Studies on the Content of Pueraria Radix in the Tea by HPLC (HPLC에 의한 한국산 칡차의 분석)

  • This studies was performed to investigate the quality control of Pueraria RadiI tea. Experimental subjects were 8 kinds of tea and wet and dry Pueraria RadiI which were collected from the Seoul area. For standards, Daidzein and Daidzin were isolated from Pueraria Radix. Analysis method was carried out by HPLC using ${\mu}-Bondapark{\;}C_{18}$ column and UV 254 nm detector. Samples were extracted with methanol by Cold method (at room temperature for 24 hours) and Hot extraction method (at $^{\circ}C$ for 4 hours) Pueraria radix contained 0.08% Daidzein and 0.66% Daidzin Most of the Pueraria Radix tea contained extract of Pueraria Radix less than the labled amounts listed.

Characterization of Quorum-Sensing Signaling Molecules Produced by Burkholderia cepacia G4

  • In many Gram-negative bacteria, autoinducers, such as N-acyl-L-homoserine lactone(acyl-HSL) and its derivative molecules, mediate the cell-density-dependnet expression of certain operons. The current study identified the autoinducers produced by Burkholderia cepacia G4, a trichloroethylene-degrading lagoon isolate, using TLC bioassays with Agrobacterium tumefaciens NT1(pDCI141E33) and Chromobacterium violaceum CVO26, and a GC-MS analysis. The ${R_f}\;and\;{R_t}$ values and mass spectra were compared with those of synthetic compounds. Based on the analyses, it was confirmed that G4 produces N-hexanoyl (C6)-, N-octanoyl (C8)-, N-decanoyl (C10)-, N-dodecanoyl (C12)-HSL, and an unknown active species. The integration of the GC peak areas exhibited a ratio of C8-HSL:C10-HSL:C12-HSL at 3:17:1 with C6-HSL and C10-HSL production at trace and micromolar levels, respectively, in the culture supernatants. Nutants partially defective in producing acyl-HSLs were also partially defective in the biosynthesis of an antibiotic substance. These results indicate that the autoinducer-dependent gene regulation in G4 is dissimilar to the clinical B. cepacia strains isolated from patients with cystic fibrosis.

  • In a closed square cavity filled with a liquid, a cooled the upper horizontal wall and a heated the lower horizontal wall, the flow isn't generated under the ground-based condition when Rayleigh number is lower than 1700. In such case the flow phenomena near an air bubble under a cooled horizontal wall were investigated. The temperature and the flow fields were studied by using the Thermo-sensitive Liquid-Crystal and the image processing. The qualitative analysis for the temperature and the flow fields were carried out by applying the image processing technique to the original data. Injecting bubble at the center point of upper cooled wall, the symmetry shape of two vortexes near an air bubble was observed. The bubble size increased, the size of velocity and the magnitude of velocity increased. In spite of elapsed time, a pair of two vortexes was the unique and steady-state flow in a square cavity and wasn't induce to the other flow in the surround region.

  • Hydrolysis of manno-oligosaccharides and galactomannan was studied with the purified Bacillus subtilis WL-7 mannanase from recombinant Eschericoli. The predominant products of hydrolysis were mannose, mannobiose and mannotriose. The enzyme could hydrolyze $\beta$-1 A-linked manno-oligosaccharides larger than mannobiose, but was not active on mannobiose. When the mannanase hydrolyzed manno-oligo saccharides of degree of polymerization(DP) 4-6, it was more active on the substrate of higher DP. Based on analysis of transient reaction products by TLC, the enzyme was found to have a preference for internal $\beta$-IA-mannosidic linkages, which are the central mannosidic bond of mannotetraose and the two middle mannosidic bonds of mannopentaose. The $\beta$-l A-mannosidic bonds situated at the second and fourth positions from the nonreducing end of mannohexaose were preferenhydrolyzed by the mannanase. Locust bean gum(LBG) was enzymatically hydrolyzed with higher efficiency than guar gum, resulting that amount of reducing sugars was liberated more efficiently from LBG than guar gum with same activity of mannanase.

Analysis of the Composition of Biochemical Components in Unossified Antlers (꽃사슴 녹용(鹿茸)의 생화학적성분(生化學的成分)의 조성(組成) 및 함량(含量))

  • The composition of biochemical components such as lipids, proteins and their amino acid components and inorganic elements in the ashes in unossified antlers from Cervus nippon Temminck var. mantchuricus grown in Korea were analyzed to obtain fundamental data for quality control. As a result, it was found that total lipids were 20.75% which was approximately similar contents with those of proteins (21.8%). Sixteen amino acids were identified from the hydrolysate of the protein fraction. Three gangliosides with very similar TLC patterns of those such as $GM_3$, $GM_1$ and $GM_{1a}$ were identified from the water soluble layer of Folch's partitions. Ash contents were revelaed to be much higher in the sponge layer (40.0%) than in the velvet layer (3.7%).

