Manganese Superoxide Dismutase (MnSOD Val-9Ala) Gene Polymorphism and Susceptibility to Gastric Cancer

  • Background: Oxidative stress caused by the generation of reactive oxygen species plays an important role in human carcinogenesis. Manganese superoxide dismutase (MnSOD) Val-9Ala in the mitochondrial target sequence is the best known polymorphism of this enzyme. The purpose of the current research was to assess the association of MnSOD Val-9Ala genotypes with the risk of gastric cancer. Materials and Methods: This case-control study covered 54 gastric cancer patients compared to 100 cancer free subjects as controls. Extraction of DNA was performed on bioptic samples and genotypes were identified with a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Results: The frequencies of MnSOD Ala/Ala, Ala/Val and Val/Val genotypes in healthy individuals were 24.3, 66.7 and 9%, respectively. However, in gastric cancer patients, Ala/Ala, Ala/Val and Val/Val were observed in 24.0, 48.0 and 28.0% (p=0.01). In patients the frequency of MnSOD Val allele was higher (52%) compared to that in controls (42%). Conclusions: The results of this study show a positive association between MnSOD Val-9Ala gene polymorphism and risk of gastric cancer disease in Iranian population.

The Antioxidant Effects of HWANSODAN on the Brain Tissue of aged Rat (환소단(還少丹) 뇌조직(腦組織)의 산화작용(酸化作用)에 미치는 영향(影響))

  • The effect of HWANSODAN(HSD), on the level of brain antioxidants was examined in aged rat. The experimental groups were divided into three groups and treated as follows ; normal group(negative control), Vt.E administrated Group(HSD). The purified microsome from brain tissue, those were measired the amounts of oxidant materials like Malonfialdehyde(MDA) and H2O2, then activities of antioxidants enzymes like superoxide dismutase, catalase, NADPH-cytochrome P-450 reductase.The results were as follows;1. In TBA reaction to measure the amount of MDA, HSD group and Vt.E group did not showed signigicant decrease.2. In the formation of Hydrogen peroxide, HSD group and Vt.E group showed a little increase.3. The activity of Superoxide dismutase was increased significantly in HSD group and Vt.E group.4. In the activity of Catalase, Vt.E group was increased significantly and HSD group a little increased. 5. The activity of NADPH-cytochrome P-450 reductase in the HSD group and Vt.E group showed significantly increase.According to the above results, it is suggested that HWANSODAN(HSD) has some antioxidant effects on the tissue of brain.

  • The effect of Samyonjihwangtang(SJT) on th level of brain antioxidants was examined in aged rat. Samyongjihwangtang(SJT) is assed Cervi Pantiri-chum Cornu, Ginseng Radix to Yukmijihwangtang. The experimental groups were divided into three groups and treated as follows ; normal group(NC), Vt.E administrated group(PC), SJT administrated Group(SJT). From the purified microsome of brain tissue, those were measures the amounts of oxidant materials like malonaldehyde(MDA) and H_2O_2, then activities of antioxidants enxymes like Superoxide dismutase, Catalase, NADPH-cytochrome P-450 reductase.The results were as follows;1. Cytotoxicity of Methanol Extract of Edible Herbs Against L1210 Cells with the Changes of Antioxidant Enzymes Activities (식용 허브 메탄올추출물의 L1210 암세포에 대한 세포독성과 항산화효소 활성 변화)

  • The methanol extracts prepared from ten kinds of culinary herbs were investigated for the cytotoxcic effect againt L1210 cancer cells and the mode of action. The substantial cytotoxic effects were observed in all cases with the most prominent effect demonstrated by lemon verbena extract showing $87{\pm}4.1%$ cytotoxicity with $100{\mu}g/ml$ concentration and 3 days culture period. The cytotoxic effect was found to be dose and culture period dependent. With respect to the mechanism of the cytotoxicity, the augmented generation of $O_2{^-}ion$ and the dramatically escalated activities of antioxidant enzymes such as superoxide dismutase(SOD) and glutathione peroixdase (GPx) with addition of the herb methanol extractw suggested that there would be the involvement of reactive oxygen species (ROS) metabolism in the course of L1210 cancer cell death by the mothanol extract of the edible herbs.

