A bacterial strain, CK214, exhibiting high motility on an LB agar (1.5%, w/v) surface was isolated from the environment. The formation of unusual agar shrinking around colonies on agar plates was observed. The strain grew on minimal media containing pure agar as a sole carbon source. The cell-free culture supernatant of CK214 generated a reduced form of sugar in the in vitro reaction with the use of pure agar as a substrate, suggesting the secretion of an agar-degrading enzyme. The CK214 strain showed swarming motility on the solid media containing a wide range of concentrations of agar (0.5, 1.0, 1.5, 2.0% w/v). Various tests, including Gram staining, API analysis, and phylogenetic analysis based on 16S rDNA sequences identified that the CK214 strain was a G(+) rod-shaped bacterium grouped in genus Paenibacillus. Electron microscopic analysis demonstrated that the P. CK214 strain is peritrichously flagellated. Through transposon random mutagenesis, several agar-degrading activity defective mutants (ADMs) were generated. These mutants will be used in the future experimentation for the study of the correlation between agar-degrading activity and motility.

Recently, many problems are occurring in the bee ecosystem due to rapid climate change. The decline in the bee population and changes in the flowering period are having a huge impact on the harvest of bee-keepers. Since it is impossible to continuously observe the beehives in the hive with the naked eye, most people rely on knowledge based on experience about the state of the hive. Therefore, interest is focused on smart beekeeping incorporating IoT technology. In particular, with regard to swarming, which is one of the most important parts of beekeeping, we know empirically that the swarming time can be determined by the sound of the queen bee, but there is no way to systematically analyze this with data. You may think that it can be done by simply recording the sound of the queen bee and analyzing it, but it does not solve various problems such as various noise issues around the hive and the inability to continuously record. In this study, we developed a system that records queen bee sounds in a real-time cloud system and analyzes sound patterns. After receiving real-time analog sound from the hive through multiple channels and converting it to digital, a sound pattern that was continuously output in the queen bee sound frequency band was discovered. By accessing the cloud system, you can monitor sounds around the hive, temperature/humidity inside the hive, weight, and internal movement data. The system developed in this study made it possible to analyze the sound patterns of the queen bee and learn about the situation inside the hive. Through this, it will be possible to predict the swarming period of bees or provide information to control the swarming period.

Argument alternation in English sentences such as “load hay onto the truck" vs. “load the truck with hay" or “Bees are swarming in the. garden" vs. “The garden swarms with bees" present an interesting dilemma for linguistic theory in several ways. Along with each kind of syntactic rearrangement of arguments goes a subtle but significant and systematic change in the verb's meaning. This has been called as different terminology such as “Double-faced", “Verbal diathesis", and most commonly as “Argument alternation", Dowty adopts terminology: Agent-subject (A-subject) form and Location-subject (L-subject) form in referring the two kinds of sentences and analyses as well as describes their different properties. In this paper, I basically follow the Dowty(200l)'s assumption while surveying several linguists's analysis and show its theoretical adequacy. and show its theoretical adequacy.
Colonization studies previously performed with a green-fluorescent-protein, GFP, labeled derivative of Bacillus amyloliquefaciens FZB42 revealed that the bacterium behaved different in colonizing surfaces of plant roots of different species (Fan et al., 2012). In order to extend these studies and to elucidate which genes are crucial for root colonization, we applied targeted mutant strains to Arabidopsis seedlings. The fates of root colonization in mutant strains impaired in synthesis of alternative sigma factors, non-ribosomal synthesis of lipopeptides and polyketides, biofilm formation, swarming motility, and plant growth promoting activity were analyzed by confocal laser scanning microscopy. Whilst the wild-type strain heavily colonized surfaces of root tips and lateral roots, the mutant strains were impaired in their ability to colonize root tips and most of them were unable to colonize lateral roots. Ability to colonize plant roots is not only dependent on the ability to form biofilms or swarming motility. Six mutants, deficient in abrB-, sigH-, sigD-, nrfA-, yusV and RBAM017410, but not affected in biofilm formation, displayed significantly reduced root colonization. The nrfA- and yusV-mutant strains colonized border cells and, partly, root surfaces but did not colonize root tips or lateral roots.

