The studies were conducted to investigate the anti-diabetic activities on the hyperglycemia induced by streptozotocin in rats, anti-hypertensive activities in SHR, anti-fatigue and decrease of body weight activities in mice by Kyung Ok-Ko water extract and drink, which is a traditional preparation in Korea. 1. The blood glucose levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease glucose levels comparing with control group. 2. The serum total cholesterol levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease total cholesterol levels comparing with control group. 3. The serum triglyceride levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease triglyceride levels comparing with control group. 4. The blood pressure in SHRs were dose-dependently lower descended by administration of Kyung Ok-Ko water extract 300, 600, and 1200 mg/kg, respectively. 5. The swimming time of Kyung Ok-Ko water extract(100, 200, 400 mg/kg) and drink(0.7, 1.4, 2.1 ml/kg) were significantly extended to all experimental group dose-dependantly. 6. The decrease of body weight of Kyung Ok-Ko water extract(100, 200, 400 mg/kg) and drink(0.7, 1.4, 2.1 ml/kg) were significantly evaluated dose-dependently in all experimental group.
Acharan sulfate (AS) is a glycosaminoglycan (GAG) prepared from the giant African snail, Achatina fulica. In this study, some biological activities of AS were evaluated on the basis of structural similarities to heparin/heparan sulfate and the biological functions of GAGs. We demonstrated that it exhibited strong immunostimulating activities as measured by carbon clearance test in mice and in vivo phagocytosis. It also exhibited a significant hypoglycemic activity in epinephrine (EP)-induced hyperglycemia as well as antifatigue effects by weight-loaded forced swimming test. And it showed hypolipidemic activities in cholesterol-rich mixture induced hyperlipidemia in rats. The above results indicate that AS has diverse biological activities and suggest therapeutically important target molecules.
Demands for sports activities for the disabled are continuously increasing. Previously, Pseudomonas plecoglossicida YJR13 and Pseudomonas putida YJR92 from a sequential screening procedure were proven to effectively control Phytophthora blight caused by Phytophthora capsici. In this study, we further investigated the anti-oomycete activities of these strains against mycelial growth, zoospore germination, and germ tube elongation of P. capsici. We also investigated root colonization ability of the bacterial strains in square dishes, including cell motility (swimming and swarming motilities) and biofilm formation. Both strains significantly inhibited mycelial growth in liquid and solid V8 juice media and M9 minimal media, zoospore germination, and germ tube elongation compared with Bacillus vallismortis EXTN-1 (positive biocontrol strain), Sphingomonas aquatilis KU408 (negative biocontrol strain), and MgSO4 solution (untreated control). In diluted (nutrient-deficient) V8 juice broth, the tested strain populations were maintained at >108 cells/ml, simultaneously providing mycelial inhibitory activity. Additionally, these strains colonized pepper roots at a 106 cells/ml concentration for 7 days. The root colonization of the strains was supported by strong swimming and swarming activities, biofilm formation, and chemotactic activity towards exudate components (amino acids, organic acids, and sugars) of pepper roots. Collectively, these results suggest that strains YJR13 and YJR92 can effectively suppress Phytophthora blight of pepper through direct anti-oomycete activities against mycelial growth, zoospore germination and germ tube elongation. Bacterial colonization of pepper roots may be mediated by cell motility and biofilm formation together with chemotaxis to root exudates.
An attempt was made to study the effects of dangguibohyultang and Ssangbohwan on the muscle recovery from exhaustion after severe exercise. Before rats were given severe swimming exercise, Dangguibohyultang and Ssangbohwan were admisterd. Then, the blood lactate level, the serum activities of LDH, CPK and the serum levels of FFA and glucose were measured. The results were as follows; 1. The level of the blood lactate were significantly decreased in the group of Dangguibohyultang. 2. The activities of serum LDH were significantly decreased in the groups of Dangguibohyultang and Ssangbohwan. 3. The activities of serum CPK were significantly decreased in the groups of Dangguibohyultang and Ssangbohwan. 4. The levels of serum FFA were significantly decreased in the groups of Dangguibohyultang. 5. The levels of serum glucose were significantly increased in the groups of Dangguibohyultang and Ssangbohwan. According to the above results, it is concluded that Dangguibohyultang was more effective on the muscle recovery than Ssangbohwan in rats.

