The aim of this study is to analyze STEAM lessons focused on the learner's learning. This study was conducted on 4th-graders in Y city, Kyung-gi province. The lessons were based on a joint teaching plan for students through the teacher learning community (TLC) with three teachers from the same school. Each of the three classes that conducted the class was selected and analyzed as the main center of observation by three students. The conclusions from this study are as follows: First, we identified that different levels of learners are learning in STEAM lessons through a learner-centered class analysis. Some students arrived on their own by taking the initiative in class, others by consulting with a group of friends, and others needed active teacher guidance to learn. Second, Depending on the level and characteristics of the students, some learning criteria were not reached. Some students need guidance at a glance level, and others need individually instructed or guided activities. Teachers need to keep an eye out for students and give them an appropriate level of guidance during class. In STEAM lessons, it appears that students of different levels and characteristics can immerse themselves in their own way, as well as the clear guidance of activity for their students.
A superoxide anion radical scavenger isolated from Capsella bursa-pastoris was characterized by infrared (IR) spectroscopy, sugar analysis, ultraviolet (UV) spectroscopy, $^{1}H$ and $^{13}C$ nuclear magnetic resonance (NMR) spectroscopies, and fast atom bombardment (FAB) mass analysis. The compound was assumed to be a flavonoid-O-glycoside from IR spectrum and UV absorption maxima. When the sugar composition of the compound was examined by thin layer chromatography (TLC) and gas chromatography (GC) of the acid hydrolysate, only glucose was detected. According to the results of UV spectrotroscopy by using shift reagents, the compound was supposed to be luteolin (5,7,3',4'-tetrahydroxy flavone) or chrysoeriol (5,7,4'-trihydroxy-3'-methoxy flavone) with glucose. Based on $^{1}H$- and $^{13}C-NMR$ spectroscopies, the compound was deduced as 7,4'-dihydroxy-5,3'-dimethoxy-${\alpha}$-6-c-glucosyl-${\beta}$-2"-o-glucosyl flavone. In FAB mass analysis the compound was finally characterized as 7,4'-dihydroxy-5,3'-dimethoxy-${\alpha}$-6-c-glucosyl-${\beta}$-2"-o-glucosyl flavone ($C_{29}H_{34}O_{16}$, M.W.=638).
Adventitious root culture was established in the Jawahar variety of Withania somnifera using MS basal medium supplemented with 0.5 (mg/l) IAA and 2.0 (mg/l) IBA. Root tips from germinated seedlings, MS0 maintained plants and adventitious roots were maintained in suspension medium (1/2 MS basal medium supplemented with 3% sucrose) for a period of 1 to 6 months. The weight gain in roots was noted and the withanolides were extracted from the dry roots using solvents petroleum ether, 50% ethanol and chloroform. The withanolides in the chloroform fractions of all root samples analyzed were compared using thin layer chromatographic analysis. Withanolide content in adventitious root sample was found to be superior compared to other roots at any given point of time during the 6month growth period.HPLC analysis of in vitro adventitious roots showed the presence of a new compound.
In this study three major curcuminoids in turmeric curcumin (1), demethoxycurcumin (2) and bismethoxycurcumin (3) were efficiently extracted by optimizing extraction condition and simultaneously identified using a fast and reliable RP-HPLC-UV-MS and TLC method. The analysis by the $C_{18}$ column was performed and the UV wavelength was fixed at 425 nm. In this result, the total extraction yield of turmeric (Curcuma longa) was increased with extraction time from 1 to 7 h. So, optimum extraction time is 4 h. Also, the highest yield of extraction amount 0.433g 8.66% was obtained by ultrasonic waves with quarter frequency kHz and an extraction time of 7 h. The experiment method was consistent with theoretical Value $r^2$ = 0.987 (1), 0.997 (2) and 0.998 (3). Moreover, LC-MS analysis provided efficiently molecular weight information of three major curcuminoids in turmeric extracts and high purity (~95%) of the curcuminoids were obtained. This work offers would be useful for chemical and biological studies of natural plants and its products.
