인삼토양 내 근권세균의 다양성 및 식물에의 유용 특성 (Molecular Diversity of Rhizobacteria in Ginseng Soil and Their Plant Benefiting Attributes)

  • 연령을 달리하는 인삼토양 내 근권세균의 다양성과 이들 세균이 갖고 있는 식물생장에 유용한 특성에 대하여 조사하였다. 인삼 근권으로부터 총 15개 속에 포함되는 143 균주를 분리하였다. 인삼 연령에 따른 세균 군집의 변화가 있었으나, 분리 균주의 58%가 Firmicutes에 속하였다. 특히 Bacillus는 연령에 관계없이 가장 우점하는 속으로 나타났다. 이들 분리 균주 중 30개의 균주를 선발하여 세포벽 분해효소 분비, 인돌아세트산 생성, siderophore 생성, 인 가용화, 식물병원성 곰팡이의 저해능 등에 대하여 평가한 결과, 다수의 균주가 식물생장을 촉진할 수 있는 기능적 특성을 갖고 있었다. 이 중에서도 모든 조사 항목에 대하여 양성 반응을 보인 B. subtilis, B. amyloliquefaciens, B. velezensis, B. licheniformis 균주는 향후 인삼 경작을 위한 생장 촉진 미생물제제로 활용가치가 높을 것으로 기대된다.

Isolation, Characterization, and Use for Plant Growth Promotion Under Salt Stress, of ACC Deaminase-Producing Halotolerant Bacteria Derived from Coastal Soil

  • In total, 140 halotolerant bacterial strains were isolated from both the soil of barren fields and the rhizosphere of six naturally growing halophytic plants in the vicinity of the Yellow Sea, near the city of Incheon in the Republic of Korea. All of these strains were characterized for multiple plant growth promoting traits, such as the production of indole acetic acid (IAA), nitrogen fixation, phosphorus (P) and zinc (Zn) solubilization, thiosulfate ($S_2O_3$) oxidation, the production of ammonia ($NH_3$), and the production of extracellular hydrolytic enzymes such as protease, chitinase, pectinase, cellulase, and lipase under in vitro conditions. From the original 140 strains tested, on the basis of the latter tests for plant growth promotional activity, 36 were selected for further examination. These 36 halotolerant bacterial strains were then tested for 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity. Twenty-five of these were found to be positive, and to be exhibiting significantly varying levels of activity. 16S rRNA gene sequencing analyses of the 36 halotolerant strains showed that they belong to 10 different bacterial genera: Bacillus, Brevibacterium, Planococcus, Zhihengliuella, Halomonas, Exiguobacterium, Oceanimonas, Corynebacterium, Arthrobacter, and Micrococcus. Inoculation of the 14 halotolerant bacterial strains to ameliorate salt stress (150 mM NaCl) in canola plants produced an increase in root length of between 5.2% and 47.8%, and dry weight of between 16.2% and 43%, in comparison with the uninoculated positive controls. In particular, three of the bacteria, Brevibacterium epidermidis RS15, Micrococcus yunnanensis RS222, and Bacillus aryabhattai RS341, all showed more than 40% increase in root elongation and dry weight when compared with uninoculated salt-stressed canola seedlings. These results indicate that certain halotolerant bacteria, isolated from coastal soils, have a real potential to enhance plant growth under saline stress, through the reduction of ethylene production via ACC deaminase activity.

Identification of New Source of Resistance to Powdery Mildew of Indian Mustard and Studying Its Inheritance

  • Powdery mildew of Indian mustard (Brassica juncea), caused by Erysiphe cruciferarum, is emerging as major problem in India. All the Indian mustard cultivars presently grown in India are highly susceptible to powdery mildew and so far no resistance source has been reported. In this study, with an aim to identify resistant source, 1,020 Indian mustard accessions were evaluated against E. cruciferarum PMN isolate, at Wellington, The Nilgiris, Tamil Nadu, India under natural hot spot conditions. The study identified one accession (RDV 29) with complete resistance against E. cruciferarum PMN isolate for the first time, which was consistent in five independent evaluations. Genetic analysis of F1, F2 and backcross populations obtained from the cross RSEJ 775 (highly susceptible) × RDV 29 (highly resistant) for two season revealed that the resistance is governed by two genes with semi-dominant and gene dosage effect. Further, a new disease rating system using six scales (0, 1, 2, 3, 4, and 5) has also been proposed in this study to score powdery mildew based on progress of fungal growth in different plant parts of the F2 population. The outcome of this study viz. newly identified powdery mildew-resistant Indian mustard accession (RDV 29), information on inheritance of resistance and the newly developed disease rating scale will provide the base for development of powdery mildew-resistant cultivars of Indian mustard.