Solid-Phase Extraction of Curcuminoid from Turmeric Using Physical Process Method (물리적 가공법을 이용한 강황으로부터 Curcuminoid의 고체상추출)

  • In order to extract the curcuminoid such as curcumin, demethoxycurcumin (DMC), and bisdemethoxycurcumin (BDMC) in turmeric (Curcuma longa), solvent extraction methods (dipping and ultrasonic extraction method) and solid-phase extraction (SPE) were used. RP-HPLC (reverse-phase high-performance liquid chromatography) and TLC (thin-layer chromatography) were used for identification and analysis the three curcuminoid. From the experimental results, it is evident that the percentage of curcuminoid extracted from turmeric by ultrasonic extraction method was higher than dipping method. The percentage of curcumin extracted from turmeric by pure methanol was higher than any aqueous methanolic composition. Moreover, the total peak area of three curcuminoid was above 92% in RP-HPLC using solid-phase extraction. These results will form a database for investigating the constituents of natural products and the resources of pharmaceutical, nutrition, and cosmetic products.

Purification and Properties of HPS (Halitosis Prevention Substance) Isolated from Cumin (Cuminum cyminum L.) Seed

  • Halitosis is mainly caused by the presence of volatile sulfur-containing compounds (VSC's) produced by proteolytic periodontopathic bacteria in the oral cavity. Various mouth-rinses have been offered on the market as solutions to reduce halitosis. The aim of this study was to find a potent substance for the prevention of halitosis. The halitosis prevention substance (HPS) from cumin seed powder was purified by solvent extraction, silica gel column chromatography and preparative TLC to yield an oil phase (0.98%). Instrumental analysis such as FT-IR, $^1H$-NMR and $^{13}C$-NMR showed that HPS contained an -OH group, -HC=CH-, -COO-, and long chain acyl group. HPS was therefore determined to be 2-hydroxyethyl-${\beta}$-undecenate. HPS inhibited the growth of Fusobacterium nucleatum and Porphyromonas gingivalis, by 72.44% and 64.37% at $1{\times}10^{-2}\;M$, and by 99.85% and 91.62% at $5\;{\times}\;10^{-2}\;M$, respectively. It also inhibited the activity of L-methionine-${\alpha}$-deamino-${\gamma}$-mercaptomethane-lyase (METase), which was produced by oral microbes. Furthermore, the VSC production by oral microbes in the human mouth air decreased with increasing HPS concentration. These results suggested that HPS from cumin seed is an efficient halitosis prevention agent.

Analysis of the Prevalence and Risk Factors of Malnutrition among Hospitalized Patients in Busan

  • This study investigated the prevalence of and risk factors for malnutrition in hospitalized patients in Busan, Republic of Korea. 944 patients (440 men and 504 women) were hospitalized in four Busan general hospitals from March through April, 2011. Nutritional status was assessed on admission by the Nutritional Risk Screening 2002. Data were collected from the electronic medical records system for the characteristics of the subjects, clinical outcomes, biochemical laboratory data, and nutrition support states. Clinical dietitians interviewed the patients using structured questionnaires involving data on weight loss and problems related to oral intakes. Malnourished patients were significantly older (P<0.001) than well-nourished patients, but the values for BMI, serum albumin, total cholesterol, TLC, hemoglobin, and hematocrit were significantly lower (P<0.001) for malnourished than for well-nourished patients. Logistic regression indicated that the main determinant factors for nutritional status were the age, length of stay, BMI, serum albumin, and total cholesterol. In order to increase therapeutic effects of hospitalized patients, clinical dietitians need to offer proper nutritional intervention based on the results of nutrition assessment and identification of malnutrition.

Changes in Pattern of Tannin Isolated from Astringent Persimmon Fruits (떫은감에서 분리한 탄닌성분의 패턴 변화)

  • This study was investigated pattern changes of tannin isolated from astringent persimmon fruits. The contents of total phenolics and soluble tannins decreased as the maturing and softening of persimmon fruits proceeded. Green and mature persimmon tannins reacted with acetaldehyde. The more contents of tannin increased, the more reactions became. And the reaction of green Persimmon tannin was more active than mature persimmon tannins. But tannin from soft persimmon fruits did not react with acetaldehyde. Tannins were more polymerized during maturing and softening of fruits. So there was a little difference in chromatography of persimmon tannins. Also green and mature persimmon tannins obtained 4 bands respectively after thin layer chromatography analysis. But there was only 2 bands in soft persimmon tannin. As softening of persimmon fruits proceeded, most of tannins reacted with acetaldehyde, so coagulated. Also the component of soluble tannins was changed during softening of persimmon fruits.

A Study on Pigments from Rhodopila globiformis by Acetone Extraction : Stability of Red Pigments (Rhodopila globiformis 로부터 Acetone 추출한 색소에 대한 연구 : 적색색소의 안정성)

  • The acetone extracted pigment from the cell mass of Rhodophila globiformis DSM 161 was generally red. In a pH 5-6 condition, the color of pigment was red, while in p/h 7-9 condition it was yellowish red. The pigment was stable at pH range between 6.0-11.0 and below 4$0^{\circ}C$. In the presence of light and oxygen , the pigment was rapidly degraded and became unstable in the presence of metal ions such as Fe3++(1.0$\times$ 10-2M, 1.0$\times$10-3 M), Al3+ (1.0$\times$10-2 M, 1.0$\times$10-3 M) and Zn2+(1.0$\times$10-2 M). But in the presence of Zn2+ (1.0$\times$10-2M). But in the presence of Zn2+ (1.0$\times$10-3 M) it was very stable. Through visible absorption scanning ,it showed five sharp absorption peaks at 358 , 385, 494, 680 ad 748 nm with three shoulder peaks at 410, 466 and 522nm. On the results of TLC analysis , it was shown to be composed of seven color fractions.
