Previous studies have demonstrated that repeated restraint stress in rodents produces increases in depression and anxiety-like behaviors and alters the expression of corticotrophin-releasing factor (CRF) in the hypothalamus. The current study focused on the impact of Bupleurum falcatum (BF) extract administration on repeated restraint stress-induced behavioral responses using the forced swimming test (FST) and elevated plus maze (EPM) test. Immunohistochemical examinations of tyrosine hydroxylase (TH) expression in rat brain were also conducted. Male rats received daily doses of 20, 50, or 100 mg/kg (i.p.) BF extract for 15 days, 30 min prior to restraint stress (4 h/day). Hypothalamic-pituitary-adrenal axis activation in response to repeated restraint stress was confirmed base on serum corticosterone levels and CRF expression in the hypothalamus. Animals that were pre-treated with BF extract displayed significantly reduced immobility in the FST and increased open-arm exploration in the EPM test in comparison with controls. BF also blocked the increase in TH expression in the locus coeruleus of treated rats that experienced restraint stress. Together, these results demonstrate that BF extract administration prior to restraint stress significantly reduces depression and anxiety-like behaviors, possibly through central adrenergic mechanisms, and they suggest a role for BF extract in the treatment of depression and anxiety disorders.
Objectives: In this research, the effect of samul-tanggahyangbuja on depression and learning in ovariectomized rats subjected to repetitive stress were assessed. Samul-tanggahyangbuja is the prescription consisting of Samul-tang and Cyperi Rhizoma. Methods: Ovariectomized rats were repeatedly stressed over a 2-week period. After being orally medicated with samul-tanggahyangbuja (100 or 400 mg/kg), rats performed the Morris water maze test and forced swimming test, and social exploration was assessed in a behavior test. As well, sucrose intake was measured and measurements of blood serum corticosterone and the change of interleukin-$1{\beta}$ (IL-$1{\beta}$) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in blood samples were made. Results: 1. In the Morris water maze test, rats medicated with 100 mg samul-tanggahyangbuja mastered the maze in a shorter time on the 4th day in comparison with the control group, while rats medicated with 400 mg samul-tanggahyangbuja mastered the maze more quickly (p<0.05 on the 3rd day ; p<0.01 on the 4th day, as compared to control). 2. Immobility time in the forced swimming test was significantly decreased in rats receiving 400 mg samul-tanggahyangbuja compared with the control group (p<0.05). 3. Sucrose intake and active social behavior of rats receiving 400 mg samul-tanggahyangbuja were markedly increased in comparison with the control group (p<0.01). 4. Blood serum corticosterone measurements revealed decreased blood serum corticosterone level after medicating with samul-tanggahyangbuja. But it was not statistically significant. 5. Treatment with either dose of samul-tanggahyangbuja significantly reduced IL-$1{\beta}$ and TNF-${\alpha}$ (p<0.05). Conclusions: These results suggest that samul-tanggahyangbuja possesses the anti-depressant and cognitive-enhancing activities related to menopause.
Silibinin, a natural flavonoid antioxidant isolated from extracts of the milk thistle herb, has recently been identified as having anti-hepatotoxic and anticancer properties. In this paper, we investigated the effects of silibinin on behavior and neuroplasticity in mice subjected to chronic unpredictable mild stress (CUMS). After 5 consecutive weeks of CUMS, the mice were treated with silibinin (100 mg/kg, 200 mg/kg and 400 mg/kg by oral gavage) for 3 consecutive weeks. The results showed that silibinin administration significantly alleviated the CUMS-induced depressive-like behavior, including the total number of squares crossed and the frequency of rearing in the open field test, the immobility time in the tail suspension test and the forced swimming test. Furthermore, silibinin treatment increased the levels of brain-derived neurotrophic factor (BDNF), serotonin (5-HT) and norepinephrine (NE) in the prefrontal cortex and hippocampus. Our study provides new insight into the protective effects of silibinin on the depressive status of CUMS mice, specifically by improving neuroplasticity and neurotransmission.
The authors reviewed the etiology and essential treatment of temporomandibular disorders. The principal etiology was thought to the the myospasm of masticatory musculatures by stress (major cause) and dental irritation. Therefore, we attempted to control the myospasm by stress management, isometric exercise and regular daily physical exercises (walking, Jogging, bycycling, rope skipping, swimming & gymnastic exercise). The result was more favorable (95% success rate) without recurrence in 123 patients, who was referred to our department of dentistry(Oral & Maxillofacial surgery), Wonju Christian Hospital.
In the present study, the anti-depressant like effect of methyl gallate (MG) isolated from the stem bark of Acer barbinerve was examined in ICR mice. Body weight (BDW) and blood glucose (BDG) levels significantly decreased in the repeated restraint stress (RRS) group (2 h/day for 14 days) compared to the no stress (NS) group. To examine the effect of MG on RS-induced BDW loss and hypoglycemia, MG (10 mg/kg) and the anti-depressant fluoxetine (10 mg/kg) were administered daily for 14 days. Orally administered MG and fluoxetine significantly attenuated the RS-induced BDW loss and hypoglycemia. Interestingly, MG administered mice showed increased BDG levels in the normal and glucose feeding condition. Chronic RS-subjected mice showed immobilized and depressed behaviors. The effect of MG on the depressed behaviors was evaluated using the tail-suspension test (TST) and the forced swimming test (FST). In both tests, RS-induced immobilized behaviors were significantly reversed in MG and fluoxetine administered groups. Taken together, MG significantly attenuated the RS-induced BDW loss, hypoglycemia, and depressed behaviors. Considering that decreased BDG levels (hypoglycemia) can cause depression, MG may exert its anti-depressant like effect by preventing hypoglycemia. Our results suggest that MG isolated from A. barbinerve can exert anti-depressant like effect, and could be used as a new and natural anti-depressant therapy.