Red ginseng (RG, the extract of Panax ginseng Meyer) has various biological and psychological activities and may also alleviate fatigue-related disorders. The present study was undertaken to evaluate what kind of fatigue red ginseng alleviate. Animals were orally administered with 50, 100, 200, 400 mg/kg of RG for 7 days. Before experiments were performed. Physiological stress (swimming, rotarod, and wire test) are behavioral parameters used to represent physical fatigue. Restraint stress and electric field test to a certain degree, induce psychological fatigue in animals. Plasma concentration of lactate and corticosterone (CORT) were also measured after these behavioral assays. RG supplementation (100 mg/kg) increased movement duration and rearing frequency of restrainted mice in comparison with control. 100 and 200 mg/kg of RG increased swimming time in cold water ($8{\pm}4^{\circ}C$) while at 100 mg/kg, RG increased electric field crossing over frequencies. 50, 100 and 200 mg/kg RG prolonged running time on the rotarod and at 100 mg/kg, it increased balancing time on the wire. RG at those doses also reduced falling frequencies. RG supplementation decreased plasma CORT levels, which was increased by stress. Lactate levels were not significantly altered. These results suggest that RG supplementation can alleviate more the damages induced by psychological than physical fatigue.
Octacosanol is known to enhance endurance activities, control cholesterol in body and improve the function of cardiopulmonary. Citrulline, which is main compound of watermelon, is known to improve angiectasia through stimulating production of nitric oxide. To improve endurance activity swimming test on rats was carried out using four samples such as 1$\%$ octacosanol, citrulline, the extracts of barks of watermelon and products, mixture of 1$\%$ octacosanol and the extracts of barks of watermelon (6 : 4). Biochemical assays on the liver and serum of tested rats were also performed using commercial analysis kits. In result, it was shown that swimming time of III group increased by 26$\%$ and that of V group was increased by 22$\%$ at the swimming test. As a result of biological assays on the liver and serum of tested rats it was possible to confirm stability of toxicity When compared with creatine kinase of control group (549.11$\pm$39.15 U/l) citrulline (644.11 $\pm$50.67 U/l) and products group (646.00$\pm$46.99 U/l) were largely increased. When compared with inorganic phosphate of control group (12.01$\pm$0.75 mg/이), citrulline (13.03$\pm$0.94 mg/dl) and products group (12.90$\pm$0.55 mg/dl) showed similar results. Also, when compared with lactic acid and glucose of control group (152.91 $\pm$ 13.45, 103.00$\pm$ 8.69 mg/dl), citrulline (125.53$\pm$15.54, 83.75$\pm$7.29 mg/dl) and products group (135.26$\pm$11.50, 78.57$\pm$9.79 mg/dl) were largely decreased. As these test results, it was determined that 1$\%$ octacosanol and extracts of barks of watermelon had some effect of improving endurance activity. Futhermore, it was thought that it could be used as source of functional food.
Previously, we identified that Cirsium japonicum (CJ) extract posses antidepressant properties. In order to look for the optimal hebal preparation, CJ extracts collected from Koheung, Yesan, Cheju, Pyungchang and Pocheon were compared. The psychopharmacological activities of each extract were investigated using the open field test, the rota-rod test and the forced swimming test. Effect of each extract on $Cl^-$ influx in neuroblastoma cells were also identified. All of the extracts were orally administered at dosages of 50, 100, 200, 400 mg/kg of body weight. Supplementation of CJ did not significantly alter psychomotor behaviors such as movement, rearing frequency and total turn angle on the open field. Supplementation of CJ's extract from Pyungchang (100 mg/kg, p.o.), Cheju (200 and 400 mg/kg p.o.) and Yesan (100 and 200 mg/kg, p.o.) significantly decreased immobile duration and increased mobile duration in the forced swimming test. All of extracts have no influence on NE uptake in HEK-293 cells, but increased $Cl^-$ influx in neuroblastoma cell. The extract from Yesan demonstrated the most potent effect on $Cl^-$ influx. These results indicate that CJ's extracts from Pyungchang, Cheju and Yesan may alleviate depression, and the extract from Yesan may be the most effective preparation.
