Mining of Biomarker Genes from Expressed Sequence Tags and Differential Display Reverse Transcriptase-Polymerase Chain Reaction in the Self-fertilizing Fish, Kryptolebias marmoratus and Their Expression Patterns in Response to Exposure to an Endocrine-disrupting Alkylphenol, Bisphenol A

  • Expressed sequence tags (ESTs) and differentially expressed cDNAs from the self-fertilizing fish, Kryptolebias marmoratus were mined to develop alternative biomarkers for endocrine-disrupting chemicals (EDCs). 1,577 K. marmoratus cDNA clones were randomly sequenced from the 5'-end. These clones corresponded to 1,518 and 1,519 genes in medaka dbEST and zebrafish dbEST, respectively. Of the matched genes, 197 and 115 genes obtained Unigene IDs in medaka dbEST and zebrafish dbEST, respectively. Many of the annotated genes are potential biomarkers for environmental stresses. In a differential display reverse transcriptase-polymerase chain reaction (DD RT-PCR) study, 56 differential expressed genes were obtained from fish liver exposed to bisphenol A. Of these, 16 genes were identified after BLAST search to GenBank, and the annotated genes were mainly involved in catalytic activity and binding. The expression patterns of these 16 genes were validated by real-time RT-PCR of liver tissue from fish exposed to bisphenol A. Our findings suggest that expression of these 16 genes is modulated by endocrine disrupting chemicals, and therefore that they are potential biomarkers for environmental stress including EDCs exposure.

CrABCA2 Facilitates Triacylglycerol Accumulation in Chlamydomonas reinhardtii under Nitrogen Starvation

  • The microalga Chlamydomonas reinhardtii accumulates triacylglycerols (TAGs) in lipid droplets under stress conditions, such as nitrogen starvation. TAG biosynthesis occurs mainly at the endoplasmic reticulum (ER) and requires fatty acid (FA) substrates supplied from chloroplasts. How FAs are transferred from chloroplast to ER in microalgae was unknown. We previously reported that an Arabidopsis thaliana ATP-binding cassette (ABC) transporter, AtABCA9, facilitates FA transport at the ER during seed development. Here we identified a gene homologous to AtABCA9 in the C. reinhardtii genome, which we named CrABCA2. Under nitrogen deprivation conditions, CrABCA2 expression was upregulated, and the CrABCA2 protein level also increased. CrABCA2 knockdown lines accumulated less TAGs and CrABCA2 overexpression lines accumulated more TAGs than their untransformed parental lines. Transmission electron microscopy showed that CrABCA2 was localized in swollen ER. These results suggest that CrABCA2 transports substrates for TAG biosynthesis to the ER during nitrogen starvation. Our study provides a potential tool for increasing lipid production in microalgae.

Precise-Optimal Frame Length Based Collision Reduction Schemes for Frame Slotted Aloha RFID Systems

  • An RFID systems employ efficient Anti-Collision Algorithms (ACAs) to enhance the performance in various applications. The EPC-Global G2 RFID system utilizes Frame Slotted Aloha (FSA) as its ACA. One of the common approaches used to maximize the system performance (tag identification efficiency) of FSA-based RFID systems involves finding the optimal value of the frame length relative to the contending population size of the RFID tags. Several analytical models for finding the optimal frame length have been developed; however, they are not perfectly optimized because they lack precise characterization for the timing details of the underlying ACA. In this paper, we investigate this promising direction by precisely characterizing the timing details of the EPC-Global G2 protocol and use it to derive a precise-optimal frame length model. The main objective of the model is to determine the optimal frame length value for the estimated number of tags that maximizes the performance of an RFID system. However, because precise estimation of the contending tags is difficult, we utilize a parametric-heuristic approach to maximize the system performance and propose two simple schemes based on the obtained optimal frame length-namely, Improved Dynamic-Frame Slotted Aloha (ID-FSA) and Exponential Random Partitioning-Frame Slotted Aloha (ERP-FSA). The ID-FSA scheme is based on the tag set estimation and frame size update mechanisms, whereas the ERP-FSA scheme adjusts the contending tag population in such a way that the applied frame size becomes optimal. The results of simulations conducted indicate that the ID-FSA scheme performs better than several well-known schemes in various conditions, while the ERP-FSA scheme performs well when the frame size is small.

