Single Dose Toxicity Study of Hwangiaegongjinbo, an Invigorator, in Mice and Rats (마우스 및 랫드에서 자양강장제 황제공진보의 단회투여독성시험)

  • The single dose toxicity of Hwangiaegongjinbo, an invigorator developed by Korea Medical Science Institute was evaluated in ICR mice and Sprague-Dawley rats. The aqueous solution of freeze-dried powder of Hwangiaegongjinbo or its original solution was once administrated orally to both sexes of mice and rats at dose of 2000 mg/kg, the recommended upper limit dose for acute toxicity. Water was administered to another group as control. after single adminstration, sign of toxicity were observed every hour for the first 6 hours and every day for 14 days. Neither sign그cant toxic sign nor death was observed during the observation period. In addition, no pathological changes were noticed in macroscopic examination at necropsy in those treated group. These results indicated that $LD_{50}$ of Hwangiaegongjinbo is greater than 2000 mg/kg in ICR mice and Sprague-Dawley rats.

  • Bioluminescent bacteria fusing the stress promoter and lux gene have been developed as a toxicity biosensor. The light emitting bioluminescent bacteria have been used to measure the toxicity of many different chemicals. In this study, specially, DPD2540 (fabA::luxCDABE) was used to detect and classify phenolic toxicity to the cells membrane fatty acids, and then the relationship between phenolic toxicity and the distribution of various phenols in the cell was determined, with a model and equations provided. In addition, to show the possibility of detecting and classifying the toxicity of a chemical mixture, which may be present in wastewater, various bioluminescent bacteria having different stress promoters were used and their distinct response to the sample mixture was measured. To extend the applicable area of these bioluminescent bacteria to field, the portable biosensor using freeze-drying methods was developed and confirmed successfully.

Silver Materials Induce Differential Cytotoxicity and Pulmonary Toxicity Based on Size and Shape

  • Silver materials may be toxic in humans because they can enter the body and accumulate, typically in the lungs. We hypothesized that the cytotoxicity of naive silver materials is affected by their size and shape. Our in vitro assays revealed that the overall toxicity was in the following order: submicro-particles>wires>micro-particles. These results contrast with previous studies, which showed that silver wires are the most toxic among the three tested materials, possibly due to differences in cell lines. Evaluations of in vivo pulmonary toxicity revealed eryptosis in the cavity lining of the lung sections. The observed eryptosis was consistent with the in vitro results. Our results indicate that silver materialinduced cytotoxicity must be measured and compared using various methods.

  • In aqueous ecosystems, the level of toxicity is highly responsive dependant to multiple variables, including rainfall, sunlight, pH, adhesion, etc. Because Korea has particularly distinct wet and dry seasons, the toxicity of pollutants in rivers or streams is dependant on the sampling season and time. In order to examine the effects of rainfall on toxicity, sediment samples were collected from five sites along the Keumho river. It was found that Microtox toxicity levels were generally higher during the dry season than the wet season. It indicated that river pollutants are carried off more quickly by the water during the wet season. As a result, it was recommended that the point sources of pollutants of the Keumho river would be placed between KH3 (Paldalgyo) and KH4(Keumhogyo), KH4(Keumhogyo) and KH5(Dasa).

Effect of Dietary Chitin and Chitosan on Cadmium Toxicity and Lipid Metabolism in Rats (Chitin과 Chitosan이 흰쥐의 Cadmium중독과 지방대사에 미치는 영향)

  • This study was performed to investigate the effect of dietary chitin & chitosan on cadmium (Cd) toxicity and lipid metabolism in rats. Forty-two male rats of Sprague-Dawley strain weighing 137$\pm$2g were blocked into 6 groups according to body weight, and were raised for 4 weeks. Cadmium chloride was given at the level of 0 or 400ppm in diet and chitin and chitosan were given at the level of 0 or 4%(w/w) of diet. The results are summarized as follow. Chitosan decreased the toxicity of Cd on liver, kidney and femur and increase the Cd content of fecal excretion. Chitosan increased the lipdi & cholesterol content of fecal excretion by combining with lipid and bile acid. Chitosan decreased lipid, cholesterol and TG content in serum and liver by combining with lipid and bile acid. Chitin was less effective than chitosan in decreasing of Cd toxicity and lipid content of rat.

