Identification of Interleukin-l Like Material in Streptornyces Culture Supernatant (Streptomyces 배양액에서의 Interleukin-1 유사물질의 동정)

  • We have identified a T cell-activating material in the culture supernatant of Streptomyces species. The factor in microbial culture supernatant (MCS) induced thymocyte proliferation in a does dependent fashion and it could be detected by immunoblot analysis using anti-interleukin-1(IL-1) antibody. The factor in MCS was slightly larger(about 21 kd) in its molecular weight than IL-1 on SDS-PAGE. When 125I-MCS was covalently coupled with homo-bifunctional cross-linking agent, disuccinimidyl-propionate to IL-1 receptor(IL-1R) on mouse thymoma cell(EL-4) and immunoprecipitated with anti-IL-1R antibody the molecular weight of this complex of 110 kd was observed.

Nitrogen and Phosphorus Recoveries from Anaerobic Digester Supernatant Using Seawater as Magnesium Source for Struvite Formation (바닷물을 이용한 struvite 형성으로 혐기성 소화액으로부터 질소, 인 회수)

  • This study was performed to evaluate the effect of pH (8-12) and molar ratio of magnesium and phosphate ($[Mg^{2+}]/[PO_4{^{3-}}]$) (0.6-1.4) on struvite crystallization of anaerobic digester supernatant using seawater as magnesium source. pH range of 9-10 is favorable for ammonium and phosphate recoveries. The recovery efficiency of ammonium was highest at $[Mg^{2+}]/[PO_4{^{3-}}]$ of 1.0 and pH 10. On the other hand, high phosphate recovery efficiency (> 99%) was achieved at ($[Mg^{2+}]/[PO_4{^{3-}}]$) of 1.4 and pH 10. The results demonstrated that seawater can be considered as low-cost magnesium source to recover phosphorus from anaerobic digester supernatant.

A Study for Antimicrobial Susceptibility of Wetlands to Eliminate Toilet Bacteria

  • Purpose: The purpose of this study is to investigate whether wetland has antimicrobial activity on pathogenic bacteria in the toilet bowl. Research design, data and methodology: Air-dried mud obtained from "Jilmoe Bog" wetland was packed and dissolved in the autoclaved saline. Antimicrobial susceptibility was assessed against three Gram-negative bacteria using disk diffusion method and broth dilution method. Identification of specific bacterium presented in wetland supernatant was performed using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Results: Incubation of three Gram-negative bacteria with wetland supernatant inhibited bacterial growth of the bacteria, otherwise increased prevalence of specific bacterium. It was confirmed that Pseudomonas putida was presented in wetland supernatant. Conclusions: The results presented in this study might provide the possibility to utilize wetland supernatant as a bioremediation of toilet bowl bacteria.

  • ScopoIamine and hyoscyamine which belong to tropane alkaloids are the pharmaceutically valuable anticholinergic drugs. In order to increase the productivities, the effects of elicitation were investigated during hairy root cultures of Scopolia. parviflora. Biotic elicitors originated from 3 fungi and 1 yeast were prepared as homogenate and supernatant and added to 3-week-old cultures. Both of homogenate and supernatant of Candida albicans elicitors increased the scopolamine production. The production of hyoscyamine was enhanced by homogenate of Fusarium solani and supernatant of C. albicans. Most of the other fungal elicitors were also improved on the tropane alkaloid production compared to non-treatment. Among the elicitors tested, C. albicans was proved the optimal biotic elicitor on tropane alkaloids production. These results will be served mass production of tropane alkaloids by large-scale production.

Control Effect of Dinoflagellate Bloom by Powder of Marine Rock and Fungus Culture Supernatant (해양암석 분말과 곰팡이 배양액에 의한 적조생물 편조류의 구제효과)

  • To see effect of marine rock powder and fungal culture supernatant, we analyzed the biodegradation rates of harmful marine dinoflagellate, Heterosigma akashiwo and Prorocentrum minimum for developing the effective control methodology of algal bloom. Relatively low removal rates were observed in the treatment of marine rock powder or buffer solution alone. However, the lysis of H. akashiwo and P. minimum was enhanced in the combined treatments of marine rock powder with fungal supernatant. The effective concentration and exposure time of fungal supernatant for the lysis of H. akashiwo and P. minimum were 5 ml/l and 30 minutes, respectively. These results suggest that the fungal supernatant may be a biocontrol agent for the control of algal blooms in seawater.

Nickel Increases Chemotactic Activity of Porcine Peripheral Blood Polymorphonuclear Cells

  • Nickel is a nutritionally essential trace element that plays an important role in the immune system of several animal species. The aim of this study was to examine the effect of nickel chloride on chemotactic activity of peripheral blood polymorphonuclear cells (PMNs) and whether this effect is associated with interleukin (IL)-8 and a nuclear factor-kappa B (NF-κB)-dependent pathway. Peripheral blood mononuclear cells (PBMCs) and PMNs were isolated by Percoll solution (Specific gravity; 1.080) and 1.5% dextran treatment, respectively. A modified Boyden chamber assay was used to measure the chemotactic activity of PMNs. The level of IL-8 in culture supernatant from PBMCs was measured by enzyme-linked immunosorbent assay (ELISA). Both of PBMCs and PMNs exhibited a low viability when cultured with concentration of greater than 1,000 μM of nickel chloride for 24 h. Thus, nickel chloride was used at concentration of 500 μM, which preserved cell viability. Treatment with nickel did not directly affect the chemotactic activity of PMNs. However, the chemotactic activity of PMNs was remarkably increased by culture supernatant from PBMCs treated with nickel chloride (500 μM) for 24 h. Recombinant porcine IL-8 polyclonal antibody (pAb) neutralized the enhancing effect on the chemotactic activity of PMNs by culture supernatant from PBMCs treated with nickel and this culture supernatant had higher IL-8 levels than the culture supernatant from untreated PBMCs. In addition, n-tosyll-phenylalanine chloromethyl ketone (TPCK), a NF-κB inhibitor, antagonized the enhancing effect on the chemotactic activity of PMNs by the culture supernatant from PBMCs treated with nickel. These results suggested that nickel stimulates porcine PBMCs to produce IL-8, which increases the chemotaxis of PMNs via NF-κB-dependent pathway.