Pseudomonas sp. NGK1, isolated by naphthalene enrichment culture technique, is capable of degrading anthracene as a sole source of carbon and energy. The organism degraded anthracene through the intermediate formation of 1,2-dihydroxyanthracene, 2-hydroxy-3-naphthoic acid, salicylate, and catechol. The intermediates were isolated and characterized by TLC, spectrophotometry, and HPLC analysis. The cell free extract of anthracene-grown cells showed activities of anthracene dioxygenase, 2-hydroxy-3-naphthylaldehyde dehydrogenae, 2-hydroxy-3-naphthoate hydroxylase, salicylate hydroxylase and catechol 2,3-dioxygenase. The formed catechol as a metabolite is degraded through meta-cleavage with the formation of ${\alpha}$-hydroxymuconic semi-aldehyde.
New simple methods for the Isolation of sesquiterpenes from Panax ginseng were developed. First, volatile compounds were isolated by simultaneous distillation and extraction (SDE) with 30% methanol and $\alpha$-hexane instead of water and ethyl ether/pentane (1:1). Secondly, head space volatiles in U-shaped tube at 7$0^{\circ}C$ were passed through C18 Sep-Pak by nitrogen gas streaming and the adsorbed volatiles were fluted by $\alpha$-hexane. TLC analysis showed that the volatile concentrates consisted mainly of terpenes when colored by vanillin-sulfuric and. GC/MS data revealed that approximately 30 sesquiterpenes of molecular weight 204 occupied 81.1% or more of the volatile concentrates isolated by those two newly developed methods. This study was carried out to establish a new efficient method for isolation and purification of ginsenosides. Silica gel column chromatography, having been used for the isolation of ginsenosides, is advantageous to obtain a large amount of ginsenosides. However, it has a disadvantage to isolate ginsenosides to their highest purity. In addition, normal-or reverse-phase HPLC method thus far reported is confined to quantitative analysis. Especially, it has not been possible to isolate racemic 20(S)- and 20(R)-ginsenoside Rg2. In this experiment, isolation and purification of ginsenosides were accomplished by Diaion HP-20 adsorption chromatography, silica gel column chromatography, recrystalization and Prep. HPLC with or without Prep. TLC. From this study, we could establish a new efficient method for isolation and purification of 9 major and/or minor ginsenosides.
An agarase was partially purified from the culture broth of marine bacterium Bacillus cereus ASK202. Optimal pH and temperature of this agarase were found to be 7.0 and 40$^{\circ}C$, respectively. The maximum productivity of agarooligosaccharides was obtained from 0.3 %(w/v) agar by using of 1 unit agarase. As the results of TLC and HPLC analysis, these oilgosaccharides consisted of neoagarobiose, neoagarotetraose and neoagarohexaose. Under the optimal reaction conditions, 77.5 %(w/v) neoagarobiose and 6.2 %(w/v) neoagarotetraose were produced from agar and the conversion yield of total agarooligosaccharides was 83.7 %(w/v) after for 2 h reaction at 40$^{\circ}C$.
Sikhye is a traditional sweet rice drink in Korea by $\beta$-amylase's saccharifing action. Sikhye has great potentiality for commercial beverage because of It's characteristic taste and flavors. But, the chemical compositions and production methods of Sikhye are little known. The total amount of carbohydrate of sixteen Sikhye selected in Korean market was 11~15%. The reducing sugar by the Somogyi-Nelson's method was measured 0.4~2% as maltose, which has little influence on the sweetness of Sikhye. While sucrose content was about 10% by the TLC and Seliwanoff analysis. It means that carbohydrate in Sikhye was almost sucrose. Furthermore the most of Sikhye has brown color and turbidity. To improve the quality of Korean Sikhye, it is necessary to increase the production of maltose and to reduce brown color and turbidity, without adding sucrose.
Three medicinal species of the genus Polygala were examined by comparative morphology, anatomy and TLC analysis. They were classified into three species, and distinctly divided into two groups in this study. Group I is composed of P. japonica, P. sibirica and Group II P. tenuifolia. Considered on the relationships between two groups by the differences of leaf shape, adnated part of petal, flower colour, fruit shape etc., Group II may have been independently evolved from the common ancestor by having one raw of palisadelike chlorenchyma under the stem epidermis through the different pathway. It is also suggested that the taxa of Group II appear to be more advanced than those of Group I.
