Species Diversity of Spontaneous Mushrooms on Jeju Island (제주도 자생버섯의 종다양성)

  • This study was conducted to survey the diversity and distribution of mushrooms growing spontaneously on Jeju Island in the last eight years. Approximately 2,000 specimens of mushrooms were collected in Gotjawal, Hallasan Natural Park, and Oreum, the habitats where a rich diversity of mushrooms was expected, from 2006 to 2013. They were identified as 74 families, 213 genera and 511 species. Thus, including 69 families, 99 genera and 561 species from the previous study conducted in 2005, a total of 83 families, 257 genera and 740 species of wild mushrooms have been recorded. This study would support new information for fungal flora in Korea and preliminary data for further studies on mushroom diversity.

Assessment of Riverine Health Condition and Estimation of Optimal Ecological Flowrate Considering Fish Habitat in downstream of Yongdam Dam (용담댐 하류의 하천건강성 평가 및 어류 서식처를 고려한 최적 생태유량 산정)

  • In this study, a comprehensive field monitoring was conducted to understand habitat conditions of fish species in the upper Geum river. Based on the monitoring data, riverine health conditions such as composition ratio of fish species, richness and dominance indexes, bio-diversity (dominance index, diversity, evenness and richness), and index of biological integrity were assessed, and optimal ecological flowrates were estimated using the habitat suitability indexes established for three fish species Coreoleuciscus splendidus, Zacco platypus and Pseudopungtungia nigra selected as icon species using the physical habitat simulation system (PHABSIM). The total number of species sampled was 20 species, and two sensitive species of C. splendidus (22.4%) and Z. platypus (22.0%) dominated the fish community. The estimated IBI values ranged from 34 to 42 with average being 38 out of 50, rendering the site ecologically fair to good health conditions. An optimal ecological flowrate of 9.0 cms was recommended for the representative fish species at the site.

  • This study analyzed how the entrepreneurial orientation of Korean CEO influenced to the performance of companies in entertainment business. The research verified the direct influence that the entrepreneurial orientation of entertainment business affected the companies' performance, and examined the diversity of TMT and the control effect of corporate governance. This study contributed to previous researches in two ways. The first, the study verified the concept of entrepreneurial orientation on entertainment business which was only focused on manufacturing or IT industry although the entrepreneurial orientation received attention as a significant research issue on middle and small venture businesses. Second, the research also verified the characteristics of CEO and executives bringing the concept of TMT's diversity in entertainment companies which were dependent on person's capacity. As a result of the research, the entrepreneurial orientation (innovation, proactiveness, risk-taking) of entertainment entrepreneurs led to a positive effect on financial performance. The research showed that the more TMT was diverse or ownership oriented management, the more broadened the positive effects.

The assessment of conservation value for agricultural pond wetland using the weighted function of modified RAM (modified RAM의 기능별 가중치 부여를 통한 농촌지역 연못형습지의 보전가치 평가)

  • The pond wetlands in agricultural landscape are important natural resources that carry out the function of bio-diversity conservation. However, recently, those have been gradually embedded as their utility value were disappeared. And, the assessment methods used for pond wetlands are insufficient. Therefore, this study was conducted to examine the conservation value of pond wetlands by using the modified RAM, and present the improvement of assesment methods. The study sites, a total of 32 pond of 4 types by land use, were selected on the basis of Ramsar Convention. Through the analysis of precedent studies, the weighted 8 functions were adjusted. According to the assessment results, pond wetlands made the largest contribution to Fishery and Herpetile Habitat function. In addition, it also made large contribution to Floral Diversity, Wildlife Habitat, and Water Quality Protection function. On the other hand, it made a small contribution to Aesthetics and Recreation, Runoff Attenuation, Shoreline /Stream Bank Protection, and Flood/Storm Water Storage function due to the characteristics of small-scale pond wetlands. In the assessment of 8 functions, house type showed the worst assessment result, and mountain type showed the best assessment result. It is thought that those are due to land use type in terms of vicinity. 10 items among 52 of the modified RAM showed the same assessment results in all land use types. Accordingly, it is required to be deleted and modified the assessment method. On the other hand, it is required to add age, interference, and water use to the assessment method. It is thought that these results can be utilized for the development and modification of assessment methods focused on pond wetlands in rural area.

