해조류 유래 호흡기 질환 천식 치료제 연구 동향 (A Review of Marine Algae-derived Therapeutic Agents for Respiratory Disease Asthma)

  • Asthma is a complex inflammatory disease of the lung characterized by variable airflow obstruction, airway hyperresponsiveness, airway inflammation, and reduction of respiratory function. Its prevalence and incidence are increasing because of the effect of various environmental and lifestyle risk factors. Steroid inhalation, long-acting agonists, and other synthetic drugs are used for the treatment of this disease. However, they have some side effects and show unsatisfied result and response after treatment. Therefore, many researchers have focused on the development of natural product-related treatment for asthma to suppress the side effects and unsatisfied results. Seaweeds contain various bioactive compounds with anti-inflammatory, antibacterial, and anti-oxidant activities. Thus, we investigated the asthma treatment-related literature using marine algae via the Google scholar search engine. Consequently, the literature is rarely investigated, but is increasing steadily. The literature was performed as a comparison study with an ovalbumin-induced group or drug-treated group, and investigated the antiasthma activity of algae ethanol extract. Although many researchers have studied marine algae-derived therapeutic agents for asthma, the amount of literature is rare compared with those of herbal medicine-derived therapeutic agents. Conclusively, we suggest that many researchers should investigate and develop algae-derived therapeutic agents for asthma treatment.

액체 크로마토그라피를 이용한 동물 근육조직 중의 합성항균제 동시 분석 (Simultaneous Determination of Residual Synthetic Antimicrobials in Animal Muscles by High Performance Liquid Chromatography)

  • 본 연구에서는 쇠고기, 돼지고기, 닭고기 중에 잔류하는 설파메라진(SMR), 설파메타진(SMT), 설파모노메톡신(SMMX), 설파디메톡신(SDMX), 설파퀴녹살린(SQX), 후라졸리돈(FZ), 죠렌(ZOL) , 에토파베이트 (EPB)등 합성항균제 8종에 대한 신속하고 간편한 동시분석 방법을 검토하였다. 동시분석파장으로 270nm, 이동상 용매로는 0.05M Oxalic acid : Acetonitile(22: 78), 시료 추출용매로 물과 디클로로메탄을 사용하였고 세정과정은 헥산/물. 물/디클로로메탄 분배를 이용하였다. 각 시료에 합성항균제 표준용액을 첨가하고 위의 과정을 거친 최종 추출액을 HPLCdp 주입하여 나타난 각 합성항균제의 분리는 좋았고, 각 시료에서의 대상 합성항균제들의 회수율은 쇠고기에서 74~99%, 돼지고기에서 73~99% 닭에서 75~96%로 나타났다. 검출한계는 SMR, SMT, SMMX, SDMX, EPB에서 5 ppb, SQX, FZ, ZOL에서 8ppb로 나타났으며 각 합성 항균제들의 흡수파장시험 및 pH 변화에 의한 피크분리실험에서 실제 업무수행 중에 검출된 약제를 다른 장비를 사용하지 않고 단지 HPLC만으로도 확인이 가능하였다.

Korean Mistletoe (Viscum album var. coloratum) Inhibits Amyloid β Protein (25-35)-induced Cultured Neuronal Cell Damage and Memory Impairment

  • The present study aims to investigate the effect of methanol extract of Korean mistletoe (KM; Viscum album var. coloratum), on amyloid $\beta$ protein ($A\beta$) (25-35), a synthetic 25-35 amyloid peptide, -induced neurotoxicity in cultured rat cerebral cortical neurons and memory impairment in mice. Exposure of cultured neurons to $10{\mu}M$ $A\beta$ (25-35) for 24 h induced a neuronal cell death, which was measured by a 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay and Hoechst 33342 staining. KM (10, 30 and $50{\mu}g/ml$) significantly inhibited the $A\beta$ (25-35)-induced apoptotic neuronal death. KM ($50{\mu}g/ml$) inhibited 10 μM Aβ (25-35)-induced elevation of intracellular calcium concentration ([Ca2+]i), which was measured by a fluorescent dye, Fluo-4 AM. Glutamate release into medium and generation of reactive oxygen species (ROS) induced by $10{\mu}M$ $A\beta$ (25-35) were also inhibited by KM (10, 30 and $50{\mu}g/ml$). These results suggest that KM may mitigate the $A\beta$ (25-35)-induced neurotoxicity by interfering with the increase of [Ca2+]i and then inhibiting glutamate release and generation of ROS in cultured neurons. In addition, orally administered KM (25 and 50 mg/kg, 7 days) significantly prevented memory impairment induced by intracerebroventricular injection of $A\beta$ (25-35) (8 nmol). Taken together, it is suggested that anti-dementia effect of KM is due to its neuroprotective effect against $A\beta$ (25-35)-induced neurotoxicity and that KM may have therapeutic role in prevention of the progression of Alzheimer's disease.