This study was performed to evaluate the effects of Koamsimsinwhan (without Cinnabarite) on depression in CMS-treated rats. Rats of trial groups were exposured to chronic mild unpredictable stress for 4 weeks. These CMS-treated rats were stratified into Koamsimsinwhan(without Cinnabarite) group and vehicle(non-drug) group. And there were control rats excluding both CMS and Koamsimsinwhan(without Cinnabarite). Forced swimming test, general activity test and immunohistochemical test(measuring the level of manifestation of c-Fos in PVN) were executed to evaluate the effects of Koamsimsinwhan(without Cinnabarite) on depression after 4 weeks of CMS treatment. The results were as follows 1. In FST, depression was induced from CMS treatment and was supressed with Koamsimsinwhan(without Cinnabarite). 2. The increased activity of nerve cell in PVN, which refers to stress, was induced from CMS treatment and normalized with Koamsimsinwhan (without Cinnabarite).

This study was undertaken to determine the effects of ginseng total saponin (GTS) on stress- induced analgesia (SIA) in mice. intermittent foot shock (FS)-SIA was antagonized not by on but by naloxone in the tail flick FS-SIA which was not antagonized by naloxone in the T.F. test. On the other hand, GTS did not antagonize the continuous FS-SIA naloxone antagonized in the T.P. test. Also GTS antagonized psychological (PSIF)-SIA which was not antagonized by naloxone in the T.F. test. However, GTS did not antagonize the PSY-SIA which naloxone antagonized in the T.P. test. Forced swimming (FSIP)-SIA was not affected by both GTS and naloxone. These results suggest that the antapeonisms of intermittent FS-SIA in the T.F. test, continuous FS-SIA and PSY-SIA by GTS are mediated by non-opioid mechanisms but the antagonism of intermittent FS-SIA in the T.P. test by GTS is mediated by opioid mechanism.
Objectives : The Bambusae Caulis in Taeniam has been used in traditional oriental medicine to treat a variety of mental disorders including anxiety and depression. The purpose of present study is to observe the changes which take place in the body in stressful situations and to compare the anti-depressive, anxiolytic and anti-stress effect of Bambusae Caulis in Taeniam in different doses. Methods : This study was performed to evaluate the effects of Bambusae Caulis in Taeniam in the immobilization stress model in rats. Twenty-five rats were divided into 5 groups of 5: control, stress, low dose administration (75 mg/kg), medium dose administration (150 mg/kg) and high dose administration (225 mg/kg). The four groups other than the control group were placed in an immobilization stress test, and distilled water (control) or Bambusae Caulis in Taeniam extract was administration orally for 2 weeks. After treatment, the despair and anxiety behavior of rats were measured by open field test, forced swimming test, weight gain, contents of 5-HT in raphe nucleus and adrenal gland weight. Results & Conclusions : As a result of evaluation by measuring five aspects, FST, OFT, weight gain, 5-HT contents and adrenal gland weight, Bambusae Caulis in Taeniam has significant antidepressant, anxiolytic, anti-stress effects.
In this study, we assessed the effects of ginsenoside Re (GRe) administration on repeated immobilization stress-induced behavioral alterations using the forced swimming test (FST), the elevated plus maze (EPM), and the active avoidance conditioning test (AAT). Additionally, we examined the effect of GRe on the central adrenergic system by observing changes in neuronal tyrosine hydroxylase (TH) immunoreactivity and brain-derived neurotrophic factor (BDNF) mRNA expression in the rat brain. Male rats received 10, 20, or 50 mg/kg GRe (i.p.) 30 min before daily exposures to repeated immobilization stress (2 h/day) for 10 days. Activation of the hypothalamic-pituitary-adrenal (HPA) axis in response to repeated immobilization was confirmed by measuring serum levels of corticosterone (CORT) and the expression of corticotrophin-releasing factor (CRF) in the hypothalamus. Repeated immobilization stress increased immobility in the FST and reduced open-arm exploration in the EPM test. It also increased the probability of escape failures in the AAT test, indicating a reduced avoidance response. Daily administration of GRe during the repeated immobilization stress period significantly inhibited the stress-induced behavioral deficits in these behavioral tests. Administration of GRe also significantly blocked the increase in TH expression in the locus coeruleus (LC) and the decrease in BDNF mRNA expression in the hippocampus. Taken together, these findings indicate that administration of GRe prior to immobilization stress significantly improved helpless behaviors and cognitive impairment, possibly through modulating the central noradrenergic system in rats. These findings suggest that GRe may be a useful agent for treating complex symptoms of depression, anxiety, and cognitive impairment.

The critical low temperature and response of the behavioral tolerance of red sea bream Pagrus major fingerlings were determined using the continuous behavior monitoring system (CBMS). The behavior of the experimental organisms was observed by decreasing the water temperature by 2.0℃ and 4.0℃ every 12 hours and 24 hours in the range of 8.0-20.0℃. An unstable behavior pattern was observed in red seabream fingerlings exposed to water temperatures below 12.0℃, in which the swimming activity decreased and repeatedly stopped, regardless of the exposure time and water temperature fluctuation. The swimming ability of the organisms exposed to 8.0-10.0℃ decreased sharply, and the behavior of staying at the bottom of the test tank was observed. Only 50 % of the organisms survived due to the low-temperature stress, and all individuals died within six hours after the cold shock. In addition, the behavior index (BI) decreased rapidly, and the amplitude change of the coefficient of variation (CV) was found to have a greater variation than the other water temperatures (p<0.05). Low-temperature stress of red sea bream is promoted at 12.0℃, and it is interpreted as the tolerance limit, which can induce a sublethal response of the organisms exposed to cold shock of 8.0-10.0℃.