Screening and Isolation of a Gene Encoding 4-Hydroxyphenylpyruvate Dioxygenase from a Metagenomic Library of Soil DNA (토양의 DNA로부터 4-Hydroxyphenylpyruvate Dioxygenase 유전자 탐색 및 분리)

  • To access the natural products of uncultured microorganisms, we constructed and screened the metagenomic DNA libraries by using a cosmid vector and DNA inserts isolated directly from soil. Initial screening of the libraries in Escherichia coli resulted in the isolation of several clones that produce a dark brown color when grown in LB medium. One of the positive clones, designed pYS85C, was transposon mutagenized and the DNA surrounding the transposon insertions in cosmids that no longer conferred the production of brown pigment to E. coli was sequenced. Annotation of the pYS85C sequence obtained from the transposon mutagenesis experiment indicated a single 393 amino acid open reading frame (ORF) with a molecular mass of about 44.5 kDa, predicted to be a 4-hydroxyphenylpyruvate dioxygenases (HPPDs), was responsible for the observed brown pigment. In a BLAST search against deposited sequence, the translated protein from this ORF showed moderate-level identity (>60%) to the other known HPPDs and was most conserved in the C-terminal region of the protein. These results show that genes involved in natural product synthesis can be cloned directly from soil DNA and expressed in a heterologous host, supporting the idea that this technology has the potential to provide novel natural products from the wealth of environmental microbial diversity and is a potentially important new tool for drug discovery.

Impact of Fish Farming on Macrobenthic Polychaete Communities (해상 가두리 양식이 저서 다모류군집에 미치는 영향)

  • Excessive input of organic matters from fish cage farms to the coastal waters has been considered as one of the major factors disturbing their benthic ecosystem. Sediment samples were taken from around the two fish cage zones (A and B) in Tongyeong coast in June and August 2003, to evaluate the ecological impacts of fish cage farming activity on the macrobenthic polychaete communities. Polychaete accounted for $81{\sim}87%$ of the total macrofauna individuals from each of the sampling stations. The number of species, abundance, diversity and dominant species of polychaete were rapidly changed with the distance from the fish cages. Within 10 m from the fish cages, Capitella capitata, which is a bio-indicator for the highly enriched sediments, was a dominant species and the lowest diversity was recorded. In particular, the maximum density (${\sim}18,410\;ind.m^2$) of C. capitata was found at Farm A where fish cages were more densely established within a semi-enclosed bay system. The sampling zone between 10 m and 15 m showed a rapid decrease of C. capitata with a rapid increase of the numbers of species, implying that this zone may be an ecotone point from a highly to a slightly enriched area. In the sampling zone between 15 m and 60 m, a transitional zone, which represents slightly enriched condition before normal one, was observed with additional increase and maintenance of the number of species and density of polychaete. In addition, the potential bio-indicators of organic enrichment, such as Lumbrineris longifolia and Aphelochaeta monilaris were the predominant species in the sampling zone. Multidimensional scaling (MDS) ordination plots and k-dominance curves confirmed the above results on the gradual changes in the macrobenthic polychaete communities. Our findings suggest that the magnitude of impact of fish cage farming activity on polychaete communities is probably governed by a distance from fish cage, density of fish cage and geomorphological characteristics around fish cage farm.

Ecological Health Assessment using Fish for the Han River and Nakdong River in Korea (한강 및 낙동강 수계 주요 하천을 중심으로 어류를 이용한 수생태 건강성 평가)

  • The present study was carried out to develop biological criteria for aquatic ecosystem health assessment using composition and diversity of collected species. The sampling sites were a total of 67 sites in the Han River (29 sites) and the Nakdong River (38 sites), May and September 2012. During the survey period fish were collected totally 93 species. In each water system, 73 and 61 species were collected in the Han River and the Nakdong River respectively. The current composition of fish species between the Han River and Nakdong River showed similar pattern. The dominant species was Zacco platypus of the Han River, and the Nakdong River was Opsariichthys uncirostris. In particular, ecological characteristics of O. uncirostris frequently appeared in general polluted waters. In conclusion, in the Nakdong River, average value of FAI (Fish Assessment Index) was averaged 41.3, indicating a "poor condition", and the Han River was 53.1, indicating a "poor condition". The aquatic health of the Nakdong River assessed based on FAI was considered to be worse than that of the Han River.

