Effect of Dietary Copper Sources (Cupric Sulfate and Cupric Methionate) and Concentrations on Performance and Fecal Characteristics in Growing Pigs

  • This study was conducted to assess the effects of organic and inorganic copper on performance in growing pigs. A total of 100 pigs, average age 63 d and initial body weight 21.46${\pm}$1.13 kg, were assigned to five treatment groups. Dietary treatments included i) CON (basal diet, 0 ppm Cu), ii) T1 (basal diet with 67 ppm Cu as cupric sulfate, $CuSO_4$), iii) T2 (basal diet with 134 ppm Cu as $CuSO_4$), iv) T3 (basal diet with 67 ppm Cu as cupric methionate, CuMet) and v) T4 (basal diet with 134 ppm Cu as CuMet). Throughout the entire experimental period, ADG (average daily gain), ADFI (average daily feed intake) and G/F (gain: feed) ratios showed no significant differences. The dry matter digestibility was improved in the T1, T2, T3, and T4 treatments (p<0.05), as compared with CON. Nitrogen digestibility was improved in the T3 treatment group as compared with CON (p<0.05). As compared with the T1 treatment group, fecal pH values were improved in the CON, T3, and T4 treatment groups (p<0.05). Fecal Cu concentrations were significantly lower in the CON, T3, and T4 treatment groups than in T1 and T2 (p<0.05). The incidence of diarrhea was reduced when the pigs were fed on the T2, T3, and T4 diets as compared with CON. In conclusion, diets supplemented with 67 or 134 ppm Cu as CuMet may prove effective in improving nutrient digestibility and fecal pH value in growing pigs, and fecal Cu concentrations may be reduced by CuMet supplementation.

Effect of Italian Ryegrass(Lolium multiflorum) and Kentucky Bluegrass(Poa pratensis) Planted Singly or in Combination with Other Forage Species on Suppressing Weeds (단파 또는 혼파초지에서 Italian ryegrass와 Kentucky bluegrass의 잡초억제 효과)

  • A field plot experiment was carried out on mixed pasture from October 1995 to October 1997 to examine the effect of weed control as affected by the difference of mixed pasture species, planning with spilt plot design which main plots of 2 treatments (companion crop Italian ryegrass Lolium multiflorum and Kentucky bluegrass Poa pratensis) are divided into subplots of 4 treatments (T1: single companion crop, T2: Tall fescue Festuca arundinasea + Orchardgrass Dactylis glomerata + Ladino clover Trifolium repens, T3 : Perennial ryegrass Lolium perenne + orchardgrass + ladino clover, T4 : orchardgrass + ladino clover). Average 2 year-dry matter yields of companion crops as whole plots, Italian ryegrass and Kentucky bluegrass, were 15,437kg/ha and 14,629kg/ha, respectively and those of T1, T2, T3 and T4 as sub plots were 14,815, 15,407, 14,629 and 15,187kg/ha respectively without statistical1y significant difference(P>0.05). Though there was no significant difference between the dry matter yields of Italian ryegrass and Kentucky bluegrass, as whole plots, at 1st harvest of second year, dry matter yields of T3 and T4, as 4,591kg/ha and 4,816kg/ha respectively, were higher than those of T1, 3,376kg/ha(P<0.05), significantly. There was a significant difference among 2nd year-average DM yields of sub plots(P<0.05), showing highest with T4 15,131kg/ha and least with T1 13,085kg/ha and T3 13,269kg/ha. Italian ryegrass, as the companion crop, was better for the control of weeds in the early spring than Kentucky bluegrass, but reached to the similar botanical composition level with Kentucky bluegrass after summer with rapid decrease of Italian ryegrass. Percentage of Kentucky bluegrass in botanical composition increased slowly, but did'nt become an important factor for the control of weeds at the end of this experiment. Using orchardgrass in mixed pasture such as T2, T3 and T4 made weeds more decreased in mixed pasture than one in single conpanion crop. There was no difference among treatments of whole or sub plots in crude protein contents of pasture taken in this study. Contents of NDF and ADF of companion crop. Italian ryegrass plots harvested in October 1997 were hither than those of Kentucky bluegrass plots(P<0.05). ADF contents of Kentucky blue grass-plots taken in July 1997 has shown to be high significantly as compared to those of Italian ryegrass plots(P<0.01). The results of this study support that the companion crop Italian ryegrass was good for weed controls only in spring of 1st year after sowing and orchardgrass in mixed pastures played good roles for increase of dry matter yield and weed control.

