헬륨 -네온 레이저자극으로 유발된 흰쥐 진통작용의 날록손 반전 (Naloxone Reversal of He-Ne Laser Stimulation Induced Analgesia in Rat)

  • The purpose of this study were to 1) determine the analgesic effect of 632.8 nm of helium-neon (He-Ne) laser stimulation on acupuncture point in rat and 2) determine the reversal of analgesic effect by naloxone injection. Eighteen Sprague-Dawley rats were devided into three groups : control (n=6) : laser (n=6), laser stimulation at $3.58\;J/cm^2$ ; and naloxone (n=6), 1 mg/kg of naloxone chloride inject into peritoneum before laser stimulation at $3.63J/cm^2$. Tail-flick latency were measured pretreat and posttreat with hot plate $(55^{\circ}C)$. Data were analyzed using one-way ANOVA and paired t-teat for tail-flick latency. No significant change was noted in the tail-flick latency in either control or naloxone groups. But significant increased in tail-flick latency in taller group. The results suggest that He-Ne laser induced analgesic effect, and endogenous opioids may be involved in He-Ne laser induced analgesia.

흰쥐 Tail Flick Reflex의 신경생리학적 기전 (Electrophysiologic Mechanism of Tail Flick Reflex in Rats)

  • Although tail flick reflex (TFR) in rats has been used as a classic model of the nociceptive test to evaluate the action of analgesics, there have been few studies on the origin of the latent period of TFR. Present study was performed to elucidate the mechanism of increase in latency of TFR by morphine in anesthetized rats. Tail skin and dorsolateral tail nerve were stimulated electrically and EMG activities were recorded from abductor caudae dorsalis muscle participating in tail flick reflex. In the case of noxious radiant heat stimulation to tail, the tail flick tension was recorded before and after administration of morphine. Then changes in latency and conduction velocity of peripheral nerve were evaluated. The results obtained were as follows: 1) The latencies of TFR evoked by the electrical stimulation of tail skin and dorsolateral tail nerve were all within 40 ms and were elongated by several milliseconds from control after the administration of morphine. Peripheral conduction velocities of tail flick afferent nerve were within the range of 10-25 m/s. 2) The conduction velocity of peripheral nerve was significantly reduced after morphine administration, therefore the afferent time (utilization time+conduction time to spinal cord) was significantly increased. But the time for central delay and efferent time was not affected by morphine. 3) The conduction velocity under room temperature $(20-25^{\circ}C)$ was significantly reduced after morphine while that under vasodilation state $(40{\sim}42^{\circ}C)$ increased, 30 min and 45 min after morphine. The conduction velocity under vasodilation state without treatment of morphine increased continuously 4) The latency in tension response of TFR evoked by electrical stimulation was elongated by several milliseconds from control while the latency evoked by noxious radiant heat was elongated by several seconds compared with that of control. From the above results, it could be concluded that: 1) the increased latency of TFR evoked by electrical stimulation of the tail after morphine administration was due to the reducton in conduction velocity of peripheral nerve, which was the secondry effect of morphine on the peripheral vasomotion and 2) increased latency of TFR evoked by noxious radiant heat was also due to the same effect of morphine and the increase in cutaneous insulation to the noxious heat.

Antioxidant Effects of Berchemia berchemiaefolia in Nerve Pain Models

  • Berchemia berchemiaefolia (BB) are climbing plants or small to medium-sized trees that live in Africa, Asia and America. We performed the present study to investigate whether oral administration of Berchemia berchemiaefolia extract (BBE) protects SD rats from pain. The SD rat experimental groups were divided into four groups. Two of the animal model groups were fed on BBE (200 mg/kg or 100 mg/kg). We performed oral acute toxicity test to determine the optimal oral dose of BBE. To explore if BBE alleviated pain in the SD rat, we undertook the tail flick latency test and formalin test. Additionally, we conducted the anti-oxidative test. The findings of the present study suggest that Berchemia berchemiaefolia extract exhibits strong antioxidant and analgesic activities.

자동가열침의 진통 효과 (The Analgesic Effects of Automatically Controlled Heating Acupuncture)

  • Objectives : The present study was conducted to evaluate the analgesic effects of automatically controlled heating acupuncture(ACHA) using 2 different pain models(acute pain and neuropathic pain) and 2 different stimulation conditions (heating $41.5^{\cdot}C$ and heating $44.5^{\cdot}C$) in Sprague-Dawley rats. Methods : Tail flick latency(TFL) to a noxious radiant heat stimulus in lightly anesthetized rats was measured before and after ACHA stimulation for 5-min at the Zusanli(ST36) acupoint. For the neuropathic surgery, the right superior caudal trunk was resected at the level between S1 and S2 spinal nerves innervating the tail. Two weeks after the nerve injury, ACHA stimulation($41.5^{\cdot}C$ or $44.5^{\cdot}C$) was delivered to Zusanli(ST36) for 5 min. The behavioral signs of warm allodynia were evaluated by the tail immersion test (i.e. immersing the tail in warm $water(40^{\cdot}C)$ and measuring the latency to an abrupt tail movement) before and after the ACHA stimulation. Results : In the TFL test, ACHA stimulations under both the conditions above produced more potent analgesic effects than plain acupuncture(PA, acupuncture needle insertion only) and control(no treatment). In the tail immersion test, ACHA stimulations under all of the conditions had markedly relieved the warm allodynia signs. Conclusion : Automatically controlled heating acupul1cture produced analgesic effecs in acute and neuropathic pains.

