In order to make an efficient prescription and cope with dementia, learning and memory functions of Sprague-Dawley model rats were tested with Morris water maze. And to evaluate the effect of the sample drug(CHM) on choline acetyltranferase and acetylcholine esterase, immunoreactive measurement and enzymatic activity measuring were carried out. Rats were injected with ibotenic acid through hippocampus CA1 and CA3 area. The results are as following. CHM improves the learning ability in the acquisition test and memory function in the retention test significantly. And CHM increases the level of AChE which is resolving acetylcholine. Though it doesn't increase the level of ChAT significantly which is synthesizing acetylcholine, but it shows the tendency of increase. So these results show that CHM improve the cholinergic catabolism and anabolism, and the increment of metabolic activity of cholinergic system. Thus it can be concluded that CHM will be helpful to cholinergic brain disease induced by primary or senile reduction of acetylcholine secretive activity.
Prenatal nicotine exposure over an entire pregnancy has been associated with an increased prevalence of hyperactivity, anxiety-like behavior and depression-like behavior in mature rats. However, the effects of maternal nicotine exposure in late gestation and lactation on the psychology and behavior of adolescent rat offspring are unclear. Thus, we investigated the effect of nicotine exposure during late gestation and lactation on anxiety-like and impulsive decision-making behavior in adolescent offspring of rat. Female rats were orally exposed to nicotine which is within range of plasma level of human chronic smokers during the period of third last period of gestation and lactation. When the offspring were weaned, we observed alterations in the anxiety-like behavior and decision-making ability of adolescent rat offspring using light/dark box test and T-maze delay-based cost-benefit decision-making task. The maternal consumption of nicotine reduced both the time spent in the light compartment and the number of transitions compared to nicotine-free rats. Moreover, such nicotine exposed adolescent offspring rats showed impulsive decision making which chose the instant reward in a decision-making situation. We found that nicotine exposure during late gestation and lactation induces an increase in anxiety-like and impulsive decision-making behavior at this developmental stage. These findings suggest that maternal nicotine-exposed offspring are at an increased risk of developing anxious and impulsive behavior.

Propofol is an anesthetic agent that gained wide use because of its fast induction of anesthesia and rapid recovery post-anesthesia. However, previous studies have reported immediate neurodegeneration and long-term impairment in spatial learning and memory from repeated neonatal propofol administration in animals. Yet, none of those studies has explored the sex-specific long-term physical changes and behavioral alterations such as social (sociability and social preference), emotional (anxiety), and other cognitive functions (spatial working, recognition, and avoidance memory) after neonatal propofol treatment. Seven-day-old Wistar-Kyoto (WKY) rats underwent repeated daily intraperitoneal injections of propofol or normal saline for 7 days. Starting fourth week of age and onwards, rats were subjected to behavior tests including open-field, elevated-plus-maze, Y-maze, 3-chamber social interaction, novel-object-recognition, passive-avoidance, and rotarod. Rats were sacrificed at 9 weeks and hippocampal protein expressions were analyzed by Western blot. Results revealed long-term body weight gain alterations in the growing rats and sex-specific impairments in spatial (female) and recognition (male) learning and memory paradigms. A markedly decreased expression of hippocampal NMDA receptor GluN1 subunit in female- and increased expression of AMPA GluR1 subunit protein expression in male rats were also found. Other aspects of behaviors such as locomotor activity and coordination, anxiety, sociability, social preference and avoidance learning and memory were not generally affected. These results suggest that neonatal repeated propofol administration disrupts normal growth and some aspects of neurodevelopment in rats in a sex-specific manner.
본 연구의 목적은 조기영어학습 경험의 유형에 따른 실행기능의 차이를 분석하는 것이다. 연구대상은 어학원에서 몰입식 영어교육 경험을 가진 23명의 유아, 어린이집에서 방과후 영어학습 경험을 가진 52명의 유아, 총 75명을 대상으로 삼았다. 연구도구는 주의통제의 경우 스트룹 과제, 인지적 융통성의 경우 카드분류과제와 숫자 따라 외우기 소검사, 정보처리의 경우 도안 유창성, 목표설정의 경우 미로 소검사를 사용하여 측정하였다. 자료분석은 전체지능지수를 공변인으로 한 공분산분석을 적용하였다. 연구결과 첫째, 주의통제와 인지적 융통성은 조기영어학습의 유형에 따라 유의한 차이를 보였다. 둘째, 정보처리와 목표설정은 조기영어학습의 유형에 따라 어떠한 차이를 보이지 않았다. 본 연구는 전일제 조기영어학습의 경험이 주의통제와 인지적 융통성에는 긍정적인 영향을 미치는 반면, 정보처리와 목표설정에는 어떠한 영향도 미치지 않음을 시사한다.