Implications of Five Laws of Library Science on Dr. S. R. Ranganathan's Colon Classification: An Explorative Study

  • There have been several milestones in the history of library classification but most of the schemes failed to meet the new challenges in the organisation of information. Dr. S. R. Ranganathan tried to revolutionise the whole thinking on classificatory approach, when he devised the Colon Classification (CC) in 1933. He developed the Colon Classification scheme with a sound theoretical background based on normative principles, Five laws of Library science, canons, etc. One important feature of CC is that, its use is not confined to information storage and retrieval alone. This paper presents an over view of different editions of the CC highlighting the salient features of the editions. Further the implication of Five Laws of Library Science has been described. The authors stressed that the features of such as greater hospitality, specificity and mixed notation has paved the way to design and develop the depth schedules on various micro level subjects and so far about 130 micro schedules have been published. The impressions by the leading LIS professionals during and after Ranganathan's time have been highlighted. The authors expressed the fear that when the library world would see the complete version of the seventh edition of CC? It may be due to lack of institutional support engaging in the research or financial constraints. The authors are of the opinion that any scheme to flourish needs a sound research body to bring out the revised editions as done in the case of Dewey Decimal Classification. The relevance of the CC in the contemporary world of Librarianship is discussed. Finally concludes that CC needs to be resuscitated as it is a precious national heritage; and still a force for the management of libraries.

Biocontrol Potential of a Lytic Bacteriophage PE204 against Bacterial Wilt of Tomato

  • Bacterial wilt caused by Ralstonia solanacearum is a devastating disease of many economically important crops. Since there is no promising control strategy for bacterial wilt, phage therapy could be adopted using virulent phages. We used phage PE204 as a model lytic bacteriophage to investigate its biocontrol potential for bacterial wilt on tomato plants. The phage PE204 has a short-tailed icosahedral structure and double-stranded DNA genome similar to that of the members of Podoviridae. PE204 is stable under a wide range of temperature and pH, and is also stable in the presence of the surfactant Silwet L-77. An artificial soil microcosm (ASM) to study phage stability in soil was adopted to investigate phage viability under a controlled system. Whereas phage showed less stability under elevated temperature in the ASM, the presence of host bacteria helped to maintain a stable phage population. Simultaneous treatment of phage PE204 at $10^8$ PFU/ml with R. solanacearum on tomato rhizosphere completely inhibited bacterial wilt occurrence, and amendment of Silwet L-77 at 0.1% to the phage suspension did not impair the disease control activity of PE204. The biocontrol activities of phage PE204 application onto tomato rhizosphere before or after R. solanacearum inoculation were also investigated. Whereas pretreatment with the phage was not effective in the control of bacterial wilt, post-treatment of PE204 delayed bacterial wilt development. Our results suggested that appropriate application of lytic phages to the plant root system with a surfactant such as Silwet L-77 could be used to control the bacterial wilt of crops.

Synergistic Effects of Arbuscular Mycorrhizal Fungi and Plant Growth Promoting Rhizobacteria for Sustainable Agricultural Production

  • Soil microorganisms play a major role in improving soil fertility and plant health. Symbiotic arbuscular mycorrhizal fungi (AMF) form a key component of the soil microbial populations. AMF form a mutualistic association with the host plant and exert a positive influence on its growth and nutrient uptake. The establishment of mycorrhizal symbioses with the host plant can positively be influenced by plant growth promoting rhizobacteria through various mechanisms such as increased spore germination and hyphal permeability in plant roots. Though there are evidences that combined interactions between AMF and PGPR can promote the plant growth however mechanisms of these interactions are poorly understood. Better understanding of the interactions between AMF and other microorganisms is necessary for maintaining soil fertility and enhancing crop production. This paper reviews current knowledge concerning the interactions between AMF and PGPR with plants and discusses on enhanced nutrient availability, biocontrol, abiotic stress tolerance and phytoremediation in sustainable agriculture.

Impact of Online Learning in India: A Survey of University Students during the COVID-19 Crisis

  • The unprecedented situation of COVID-19 caused the government of India to instruct educational institutions to switch to an online mode to mitigate the losses for students due to the pandemic. The present study attempts to explore the impact of online learning introduced as a stop-gap arrangement during the pandemic in India. A survey was conducted (N=289), via Facebook and WhatsApp, June 1-15, 2020 to understand the accessibility and effectiveness of online learning and constraints that students of higher education across the country faced during the peak times of the pandemic. The analysis and interpretation of the data revealed that the students acclimatized in a short span of time to online learning, with only 33.21% saying they were not satisfied with the online learning mode. However, the sudden shift to online education has presented more challenges for the socially and economically marginalized groups, including Scheduled Caste (SC), Scheduled Tribes (ST), Other Backward Class (OBC), females, and students in rural areas, due to factors like the price of high-speed Internet (78.20% identified it as a barrier to online learning), insufficient infrastructure (23.52% needed to share their device frequently or very frequently), poor Internet connectivity, etc. According to 76.47% of respondents, the future of learning will be in "blended mode." A total of 88.92% of the respondents suggested that the government should provide high-quality video conferencing facilities free to students to mitigate the division created by online education in an already divided society.