Effects of Paraquat on Ascorbic Acid and Malondialdehyde Contents, and Superoxide Dismutase Activity in Spinach Chloroplasts under Light and Dark

  • The amounts of ascorbic acid in chloroplasts treated with light and light+paraquat (PQ) were reduced by 81 and $82\%$ of initial level, respectively at 24 hr at incubation. And those treated with dark and dark+PQ were decreased by 46 and $55\%$ of the original level, respectively. Malondialdehyde (MDA) contents at 24 hr of dark and dark+PQ treatment were increased by 6 and $31\%$ of the initial level, respectively. When chloroplasts were treated with light and light+PQ, MDA contents after 24 hr were increased by 88 and $146\%$ of the initial level, respectively. SOD activities treated with light and light+PQ were increased by 10 and $20\%$ of the initial level, respectively for 3 hr and thereafter reduced by 46 and $49\%$ of the original level, respectively at 24 hr. However, the SOD activities treated with dark and dark+PQ were decreased by 37 and $30\%$ of the initial level, respectively, It is considered that PQ triggers the oxidation of ascorbic acid, the induction of lipid peroxidation and the inactivation of SOD under light so that PQ has inhibitory effect on the pathway of plant metabolism.

Protective Effect of an Aged Garlic-bamboo Salt Mixture on the Rat with the Alcohol-salicylate Induced Gastropathy (마늘-죽염 제제가 위장 장애 유발 흰쥐의 항산화 효소활성에 미치는 영향)

  • Garlic has been known to be effective against the gastrointestinal diseases which can be induced by production of oxygen-derived free radical. It has been shown that bamboo salt is effective on the treatment and prevention of various gastrointestinal disorders. Bamboo salt is a processed salt invented by a Korean, ll-Hoon Kim. It has been reported that garlic and bamboo-salt are useful to the treatment of gastric disorders in Korea. To clarify the protective mechanism of the garlic-bamboo salt mixture, the gas-tropathy was induced in rats with alcohol-salicylate and the activities of the free radical scavenging enzymes were examined. In this study, we found that the garlic-bamboo salt mixture reduced the severity of hemorrhagic lesion in gastric mucosa in the rats. In addition, the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GR) were significnatly increased and the total content of GSH was recovered. From those results, we concluded that the protective effect of the garlic-bamboo salt mixture on gastropathy in rats is its ability to recover the level of GSH and to increase the activities of the free radical scavenging enzymes (SOD, GPx, GR).

Effects of Bombycis corpus on Amyloid-induced Lipid Peroxidation Antioxidative Enzymes and NO Synthesis in Rat Astrocytes (흰쥐의 뇌 astrocyte에서 $amyloid-{\beta}25-35$로 유발된 지질의 과산화와 항산화 효소계 및 NO 생성에 미치는 백강잠의 효과)

  • 목적 : 본 연구는 식풍해경(熄風解痙), 소산풍열(疏散風熱) 효능이 있는 백강잠이 치매에 미치는 영향을 알아보기 위하여 실험을 행하였다. 방법 : 치매 유발물질인 $amyloid{\beta}(A{\beta})$ 25-35를 흰쥐의 뇌 astrocyte에 처리한 후 대표적인 항산화 효소인 catalase, superoxide dismutase,glutathione peroxidase 및 glutathione-S-transferase의 활성 변화와 NO 생성 변화를 관찰하였다. 결과 $A{\beta}$ 25-35 처리로 catalase와 superoxide dismutase 활성이 현저히 감소하였으나 백강잠을 처리한 경우는 이들 효소 활성이 크게 증가하였다. 그리고, $A{\beta}$ 25-35의 농도에 의존적으로 증가된 NO 생성은 백강잠의 농도에 의존적으로 유의성 있게 억제되는 것으로 나타났다. 결론 : 백강잠은 항산화계 효소의 활성화 및 $A{\beta}$처리와 같은 치매유발 물질의 독성에 대한 적응능력 향상을 통하여 astrocyte를 보호하는 효능을 가지는 것으로 사료되며, 아울러 노인성 치매 등 임상적 응용에 그 효과가 기대된다.