Objectives: The object of this study is to observe the cognition and motor function recovery effects of Joojakwhan (JJW), a traditional Korean poly-herbal formula for treating various neuropsychiatric diseases such as dementia, for the mildly stroke rats, with 60 minutes of reperfusion transient middle cerebral artery occlusion (tMCAO). Methods: In the present study, 125, 250 and 500 mg/kg of JJW were orally administered, once per day for 10 continuous days 2 hours after the tMCAO. The body weight changes, infarct sizes under 2% 2, 3, 5-triphenyl tetrazolium chloride (TTC) stain, sensorimotor functions and cognitive motor behavior tests were serially monitored with cerebral caspase-3 and cleaved poly (ADP-ribose) polymerase (PARP)-immunoreactivities and histopathological changes. The effects of tMCAO on sensorimotor functions were evaluated by using of limb placing and body-swing tests, and the cognitive motor behaviors were also observed with water maze tests. Results: From the results of tMCAO, with marked decreases of body weights, disorders of sensorimotor functions increases the limb placing test scores, and decrease the numbers and percentages of body swings to the ipsilateral sides. The cognitive motor behaviors increases the distances and time to reach the escape platform which included the inhibitions of the decreases with repeated trials that were observed with focal cerebral cortex infarct volumes. In addition, the marked increases of the atrophy, numbers of degeneration, caspase-3- and PARP-immunoreactive cells around peri-infarct ipsilateral cerebral cortex were also observed in tMCAO controls when compared with the sham control rats, respectively. Conclusions: The results obtained from this study suggest that oral administrations of JJW indicate obvious cognitions and motor function recoveries of the rats with tMCAO, mild strokes, which are mediated by neuro-protective effects through known antioxidant effects of components.

Background: As a complication of Type II Diabetes Mellitus (T2DM), the etiology, pathogenesis, and treatment of cognitive dysfunction are still undefined. Recent studies demonstrated that Ginsenoside Rg1 (Rg1) has promising neuroprotective properties, but the effect and mechanism in diabetes-associated cognitive dysfunction (DACD) deserve further investigation. Methods: After establishing the T2DM model with a high-fat diet and STZ intraperitoneal injection, Rg1 was given for 8 weeks. The behavior alterations and neuronal lesions were judged using the open field test (OFT) and Morris water maze (MWM), as well as HE and Nissl staining. The protein or mRNA changes of NOX2, p-PLC, TRPC6, CN, NFAT1, APP, BACE1, NCSTN, and Ab1-42 were investigated by immunoblot, immunofluorescence or qPCR. Commercial kits were used to evaluate the levels of IP3, DAG, and calcium ion (Ca2+) in brain tissues. Results: Rg1 therapy improved memory impairment and neuronal injury, decreased ROS, IP3, and DAG levels to revert Ca2+ overload, downregulated the expressions of p-PLC, TRPC6, CN, and NFAT1 nuclear translocation, and alleviated Aβ deposition in T2DM mice. In addition, Rg1 therapy elevated the expression of PSD95 and SYN in T2DM mice, which in turn improved synaptic dysfunction. Conclusions: Rg1 therapy may improve neuronal injury and DACD via mediating PLC-CN-NFAT1 signal pathway to reduce Aβ generation in T2DM mice.
심방세동은 가장 흔한 부정맥 질환으로서,특히 승모판막 질환이 있는 경우에는 60%에 이르는높은 빈도의 유병율을 보이는데,심방세동 환자의 약 113에서 혈전 색전증을 일으키고,혈전 색전증이 생긴 환자의 약 60%에서는 사망에 이르거나 심각한 합병증이 초래되므로 심장질환에 대한 수술시,동반 심 방세동에 대한 적극적 인 치료가 고려되어야 한다. 서울대학교병원 흉부외과학교실에서는 1994년 4월부터 1995년 6월까지 심방세동을 동반한 심장질환 을 가진 20명의 환자에서 Maze 술식을 포함한 개심술을 시행하였다 대상환자들의 남녀 성비는 남 '녀 : 6 : 14 이 었으며, 평균연령은 48$\pm$11세 (31 ~66세) 였다. 1년이상 지 속된 만성 심방세동이 14례(70%)였고, 1년미만인 경우가 6례(30%)였으며,심방세동의 과거력은 평균 36$\pm$42개월(1~132개월)이었다. 수술전 혈전전색증의 과거력이 있었던 경우가 7례(35%), 좌심방내에 혈전이 있었던 경우가 9례(45%)였다. 동반 심장질환으로는 판막질환이 19례,심실중격결손증이 1례였 으며, Maze술식과 더불어 승모판막 및 대동맥판막 치환술이 5례, 승모판막 치환술을 시행한 경우가 4 fl, 승모판막 치환술 및 삼첨판\ulcorner 성형술 4례, 승모판막 성형술 3례, 승모판막 성형술 및 삼첨판막 성형 술이 1례, 승모판막 치환술 및 관상동맥 우회술이 1례, 대동맥판막 치환술 1례, 심실중격결손봉합술이 1례 였다. 대동맥차단시간은 평균 175 :41분(116~270분)이었다. 수술과 관련된 사망은 없었으며, 수술 후 심방세동의 재발이 16명(80%)에서 있었으나, 수술후 평균 41일째 규칙적인 심박동 소견을 보였다. 수술후 합병증으로서는 저심박출증을 보였던 경우가 3례 (15%), 술전 존재하였던 반신불수의 악화가 1 례, 그리고 급성 신부전이 1례씩 관찰되었다. 20명의 외래추적 관찰기간은 평균 16.5개월(10.5~24개월) 이었는데, 외래 추적기간 중 모든 환자에서 규칙적인 심박동의 소견을 보였으며, 정상 동방결절리듬을 보인 경우 17례 (85%)중에서 항부정맥제의 투여가 필요 없는 경우가 13례 (76%)이고 나머지 4례에서는 항부정맥제를 투여중이며, 접합부 율동을 보이는 3례 (15%) 중 2례에서는 항부정맥제를 투.i중이고, 1 례는 접합부 서맥으로 인공심박동기 (DDD-R type : AAI mode)의 삽입이 필요했다. 추적기간 중에 심에 코검사는 19명에서 시행하였는데, 우심방 수축력이 보이는 경우가 1 례 (95%) 이었고, 좌심방 수축력은 12례 (63%)에서 명백히 관찰되 었다. 심장질환에 대한 개심술시 Maze술식을 동반시행할 경우심장허혈시간이 길어지는 단점이 있으나, 최근의 발달된 심근보호법의 적용으로 수술에 따른 위험을 최소화할 수 있으므로 심방세동의 적극적 인 치료를 고려하여야 할 것으로 사료된다.