스파르가눔 총체에서 분리한 cysteine proteinase의 정제 및 부분 특성 (Isolation and Partial Characterization of Cysteine Proteinase from Sparganum)

  • 한국산 유혈목이에서 스파르가눔 충체를 수집하고, 이들 충체의 추출액에서 ion-exchange chromatography와 affinity chromatography를 실시하여 cysteine proteinase를 순수 정제하였다. 경제된 효소의 최적 pH는 5.5이었고, 최적 mole 농도는 0.IM (0.1M sodium acetate, pH5.5) 이었다. 정제된 대소는 thiol-dependent이고, $4^{\circ}C$에서 pH 5.0일 때 24시간 동안 안전성을 보였다. 효소의 환성도는 저분자 합성기질인 CBZ-phe-arg-AFC에 대 해 활성이 높았다. 정제된 효소는 척추동물의 산성 cysteine proteinase의 억제인자에 감수성을 보였다. UItrogel AcA54 column chromatography로 정제된 cysteine proteinase의 분자량을 측정한 결과 28,000 dalton이었다. 정제된 효소는 collagen type I과 hemoglobin을 분해하였다. Immunoblot한 결과 정제된 효소는 스파르가눔증 환자의 혈청과 반응하였다. 이상의 결과에서 스파르가눔의 cysteine proteinase는숙주 체내이동, 조직침수성 및 영양소 섭취에 관여할 것이라 추정되며, 정제된 효소는 스파르가눔 현중의 혈청학적 진단에 이용될 수 있을 것으로 생각된다.

북극 스발바드 콩스피오르덴 해역에서 L 밴드 PALSAR 데이터를 이용한 눈과 부빙에 의한 다중편파 산란특성 해석 (Polarimetric Scattering of Sea Ice and Snow Using L-band Quad-polarized PALSAR Data in Kongsfjorden, Svalbard)

  • This study describes measurements of fast ice recorded on May 23, 2009, in Kongsfjorden (translated as 'Kongs Fjord'), an inlet on the west coast of Spitsbergen in the Svalbard Archipelago. Seasonal fast ice is an important feature for Svalbard fjords, both in relation to their physical environment and also the local ecosystem, since it grows seaward from the coast and remains in place throughout the winter. Ice thickness, snow, ice properties, and wind speed were measured, while SAR (Synthetic Aperture Radar) data was observed simultaneously observed two times from ALOS-PALSAR (L-band). Measured ice thickness was about 25-35 cm while the thickness of ice floe broken from fast ice was measured as 10-15 cm. Average salinity was 1.9-2.0 ppt during the melting period. Polarimetric data was used to extract H/A/alpha-angle parameters of fast ice, ice floe, snow and glacier, which was classified into 18 classes based on these parameters. It was established that the area of fast ice represents surface scattering which indicates low and medium entropy surface scatters such as Bragg and random surfaces, while fast ice covered with snow belongs to a zone of low entropy surface scattering similar to snow-covered land surfaces. The results of this study will contribute to various interpretations of interrelationships between H/A/alpha parameters and the wave scattering Phenomenon of sea ice.

Astaxanthin과 Capxanthin의 급여가 난황의 착색에 미치는 효과 (Effect of Dietary Supplementation of Astaxanthin and Capxanthin on the Pigmentation of Egg Yolk)

  • 63주령 ISA Brown 산란계 총 96수를 사용하여, 12일 동안 난황에서의 biological astaxanthin과 chemical astaxanthin 및 capxanthin의 축적을 통한 착색 효과를 비교하였다. 대조구 사료에는 착색제를 첨가하지 않았고, 효모 Phaffia rhodozyma 3%를 대조구 사료에 첨가하여 astaxanthin 함량이 22.5 mg/kg 되게 한 후, 화학적으로 합성한 astaxanthin을 대조구 사료에 45 mg/kg 첨가한 후 및 paprika로부터 추출한 capxanthin을 대조구 사료에 45 mg/kg 첨가구 모두 4개의 처리구들을 두었다. 처리당 6반복, 반복당 4수씩을 임의로 배치하였다. Astaxanthin은 생물학적 및 화학적 급원 모두 급여 9일째까지 축적이 이루어졌다. Capxanthin의 경우, 난황에서 축적되지는 않았고 이로부터 파생되어진 물질들이 급여 $6{\sim}9$일 후에 미미하게 검출되었다. 난황에서의 astaxanthin 축적 정도는 급여한 농도에 비례하였다. 난황의 색깔을 제외하면, 달걀의 다른 품질 요인들은 처리구들 간의 차이가 거의 없었다.

