Comparison of Mutant Frequencies Induced by ${\gamma}$-radiation and Pentachlorophenol at hprt Locus in Human T-lymphocytes(I) (인체 T-림프구 hprt 유전자에서 방사선 및 pentachlorophenol에 의한 돌연변이 빈도의 비교(I))

  • In vitro somatic mutation induced by ${\gamma}$-radiation and pentachlorophenol(PCP) which is representative of chemical pollutant was measured at the hypoxanthine-guanine phosphoribosyl transferase(hprt) locus in human T- lymphocytes by a cell cloning assay. Mutant cells were selected by their ability to form a clone in the presence of purine analogue 6-thioguanine. The mutant frequencies by ${\gamma}$-irradiation to a dose of 1.0 Gy, 2.0 Gy and 3.0 Gy were 40%, 400% and 750% higher than those in controls. Significant changes were not observed in mutant frequencies in the 0.2 Gy and 0.5 Gy irradiated groups. When the doses of PCP were 15 ppm, 25 ppm and 50 ppm, the mutant frequencies increased by 30%, 90% and 520%, respectively. No changes were observed in the 10 ppm treated group. Similar types of dose-response relationship were shown in the two different mutagens. Reverse transcriptase/polymerase chain reaction technique(RT/PCR) was needed for the mutation spectrum to discriminate combined exposure to radiation and chemicals.

PCR for Diagnosis of Male Trichomonas vaginalis Infection with Chronic Prostatitis and Urethritis

  • The aim of this study was to assess the usefulness of PCR for diagnosis of Trichomonas vaginalis infection among male patients with chronic recurrent prostatitis and urethritis. Between June 2001 and December 2003, a total of 33 patients visited the Department of Urology, Hanyang University Guri Hospital and were examined for T. vaginalis infection by PCR and culture in TYM medium. For the PCR, we used primers based on a repetitive sequence cloned from T. vaginalis (TV-E650). Voided bladder urine (VB1 and VB3) was sampled from 33 men with symptoms of lower urinary tract infection (urethral charge, residual urine sensation, and frequency). Culture failed to detect any T. vaginalis infection whereas PCR identified 7 cases of trichomoniasis (21.2%). Five of the 7 cases had been diagnosed with prostatitis and 2 with urethritis. PCR for the 5 prostatitis cases yielded a positive 330 bp band from bothVB1 and VB3, whereas positive results were only obtained from VB1 for the 2 urethritis patients. We showed that the PCR method could detect T. vaginalis when there was only 1 T. vaginalis cell per PCR mixture. Our results strongly support the usefulness of PCR on urine samples for detecting T. vaginalis in chronic prostatitis and urethritis patients.

Draft Genome of Toxocara canis, a Pathogen Responsible for Visceral Larva Migrans

  • This study aimed at constructing a draft genome of the adult female worm Toxocara canis using next-generation sequencing (NGS) and de novo assembly, as well as to find new genes after annotation using functional genomics tools. Using an NGS machine, we produced DNA read data of T. canis. The de novo assembly of the read data was performed using SOAPdenovo. RNA read data were assembled using Trinity. Structural annotation, homology search, functional annotation, classification of protein domains, and KEGG pathway analysis were carried out. Besides them, recently developed tools such as MAKER, PASA, Evidence Modeler, and Blast2GO were used. The scaffold DNA was obtained, the N50 was 108,950 bp, and the overall length was 341,776,187 bp. The N50 of the transcriptome was 940 bp, and its length was 53,046,952 bp. The GC content of the entire genome was 39.3%. The total number of genes was 20,178, and the total number of protein sequences was 22,358. Of the 22,358 protein sequences, 4,992 were newly observed in T. canis. Following proteins previously unknown were found: E3 ubiquitin-protein ligase cbl-b and antigen T-cell receptor, zeta chain for T-cell and B-cell regulation; endoprotease bli-4 for cuticle metabolism; mucin 12Ea and polymorphic mucin variant C6/1/40r2.1 for mucin production; tropomodulin-family protein and ryanodine receptor calcium release channels for muscle movement. We were able to find new hypothetical polypeptides sequences unique to T. canis, and the findings of this study are capable of serving as a basis for extending our biological understanding of T. canis.