본 연구는 뉴로피드백 훈련이 유아의 뇌 기능과 지능에 미치는 영향에 관한 연구로 첫째, 뉴로피드백 훈련이 유아의 뇌 기능에 어떤 변화를 주는지를 검증하였으며, 둘째, 한국 웩슬러유아지능검사(K-WPPSI: Korean Wechsler Preschool and Primary Scale of Intelligence)를 통해 뉴로피드백 훈련이 유아의 지능 발달에 어떤 영향을 미치는지를 분석하였다. 본 연구는 천안 소재의 J 유치원에 재원중인 만5세 유아 60명을 대상으로, 2008년 4월 21일부터 12월 12일에 걸쳐 뉴로피드백 훈련 지침에 따라 훈련을 실시하였다. 뉴로피드백 훈련 전과 후의 뇌기능지수와 지능을 분석하였으며, 통계분석은 SPSS for Window 13.0 패키지를 사용하여 t-검증을 통해 분석하였다. 분석 결과 첫째, 뇌기능 지수 전체를 종합 평가한 브레인지수는 유의미한 영향을 미치는 것으로 나타나, 뉴로피드백 훈련이 유아의 뇌 기능 발달에 매우 효과적인 방법임을 알 수 있었다. 둘째, 뉴로피드백 훈련이 동작성 지능 지수 중 도형 영역, 미로 영역, 빠진 곳 찾기 영역은 유의미한 영향을 미치는 것으로 나타났으며, 종합적인 평가 지수인 동작성 지능 지수도 유의미한 영향을 미치는 것으로 나타났다. 따라서 이러한 실험 결과는 뉴로피드백 훈련이 동작성 지능 지수 발달에 매우 효과적인 방법임을 알 수 있었고, 세째, 뉴로피드백 훈련은 언어성 지능 지수 중 공통성 영역만 유의미한 영향을 미치는 것으로 나타났다.
This research was done to make the effective prescription and cope with various senile dementia. So Sprague-Dawley rats were injected with ibotenate to make a damage on learning and memory functions. At first acquisition test and retention rest were done in the Morris water maze. And to evaluate the effects of the sample drug(TBM) on choline acetyltranferase and acetylcholine esterase, immunoreactive measurement and enzymatic activity measuring were carried out. The ibotenic acid were injected to hippocampus CA1 and CA3 area. The results were as following. TBM improved the learning ability in the acquisition test and memory function in the retention test significantly. And TBM increased the level of ChAT which is synthesizing acetylcholine in CA3 area, and at the same time it increased the level of AChE which is resolving acetylcholine. These results show that T8M improved the cholinergic catabolism and anabolism, and the increment of metabolic activity of cholinergic system. In other words, it contributes to the recovery of damaged learning and memory function by ibotenic acid. So it can be concluded that TBM will be helpful to cholinergic brain damage induced by primary or senile reduction of acetylcholine secretive activity.
우리는 도망자-추적자 게임 풀이에 관심이 많다. 도망자가 추적자를 피해 미로를 탈출하는 게임이다. 도망자가 추적자를 유인하기도 하고 벽을 이용해 교묘히 피하기도 한다. 경험에 의하면 수작업으로 풀기에는 매우 어려운 단계들도 있었다. 게임을 풀기 위한 방법으로 모델 체킹 기법을 사용하였다. 모델 체킹은 게임의 모든 상태 공간을 넓이 우선 방식으로 철저하게 탐색하기 때문에 게임을 풀 수 있는 가장 짧은 경로인 최적의 답을 구할 수 있다. 다행히 풀이 과정에서 상태 폭발 문제는 일어나지 않았고, 게임 풀이 결과를 임베디드 시스템인 레고 마인드스톰에 응용하였다. 도망자, 추적자에 해당하는 두 대의 에이전트를 만들어 게임을 구현하고 실험하여 풀이가 정확한 지를 실제 확인할 수 있었다.
