Seasonal Distribution of T Serotyping and emm Genotyping of Group A Streptococci Obtained from Children with Streptococcal Infections in Masan, Korea, 2003~2004 (2003~2004년 경남 마산 지역에서 분리된 A군 연쇄구균의 T 혈청형과 emm 유전자형의 계절별 분포)

  • Purpose : The aim of this study is to know seasonal distribution of group A streptococci obtained from one center using emm genotyping and T serotyping in Masan from 2003 through 2004. Methods : Among children who visited the Changwon Fatima Hospital at Masan, Korea from June 2003 through February 2004, 100 patients who had clinical findings of acute pharyngitis, scarlet fever, and cellulitis were confirmed as GAS by culture, and were enrolled in our study. All obtained GAS were sent to the WHO Collaborative Center for Reference and Research on Streptococci, University of Minnesota, Minneapolis for T serotyping and emm genotyping. We classified these results again according to seasonal and disease's entities. Results : 19 different T serotypes was typed. T4(27.5%), T1(17.6%), T6(13.7%), and T12(13.7%) serotypes were relatively common in summer, while T4(28.3%), T12(15.2%), and T12/B3264(8.7%) were common in winter. T4 and T12 were persistent all year around. Distribution of T serotypes in 89 patients with pharyngotonsillitis were T4(26.7%), T12(14.0%), T1(12.8%), and T6(11.6%) in order of frequency. 15 different emm genotypes was typed. The number of emm 1, emm 6, emm 9, and emm 44 genotypes decreased or disappeared in winter, and the number of emm 3, emm 12, and emm 89 genotypes increased or reappeared in winter. Conclusion : Because T serotyping and emm genotyping are useful tools for evaluating epidemiology and pathogenesis of group A streptococci, we should monitor these strains every year, and should serotype and genotype GAS obtained from the invasive streptococcal infections.

  • This pot experiment was conducted in order to find out the effects of application of combined micronutrients(T$_1$: control. T$_2$; Fe, T$_3$; Fe+Mn, T$_4$; Fe+Mn+Cu, T$_{5}$; Fe+Mn+Cu+Zn, T$_{6}$;Fe+Mn+Cu+Zn+Mo, T$_{7}$; Fe+Mn+Cu+Zn+Mo+B) on forage performance of pure and mixed cultures of orchardgrass and white clover The 2nd part was concerned with the changes in the forage yields and concurrence index. The results obtained are summarized as follows: 1. The effects of combined micronutrient applications on the forage yields were different according to the forage species, whether it was a pure or mixed cultures, and additional fertilization(especially N). The effects of them on the forage productivity and botanical composition were more obvious in white clover, especially in mixed culture, than in orchardgrass. By the significant role of B as a regulator, the yields of both forages were best in the T$_{7}$, respectively. 2. In the pure culture, the high yields of both forages were obtained by the T$_{7}$ and T$_2$, whereas the T$_{6}$ and T$_3$resulted in the low yields. The best yields of both forages were obtained by the T$_{7}$ with relatively optimum ratios among the micronutrients as follows; Fe/Mn/Cu/Zn, Fe/Mo, Mo/B, and ∑ cation/∑anion. It was observed the multiple interaction of Fe${\times}$Mn${\times}$Mo${\times}$B, and the significant role of B as a regulator. The effects of them on white clover were more distinct at no additional fertilization than at the additional fertilization(especially N). 3. In mixed culture, the optimum applications of them resulted in the positive increase of yield and botanical composition of white clover, whereas orchardgrass tended to be inversely except the T$_{7}$.X> 7/. 7/.

  • To develop Grifola frondosa as a new commercial product, we investigated the optimal primordium formation method for bottle cultivation. Primordium formation was initiated on the sixth day in all treatments, but in the T7 treatment, which consisted of removing old spawn and turning the bottle upside-down, was initiated on the ninth day. The ratio of primordium formation was at its highest value, 98.9%, in T3 and T7. Among the treatments, T7 produced the highest quality fruiting bodies and a mushroom yield of 112.8 g in each 850 mL bottle, but its crop cycle time was somewhat longer than those of the other treatments. These results indicate that appropriate methods for primordium formation in bottle culture could enhance mushroom quality and productivity.

  • This study was conducted to evaluate effect of liquid fertilizer contained medium of Lactobacillus confusa and Pichia anoala on growth of creeping bentgrass(Agrostis palustris Huds. CV. Pennlixs). By application method of liquid fertilizer, the experiment plot was designed as following; NF was non-fertilized treatment, control was applied with compound fertilizer and treatment 1(T-1), treatment 2(T-2) and treatment 3(T-3) were applied with compound fertilizer and liquid fertilizer solution diluted 500, 300 and 100 folds, respectively. The every treatments was arranged a randomized complete block with three replications. Compared with NF, leaf color index of control, T-1, T-2 and T-3 in creeping bentgrass was increased 7.4%, 7.5%, 77% and 7.2%, respectively and chlorophyll content of T-1, T-2 and T-3 in creeping bentgrass was increased 45.7%, 45.6%, 52.1% and 49.6%, respectively, and T-2 and T-3 was increased 4.4% and 2.7% more than control. Dry weight of T-1, T-2 and T-3 was increased 2.7%, 13.8% and 13.9% more than control, when compared to control. These results were found that turfgrass qualities and growth was improved in treatment applied to compound fertilizer and solution of liquid fertilizer diluted 300 folds.