Alternative Input Lower Weight Information Method Error to Reduce Specific Absorption Rate in MRI (자기공명영상 검사 시 환자정보의 체중을 낮게 입력하여 전자파흡수율을 감소시키는 대안의 오류)

  • The purpose of this study is to correct the error of lower weight input method as an alternative to reduce the specific absorption rate(SAR) in MRI. In order to prove that the SAR values not change according to the weight entered into the patient information, the 50kg phantom is placed in the coil and the input weight is changed from 10 to 100 in 10kg units to compare the SAR values. As a result, T1-weighted images had a SAR rate of 0.2W/kg and T2-weighted images had an average of 0.4W/kg. In conclusions, the SAR does not change according to the weight input by the technician before the scan, a lower weight when inputting patient information cannot be an alternative to reduce the SAR.

Growth Response and Arsenic Uptake of White Clover (Trifolium repens) and Evening Primrose(Oenothera odorata) Colonized with Arbuscular Mycorrhizal Fungi in Arsenic-Contaminated Soil

  • A greenhouse experiment was conducted to investigate the role of the arbuscular mycorrhizal(AM) fungus, Glomus mosseae(BEG 107) in enhancing growth and arsenic(As) and phosphorus(P) uptake of white clover(Trifolium repens) and evening primrose(Oenothera odorata) in soil collected from a gold mine having concentrations of 381.6 mg total As $kg^{-1}$ and 20.5 mg available As $kg^{-1}$. Trifolium repens and O. odorata are widely distributed on abandoned metalliferous mines in Korea. The percent root colonization by the AM fungus was 55.9% and 62.3% in T. repens and O. odorata, respectively, whereas no root colonization was detected in control plants grown in a sterile medium. The shoot dry weight of T. repens and O. odorata was increased by 323 and 117% in the AM plants compared to non-mycorrhizal(NAM) plants, respectively. The root dry weight increased up to 24% in T. repens and 70% in O. odorata following AM colonization compared to control plants. Mycorrhizal colonization increased the accumulation of As in the root tissues of T. repens and O. odorata by 99.7 and 91.7% compared to the NAM plants, respectively. The total uptake of P following AM colonization increased by 50% in T. repens and 70% in O. odorata, whereas the P concentration was higher in NAM plants than in the AM plants. Colonization with AM fungi increased the As resistance of the host plants to As toxicity by augmenting the yield of dry matter and increasing the total P uptake. Hence, the application of an AM fungus can effectively improve the phytoremediation capability of T. repens and O. odorata in As-contaminated soil.

  • This study was conducted to detennine the effect of dietary supplements of mineral powder (pegmatite) on the physico-chemical properties of M longissimuss for 24 Hanwoo steers. Each eight Hanwoo steers were randomly allotted to three treatments ; Control $(0\%)$, T1 (supplemented with pegmatite $2.0\%$ and vitamin A $0.2\%)$, T2 (supplemented with pegmatite $2.0\%$). Cooking loss was highest in control, while water holding capacity (WHC) was the highest in T2 (56.38) than the others. However, effect of the diets on meat color was not significantly different (p>0.05) Panel test scores of juiciness, tenderness and flavor of M longissinus were the highest for T1 $(4.6\~5.0)$ between the treatment group (p<0.05). The content of total fatty acid in M longissimus was the highest in T2 $(50.80\%)$ among the treatment group. Palmitic acid and oleic acid were siginificantly higher in T1 and control than those for the others(p<0.05). Unsaturated fatty acid (UFA) was highest in T2 $(58.57\%)$ than the others$(55.09\~55.64\%)$ (p<0.05).

Effects of TDN/CP Ratio on Nutrient Intake, Digestibility, Ruminal Fermentation and Blood Characteristics of Replacement Dairy Heifers (TDN/CP 비율에 의한 영양소 공급이 젖소 육성우의 영양소 섭취량, 소화율, 반추위 발효 및 혈액특성에 미치는 영향)