  • 본 연구에서는 $CD4^+$ T 세포를 일회성으로 감염시킬 수 있고 $CD4^+$ T 세포에 HIV-1 envelope 유전자를 전달할 수 있는 바이러스 입자를 생산하는 HIV-1 complementation system을 개발하였고 이 system을 이용하여 $CD4^+$ T 세포에 선택적으로 HIV-1 envelope 당단백질을 발현시켰다. 이 system은 Gag/Gag-Pol expressor와 Env expressor로 구성되어있다. Gag/Gag-Pol expressor는 바이러스 입자 생산에 필요한 구조단백질과 기능단백질을 발현시키지만 packaging signal이 결핍되어 바이러스 입자로 유전자가 들어가지 못하도록 제조되었다. Env expressor는 Tat, Rev와 envelope 당단백질을 발현시키고 packaging signal을 갖고 있어 바이러스 입자로 envelope 유전자가 들어가도록 제조되었다. Gag/Gag-Pol expressor로부터 Gag와 Gag-Pol의 발현은 Rev 단백질을 요구하였고 Env expressor로부터 Rev 단백질 이 제공될 때 Gag와 Gag-Pol 단백질은 효율적으로 발현되었다. Gag/Gag-Pol과 Env expressor로 cotransfection된 COS-1 세포에서 $CD4^+$ T 세포를 일회성으로 감염시킬 수 있는 바이러스 입자가 생산되었다. 생산된 바이러스 입자는 $CD4^+$ T 세포에 HIV-1 envelope 유전자를 전달하여 envelope 당단백질을 발현시켰고 복제 가능한 자손 바이러스의 생성을 유도하지 못하였다. 본 연구에서 개발된 방법은 $CD4^+$ T 세포에서 envelope 당단백질의 기능을 분석하고 관심 있는 유전자를 $CD4^+$ T 세포에 전달하는 바이러스 입자의 생산에 이용할 수 있다.

Comparative Analysis of T4SS Molecular Architectures

  • The recently published high-resolution R388 T4SS structure provides exciting new details about the complete complex of T4SS, including the components making up the stalk and arches, numerous symmetry mismatches between regions of the complex, and an intriguing interpretation of the closed stalk and radial symmetry of the inner membrane complex, which is related to pilus biogenesis assembly. However, there are a few unidentified densities in the electron microscopy map and portions of the identified component sequences for which the structure is not yet known. It is also unclear how well this minimized DNA-transporting T4SS predicts the structure of other T4SSs, such as expanded systems and those that transport proteins rather than DNA. In this review, we evaluate what can be inferred from the recent high-resolution structure of the R388 T4SS with respect to the Cag and Dot/Icm systems. These systems were selected because, given what is currently known about these systems, we expect them to present most structural differences compared to the R388 T4SS structure. Furthermore, we discuss bacterial physiology and diversity, the T4SS structures and their variations between different bacterial species. These insights may prove beneficial for researchers who elucidate the structure and functions of T4SS in different bacterial species.

Optimum Harvest Stage of Italian Ryegrass 'Kowinearly' According to One and Two Harvests During Spring Season (이탈리안 라이그라스 '코윈어리'의 봄철 1회 및 2회 이용에 따른 수확적기 구명)

