Morphological and Genetic Species Identification in the Chironomidae Larvae Found in Tap Water Purification Plants in Jeju (제주 정수장에서 출현한 깔따구과 유충의 형태 및 유전학적 분석)

  • The Chironomidae is a benthic macroinvertebrate commonly found in freshwater ecosystems, along with Ephemeroptera and Trichoptera, which can be used for environmental health assessments. There are approximately 15,000 species of Chironomidae worldwide, but there are limited studies on species identification of domestic Chironomidae larvae. In the present study, we carried out species classification of the Chironomidae larvae that found in Jeju's tap water purification plants using morphological characteristics and genetic identification based on cytochrome c oxidase subunit I (COI) gene of the mitochondrial DNA. Body shape, mentum, antenna, mandible in the head capsule, and claws were observed in the larvae for morphological classification. Analysis of 17 larvae collected from faucets and fire hydrants of domestic tap water purification plants revealed the presence of two species, including 14 Orthocladius tamarutilus and 3 Paratrichocladius tammaater. These results will aid the use of the criteria information about species classification of the Chironomidae for water quality management in water purification plants and diversity monitoring of freshwater environments.

A Study on the Distribution Characteristics of Bromide and Bromate in Drinking Water in Northern Gyeonggi Area (경기북부지역 먹는 물 중 브롬이온 및 브롬산염의 분포특성에 관한 연구)

  • Objectives: The purpose of this study was the investigation of bromide and bromate in drinking water of water supply plants, mineral springs and small water supply system located in northern area of Gyeonggi province. Methods: Analytical method was based on EPA 326.0 to use Postcolumn reaction (PCR). The instrument was 887 professional UV/VIS detector IC manufactured in Metrohm. Results: Bromate was detected at $0.5{\sim}2.4{\mu}g/L$ in tap water from 5 water supply plants. These plants were used as disinfection method for sodium hypochlorite and on-site chlorine that causes generate bromate as a by products even if not used ozone. Conclusions: The bromate was detected up to $2.5{\mu}g/Lin$ drinking water in northern Gyeonggi area that showed within $10{\mu}g/L$ for standard of tap water. However, the continuous monitoring of bromate is necessary in drinking water.

It is possible to Prevent contamination by virus owing to development of antibiotic but not to dissolve basely health effect by hazardous chemical substances because it takes very long time, about 5-40 years, average 20 years, in appearing a disease after exposure by then To maintain human life for 75 years, it need 55 ton of water. In the experiment of Public Health Institute, Tokyo, they reported that over 200 chemical substances can be detected in case of human being to drink tap water of Tokyo(1989) continuously. In our country, elevation of life style and development of industry result in increase of water use and overappreciation of pesticides in farm land a(sects drinking water quality. We estimated analytical item of drinking water hygienically by dividing into 5 groups such as physical effect item health hazardous inorganic item and health hazardous organic item ect. based on hygienic purpose.

Comparison of Clinical Outcomes Using Various First-Aid Cooling Method for Burn (다양한 화상 응급처치의 치료효과 비교)

  • Purpose: Currently, the most recommended method for burn first aid is the cool running water for 20 minutes (CRW20). If CRW20 is not practicable, cooling dressing products with tea tree oil are used as substitutes. In this study, we analyze the effect of various burn first aids, including CRW20, cooling products, on the treatment period. Methods: This study was conducted on patients who suffered burns and visited Hangangsoo Hospital from March 2019 to March 2020. The study conducted in a retrospective method. The duration of treatment was analyzed by dividing the experimental group. Results: The treatment period was shortened when first aid was given (P<0.001). Cold water, ice, and ice packs resulted in reduced treatment periods (P<0.001, P=0.004). The treatment period was reduced when cooling dressing products were used in all groups (P=0.041). The implementation of first aid has reduced the duration of burn treatment. This means that cooling helps burn treatment. CRW20 did not show statistically meaningful results. This is the result of a failure to fully control the temperature and time of tap water. In addition, the change in temperature of tap water according to season, the use of water is higher than that of animal testing, and the general lack of information on proper burn first aid may have affected. Conclusion: The treatment period was statistically significantly reduced in the group that performed first aid. Among the various first aid methods, cold water and ice reduced the treatment period.