Constructing a Metadata Database to Enhance Internet Retrieval of Educational Materials

  • This paper reports the GEM (Gateway to Educational Materials) project whose goal is to develop an operational framework to provide the K-12 teachers in the world with 'one-stop/any-stop' access to thousands of lesson plans, curriculum units and other Internet-based educational resources. To the IS-element Dublin Core base package, the GEM project added an 8-element, domain-specific GEM package. The GEM project employed the conceptual data modeling approach to designing the GEM database, used the Sybase relational database management system (RDBMS) to construct the backend database for storing the metadata of educational resources, and also employed the active server page (ASP) technology to provide Web interfaces to that database. The consortium members catalog lesson plans and other Internet-based educational resources using a cataloging module program that produces HTML meta tags. A harvest program collects these meta tags across the Internet and outputs an ASCII file that conforms to the standard agreed by the consortium members. A parser program processes this file to enter meta tags automatically into appropriate relational tables in the Sybase database. The conceptual/logical schemas of Dublin Core and GEM profile are presented. The advantages of conceptual modeling approach to manage metadata are discussed. A prototype system that provides access to the GEM metadata is available at http://lis.skku.ac.kr/gem/.

RFID Tag Number Estimation and Query Time Optimization Methods (RFID 태그 개수 추정 방법 및 질의 시간 최소화 방안)

  • An RFID system is an important technology that could replace the traditional bar code system changing the paradigm of manufacturing, distribution, and service industry. An RFID reader can recognize several hundred tags in one second. Tag identification is done by tags' random transmission of their IDs in a frame which is assigned by the reader at each round. To minimize tag identification time, optimal frame size should be selected according to the number of tags. This paper presents new query optimization methods in RFID systems. Query optimization consists of tag number estimation problem and frame length determination problem. We propose a simple yet efficient tag estimation method and calculate optimal frame lengths that minimize overall query time. We conducted rigorous performance studies. Performance results show that the new tag number estimation technique is more accurate than previous methods. We also observe that a simple greedy method is as efficient as the optimal method in minimizing the query time.

Design and Implementation of Electronic Shelf Label System using Technique of Reliable Image Transmission (신뢰성 있는 이미지 전송 기법을 적용한 전자 가격표시 시스템의 설계 및 구현)

  • Recently, in distribution market, demand for electronic shelf label system is increasing gradually to provide the accurate price immediately and detailed product information to consumers and reduce operation costs. Most of electronic shelf label system companies develop the full-graphic display device to display a wide variety of product information as well as the exact price. Our system had introduced Go-Back-N retransmission method in the early. However, we encountered performance problems that it delayed updating of the electronic shelf label system and exhausted the battery life time. Proposed adaptive image retransmission technique based on the selective scheme is that tags of electronic shelf label system recognize idle time of transmission cycle and require partial image retransmission to sever by itself. As a result, it can acquire much more opportunities of partial image retransmission within the same period and increase reception rate of full image for each tags. The experimental result shows that adaptive image retransmission technique's reception rate of full image for each tags is approximately 4% higher than existing previous works. And total battery life time increases 30 hours because tag reduce wake-up time as it receive only lost data instead of whole data.