  • This study was carried out to determine the optimum harvest stage of Italian ryegrass (Lolium multiflorum Lam., IRG) for maximum forage production during the spring season in Suwon, 2010. The variety of IRG was the early maturity type, 'Kowinearly', and six harvest stages (treatments) were first heading (T1), heading (T2), late heading to early bloom (T3), bloom to late bloom (T4), ripeness (T5), and late ripeness stage (T6). The dates of the first heading and heading of 'Kowinearly' were seen on 4 to 5 May, and 14 May, respectively. Plant length and dry matter (DM) percentage at first harvest were from 69 cm and 14.8% at T1 stage to 103 cm and 35.0% at T6 stage, respectively. The content of crude protein (CP) and in vitro DM digestibility (IVDMD) of T1, T2, T3, T4, T5 and T6 at first harvest were 15.6%, 10.6%, 10.1%, 8.1%, 7.3% and 5.4%, and 81.8%, 72.1%, 64.8%, 63.8%, 61.4% and 59.0%, respectively. The content of neural detergent fiber (NDF) and acid detergent fiber (ADF) were increased continuously with delayed harvest. A significantly higher yield of DM, CP and in vitro digestible DM (IVDDM) were observed for T3, and T4 (p<0.05). DM yield of 3,526 kg, 6,278 kg, 7,842 kg, 8,984 kg, 8,346 kg and 8,008 kg/ha, CP yield of 549 kg, 665 kg, 795 kg, 725 kg, 608 kg and 430 kg/ha, and IVDDM of 2,883 kg. 4,526 kg, 5,083 kg, 5,728 kg, 5,124 kg and 4,722 kg/ha at first harvest were recorded in T1, T2, T3, T4, T5 and T6, respectively. Regrowth yield of DM, CP and IVDDM were shown to be higher at T1 and T2 (p<0.05). However, no significant differences were observed between the two stages. Daily DM and DDM production of regrowth IRG were higher at T2, followed by T1. The total yield (at first and at regrowth) of DM, CP and IVDDM were significant higher for T2, followed by T3, T4 and T1 in order. At T2 stage, the yield was 11,089 kg, 1,254 kg, and 7,669 kg/ha in DM, CP, and IVDDM. In conclusion, the late heading to bloom stage was determined to be the optimum harvest stage for a single harvest, while the heading stage was a suitable stage of first harvest of 'Kowinearly' where two harvests were sought in a single year.

Effects of Dietary Supplementation of Mulberry Leaves and Dandelion Extracts on Performance and Blood Characteristics of Chickens (뽕잎과 민들레 추출물 급여가 육계의 생산성 및 혈액성상에 미치는 영향)

  • This study was conducted to investigate the effects of dietary supplementation of mulberry leaves and dandelion extracts on performance, proximate composition, heating loss, drip loss, and blood characteristics of chickens. One hundred sixty broiler chicks were fed diets for five weeks containing 1% mulberry leaves extracts (T1), 2% mulberry leaves extracts (T2), 1% dandelion extracts (T3), and 2% dandelion extracts (T4). At the end of five week feeding experiment, broiler were slaughtered, and stored at $4^{\circ}C$. T2 resulted in much better weight gain and feed conversion than other treatment groups. As storage time increased, all treatment groups resulted in increased heating loss and drip loss (P<0.05) but no significant difference were observed among the treatment groups. In blood composition, GOT (glutamic oxaloacetate transaminase), GPT (glutamic pyrubic transaminase), total cholesterol, and triglyceride were significantly decreased by the supplementation of mulberry leaves and dandelion extracts compared to the control (P<0.05). Especially, T4 was significantly (P<0.05) more effective in improving blood composition compared to other treatment groups. However, no significant difference (P>0.05) were found in LDL-cholesterol among all treatment groups. In conclusion, these data indicate that compared to other treatments, supplementation of 2% dandelion extracts (T4) were most effective in decreasing total cholesterol and triglyceride and increasing HDL-cholesterol.

Effects of Feeding Dietary Different Oil and Tocopherol on Physico-Chemical Characteristics of Pork (오일과 토코페롤 급여가 돈육의 이화학적 성질에 미치는 영향)

  • The pork quality was determined with following treatments. The meat samples were obtained from pigs which had been fed finishing pig diets containing 5% beef tallow(Control), 3% beef tallow and 2% perillar seed oil(T1), 250 ppm vitamin E(a-tocopheryl acetate) in T1(T2), 3% beef tallow and 2% squid viscera oil(T3), 250 ppm vitamin E in T3(T4), 3% beef tallow and 2% CLA(Conjugated linoleic acid, T5). T1 had the lowest sarcomere length, salt solubility and total protein contents among the treatments. Salt solubility and total protein content of T2 and T4 which had been fed diets containing Vit. E were higher than those of T1 and T3 which had not been fed diets without Vit. E. pH and water holding capacity(WHC) values of control were higher than those of T1, T3 and T5, while WHC of T2 and T4 was higher than those of T1, T3 and T5. The hunter L value of meat and a value of fat showed higher in T5 than those in control, T, T3. The adhesiveness of T3 and the springiness of T5 in cooked meat showed higher level than other treatments.