초음파경혈요법의 진통소염효과 연구 (Analgesic and Anti-inflammatory Effects of Sono-acupoint Therapy)

  • Objective : Sono-acupoint (SA) therapy is a new therapeutic technique that combined with acupuncture points, herbal medicine and ultrasound therapy. This study was carried out to investigate the analgesic and anti-inflammatory effects of sono-acupoint therapy. Methods : We performed the tail-flick test with normal rats to examine the tail-flick latency (TFL), and the Freund's adjuvant-induced arthritis rat model to examine the edema, skin temperature and serum concentration of c-reactive protein and rheumatoid factor (RF). Herbal SA (HSA) treatment was performed at bilateral Zusanli (ST36) with the hanbang-gel made of several selected herbal drugs in Sprague-Dawley rats (male, $250{\pm}30g$). General SA (GSA) treatment was performed at bilateral Zusanli (ST36) with the gel used in ultrasound therapy. In arthritis rat model, Freund's adjuvant (50mg/ml) was injected in dorsal part of right foot, and these treatments were performed after 15 days. Results : TFL was lengthened after SA treatments. Skin temperature and RF concentration that were the evidence of arthritis in rats were decreased by HSA treatment (P < 0.05). Conclusion : These results indicate that HSA has the analgesic and anti-inflammatory effects in rats, and further developments will produce the advance of this new therapeutic skill.

Capsaicin에 의한 척수 수준에서의 급성 진통효과와 Norepinephrine의 변화 (Capsaicin Induces Acute Spinal Analgesia and Changes in the Spinal Norepinephrine Level)

  • Capsaicin의 중추신경계에 대한 진통효과를 척수 수준에서 규명하고, 이에 대한 norepinephrine (NE)계의 역할을 규명하기 위하여 tail flick reflex (TFR) latency time의 증가와 척수 내 NE 양의 변화를 측정하였다. 웅성의 Sprague-Dawley 백서를 pentobarbital sodium으로 마취를 하여 femoral vein cannulation과 intrathecal catheterization을 하였고, 이를 통하여 시험약제를 투여하였다. TFR은 capsaicin 또는 용매 투여전, 투여후 10분, 30분, 60분, 때로는 2시간에 측정하였다. MK-801은 capsaicin 투여전 20분에 femoral vein을 통하여 주입하였다. $35{\sim}150{\mu}g$의 capsaicin을 intrathecal로 주었을 때, TFR latency가 최고치인 7초를 넘어 정상인 2.9초에 비해 100% 이상의 증가를 보였고, 척수 내 NE은 16 ng/mg protein에서 7 ng/mg protein으로 50% 이상 감소하였다. 이와 같은 TFR latency의 증가와 NE의 감소는 전 실험기간 (capsaicin 투여후 2시간)에 걸쳐 관찰되었다. 반면, 50mg/kg의 capsaicin을 피하로 전신투여 하였을 경우에 척수 NE은 같은 변화를 보였으나 TFR latency의 차이는 없었다. 또한, 0.5 mg/kg의 MK-801를 i.v.로 전처치 하였을 때, intrathecal capsaicin에 의한 TFR latency의 증가와 척수 내 NE의 감소가 모두 억제되었다. 이상의 결과는 capsain이 excitatory aminoacid (EAA)계 - NE계 - 척수 dorsal horn으로 이루어진 동통전달의 하행성 억제계 축의 활성을 항진시켜 진통효과를 가져옴을 보여준다. 또한, capsaicin의 피하 주사시에 진통효과를 볼 수 없음을 말초 동통전달신경의 흥분에 의한 중추 동통전달계 환성의 촉진이 진통 효과를 상쇄함을 암시한다.

전침(電鍼)의 collagen 유발(誘發) 관절염(關節炎)에 대한 진통(鎭痛) 효과(效果) 및 그 기전(機轉)에 관한 연구(硏究) - opioidergic mechanism을 중심으로 - (The Analgesic Effect and Its Opioidergic Mechanism of Electroacupuncture on Inflammatory Pain in the Type II Collagen-induced Arthritis Rats)