Suitability Assessment of Domestic Natural Waters as a Culture Medium for Daphnia magna (Daphnia magna의 배양배지로서 국내 자연수의 적절성 평가)

  • The present study was conducted to assess the suitability of domestic natural waters as a Daphnia magna culture medium. In order to assess survival rate and reproductive output, young female daphnids (parent animals), aged less than 24 hours at the start of the test and produced in the Elendt M4 medium, were exposed to Elendt M4 medium, de-chlorinated tap water, and natural mineral water for 21 days. D. magna cultured in Elendt M4 medium (reference medium) and natural mineral water met the criteria of OECD No. 211, Daphnia magna Reproduction Test Guidelines in terms of percent adult survival, first day of reproduction, and average young production. However, the mortalities of adult daphnids observed in de-chlorinated tap water were more than 20% in two reproduction tests for 21 days. Mortality was observed on exposure days 13, 15, and 18 in de-chlorinated water. The use of D. magna is recommended in water of hardness >80 mg $CaCO_3\;L^{-1}$. However, the hardness of de-chlorinated tap water used in the present study was 50~53 mg $CaCO_3\;L^{-1}$. Therefore, it is judged that the delayed mortalities observed in de-chlorinated tap water were caused by a rapid decreased in hardness when the medium was changed from Elendt M4 to de-chlorinated tap water. When D. magna is cultured using domestic natural waters (underground water, surface water, and de-chlorinated water), the quality-control (QC) data should be maintained through a standardization for health assessment method, toxicity test method using reference chemical, test intervals of reference toxicant toxicity test, and data treatment and interpretation. In the long term, national research programs are needed for the development of test species which are representative of domestic aquatic environmental conditions among indigenous daphnids.

Methods for the Extraction of DNA from Water Samples for Polymerase Chain Reaction

  • Methods for the extraction of DNA from water sample were approximated. Four different procedures of DNA extraction were carried out with pellets obtained from centrifugation of 4 liter water samples. The recovery efficiency and purity of DNA extracted by each method from different sources were compared. DNA yield varied with extraction methods, Method I, which involves enzymatic and freeze-thaw lysis steps and phenol and phenol-chloroform purification of extracted nucleic acid, showed a significantly higher yield and purity than the other methods. The use of glass beads in the DNA extraction methods improved the purity of DNA suitable for PCR. Bovine serum albumin in the PCR reaction mixture was useful in reducing inhibitory effects of contaminants. The efficiency of an extraction method was determined by the detection of the aer of Aeromonas hydrophila with PCR. The lower limit of detection of A. hydrophila from seeded tap water was 2 CFU/ml in PCR when method I was used for DNA preparation.

Effect of intracanal medications on the interfacial properties of reparative cements

  • Objectives: The purpose of the present study was to evaluate the effect of calcium hydroxide with 2% chlorhexidine gel (HCX) or distilled water (HCA) compared to triple antibiotic paste (TAP) on push-out bond strength and the cement/dentin interface in canals sealed with White MTA Angelus (WMTA) or Biodentine (BD). Materials and Methods: A total of 70 extracted human lower premolars were endodontically prepared and randomly divided into 4 groups according to the intracanal medication, as follows: group 1, HCX; group 2, TAP; group 3, HCA; and group 4, control (without intracanal medication). After 7 days, the medications were removed and the cervical third of the specimens was sectioned into five 1-mm sections. The sections were then sealed with WMTA or BD as a reparative material. After 7 days in 100% humidity, a push-out bond strength test was performed. Elemental analysis was performed at the interface, using energy-dispersive spectroscopy. The data were statistically analyzed using analysis of variance and the Tukey test (p < 0.05). Results: BD presented a higher bond strength than WMTA (p < 0.05). BD or WMTA in canals treated with calcium hydroxide intracanal medications had the highest bond strength values, with a statistically significant difference compared to TAP in the WMTA group (p < 0.05). There were small amounts of phosphorus in samples exposed to triple antibiotic paste, regardless of the coronal sealing. Conclusions: The use of intracanal medications did not affect the bond strength of WMTA and BD, except when TAP was used with WMTA.

