In order to examine whether arsenic, one of environmental stress, contribute to augumentation and relaxation of rat aorta, this study was performed in vivo and in vitro, using intacted or denuded rats aorta ring preparation, respectively. The carotid arterial pressure was recorded on an ink-writing physiograph(Grass Co. 79E) connected to strain gauge. The contractile response of vascular ring with or without endothelium preparation isolated from rat were determined in organ bath and was recorded on physiograph connected to isometric transducer. Vasopressin-,and phenylephrine- induced increase in arterial pressure significantly enhanced in arsenic-treated rats; increase of 19.1%, and 46.6%, respectively. Vascular contractile response was measured in vitro preparations exposed to 0, 0.5, 1, 2 and 4 mM of arsenic for 1, 3, 5 and 8 hours. The dose-vascular responses of phenylephrine augmented by increasing dose of arsenic in the strips exposed to arsenic for 8 hours, and did not augmented for 1, 3, 5 hours. The phenomenon was not affected by strips denuded endothelium. And the response of relaxation of rat aorta induced by nitroprusside was not influenced by arsenic stress, but acetylcholine was a little increased. compared to that of control. There were no significant difference in relaxation between control and arsenic treated rings with endothelium or denuded. All of the results, phenyleprine-induced vascular contraction was significantly enhanced in 4 mM arsenic-treated rat aortic rings compared with control, whether endothelium was present or denuded at 8 hours after arsenic treatment. It may be a mechanism by which long-term arsenic stresses play a role in development of hypertension.

The purpose of this study is to describe the Non-Destructive Testing(NDT) of the rockbolt and investigate the applicability of the NDT methods to estimate the integrity of the rockbolt. To examine the rockbolt integrity including rockbolt itself and grouting material, two methods are adopted: numerical and experimental methods. In the numerical method, the numerical code DISPERSE is used to analyze the dispersion of the rockbolt. The dispersion curve shows the effects of the thickness and stiffness of grouted materials on the embedded rockbolt. Therefore, the optimal frequency for the integrity test of the rockbolt is obtained: 20~120kHz in L(1,0) mode. In the experimental methods, destructive and non-destructive tests are carried out in a laboratory. In the non-destructive test, the low frequency mode generated by an impact and t he high frequency mode generated by an ultrasonic transducer seem to characterize the rockbolt condition readily. The experimental results show that the guided waves attenuate more significantly when the stiffness of the grouted material increases and/or the zone of the defect increases. Meanwhile, the ultimate capacity of rockbolt was evaluated through the pull-out tests and is compared to the NDT results. This study demonstrates that the NDT is a valuable tool for the rockbolt integrity evaluation.
Background: Anticholinesterase drug inhibits acetylcholinesterase(AChE), induce accumulation of acetylcholine(ACh) near cholinergic receptors and cholinergic stimulation. This experiment was performed to study the effects of anticholinesterase drugs on gastric motility and the effect of ethanol on anticholinesterase drug-induced motility change. Materials and Methods: After excision of stomach, $2{\times}10mm$ circular muscle strips were made, which were then fixed to the isolated muscle chamber. An isometric tension transducer was used to measure the contraction change of the gastric smooth muscle strips after drug addition. Results: Fenthion, an irreversible anticholinesterase drug, increased ACh induced contraction of gastric smooth muscle strips and PAM, a cholinesterase activator, antagonized this action. Physostigmine, a reversible anticholinesterase drug, also increased the ACh induced contraction. The gastric motility was decreased by PAM. Ethanol, which is known to induce smooth muscle relaxation, inhibited the increase of contraction by fenthion. Conclusion: These results indicate that irreversible and reversible anticholinesterase drugs increase gastric motility and antagonized by cholinesterase activating drugs. And when exposed to both ethanol and anticholinesterase drug, gastric motility was decreased by the smooth muscle relaxation effect by ethanol.
Beak trimming in laying hens is a routine practice in which about 1/3-1/2 of the upper and lower beak is removed with the aim of reducing cannibalism. This experiment aimed to identify if this procedure causes pain by examining self-administration of an analgesic (carprofen) and pecking behaviour in 80 laying pullets beak-trimmed by two different methods at one day of age using hot-blade cauterisation or infra-red cauterisation. We also tested a control treatment, pullets with intact beaks, and a positive control treatment of pullets beak trimmed at 10 weeks of age which were expected to experience some pain due to recent severing of the underlying nerves in the beak. At 11 weeks of age birds trimmed at 10 weeks of age pecked more (p<0.001) gently ($0.6{\pm}0.06N$) at a disc attached to a force-displacement transducer than birds trimmed at 1 day of age with an infra-red machine ($0.9{\pm}0.1N$) or a hot blade ($1.1{\pm}0.07N$) and intact birds ($1.2{\pm}0.1N$). Maximum force of pecks recorded was also lower (p<0.001) in birds trimmed at 10 weeks of age than birds trimmed at 1 day of age with an infra-red method or a hot blade and intact birds. However, the pecking force in birds trimmed at 10 weeks of age was not increased by providing them with analgesic-treated feed, though birds that ate more carprofen had a higher maximum force of peck (p = 0.03). Pecking force in birds beak-trimmed at 1 day of age was the same as the pecking force of intact birds, and was unaffected by feeding pullets carprofen. A method of self-administration of an analgesic had previously revealed that chickens in neuromuscular pain arising from lameness consumed more of a feed containing carprofen than healthy chickens. However, we found no evidence that beak-trimmed pullets consumed more carprofen-treated feed than pullets with an intact beak. It should be noted that the three beak trimming methods resulted in an average 34% reduction in beak length, considered a light trim, and is perhaps not representative of commercial birds where greater portions of the beak are removed. We conclude that although carprofen has been reported to have an analgesic effect on neuromuscular pain in chickens, it appears to have no analgesic effect on potential neuropathic pain arising from the nerves severed by a light beak trim.