Cloning and Expression of K11 Phage RNA Polymerase (K11 RNA 중합효소의 Cloning 및 발현)

  • Using the PCR(polymerase chain reaction method), gone 1 of phage K11 coding for K11 phage RNA polymerase has been cloned and expressed under the control of lac promoter. K11 phage RNA polymerase was conventionally purified through the DEAE-sephacel and Affigel blue column chromatographies. The 0.2-0.3 M $NH_4Cl$ fractions of DAEA-sephacel column chromatography showed K11 phage RNA polymerase activity and further purification with Affigel blue column chromatography showed nearly single protein band on SDS-polyacryl amide gel. K11 phage RNA polymerase, which is one of the T7 group phage RNA polymerase (E. coil phage T7, T3 and Salmonella tyhimurium phage SP6 RNA polymerase), shares high degrees of homology with the other T7 group phage RNA polymerase. Previously we constructed T7 and SP6 promoter variants and revealed promoter specificity of T7 and SP6 RNA polymerase (Lee and Kang, 1993). To investigate the promoter specificity of K11 RNA polymerase in vitro K11 promoter activity was measured with SP6 promoter variants. The SP6 promoter variant share highest degrees of sequence homology with K11 promoter sequence show strongest promoter activity.

Effects of Water Temperature on Egg Development, Hatching and Laval Growth Rearing of the Pacific Cod Gadus macrocephalus (태평양 대구 Gadus macrocephalus 채란, 부화 및 자어성장에 미치는 수온의 영향)

  • We investigated egg collection and the effects of water temperature (4, 7, 10, 13 and $16^{\circ}C$) on egg development, hatching and larval growth of Pacific cod Gadus macrocephalus under laboratory conditions. Fertilized eggs were round in shape ($1.01{\pm}0.03\;cm$) and adhesive to nylon nets. Fertilization rate was 68% by wet method. The time of egg development was negatively proportional to water temperature with the range of $4^{\circ}C$ to $13^{\circ}C$. Eggs hatched only at $7^{\circ}C$ after 288 hours of fertilization and $10^{\circ}C$ after 192 hours. Hatching rate was highest as 65% at $7^{\circ}C$ followed by $34.4^{\circ}C$ at $10^{\circ}C$. Survival rate was 18.3% at $7^{\circ}C$ and 5.2% at $10^{\circ}C$.

CCR7 Ligands Induced Expansion of Memory CD4+ T Cells and Protection from Viral Infection (CCR7 Ligand의 Memory CD4+ T 세포 증가유도 및 바이러스 감염에 대한 방어효과)

  • Background: CC chemokine receptor (CCR) 7 and cognate CCR7 ligands, CCL21 (formerly secondary lymphoid tissue chemokine [SLC]) and CCL19 (formerly Epstein-Barr virus-induced molecule 1 ligand chemokine [ELC]), were known to establish microenvironment for the initiation of immune responses in secondary lymphoid tissue. As described previously, coadministration of DNA vaccine with CCR7 ligand-encoding plasmid DNA elicited enhanced humoral and cellular immunity via increasing the number of dendritic cells (DC) in secondary lymphoid tissue. The author hypothesized here that CCR7 ligand DNA could effectively expand memory CD4+ T cells to protect from viral infection likely via increasing DC number. Methods: To evaluate the effect of CCR7 ligand DNA on the expansion of memory CD4+ T cells, DO11.10.BALB/c transgenic (Tg)-mice, which have highly frequent ovalbumin $(OVA)_{323-339}$ peptide-specific CD4+ T cells, were used. Tg-mice were previously injected with CCR7 ligand DNA, then immunized with $OVA_{323-339}$ peptide plus complete Freund's adjuvant. Subsequently, memory CD4+ T cells in peripheral blood lymphocytes (PBL) were analyzed by FACS analysis for memory phenotype ($CD44^{high}$ and CD62 $L^{low}$) at memory stage. Memory CD4+ T cells recruited into inflammatory site induced with OVA-expressing virus were also analyzed. Finally, the protective efficacy against viral infection was evaluated. Results: CCR7 ligand DNA-treated Tg-mice showed more expanded $CD44^{high}$ memory CD4+ T cells in PBL than control vector-treated animals. The increased number of memory CD4+ T cells recruited into inflammatory site was also observed in CCR7 ligand DNA-treated Tg-mice. Such effectively expanded memory CD4+ T cell population increased the protective immunity against virulent viral infection. Conclusion: These results document that CCR7 and its cognate ligands play an important role in intracellular infection through establishing optimal memory T cell. Moreover, CCR7 ligand could be useful as modulator in DNA vaccination against viral infection as well as cancer.