Introduction of Corrosion Index System for Stability of Drinking Water Quality (음용수질의 안정성을 위한 부식지수제도의 도입)

  • Replacement of old water distribution pipes for protecting water quality induced by pipe corrosion requires enormous budget. Even after the replacement, however, corrosion can occur again at any times and, therefore, inhibitive measure of the corrosion will be not only economical but needed to diminish the consumers' distrust on tap water quality. In 2008, National Environmental Research Institute did a survey on 8 major drinking water source and proposed to establish the Langelier Saturation Index(LI) as a corrosion index in Drinking Water Quality Criteria. Among the water industries of Korea, K-Water is the only one that set up the level of pH over 7.0 and LI above -1.5 on yearly average basis. However, no systematic regulation including LI to inhibit the corrosive tendency has been established yet. In this paper, LI values out of 31 drinking water treatment plants were analyzed and two-stage control of LI value as a measure of corrosive tendency of water is proposed. Primarily, water treatment facilities may operate the system at a target LI value below -1.5. Following the investigation on the effect caused by adjusting the LI value on water quality and corrosiveness, it will be desirable to improve LI value below -1.0 in the long run. In addition to the LI, supplemental use of Larson's modified ratio (LMR) which incorporates hydraulic detention time will be necessary. Several methods to prove the inhibitive effect of improving the LI value on water quality have been also suggested.

Mathematical Model Simulations Assessing the Effects of Temperature on Residual Chlorine Concentrations in Water Storage Tanks (온도 변화에 따른 수돗물 저장 저수조 내 잔류염소에 관한 수학적 모형 시뮬레이션)

  • To ensure hygienic safety of drinking water in a water storage tank, the concentrations of residual chlorine should be above a certain regulation level. In this study, we conducted model simulations to investigate the effects of temperature on residual chlorine in water storage tank conditions typically used in Seoul. For this, values of model parameters (decomposition rate constant, sorption coefficient, and evaporation mass transfer coefficient) were experimentally determined from laboratory experiments. The model simulations under continuous flow conditions showed that the residual chlorine concentrations were satisfied the water quality standard level (0.1 mg/L) at all the temperature conditions ($5^{\circ}C$, $10^{\circ}C$, $15^{\circ}C$, $20^{\circ}C$ and $25^{\circ}C$). Meanwhile, when the tanks had a no flow condition (i.e., no tap-water influent due to a sudden shut-down), the concentrations became lower than the regulatory level after certain periods. The findings from this modeling works simulating Seoul's water storage tanks suggested disappearance rate of residual chlorine could be reduced through the tanks design optimization with maintenance of low water temperature, minimization of air flow and volume, suppression of dispersion and the use of wall materials with low sorption ability.

Recent Trends of Domestic and International Management and Research of Natural Mineral Water Used for Bottled Water (먹는샘물과 병입수로 이용되는 천연 광천수의 국내외 관리와 연구 동향 분석)

  • In recent years, the sales of bottled spring water (BSW) have been drastically increasing in Korea and other countries, which accompanied great interests in conservation and reclamation of natural mineral water (NMW). In this study, management and research activities on NMW in Europe, USA, and Codex Alimentarius were reviewed. In each region, NMW is regulated with its own quality standards that differ from ordinary drinking water, and management actions are strictly implemented to protect water resources and to secure quality of NMW. Many studies on NMW were carried out for monitoring inorganic constituents including major and trace elements in national levels for bottled NMW, groundwater, and tap water in other countries. In Korea, NMW became commercialized in 1995 when BSW was legally approved as a drinking water. To further promote utilization of various types of NMW in Korea, regulations and policies for NMW need to be revised in accordance with international NMW management trends. Further, studies of NMW that compile a comprehensive set of physical and chemical parameters of NMW are also needed to properly understand occurrences, hydrogeological and geochemical processes of NMW, as well as to evaluate its potential use as a natural resource.