  • This study was conducted to investigate a proper management and nutrient supply for raising replacement heifers to improve lactating performance. In order to determine a proper TDN/CP ratio that would be suitable for domestic use, 13-month-old replacement heifers were fed in the ratios of 4.6:1(T1) and 4.3:1(T2), respectively, to examine the changes in nutritional intake, digestibility, characteristics of rumen fluid and blood compositions. While dry matter intake was same at 9.0kg/day, CP intake did not show any significant difference, though T2(1.45kg/day) was slightly higher than T1(1.34kg/day). In fiber intake and digestibility, two results showed no significant difference, though T1 was slightly higher than T2. The pH, NH3-N and VFA densities in rumen fluid examined at the fourth hour after feed intake did not show any significant difference either. Overall level of VFA was not influenced largely by the TDN/CP ratio, but the iso-butyrate and iso-valerate levels and the A/P ratio were significantly high in T1(p<0.05). In blood characteristics, the treatments showed no significant difference. In conclusion, the TDN/CP ratios of 4.3:1 and 4.6:1 did not significantly affect the characteristics of the ruminant stomach, but in comparison with digestibility of 13-month-old heifers, the ratio of 4.6:1 was more appropriate for increased intake of fiber and higher digestibility.

Improvement of DS System using Transversal Filter for Advanced Duty Rate on Meteor Burst Channel (유성 버스트 채널 도통율 향상을 위해 Transversal Filter를 적용한 DS시스템의 개선)

  • In spite of many advantages of Meteor Burst Communications(MBC) on its transmission channel, the fact that its duty rate is less than 10 percents is a considerable deficiency of MBC. To overcome with this deficiency without paying large cost, we use a direct sequence(DS) simple reception system. This method doesn't need to add the components for acquisition or tracking the signal so that it keeps the cost efficiency of the MBC. But it has a disadvantage that its duty rate shows a large decline by noise. For this problem we adopt the proposed system applying a transversal filter circuit. The improvement by this method is more significant when the received signal is weak Simulation results show as t1 is 100ms, then the burst length would be increased by 35% when the initial value of the received signal power increases by 3 dB, t2 is 135ms. If the power increases by 6dB the burst length would be 170ms thus increased by 70%, which shows a great enhancement. Suppose now that t1 is 50ms and the power is increased by 3dB, then t2 would be increased by 70% to 85ms, and increasing the power by 6dB it would be increased by 130% to 115ms.

Membrane distillation of power plant cooling tower blowdown water

  • The objective of this study was to examine the recovery of the power plant cooling tower blowdown water (CTBD) by membrane distillation. The experiments were carried out using a flat plate poly vinylidene fluoride (PVDF) membrane with a pore diameter of $0.22{\mu}m$ by a direct contact membrane distillation unit (DCMD). The effects of operating parameters such as transmembrane temperature difference (${\Delta}T$), circulation rate and operating time on permeate flux and membrane fouling have been investigated. The results indicated that permeate flux increased with increasing ${\Delta}T$ and circulation rate. Whereas maximum permeate flux was determined as $47.4L/m^2{\cdot}h$ at ${\Delta}T$ of $50^{\circ}C$ for all short term experiments, minimum permeate flux was determined as $7.7L/m^2{\cdot}h$ at ${\Delta}T$ of $20^{\circ}C$. While $40^{\circ}C$ was determined as the optimum ${\Delta}T$ in long term experiments. Inorganic and non-volatile substances caused fouling in the membranes.

Stability and Modification of Aspergillus awamori $\alpha$-Glucosidase with $IO_4$-oxidized Soluble Starch (과요오드산-산화 가용성 전분에 의한 Aspergillus awamori $\alpha$-Glucosidase의 안정성 및 변형)

  • Periodate-oxidized soluble starch increased pH stability of Aspergillus awamori a-glucosidase. After incubation for two hours, the enzyme in the absence of oxidized soluble starch was stable in the range of pH 3-7 at 40℃, pH 3-6 at 50℃ and the enzyme in the presence of oxidized soluble starch was stable in the range of pH 3-9 at 40℃, pH 3-8 at 50℃. At 60℃, the enzyme was stable in pH 3-6 regardless of the presence or absence of IO₄-oxidized soluble starch, but when IO₄-oxidized soluble starch existed in pH 5-6, remained activity of the enzyme increased 20% more than when it didn't exist. The enzyme modified with IO₄-oxidized soluble starch remained 70% of activity in pH 9, but native enzyme didn't remain, showing the increase of stability due to modification. In thermal stability, modified enzyme remained 12% at 50℃ and 7% at 80℃. But native enzyme remained 8% at 50℃ and didn't remain at more than 70℃. The result of HPLC analysis revealed the subunit of the enzyme at under pH 2 or over pH 9 was separated or the enzyme was denatured and conjugated. Protein structure of native enzyme was denatured by acidic and basic pH but was stable in the presence of IO₄-oxidized soluble starch.