오염된 토양의 중금속 분포와 독성상관 관계 연구

  • In this study, the relationship between metals (Cu, Cr, Cd, Zn. Pb, and Fe) 5 fractions and toxicity of soil samples from various contaminated sites in Korea were investigated. Metal toxicity of soils was tested using MetPLATE$^{TM}$ test kit, which is known as metal sensitive and organic insensitive. Significant amount of Fe was found in soils, and metal contents were in the order of Fe>Zn>Pb>Cu>Cr>Cd. Metal levels in organic fraction were rather high for all metals except Fe, and quite high percentages (35~79%) for residual fraction were observed for all metals. There were no significant relationships between the content of each metal fraction and toxicity which showed regression $R^2$in the range of 0.0003~0.414. However, correlation between toxicity and total metal contents showed regression coefficient $R^2$= 0.72. These results showed that the risk evaluation of metals in contaminated sites should be difficult to assess only by the contents of metal distribution because of the complexity of mixture of various metals.s.

  • The acute and subacute toxicity of fumomsin $B_1$ was evaluted in fingerlings of common carp, Cyprinus carpio. Dipping of fish for acute toxicity was performed for a period of 48 hours, and the $TLm_{48h}$ value(median tolerance limit) was more than 1000 ppm in common carp. Severe damages were observed in various organs and among them, clubbing of gill lamella, lytic degeneration and vacuolation of liver cells, and epithelial edema of renal tubules were relatively prominent. The most significant changes were hyperbasophilic foci of liver cells in subacute toxicity test and these can imply the possibility of hepatocarcinogenecity of fumonisin $B_1$.

  • In this study, we investigated the single dose toxicity and safety about water-extracted Galgeuntang. To evaluate single dose toxicity, 20 male and 20 female ICR mice were observed survival rates, general toxicity, changes of body weight for 14 days after single oral administration of 0 (control group), 1250, 2500 and 5000mg/kg Galgeuntang. And after 14 day, We observed autopsy. Compared with the control group, we could not find any toxic alteration in all treated groups (1250, 2500 and 5000 mg/kg). LD50 of Galgeuntang might be over 5000mg/kg and it is very safe to ICR mice.

  • Acute toxicity study was conducted on a Hepatitis B vaccine (Hepaccine-B-inj.) with mice, guinea pigs, and rabbits, in accordance with the norms suggested by the F.D.A. in U.S.A. Dose ranges were 2 doses/mouse, 5 doses/guinea pig, 10 doses/rabbit. They received the vaccine subcutaneously and intraperitoneally. Thereafter, all animals injected were observed of general signsdaily, and of body weight for two weeks. At the end of the observation period (or at the time of death), all animals received the highest dose group were autopsied and gross observation was made on various organs and tissues. No significant toxicity was noted.

  • In crustaceans, as in other arthropods, the molt cycle and the physiological process of growth are controlled by molting hormones (MH) which are steroid hormones, the ecdysteroids. Ecdysteroids are major arthropod hormones which control both development (embryonic and larval molts, metamorphosis) and reproduction. The purpose of the present study was to evaluate both fenarimol and methoprene for embryotoxicity to daphnids. The embryotoxicity associated with each compound was assessed to discern whether the embryotoxicity of methoprene might be due to ecdysone agonist and the ecdysone antagonistic effects of fenarimol on Daphnia embryo. Exposure of daphnids for three weeks to 50 M methoprene resulted in a significantly high incidence of offspring that exhibited general toxicity. This exposure concentration had significant effects on the overall number of embryo death. However, exposure to 3 or 1 $\mu$M fenarimol were no significant effects on the embryo toxicity. The incidence of both of these toxicity increased with methoprene exposure. This observation suggest that methoprene showed embryonic general toxicity during embryo development, while, only fenarimol showed weak general toxicity with early stages of embryonic development.