Protective effects of Hydro-alcoholic extract of the roots of Kasondi (Cassia occidentalis L.) against gentamicin-induced nephrotoxicity in albino Wistar rats

  • Objectives: Cassia occidentalis L. is a weed belonging to the Caesalpiniaceae family. The root of this medicinal plant is used for the treatment of various ailments, including kidney diseases. The present study was aimed at evaluating the nephroprotective effects of HAE of the roots of Cassia occidentalis L. against gentamicininduced renal toxicity in albino Wistar rats. Methods: The renal toxicity was induced by subcutaneous administration of gentamicin at 100 mg/kg in the rats belonging to the disease control and treatment groups from the 4th to the 8th day. The rats in the treatment group received HAE of the roots of Cassia occidentalis L. at 67 mg/kg b. w. orally for 8 days, while no treatment was given to the rats in the disease control and plain control groups. At the end of the experiment, renal biomarkers viz; s. creatinine, b. urea, and s. uric acid, were investigated. The histopathological examination of the kidney specimens was also carried out. Results: The results of the present study revealed that renal function biomarkers such as s. creatinine, b. urea, and s. uric acid were significantly reduced in the rats of the treatment group as compared to those of the disease control group. Moreover, the histoarchitecture reports of the treatment group's kidney specimens showed significant improvements. Conclusion: The results suggested that the HAE of Cassia occidentalis L. roots promisingly prevented kidney injury in gentamicin-induced nephrotoxic rats. This effect might be due to improved clearance of gentamicin from the renal tubule and decreased generation of reactive oxygen species (ROS).

질소고정균(Rhizobium)과 인산가용화균(Bacillus megaterium)의 동시배양을 위한 배양조건 탐색 (Effect of a Common Medium on the Growth of Nitrogen Fixer Rhizobium and Phosphate Solubilizer Bacillus megaterium)

  • 다양한 기능을 보유하고있는 농업용 유용미생물을 경제적으로 대량생산하기 위하여는 동시에 다양한 미생물이 상호간에 길항관계를 갖지않고 잘자랄수있는 배지의 선택이 필수적으로 요망된다. 본실험은 질소고정균 (Rhizobium)과 인산가용화균 (Phosphobacteria)을 yeast extract mannitol변형배지를 포함한 다양한배지에서 함께 배양시 각각의 성장을 극대화할 수 있는 접종시차를 규명하고 각 조건에서 생산된 미생물의 효능유지기간을 비교하였다. 인산가용화균과 질소고정균을 동시배양시 최대 수율을 얻기위한 조건은 인산가용화균을 먼저접종한 후 48시간후에 질소고정균을 접종한 후 60시간 배양하는것이었다. 이러한 조건으로 배양하였을 때 생산된 균의 효능유지기간은 각각의 균을 단독배양한것과 비교하여 차이가 나타나지 않았다. 또한, 이러한 조건에서 생산된 미생물의 개체수는 담체에 고정시킨후에도 60일까지 변화없이 유지되었다.

Comparisons of Developmental Potential and Gene Expression Level in Porcine Nuclear Transfer, Parthenogenetic and Fertilized Embryos

  • This study was conducted to detect the apoptosis incidence in blastocysts and to compare the abundance of Bax, Bcl2L1, VEGF and FGFR2 in in vitro fertilized (IVF), parthenogenetic (PAT) and nuclear transfer (NT) embryos. Oocytes matured for 40 hr were enucleated and reconstructed with confluenced fetal fibroblasts (FFs) derived from a ${\sim}45$ day fetus. Reconstructed eggs were then fused with 2 DC pulses (2.0 kV/cm, $30{\mu}sec$) and cultured with $7.5{\mu}g/ml$ cytochalasin B for 3 hr. Parthenotes (PAT) were produced with the same electric strength and culture for NT eggs. The embryos were cultured in NCSU-23 medium at $39^{\circ}C,\;5%\;CO_2,\;5%\l;O_2$ in air. In 3 runs, set of 10 embryos at the 4-cell to blastocyst stages were used to extract total RNA for analyzing the gene expression patterns of pro-apoptotic (Bax), anti-apoptotic (Bcl2L1), vasculogenesis (VEGF), implantation (FGFR2III) using real-time quantitative PCR. Cleavage and blastocyst rates were significantly higher (P<0.05) in IVF and PAT ($79.3{\pm}8.5\;and\;25.5{\pm}6.1,\;and\;85.0{\pm}6.4\;and\;38.6{\pm}5.5$, respectively)than NT counterparts ($65.1{\pm}5.2\;and\;15.6{\pm}3.0$, respectively). Significantly higher (P<0.05) total cells were observed in IVF controls and PAT ($34.7{\pm}5.8\;and\;38.1{\pm}4.1$) than NT embryos ($24.8{\pm}3.2$). Apoptosis index was significantly lower (P<0.05) in IVF than NT embryos. The Relative abundances (RA) of Bax and VEGF were significantly higher (P<0.05) at blastocyst stage in NT than IVF control. The RA of Bcl2L1 and FGFR2III were significantly higher (P<0.05) at blastocyst stage in IVF than NT. The present study observed the abnormal gene expressions in NT embryos at various developmental stages, suggesting certain clues to find out the cause of the low efficiency of NT to term.