First report of three Filosporella species isolated from freshwater ecosystem in Korea (담수생태계로부터 분리된 Filosporella 3종의 국내 최초보고)

  • To investigate the diversity of aquatic fungi, we collected deposits of soil, plants, and plant litter from ponds and streams. NNIBRFG1552 was isolated from soil deposits and NNIBRFG3013 from plant deposits in Namsaengi-mot in Jeju, Korea in 2016; NNIBRFG5472 was isolated from plant litter in Bocheong-cheon, Boeun, Korea in 2018. Based on morphological characteristics and phylogenetic analysis using rDNA sequences of the internal transcribed spacer (ITS), NNIBRFG1552, NNIBRFG3013, and NNIBRFG5472 were identified as Filoporella exilis (100% similarity with KC834046), F. fistucella (99.8% with KC834047), and F. cf. annelidica (100% with KC834044), respectively. Furthermore, cultural and morphological characteristics were analyzed to confirm the molecular identification. No species of the genus Filosporella has yet been reported in Korea.

A study on the development of multi-purpose fisheries training ship and result of seakeeping model test (다목적 어업실습선 개발과 내항성능 시험 결과)

  • According to the recent presentation by the Korean Maritime Safety Tribunal, about 70% of marine accident occurs from fishing vessel, and 90% of cause of entire marine accidents attributes to human error. As fishing vessels require basic operations, fishing operations, other additional operations and techniques such as fish handling, cultivating excellent marine officer to prevent marine accident and develop industry is very important. A fisheries training ship is still very difficult to satisfy the demand for diversity of fishery training and sense of realism of the industry. As the result of employment expectation by category of business survey targeting 266 marine industry high school graduates who hope to board fishing vessels for the last four years, tuna purse seine was the highest with 132 cadets (49.6%), followed by offshore large purse seine (65 cadets, 22.4%), and tuna long line (35 cadets, 13.2%). The Korea Institute of Maritime and Fisheries Technology (KIMFT) has replaced old jigging and fish pot fishery training ships and proceeded developing and building multi-purpose fisheries training ships considering the demand of industry and the promotion of employment; however, the basic fishing method was set for a tuna purse seine. As a result of seakeeping model test, it can conduct the satisfiable operation at sea state 5, and survive at sea state 8.

Distribution of Legionella species from water systems and genetic diversity of L. pneumophila serogroup 1 in Gyeonggi-do (경기도내 수계시설에서 분리된 레지오넬라균의 분포현황 및 Legionella pneumophila serogroup 1의 유전학적 다양성 연구)

  • Legionnaires' disease (LD) is a severe and potentially fatal pneumonia caused by colonization of human-made water system and subsequent aerosolization and inhalation of Legionella bacteria. A total of 147 Legionella strains was isolated from environmental water sources from public facilities in Gyeonggi-do, South Korea. The distribution of Legionella isolates was investigated according to facility type, and sample type. L. pneumophila was distributed broadly throughout Gyeonggi-do, accounting for 85.7% of the isolates, and L. pneumophila serogroup (sg) 1 predominated in all of the public facilities. L. wadsworthii predominated among non-L. pneumophila species. We performed comparative analyses of L. pneumophila sg 1 isolated from environment water of public facilities in Gyeonggi-do by pulsed field gel electrophoresis (PFGE) and sequence-based typing (SBT). Thirty-two isolates were classified into 22 types by PFGE and 9 sequence types (STs) by SBT and categorized into 3 groups. ST1 was the most prevalent sequence type and two STs obtained in this study had unique allelic profiles. The use of SBT data from different countries for epidemiology study of LD constitutes a technically uncomplicated and relatively easy method for strain subtyping, especially compared to other contemporary techniques.