Biochemical Changes in Brassica Seedlings Due to Uniconazole Treatment (Brassica속 작물 유묘에서 생장억제제 Uniconazole 처리에 따른 생화학적 변화)

  • In order to obtain the basic data for clarifing the mechanism of cold tolerance in crops, we analyzed various biochemical changes according to the Uniconazole treatment in Brassica seedling. Peroxidase activity in the root fraction of Brassica seedling was about 3 to 4 times higher than that in hypocotyl fraction, while catalase activity in those fractions showed opposite trend to the peroxidase activity. The content of hydrogen peroxide in root fraction was higher than that of hypocotyl fraction as being a reciprocal proportion with catalase activity. Especially in all fractions, peroxidase· activity of 'Sandongchae' (B. campestris) seedling, known as cold tolerant, was two-fold higher than that of cold sensitive rape(B. napus). The elongation rate of hypocotyl after germination was faster in B. napus than in B. campestris. The application of Uniconazole at 0.3 to 1.0 ppm to B. napus suppressed 43 to 46% of hypocotyl elongation and increased 65 to 73% of peroxidase activity in hypocotyl fraction. The shortening rate of hypocotyl length due to Uniconazole treatment was positively correlated with the increasing rate of peroxidase activity in hypocotyl fraction. Superoxide dismutase was not induced upon Uniconazole treatment and has only 3 isozymes in any fractions. Its activity was observed in the order of cotyledon>root>hypocotyl fraction.

  • To study effect of dietary polyunsaturated fatty acid and $\omega$-tocopherol content on lipid peroxidation in rat liver, rats were fed for 3, 6 and 9 weeks with normal tocopherol diet added 40mg of DL-$\alpha$-tocopherol/kg of diet (PF group), high tocopherol eit 200mg of DL-$\alpha$-tocopherol/kg of diet(PFE group), low tocopherol diet without addition of DL-$\alpha$-tocopherol to diet(PFO group), and control diet added 40mg of DL-$\alpha$-tocopherol/kg of diet(control group). Each diet group supplied 45% of total calorie from corn oil except control group which supplied 12% of total calorie from corn oil. After each feeding period, lipid peroxide and tocopherol contents were measured in the liver as well as activities of glutathione peroxidase and superoxide dismutase. PF group had almost the same contents of liver peroxide, tocopherol contents and activities of glutathione peroxidease and superoxide dimutase as control group, while PFE group had higher tocopherol content and glutathione peroxidase activity and lower lipid peroxide contents and superoxide dismutase activity than control group. On the other hand, changes in the values of PFO group were opposite to those of PFE group. Differences in the values among groups were more pronounced as feeding period became longer.

The Effect of UV Blocking Lens on the Denaturation of Antioxidative Enzymes Induced by UV-A (UV-A로 유발된 항산화효소의 변성에 대한 자외선 차단렌즈의 작용)

  • This study was investigated to find the proper UV-A blocking percentage that could protect the denaturation of catalase and superoxide dismutase (SOD), antioxidative enzymes in eye, induced by UV-A. Catalase or SOD were irradiated at 365 nm for 1, 3, 6, 24, 96 hr and the extent of denaturation was evalutated by polyacrylamide gel electrophoresis. Furthermore, it was investigated whether blocking of UV-A by 20, 50, 80 and 99% eyeglass lens could protect the denaturation of catalase and SOD or not. Catalase became to denature when catalase were irradiated by UV-A for more than 3 hours. However, the denaturation of SOD was induced by more than 6 hours irradiation. The denaturation of catalase induced by irradiation for 3 hr could be perfectly protected by 99% UV-A blocking lens. But, when the irradiation time became longer than 3 hr or the blocking percentage of lens were lower than 99%, the denaturation of catalase was not perfectly protected but partially protected. Although 50% UV-A blocking lens had partial protecting effects, lenses having 80 or 99% UV-A blocking effect could perfectly prevent the denaturation of SOD induced by 96 hr irradiation.