Induction of CD4+ Regulatory and Polarized Effector/helper T Cells by Dendritic Cells

  • Dendritic cells (DCs) are considered to play major roles during the induction of T cell immune responses as well as the maintenance of T cell tolerance. Naive CD4+ T cells have been shown to respond with high plasticity to signals inducing their polarization into effector/helper or regulatory T cells. Data obtained from in vitro generated bone-marrow (BM)-derived DCs as well as genetic mouse models revealed an important but not exclusive role of DCs in shaping CD4+ T cell responses. Besides the specialization of some conventional DC subsets for the induction of polarized immunity, also the maturation stage, activation of specialized transcription factors and the cytokine production of DCs have major impact on CD4+ T cells. Since in vitro generated BM-DCs show a high diversity to shape CD4+ T cells and their high similarity to monocyte-derived DCs in vivo, this review reports data mainly on BM-DCs in this process and only touches the roles of transcription factors or of DC subsets, which have been discussed elsewhere. Here, recent findings on 1) the conversion of naive into anergic and further into Foxp3- regulatory T cells (Treg) by immature DCs, 2) the role of RelB in steady state migratory DCs (ssmDCs) for conversion of naive T cells into Foxp3+ Treg, 3) the DC maturation signature for polarized Th2 cell induction and 4) the DC source of IL-12 for Th1 induction are discussed.

Biomass and Net Primary Productivity in Natural Forests of Quercus mongolica and Quercus variabilis (신갈나무와 굴참나무 천연림(天然林) 생태계(生態系)의 현존량(現存量) 및 물질(物質) 생산성(生産性)에 관한 연구)

  • A study has been made to estimate biomass and NPP based on equation form of $Wt=aD^bH^c$ for Quercus variabilis and Quercus mongolica natural stands(Mean age; 67, 62yrs old) in Chungju. Equation form of $Wt=aD^bH^c$ was more adequate than $Wt=a(D^2H)^b$ and $Wt=aD^b$ for the estimation of the biomass and NPP. Individual biomass was compared using a paired t-test by tree component which showed no significant differences. Total aboveground biomass of Quercus mongolica was 130.6 t/ha and that of Quercus variabilis was 137.4 t/ha. Biomass of Q. mongolica was composed of foliage 5.1 t/ha(3.9%), dead branch 3.5 t/ha(2.7%), live branch 29.7 t/ha(23.0%), bolebark 16.2 t/ha(12.5%), and bolewood 74.9 t/ha(58.0%), and that of Q. variabilis was composed of foliage 3.8 t/ha(2.9%), dead branch 2.9 t/ha(2.2%), live branch 24.3 t/ha(18.4%), bolebark 20.4 t/ha(15.5%), and bolewood 80.4 t/ha(61.0%). Net primary production was 10.0 t/ha/yr in the Q. mongolica stand and 8.6 t/ha/yr in the Q. variabilis stand, respectively. Net primary production of Quercus forest in Chungju was very close to the mean NPP of the broadleaved forest of temperate zone.

  • At the aim of finding a Fehased amorphous alloy with a wide supercooled liquid region (${\Delta}T_{x}=T_{x}-T_{g}$) before crystallization, the changes in glass transition temperatudfI$T_{g}$ and crystallization temperature ($T_{x}$) by the dissolution of additional M elements were examined for the $Fe_{80}P_{10}C_{6}B_{4}$(x~6at%. M= transition metals) amorphous alloys. The ${\Delta}T_{x}$ value is 27K for the Fe,,,P,,,C,,R, alloy and increases to 40K for the addition of M=4at%Hf, 4at%Ta or 4at%Mo. The increase in ${\Delta}T_{x}$ is due to the increase of $T_{x}$ exceeding the degree in the increase in $T_{g}$. The $T_{g}$ and $T_{x}$ increase with decreasing electron concentration (e/a) from about 7 38 to 7.05. The decrease of e/a also implies the increase in the attractive bonding state between the M elements and other constitutent elements. It is therefore said that $T_{g}$ and $T_{x}$ increase kith increasing attractive bonding force.