A STUDY ON THE DENTIN BONDING OF ONE-STEP BONDING AGENT (ONE-STEP 접착제의 상아질 접착에 관한 연구)

  • The purpose of this study was to observe the morphologic change of dentinal surface, adhesion in interface between dentin and bonding agents, and penetration pattern of resin tags into dentinal tubles according to bonding procedure of ONE-STEP universal adhesive system. Ten extracted human molars were mounted in dental stone and sectioned to expose mid-coronal occlusal dentin and again sectioned tooth crown apically. Specimens were randomly assigned to three groups for dentin conditioning with 32% phoshoric acid, two coats of bonding agents after dentin conditioning, and bond of composite resin. The surfaces of dentin were treated with etch ant and applied bonding agent, and bonded composite resin according to the directions of manufacturer. Specimens which were boned composite were sectioned longitudinally for observing interfaces between resin and dentin. Two of specimens which were sectioned longitudinally were immersed in 6 N HCL for 30 seconds and 1% NaOCL for 12 hours to partially demineralize and deproteinize the dentin substrate. Each specimen was mounted on a brass stub, sputter-coated with gold and observed under SEM. The result were as follows : 1. On the dentinal surface which was conditioned with 32% phosphoric acid. the smear layer was completely removed. orifices of dentinal tubules were opened 3-$5{\mu}m$ wide. and dentinal surface was irregular. 2. On the dentinal surface which was applied ONE-STEP. bonding agent. resin particles were observed on the orifices of dentinal tubules and intertubular dentin. 3. There were close adaptation between dentin and resin and were the pattern which composite invaded into dentin. 4. 1-$3{\mu}m$-wide hybrid layer was visible in the interface between dentin and resin. 5. Long and funnel shaped resin tags were observed in demineralized specimens. and the surfaces of tags were rough.

A Message Reduction Method for Performance Improvement of the ISO/IEC 18000-7 based Active RFID System (ISO/IEC 18000-7 기반 능동형 RFID 시스템의 성능 개선을 위한 메시지 감소 기법)

  • In this paper, we propose a novel method for improving the tag collection performance in active RFID systems by modifying the tag collection algorithm in the ISO/IEC 18000-7 standard. The proposed method enables to reduce the time slot size by reducing the response message size from the tag and to decrease the number of command messages from the reader throughout the tag collection process. This results in reducing the time required for tag collection and the battery consumption on tags by decreasing the total amount of messages. Via the simulation experiments, we evaluated the performance of the tag collection applied with the proposed method, compared with that of the basic tag collection complying with the standard. The simulation results showed that the proposed method could decrease the total amount of messages between the reader and tags dramatically and reduce the average tag collection time by 19.99% and 16.03% when the reader requested the additional data of 50 bytes and 100 bytes from the tags, respectively.

HTML Text Extraction Using Frequency Analysis (빈도 분석을 이용한 HTML 텍스트 추출)

  • Recently, text collection using a web crawler for big data analysis has been frequently performed. However, in order to collect only the necessary text from a web page that is complexly composed of numerous tags and texts, there is a cumbersome requirement to specify HTML tags and style attributes that contain the text required for big data analysis in the web crawler. In this paper, we proposed a method of extracting text using the frequency of text appearing in web pages without specifying HTML tags and style attributes. In the proposed method, the text was extracted from the DOM tree of all collected web pages, the frequency of appearance of the text was analyzed, and the main text was extracted by excluding the text with high frequency of appearance. Through this study, the superiority of the proposed method was verified.

Understanding Collaborative Tags and User Behavioral Patterns for Improving Recommendation Accuracy (추천 시스템 정확도 개선을 위한 협업태그와 사용자 행동패턴의 활용과 이해)

  • Due to the ever expanding nature of the Web, separating more valuable information from the noisy data is getting more important. Although recommendation systems are widely used for addressing the information overloading issue, their performance does not seem meaningfully improved in currently suggested approaches. Hence, to investigate the issues, this study discusses different characteristics of popular, existing recommendation approaches, and proposes a new profiling technique that uses collaborative tags and test whether it successfully compensates the limitations of the existing approaches. In addition, the study also empirically evaluates rating/tagging patterns of users in various recommendation approaches, which include the proposed approach, to learn whether those patterns can be used as effective cues for improving the recommendations accuracy. Through the sensitivity analyses, this study also suggests the potential associated with a single recommendation system that applies multiple approaches for different users or items depending upon the types and contexts of recommendations.