There are many tube and pipeline in nuclear power plant under high temperature and high pressure. Erosion and corrosion defects were expected on these tube and pipe-line by environmental and mechanical factors. These erosion and corrosion defects ran be evaluated by ultrasonic technique. In these study, Scanning Laser Source(SLS) technique was applied to detect defect and construct image. This technique also makes detection possible on rough and curved surfaces such as tube and pipe-line by scanning. Conventional ultrasonic scanning technique requires immersion of specimen or water jet for transferring ultrasonic wave between transducer and specimen. However, this SLS technique does not need contacting and couplant to generate surface wave and to get flaw images. Therefore, this SLS technique has several advantages, for complicated production inspection, non-contact, remote from specimen, and high resolution. In this study, SLS images were obtained with various conditions of generation laser ultrasound and receiving in order to enhance detectability of flaws on the tube. Stress corrosion cracks were produced on tube and images of stress corrosion cracks were constructed by using SLS technique.

Failure of small bore piping welds is a recurring problem at nuclear power plants. And the socket weld cracking in small bore piping has caused unplanned plant shutdowns for repair and high economic impact on the plants. Consequently, early crack detection, including the detection of manufacturing defects, is of the utmost importance. Until now, the surface inspection methods has been applied according to ASME Section XI requirements. But the ultrasonic inspection as a volumetric method is also applying to enforce the inspection requirement. However, the conventional manual ultrasonic inspection techniques are used to detect service induced fatigue cracks. And there was uncertainty on manual ultrasonic inspection because of limited access to the welds and difficulties with contact between the ultrasonic probe and the OD(outer diameter) surface of small bore piping. In this study, phased array ultrasonic inspection technique is applied to increase inspection speed and reliability. To achieve this object, the 3.5 MHz phased array ultrasonic transducer are designed and fabricated. The manually encoded scanner was also developed to enhance contact conditions and maintain constant signal quality. Additionally inspection system is configured and inspection procedure is developed.
Suppressors of cytokine signaling (SOCS) proteins were originally identified as negative feedback regulators of cytokine signaling and include the Janus kinase/Signal transducer and activator of transcription (JAK/STAT) pathways. Recent studies have shown that SOCS proteins negatively regulate the receptor tyrosine kinase (RTK) pathway including the insulin receptor (IR), EGFR, and KIT signaling pathways. In addition, SOCS1 and SOCS3 have been reported to have anti-tumor effects in human hepatocellular carcinoma (HCC). However, it is uncertain whether other members of the SOCS family are associated with tumor development and progression. In this study, to investigate whether SOCS6 is aberrantly regulated in HCC, we examined the expression level of SOCS6 in HCC by Western blot analysis and immunohistochemical staining. The results showed that SOCS6 was down-regulated in all examined HCCs compared to the corresponding normal tissues. In addition, expression of SOCS6 was observed in the cytoplasm of most normal and precancerous tissue, but not in the HCCs by immunohistochemical staining. This is first report to demonstrate that SOCS6 is aberrantly regulated in HCC. These findings suggest that underexpression of SOCS6 is involved in hepatocarcinogenesis, and SOCS6 may play a role, as a tumor suppressor, in HCC development and progression.

