오늘날 섬유 염색공정에서 발생하는 염색폐수로 인해 많은 문제점들을 야기한다. 염색폐수에는 난분해성의 EG, PVA, TPA, 여러 종류의 계면활성제가 포함되어 있어 완전하게 처리하기가 어렵다. 현재 일반적으로 사용하고 있는 물리화학적 처리와 활성오니공정의 처리성을 향상하기 위하여 jet-loop reactor (JLR)와 물리화학적 처리를 결합한 새로운 공정을 개발하였다. JLR의 부피산소전달계수는 air-lift 반응기에 비해 아주 우수하였다. 또한 좀더 효율적인 처리를 위하여 JLR에 활성탄 담체를 적용하여 (JLRAS) 실험하였다. 체류시간이 8시간일 때 BOD, $COD_{Mn}$, $COD_{Cr}$, 색도 제거율은 각각 99, 86, 84, 83%로 우수하였다. JLRAS 성능은 $COD_{Mn}$의 부하 변동에도 아주 빠르게 회복되었다. 2차 물리화학적 응집처리의 최적 응집제로는 폴리황산제2철이며 130 mg/L 투입하였을때 $COD_{Mn}$과 색도의 제거율은 자각 85, 73%였다. 결론적으로 이 공정으로 난분해성의 많은 유기물을 효율적으로 처리할 수 있으며 운전비용도 감소시킬 수 있었다.
급성 간부전 환자나 간이식 대기 상태의 환자의 생명을 간이 재생되거나 간 이식때 까지 연장시키기 위하여 생인공간이 많이 연구되고 있다. 본 연구에서는 적당한 cell opening을 가지는 폴리우레탄 폼을 15% NCO-prepolymer를 이용하여 직접 제조하였으며 이를 직경 3~4 mm 크기의 정육면체로 절단하여 충진형 반응기를 제조하였다. 분리된 일차간세포를 원심분리 방법을 이용하여 PUF 1 $cm^3$ 당 5.5$\pm$1.1$\times$$10^6$개의 세포를 접종하였으며 일주일간 관류 배양하면서 생인공간으로서의 성능을 측정하였다. 접종된 간세포는 구상체를 형성하였고 안정된 암모니아 제거능력과 요소 분비율을 나타내었다. 알부민 분비율은 배양 4일 이후 감소하는 경향을 보였다. 이상의 결과를 볼 때 PUF 충진형 간세포 반응기는 생인공간으로 개발되기에 충분한 가능성을 가진 것으로 판단된다.
생명공학분야에서 매우 중요한 효소 중에 하나인 lipase는 여러 산업에 유용하게 사용되고 있다. lipase를 선별하기 위해서는 최적화된 발현 시스템이 필요하다. 많은 발현 시스템중에 E.coli 발현 시스템은 바람직한 특성을 갖는 효소를 스크리닝하거나, 선별된 변이체들의 특성을 확인하는 데에 소요되는 시간과 비용을 단축시켜 줄 것이다. 본 연구에서는 그 중에 BL21와 OrigamiB에서 CalB를 발현하였다. 그 결과 BL21 균주에서 발현된 CalB는 대부분이 불용성의 inclusion body를 형성하고, 전혀 활성을 나타내지 않았다. 이전의 타 연구와 더불어 이 결과에서 E.coli 균주에서 CalB의 기능적 발현이 상당히 어렵다는 것을 알 수 있다. 특히 불용성의 inclusion body형성과 lipase의 세포에 대한 유독성이 원인이 될 수 있다. 그러나 BL21와 비교해보면, OrigamiB에서 발현된 CalB 또한 많은 양의 inclusion body를 형성하지만, lipase의 주요 특성중의 하나인 가수분해 활성이 상당하게 나타나는 것을 알 수 있다. lipase의 구조 형성을 도와주는 변형된 OrigamiB와 저온유도시스템인 pCold 플라스미드를 사용했기 때문이다. 이처럼 균주나 플라스미드의 선택, 유도조건의 변경 등의 여러 연구를 통하여 유용한 효소를 선별할 수 있다.
