significantly different between the two groups. These results indicate that cigarette smoking in adolescence decreases serum levels of HDL-cholesterol, vitamin C and vitamin E even thoughth their smoking history is very short. • Materials used for education are materials such as SM20C, Al6061, and acrylic. SM20C materials are carbon steel and are often used in certification tests and functional competitions, but are also widely used in industrial sites. The Al6061 material is said to be a material that has lower hardness and stronger flexibility than carbon steel, so it is a material that generates a lot of compositional selection of tools. If students are taught practical training using acrylic materials, vibration occurs due to excessive cutting in some parts and damage to the tool occurs. In this process, we examine to what extent the impact on the 2NC head, which is a five-axis equipment, can affect precision control. The weakest part of the five-axis equipment can be said to be the weakest part of the head that controls the AC axis. When the accuracy and cumulative tolerance of this part occur, the accuracy of all products decreases. Therefore, the core part of the 2NC head, the spindle housing, was carried out using an Al7075 T6 (Alcoa, USA) material. In the process of vibration and cutting applied to this material, the analysis was conducted to find out the value applied to the finite element analysis under extreme conditions. It is hoped that this analysis data will help students see and understand the structure of 5-axis machining rather than 5-axis cutting.

  • Most student misconceptions about evolution are similar to misconceptions and disputes which early scientists had in science history. The aim of this study was to analyze student evolution conceptions based on science history, there by revealing for effectively teaching strategies on evolution. A test was developed according to Lee's three dimensional framework (2004) on evolution concept changes. Lee's framework had been constructed according to 4 stages of evolution concept changes in history in three-dimensional aspects such as mechanism, time, and subject: before Lamarck (stage 1), Lamarck (stage 2), Darwin (stage 3), and after Darwin (stage 4). Major results were as follows. First, the evolution conceptions of students appeared fixed to stage 2 regardless of grade. Moreover, students usually possessed Lamarckian thought and did not show consistency in evolution concepts among the three dimensional aspects of mechanism, time, and subject. Therefore, students were found to apply different conceptions of evolution to each different situation.