Purpose: Ultrasound is of proven accuracy in abdominal and thoracic trauma and may be useful for diagnosing extremity injury in situations where radiography is not available, such as disasters and military and space applications. However, the diagnosis of fractures is suggested by history and physical examination and is typically confirmed with radiography. As a alternative to radiography, we prospectively evaluated the utility of extremity ultrasound performed by trained residents of emergency medicine (EM) one patient with wrist and ankle extremity injuries. Methods: Initially, residents of EM performed physical examinations for fractures. The emergency ultrasound (EM US) was performed by trained residents, who used a portable ultrasound device with a 10- to 5-MHz linear transducer, on suspected patients before radiography examination. The results of emergency ultrasound and radiography and the final diagnosis were recorded, and correlation among them were determined by using Kappa s test Results: Thirty-nine patients were enrolled in our study. The average age was $36.6\;{\pm}\;19.3$ years. There were radius Fx. (n=21), radius-ulna Fx. (n=1), ulna Fx. (n=1), and contusion (n=2) injuries among the wrist injury and lat.-med. malleolar Fx. (n=13), lat. malleolar Fx. (n=6), and med. malleolar Fx. (n=3) injuries among the ankle injury. Comparing EM US with radiography, we found the sensitivity, specificity, positive predictive value, and negative predictive value of EM US for Fx. diagnosis to be 100%, 66.7%, 97.3%, 100% and those of radiography to be 97.2%, 100%, 100%, and 75%, respectively. Kappa s test for a correlation between the Fx. diagnosis of EM US and the final diagnosis of Fx was performed, and Kappa's value was 0.787 (P = 0.004).Conclusion: EM US for Fx. can be performed quickly and accurately by EM residents with excellent accuracy in remote locations such as disaster areas and in military and aerospace applications. EM US was as useful as radiography in our study and had a high correlation to the final diagnosis of Fx. Therefore, ultrasound should performed on patients with extremity injury to determine whether extremity evaluation should be added to the FAST (focused abdominal sonography trauma) examination.
The effect of ${\alpha}_1$-adrenoceptor(${\alpha}_1$-AR) stimulation on intracellular pH($pH_i$), $Na^+$ activity($a_{Na}{^i}$) and contractility were investigated in isolated papillary muscles of euthyroid or hyperthyroid guinea pig with conventional microelectrode, $Na^+$ or $H^+$-selective microelectrodes, and tension transducer. Stimulation of the ${\alpha}_1$-AR by phenylephrine produced a decrease in $a_{Na}{^i}$ in euthyroid preparations. This decrease in $a_{Na}{^i}$ was abolished in presence of PKC activator, phorbol dibutyrate, and increased contrary to decrease. Phenylephrine also increased $a_{Na}{^i}$ in hyperthyroid ones. However, phenylrephtine produced an increase in $pH_i$ in both euthyroid and hyperthyroid ones. These changes were blocked by prazosin, an antagonist of ${\alpha}_1$-AR. These findings suggest that the changes in $a_{Na}{^i}$ and $pH_i$ are mediated by a stimulation of $Na^+-H^+$ exchange via ${\alpha}_1$-AR stimulation. This study focused on the increase in $a_{Na}{^i}$, $pH_i$ and contractility. The increase in $pH_i$ was blocked by amiloride or EIPA, $Na^+-H^+$ exchange inhibitors. Therefore, the increase in $a_{Na}{^i}$ and $pH_i$ mediated by ${\alpha}_1$-AR appeared to be due to an influx of $Na^+$ and a reduction of $H^+$ through $Na^+-H^+$ exchange. This study also revealed that the increase in $pH_i$ and $a_{Na}{^i}$ might be related to the sustained positive inotropic response. The $a_{Na}{^i}$ increase may contribute to the intracellular $Ca^{2+}$ through the $Na^+-Ca^{2+}$ exchange, and the $pH_i$ increase could cause an increase in the $Ca^{2+}$ sensitivity of myofilaments and may augment the ${\alpha}_1$-AR-mediated positive inotropic response.

Hydroacoustic surveys were conducted in the East China Sea, in the summers of 1990-1991 to investigate the distribution and migration behavior of fish, such as the diel changes in the distributions of fish, the thermoselection behavior of fish, and the fish reactions to a surveying vessel. The hydroacoustic observations were taken with a scientific echo sounder operating at 50kHz and a microcomputer-based echo processor. Fish samples were collected by bottom trawling and temperature was measured with a DBT system. The patterns of fish distributions were compared with the vertical profiles of water temperature. The results obtained can be summarized as follows: 1. The time series of hydroacoustic observations on the downward migration at dawn in the central region of East China Sea showed that the migration rate was 0.24 m/min. In this region, most of the fish collected by bottom trawling during the surveys were squid (Photologio edulis f. budo) and horse mackeral (Trachurus japonicus). 2. The hydroacoustic observations, which together associated with temperature data, indicated that squid and horse mackeral concentrated nocturally in a narrow vertical band of thermocline, particularly in summer when the survey region was strongly stratified. This result suggest that the nocturnal vertical distribution of these species evidently is controlled by temperature. 3. When the survey vessel, that is running at a speed of 11 knots, has suddenly stopped, the fish which concentrated beneath the transducer showed a strong avoidance reaction in the form of rapid downward migration. After a few minutes, the descending reaction disappeared and the fish slowly began the upward migration toward their original swimming positions. Trawl data suggest that the fish, which showed the avoidance reaction, was Liparis tessellatus.