Chemical compositions and biological functions of brown colorants extracted from pine bark(Pinus densiflora) have been studied. Dyeing test using multifiber fabrics with extracted colorants were preliminary carried out. Dyeing conditions and fastness tests of selected fabrics have been also studied. The brown colorants were produced 1.5% concentrations by solvent extraction from milled pine bark using methanol. The colorants were extracted with 80% methanol as best choice by a criteria of solid quantity and dyeability on fabrics. The chemical compositions were identified as mixtures of taxifolin epicatechin and procyanidin by LC/MS analysis. The brown colorants could be dyed not only natural fibers such as cotton, silk and wool but also synthetic fiber as nylon and semi-synthetic fiber as viscose rayon. Maximum K/S values was shown at 400 nm according to different fiber with color appearance of redish brown. Optimum pH and temperature of dyeing conditions was 4 and above $80^{\circ}C$, respectively. The brown colorants had a strong antioxidant activity compared to Butylated hydroxyanisole as standard and weak antimicrobial activity against E. coli. compared to kanamycin. Washing, rubbing, perspiration, dry cleaning and light fastness for cotton, nylon and silk dyed with the brown colorants were carried out by KS K method. Most of color fastness such as washing, rubbing, perspiration, and dry cleaning were represented as 4-5 grade. However, light fastness was reported as 2-3 grade. From this studies, brown colorants produced pine bark have a high potentials for natural dyeing on fabrics with antioxidant activity.
Soybean plants infected with Bean pod mottle virus (BPMV) develop acute symptoms that usually decrease in severity over time. In other plant-virus interactions, this type of symptom recovery has been associated with degradation of viral RNAs by RNA silencing, which is accompanied by the accumulation of virus-derived small interfering RNAs (siRNAs). In this study, changes in the accumulation of BPMV siRNAs were investigated in soybean plants infected with BPMV alone, or infected with both BPMV and Soybean mosaic virus (SMV) and in transgenic soybean plants expressing SMV helper component-protease (HC-Pro). In many potyviruses, HC-Pro is a potent suppressor of RNA silencing. In plants infected with BPMV alone, accumulation of siRNAs was positively correlated with symptom severity and accumulation of BPMV genomic RNAs. Plants infected with both BPMV and SMV and BPMV-infected transgenic soybean plants expressing SMV HC-Pro exhibited severe symptoms characteristic of BPMVSMV synergism, and showed enhanced accumulation of BPMV RNAs and siRNAs compared to plants infected with BPMV alone and nontransgenic plants. Likewise, SMV HC-Pro enhanced the accumulation of siRNAs produced from a silenced green fluorescent protein gene in transient expression assays, while the P19 silencing suppressor of Tomato bushy stunt virus did not. Consistent with the modes of action of HC-Pro in other systems, which have shown that HC-Pro suppresses RNA silencing by preventing the unwinding of duplex siRNAs and inhibiting siRNA methylation, these studies showed that SMV HC-Pro interfered with the activities of RNA-induced silencing complexes, but not the activities of Dicer-like enzymes in antiviral defenses.

To perform the prophylactic study of a vaccine derived from human papillomavirus (HPV) using Balb/c mice, we produced virus like particles consisting of HPV capsid protein L1 which has been reported to induce significant humoral and cellular immunity using various animal model systems. In order to produce HPV16 VLPs, the cDNA of L1 capsid protein in HPV type 16, obtained by polymerase chain reaction, was inserted into yeast expression vector, YEG$\alpha$-HIR525 under the control of GAL10 promoter. The transformation of YEG$\alpha$-HPV16 L1 was performed into the yeast Saccharomyces cerevisiae Y2805 by the lithium acetate method and the yeast clone expressing the highest level of L1 capsid protein of human papillomavirus type 16 was selected by Western blot analysis using anti-HPV16 L1 antibody. The purification of HPV16 VLP has been performed by the ultracentrifugation and gel-filtration methods. To validate the vaccine efficacy of the purified HPV16 VLPs and investigate the properties of HPV16 VLPs to induce humoral immunity, ELISA assay was performed. A significantly increased production of anti-HPV16 VLP antibodies was observed in sera from immunized mice. The neutralization activity of antibodies in the sera from the vaccinated mice was demonstrated by a rapid and simple assay to detect hemagglutihation inhibition activity.