In order to achieve faster content distribution speed and stronger fault tolerance, a P2P peer can connect to multiple peers in parallel and receive chunks of the data simultaneously. A critical issue in this environment is selecting a set of nodes participating in swarming sessions. Previous related researches only focus on performance metrics, such as downloading time or the round-trip time, but in this paper, we consider a new performance metric which is closely related to the network and propose a peer selection algorithm that produces the set of peers generating optimal worst link stress. We prove that the optimal algorithm is practicable and has the advantages with the experiments on PlanetLab. The algorithm optimizes the congestion level of the bottleneck link. It means the algorithm can maximize the affordable throughput. Second, the network load is well balanced. A balanced network improves the utilization of resources and leads to the fast content distribution. We also notice that if every client follows our algorithm in selecting peers, the probability is high that all sessions could benefit. We expect that the algorithm in this paper can be used complementary to existing methods to derive new and valuable insights in peer-to-peer networking.
Objectives: The quorum-sensing-inhibitory and anti-biofilm activities of the methanol extract of E. globulus leaves were determined against clinically isolated multidrug-resistant Pseudomonas aeruginosa. Methods: The preliminary anti-quorum-sensing (AQS) activity of eucalyptus was investigated against a biosensor strain Chromobacterium violaceum ATCC 12472 (CV12472) by using the agar well diffusion method. The effect of sub-minimum inhibitory concentrations (sub-MICs) of the methanol extract of eucalyptus on different quorum-sensing-regulated virulence factors, such as swarming motility, pyocyanin pigment, exopolysaccharide (EPS), and biofilm formation, against clinical isolates (CIs 2, 3, and 4) and reference PA01 of Pseudomonas aeruginosa were determined using the swarm diameter (mm)-measurement method, chloroform extraction method, phenol (5%)-sulphuric acid (concentrated) method, and the microtiter plate assay respectively, and the inhibition (%) in formation were calculated. Results: The preliminary AQS activity (violacein pigment inhibition) of eucalyptus was confirmed against Chromobacterium violaceum ATCC 12472 (CV12472). The eucalyptus extract also showed concentration-dependent inhibition (%) of swarming motility, pyocyanin pigment, EPS, and biofilm formation in different CIs and PA01 of P. aeruginosa. Conclusion: Our results revealed the effectiveness of the E. globulus extract for the regulation of quorum-sensing-dependent virulence factors and biofilm formation at a reduced dose (sub-MICs) and suggest that E. globulus may be a therapeutic agent for curing and controlling bacterial infection and thereby reducing the possibility of resistance development in pathogenic strains.
Pseudomonas aeruginosa is a Gram (-) opportunistic human pathogen causing a wide variety of infections on lung, urinary tract, eyes, and burn wound sites and quorum sensing (QS), a cell density-sensing mechanism plays an essential role in Pseudomonas pathogenesis. In order to investigate the importance of QS in the Pseudomonas infections of Korean patients, we isolated 189 clinical strains of P. aeruginosa from the patients in Pusan Paik Hospital, Busan, South Korea. The QS signal production of these clinical isolates was measured by signal diffusion assay on solid media using reporter strains. While most clinical strains (79.4%) produced the QS signals as similar level as a wild type strain, PAO1 did, where LasR, the initial QS signal sensor-regulator was fully activated, a minority of them (4.2%) produced much less QS signals at the level to which LasR failed to respond. Similarly, while 72.5% of the clinical isolates produced QS signals enough to activate QscR, an another QS signal sensor-regulator, some few of them (9%) produced the QS signals at much lower level where QscR was not activated. For further analysis, we selected 74 clinical strains that were obtained from the patients under suspicion of Pseudomonas infection and investigated the total protease activity that is considered important for virulence. Interestingly, significant portion of them showed very low protease activity (44.6%) or no detectable protease activity (12.2%). When the biofilm-forming ability that is considered very important in chronic infection was examined, most isolates showed lower biofilm-forming activity than PAO1. Similarly, significant portion of clinical isolates showed reduced motility (reduced swarming activity in 51.4% and reduced twitching activity in 41.9%), or non-detectable motility (swarming-negative in 28.4% and twitching-negative in 28.4%). Our result showed that the clinical isolates that produced QS signals at the similar level to wild type could have significantly reduced activities in the protease production, biofilm formation, and motility, and some clinical isolates had unique patterns of motility, biofilm formation, and protease production that are not correlated to their QS activity.

Pseudomonas syringae pathovar tabaci is a plant pathogenic bacterium that causes wildfire disease in tobacco plants. In P. syringae pv. tabaci, PsyI, a LuxI-type protein, acts as an AHL synthase, while primary and secondary sequence analysis of PsyR has revealed that it is a homolog of the LuxR-type transcriptional regulator that responds to AHL molecules. In this study, using phenotypic and genetic analyses in P. syringae pv. tabaci, we show the effect of PsyR protein as a quorum-sensing (QS) transcriptional regulator. Regulatory effects of PsyR on swarming motility and production of siderophores, tabtoxin, and N-acyl homoserine lactones were examined via phenotypic assays, and confirmed by quantitative real-time reverse transcription-polymerase chain reaction (qRT-PCR). Further qRT-PCR showed that PsyR regulates expression of these virulence genes in response to environmental signals. However, an upstream region of the gene was not bound with purified MBP-PsyR protein; rather, PsyR was only able to shift the upstream region of psyI. These results suggested that PsyR may be indirectly controlled via intermediate-regulatory systems and that auto-regulation by PsyR does not occur.

Quorum sensing, a gene expression in response to population density, is regulated by chemical signals, most of which are acylated homoserine lactones (AHLs). The AHL derivatives have been reported to regulate bioluminescence, virulence factors and / or swarming motility in bacteria. It is hypothesized that higher organisms may have evolved specific means to interfere with bacterial communication as exemplified in the AHL-antagonistic activity of halogenated furanones isolated from the Australian macroalga Delisea pulchra.