The Use of Pedicle Screw-Rod System for the Posterior Fixation in Cervico-Thoracic Junction

  • Objective : In cervico-thoracic junction (CTJ), the use of strong fixation device such as pedicle screw-rod system is often required. Purpose of this study is to analyze the anatomical features of C7 and T1 pedicles related to screw insertion and to evaluate the safety of pedicle screw insertion at these levels. Methods : Nineteen patients underwent posterior CTJ fixation with C7 and/or T1 included in fixation levels. Seventeen patients had tumorous conditions and two with post-laminectomy kyphosis. The anatomical features were analyzed for C7 and T1 pedicles in 19 patients using computerized tomography (CT). Pedicle screw and rod fixation system was used in 16 patients. Pedicle violation by screws was evaluated with postoperative CT scan. Results : The mean values of the width, height, stable depth, safety angle, transverse angle, and sagittal angle of C7 pedicles were $6.9{\pm}1.34\;mm$, $8.23{\pm}1.18\;mm$, $30.93{\pm}4.65\;mm$, $26.42{\pm}7.91$ degrees, $25.9{\pm}4.83$ degrees, and $10.6{\pm}3.39$ degrees. At T1 pedicles, anatomic parameters were similar to those of C7. The pedicle violation revealed that 64.1% showed grade I violation and 35.9% showed grade II violation, overall. As for C7 pedicle screw insertion, grade I was 61.5% and grade II 38.5%. At T1 level, grade I was 65.0% and grade II 35.0%. There was no significant difference in violation rate between the whole group, C7, and T1 group. Conclusion : C7 pedicles can withstand pedicle screw insertion. C7 pedicle and T1 pedicle are anatomically very similar. With the use of adequate fluoroscopic oblique view, pedicle screw can be safely inserted at C7 and T1 levels.

Changes in the Physicochemical Properties of Ground Pork Meat Containing Persimmon Peel during Refrigerated Storage (감 껍질을 함유한 분쇄 돈육의 냉장 저장 중 이화학적 특성 변화)

  • This study was carried out to investigate the effect of persimmon peel on the physicochemical properties of ground pork stored at $4^{\circ}C$ for 10 days. Four types of ground pork were evaluated: T0 without dried persimmon peel powder, and T1 with 0.3%, T2 with 0.7%, and T3 with 1.0% dried persimmon peel powder. The pH increased during storage, with the pH of T3 being the lowest (p<0.01). The L-value and b-value were not significantly changed, but the a-value decreased during storage. On the 10th day of storage, the a-values were significantly higher for T2 and T3 than for T0 and T1 (p<0.001). TBARS significantly increased during storage, with lower values for T2 and T3 than for T0 and T1 (p<0.001). DPPH free radical scavenging activity decreased during storage, with T0 having the lowest value (p<0.001). The VBN content increased during storage, and the VBN content of T0 was the highest at the 10th day (p<0.05). The water-holding capacity decreased and cooking loss increased during storage. Hardness and chewiness increased, while springiness and gumminess decreased during storage. The results of this study showed that the addition of persimmon peel during the process of making ground pork had antioxidant effects that maintained redness and physical quality. A 0.7% addition was the most appropriate.

Spreading Pattern of Epidurally-Administered Contrast Media in Rabbits (토끼에서 경막외강으로 투여한 조영제의 분포양상)

  • Background: The aim of this study was to examine the precise spreading pattern of contrast media in small increments in rabbits. Following pentobarbital anesthesia, the epidural puncture was done surgically with a blunt hook. Methods: The tip of epidural catheter was located at the mid-portion of T7 and T12, in the T7 group (n=7) and T12 group (n=8), respectively. Injection of the contrast media was started at 0.1 mL/kg and increased by 0.1 mL/kg up to a maximum of 0.6, mL/kg, under fluoroscopy. Results: In both groups, the extent of spread increased continuously as a Starling resistor with increasing injected volume(T7 group: $y=4.0+41.8x-28.1x^2$, T12 group: $y=0.2+57.7x-43.5x^2$) the total spread of contrast media was similar. The contrast media spread equally, both rostral and caudal, from catheter tip in T7 group; media spread approximately twice as far rostral as compared to caudal in T12 group (P<0.05). Conclusions: In rabbits, the position of epidural catheter tip should be positioned 2~3 segments below the aimed segment in lower thoracic or lumbar region, whereas in mid-thoracic region it should be positioned close to the level of aimed segment. Rabbits have relatively small epidural space therefore, the volume of injectant should be carefully determined with the suggested equations of this study.