Vasicinone, a quinazoline alkaloid from Adhatoda vasica Nees. is well known for its bronchodilator activity. However its anti-proliferative activities is yet to be elucidated. Here-in we investigated the anti-proliferative effect of vasicinone and its underlying mechanism against A549 lung carcinoma cells. The A549 cells upon treatment with various doses of vasicinone (10, 30, 50, $70{\mu}M$) for 72 h showed significant decrease in cell viability. Vasicinone treatment also showed DNA fragmentation, LDH leakage, and disruption of mitochondrial potential, and lower wound healing ability in A549 cells. The Annexin V/PI staining showed disrupted plasma membrane integrity and permeability of PI in treated cells. Moreover vasicinone treatment also lead to down regulation of Bcl-2, Fas death receptor and up regulation of PARP, BAD and cytochrome c, suggesting the anti-proliferative nature of vasicinone which mediated apoptosis through both Fas death receptors as well as Bcl-2 regulated signaling. Furthermore, our preliminary studies with vasicinone treatment also showed to lower the ROS levels in A549 cells and have potential free radical scavenging (DPPH, Hydroxyl) activity and ferric reducing power in cell free systems. Thus combining all, vasicinone may be used to develop a new therapeutic agent against oxidative stress induced lung cancer.

Prolonged alcohol consumption causes alcoholic liver damage due to the generation of reactive oxygen species, the accumulation of fatty acids, and an increase in inflammatory cytokines in the liver. In this study, the protective effect of a fruit extract of Paeonia anomala (FEPA) against chronic alcohol-induced liver damage was evaluated in Sprague-Dawley rats fed an ethanol or a control Lieber-DeCarli diet for 5 weeks to induce alcoholic liver damage. FEPA (50, 25, and 10 mg/kg body weight/day) as well as the reference control silymarin (25 mg/kg body weight/day) were administered along with the ethanol diet. FEPA protected against increases in alanine aminotransferase and aspartate aminotransferase in serum and attenuated alcohol-induced increases in triglycerides, tumor necrosis factor alpha, thiobarbituric acid-reactive substances, and cytochrome P450 2E1 enzyme activity in the liver compared with the group treated with ethanol only. Anti-oxidative defenses such as the total glutathione level and glutathione peroxidase activity were increased by FEPA treatment. These results suggest that FEPA exerts protective effects against chronic alcohol-induced liver damage by attenuating hepatosteatosis and pro-inflammatory cytokine production and enhancing anti-oxidative defense mechanisms in the liver.

This peptide has antibacterial activity against several Gram-positive and Gram-negative bacteria. Bombyx mori cecropinB1(BmCecB1) is antimicrobial peptides from Bombyx mori and belongs to cecropin family. Antimicrobial peptides are important components of the innate immune systems in all living organism. To produce the BmCecB1 antimicrobial peptide, we constructed transgenic silkworm that expressed BmCecB1 gene under the control BmA3 promoter using piggyBac vector. The use of the 3xP3-driven EGFP cDNA as a marker allowed us to rapidly distinguish transgenic silkworm. Mixtures of the donor vector and helper vector were micro-injected into 600 eggs of bivoltin silkworms, Baegokjam. In total, 49 larvae (G0) were hatched and allowed to develop into moths. The resulting G1 generation consisted of 22 broods, and we selected 2 broods containing at least 1 EGFP-positive embryo. The rate of successful transgenesis for the G1 broods was 9%. We identified 9 EGFP-positive G1 moths and these were backcrossed with wild-type moths. With the aim of identifying a BmCecB1 as antimicrobial peptide, we investigated the Radical diffusion Assay (RDA) and then demonstrated that BmCecB1 possesses high antibacterial activities against Gram-negative bacteria.
The majority of laboratory animals were transported from commercial breeders to a research facility by ground transportation. During the transportation, many biological functions and systems can be affected by stress. In this experiment, the change of body weight during the transportation was measured and the recovery periods from the transportation stress established based on the body weight changes. Total 676 laboratory animals which were aged between 3 to 9 wk old were studied. The transportation time taken from container packing to unpacking the container was approximately 24 h. The temperature of animal container was constantly maintained by air-conditioning and heating equipment. Rats were found to be more sensitive than mice. The body weight of rats was significantly decreased 3.71% (p<0.05) compared to the body weight of mice which decreased 0.9% There was no significant difference between the strains in the same species. When the changes of body weights were compared between delivery days, C57BL/6 mice showed the most variable changes compared to other species and strains. Consequently, C57BL/6 was more sensitive to stress than the other strains and the transportation process needs to be standardized to reduce between day variability. To establish the recovery periods from transportation stress, the body weight changes were measured during the acclimation period. Although the body weight of animals decreased during transportation, animals recovered their weight loss after the next day.