  • Objectives : The aim of this study is to evaluate the analgesic effect of electroacupuncture on Jogsamni (ST36) in the collagen-induced arthritis rats and investigate the role played by opioid receptor subtypes $({\mu},\;{\delta},\;{\kappa})$ in the antinociceptive effect of electroacupuncture (EA) In the thermal hyper algesia test. Methods : Immunization of male Sprague-Dawley rats with bovine type H collagen emulsified in incomplete Freund's adjuvant, followed by booster injection 2 weeks later induced collagen-induced arthritis (CIA). The thermal hyperalgesia was evaluated weekly with tail flick latency (TFL). In the fourth week after first immunization, EA stimulation (2 Hz, 0.07 mA, 0.3 ms) was delivered into Jogsamni (5736) for 20 minutes. Analgesic effect was evaluated by using the tail flick latency (TFL) after intraperitoneal injection of normal saline, naloxone, naltrindole and nor-binaltorphimine respectively to CIA rats. Results : The results were as follows; 1. The TFL were gradually decreased in CIA as time elapsed after e immunization of arthrogenic collagen and the maximum value was reached between the third to fifth week. 2. EA stimulation on 5736 inhibited chronic inflammatory pain induced by CIA. 3. The analgesic effect of EA was inhibited by pretreatment of ${\mu}-receptor$ antagonist (naloxone),${\delta}-receptor$ antagonist (naltrindole) and ${\kappa}-receptor$ antagonist (nor-binaltorphimine) respectively. Conclusion : Electroacupuncture has an analgesic effect on the CIA rat and has an antinociception mediated by 8, 5, H receptors.

펜타조신의 진통작용 및 내성형성에 미치는 인삼 사포닌의 효과 (Effects of Ginseng Total Saponins on the Analgesia and Tolerance Development of Pentazocine)

  • This study examined the Influence of ginseng total saponins (GTS) on the analgestic action and tolerance development of pentazocine in mice. Pentazocine prolonged the latency to response in the tail flick rather than in the tail pinch test. The analgesic effect of pentaEocine was antagonized by naloxone and completely eliminated by pretreatment u·ith f-chlorophenylalanine (PCPA). GTS provented the pentasocine-incuced analgesia ann inhibited the development of tolerance to pentazocine. The antagonistic effect of GTS on the pentazocine-induced analgesia was abolished by 5-HTP, but not by L-DOPA. These results suggest that GTS inhibits the analgesic action of pentazocine by the interaction with serotonergic neuron.

The Analgesic Effect of Bee Venom Acupuncture and Its Mechanism on the Type II Collagen-Induced Arthritis Rats

  • Objectives : to evaluate the analgesic effect of bee venom acupuncture on Choksamni (ST36) in the collagen-induced arthritis rats and investigate the role played by serotonergic receptor subtypes (5-HT1a, 5-HT2a) in the antinociceptive effect of bee venom acupuncture in a thermal hyperalgesia test Methods : Experiments were performed on 5 week-aged 60 male Sprague-Dawley rats according to National Institute of Health guidelines and the ethical guidelines of the International Association for the Study of Pain (IASP). Arthritis was induced with arthrogenic collagen emulsion (Bovine type II collagen ${\mu}g$ with incomplete Freund's adjuvant $100\;{\mu}g$). The onset of arthritis was considered to be present when erythema and swelling were detected in at least one joint. The thermal hyperalgesia was evaluated weekly with tail flick test in the rats of severity grade 3 without any injury at tail and foot (including inflammation, ulceration, snap). In the fourth week after first immunization, the analgesic effect of bee venom acupuncture (Choksamni, ST36) was measured with consecutive tail flick latency after intraperitoneal injection of spiroxatrine (1mg/kg) and spiperone (1mg/kg). Results : Chronic inflammatory pain was induced as time elapsed after the immunization of arthrogenic collagen and the maximum value was reached from third to fifth week. Chronic inflammatory pain induced by CIA was inhibited by bee venom acupuncture on the left ST36. The analgesic effect of bee venom acupuncture was inhibited by intraperitoneal injection of 5-HT1a antagonist spiroxatrine and 5-HT2a antagonist spiperone. Conclusions : Therefore, a conclusion. that the analgesic effect of bee venom acupuncture in the chronic inflammatory pain is partially mediated by 5-HT1a and 5-HT2a receptors can be made.

Loxoprofen sodium 플라스타의 소염, 진통 작용 및 혈중 약물 농도에 대한 연구 (Anti-inflammatory and Analgesic Activities, and Plasma Concentration of Loxoprofen Sodium Plasters)

  • Loxoprofen-Na (sodium 2-〔4-(2-oxocyclopentylmethyl)pheny)propionate dihydrate) is a potent analgesic drug. We developed loxoprofen-Na plasters to extend duration time of analgesic activity and to reduce side effect on gastrointestinal tract. Analgesic effect of Loxoprofen-Na plasters was investigated. Loxoprofen-Na plaster had good analgesic effect in rat paw pressure test, Tail-flick latency test and acetic acid-induced writhing test. Also, it had anti-inflammatory effect on carrageenan-induced rat hind paw edema. In pharmacokinetic study of Loxoprofen-Na, plasters dosage form showed that plasma drug concentration was prolonged up to 14 hours. So, we can conclude that loxoprofen-Na plasters, when applied on skin, will be a new type of drug for controlling the various local pain or inflammation.