Classification of Water Areas from Satellite Imagery Using Artificial Neural Networks

  • Every year, several typhoons hit the Korean peninsula and cause severe damage. For the prevention and accurate estimation of these damages, real time or almost real time flood information is essential. Because of weather conditions, images taken by optic sensors or LIDAR are sometimes not appropriate for an accurate estimation of water areas during typhoon. In this case SAR (Synthetic Aperture Radar) images which are independent of weather condition can be useful for the estimation of flood areas. To get detailed information about floods from satellite imagery, accurate classification of water areas is the most important step. A commonly- and widely-used classification methods is the ML(Maximum Likelihood) method which assumes that the distribution of brightness values of the images follows a Gaussian distribution. The distribution of brightness values of the SAR image, however, usually does not follow a Gaussian distribution. For this reason, in this study the ANN (Artificial Neural Networks) method independent of the statistical characteristics of images is applied to the SAR imagery. RADARS A TSAR images are primarily used for extraction of water areas, and DEM (Digital Elevation Model) is used as supplementary data to evaluate the ground undulation effect. Water areas are also extracted from KOMPSAT image achieved by optic sensors for comparison purpose. Both ANN and ML methods are applied to flat and mountainous areas to extract water areas. The estimated areas from satellite imagery are compared with those of manually extracted results. As a result, the ANN classifier performs better than the ML method when only the SAR image was used as input data, except for mountainous areas. When DEM was used as supplementary data for classification of SAR images, there was a 5.64% accuracy improvement for mountainous area, and a similar result of 0.24% accuracy improvement for flat areas using artificial neural networks.

광자 계수 집적 영상 현미경을 사용한 마이크로 물체의 3차원 시각화와 인식 (Three-Dimensional Visualization and Recognition of Micro-objects using Photon Counting Integral Imaging Microscopy)

  • 본 논문에서는 광자 계수 집적 영상 현미경을 사용하여 광자가 희박한 조건에서 마이크로 물체의 3차원 시각화와 인식에 대한 기술을 제안한다. 제안하는 방법에서는 고해상도의 서로 다른 원근감을 가지는 2차원 영상을 획득하기 위해 합성조리개 집적 영상을 사용한다. 그리고 영상으로부터 광자를 추출하기 위해 광자계수 영상 시스템의 수학적 모델인 포아송 분포를 사용하며 통계적 추정법으로 부터 3차원 영상을 추정한다. 따라서, 광자가 희박한 조건에서 마이크로 물체가 손상되지 않으면서 그에 대한 3차원 영상을 획득하고 시각화할 수 있다. 추가적으로, 비선형 상관 필터를 사용하여 3차원 물체의 인식도 가능하다. 본 기술의 유용성을 증명하기 위해, 광학적 실험을 수행하였다.

cDNA Cloning and Expression of Human Rotavirus Outer Capsid Protein VP7 in Insect Cells

  • Rotavirus is a major cause of severe gastroenteritis in young children and animals throughout the world. The VP7 of rotavirus is thought to induce the synthesis of neutralizing antibodies and to be responsible for determining viral serotypes. The cDNA coding for the VP7 capsid protein of human rotavirus, obtained from Korean patients (HRV-Y14), was cloned and its nucleotide sequence was determined. Comparative analysis of the nucleotide sequences between VP7 of Y14 and that of other foreign isolates showed $92.7~95.2\%$ homology to G1 serotypes (RV-4, KU, K8, WA), $74.2\%$ homolgy to G2 serotype HU-5, $76.4\%$ homology to G3 serotype SA-11, and $77.6\%$ homology to G4 serotype A01321. These data suggest that HRV-Y14 can be classified as a G1 serotype. cDNA coding for VP7 of HRV-YI4 was subcloned into the baculovirus vector and the VP7 glycoprotein was expressed in insect cells. The expressed proteins in Sf9 cell extract and tissue culture fluid were separated on SDS-PAGE, and Western blot analysis with monoclonal antibody raised against the synthetic peptide containing 21 amino acids within the VP7 conserved region was performed. The molecular weight of recombinant VP7 was estimated to be 36 kDa which is about the same size as the native VP7. Addition of tunicamycin in the culture media caused a reduction of the molecular weight of the recombinant VP7 indicating that the expressed protein was glycosylated.

COASTLINE DETECTION USING COHERENCE MAP OF ERS TANDEM DATA

  • A coastline is the boundary between land and ocean masses. Knowledge of coastline is essential for autonomous navigation, geographical exploration, coastal erosion monitoring and modelling, water line change, etc. Many methods have been researched to extract coastlines from the synthetic aperture radar (SAR) and optic images. Most methods were based on the intensity contrast between land and sea regions. However, in these methods, a coastline detection task was very difficult because of insufficient intensity contrast and the ambiguity in distinguishing coastline from other object line. In this paper, we propose an efficient method for the delineation of coastline using interferometric coherence values estimated from ERS tandem pair. The proposed method uses the facts that a tandem pair of ERS is acquired from a time interval of an accurate day and that the coherent and incoherent values in coherence map are land and water, respectively. The coherence map was generated from ERS tandem pair, filtered by MAP filter, and divided into land and water by the determination of threshold value that is based on the bimodality of the histogram. Finally, a coastline was detected by delineating the boundary pixels. There was a good visual match between the detected coastline and the manually contoured line. The interferometric coherence map will be helpful to identify land and water regions easily, and can be used to many applications that are related with a coastline.