Correlation between operation factors and nitritation using anaerobic digester supernatant at ordinary temperature (상온 조건에서 혐기 소화 상징액을 이용한 아질산화 반응과 운전 인자의 상관성 분석)

  • Anaerobic digester supernatant including high concentrations of nitrogen is recycled to water treatment line and make pollutant load increase in municipal wastewater treatment plant(MWTP). To treat nitrogen in anaerobic digester supernatant is suggested the method of MWTP retrofit. In this study, the lab scale reactor was operated about 200 days using supernatant of anaerobic digester. The results could draw operation condition that ammonium nitrogen removal efficiency more than 90% and nitrite conversion efficiency over 70%. Correlation between operation efficiency and operation factors was analyzed based on the operation results. Ammonium nitrogen remove efficiency and nitrite conversion efficiency were related to solid retention time (SRT), ammonium nitrogen load and ammonium nitrogen loading per unit mixed liquer suspended solid (MLSS). Results of this study can be used effective data on nitritation of supernatant of anaerobic digester, and be expected to increase availability of nitritation.

Zinc Increases Chemotactic Activity of Porcine Peripheral Blood Polymorphonuclear Cells

  • Zinc is necessary for normal functions in the immune system. The objective of the study is to examine the effect of zinc on the chemotactic activity of porcine peripheral blood polymorphonuclear cells (PMNs). A modified Boyden chamber was used to determine the directional migration distance of PMNs. Various concentrations of zinc showed no chemotactic activity to PMNs. However, culture supernatant from peripheral blood mononuclear cells (PBMCs) treated with zinc remarkably increased the chemotactic activity of PMNs when compared with culture supernatant from PBMCs treated without zinc. Culture supernatant from PBMCs treated without zinc also increased the migration distance of PMNs relative to vehicle control (medium alone). Increasing effect in chemotactic activity of PMNs by culture supernatant from PBMCs treated with zinc was inhibited by treatment of porcine anti-interleukin (IL)-8 polyclonal antibody (pAb). This effect was not affected by heat treatment ($4-85^{\circ}C$). This corresponded with heat stable physical characteristics of IL-8. These results suggest that zinc can upregulate the chemotaxis of PMNs, which is primary mediated by IL-8 chemotactic factor released from PBMCs treated with zinc.

A study on the diatomaceous earth filtration of recycling basin supernatant in the water treatment plant

  • This study performed the research about the recycling basin supernatant by pre-coat filtration in the D water treatment plant at Gwangju. Choice the prompt conditions with diatomaceous earth filtration which makes contaminant reduced in the basin supernatant. Element disk of candle used in this experiment are pore size 10$\mu\textrm{m}$(R), 20$\mu\textrm{m}$(B) and 40$\mu\textrm{m}$(Y). Diatomaceous earth are cake pore size 3.5$\mu\textrm{m}$(A), 7$\mu\textrm{m}$(B) and 17$\mu\textrm{m}$(C). The filtrate concentrations were from 0.18 to 0.92$\mu\textrm{g}$/1 of Chlorophyll-a. And then, removal rate percentage were from 78.30 to 95.57(R-A). In addition SS 80%, CODMn32% COD 61%, T-N 10% and T-P 39% on the D water treatment plant. The R(40$\mu\textrm{m}$) C(17$\mu\textrm{m}$) process can be substituted of reusing the recycled water of recycling basin supernatant view of capacity and removal rate of filtrate.

Inhibition of Aflatoxin Production of Aspergillus flavus by Lactobacillus casei

  • Lactobacillus casei KC-324 was tested for its ability to inhibit aflatoxin production and mycelial growth of Aspergillus flavus ATCC 15517 in liquid culture. flatoxin $B_{1}$ biosynthesis and mycelial growth were inhibited in both simultaneous culture and individual antagonism assays, suggesting that the inhibitory activity was due to extracellular metabolites. produced in cell-free supernatant fluids of the cultured broth of L. casei KC-324. In cell-free supernatant fluids of all media tested, deMan, Rogosa and Sharpe broth, potato dextrose broth, and Czapek-Dox broth+1% yeast extract showed higher antiaflatoxigenic activity. In these case, fungal growths, however, was not affected as measured by mycelial dry weight. The antiaflatoxigenic metabolites from L. casei KC-324 were produced over wide range of temperatures between $25^{\circ}C$ and $37^{\circ}C$. However, these metabolites were not thermostable since the inhibitory activity of the supernatant was inactivated within 30 minutes at $100^{\circ}C$ and $121^{\circ}C$. The inhibitory activity was not influenced by changing pH of supernatant between 4 and 10. However, the antiaflatoxigenic activity was slightly reduced at pH 10.
