초임계수 처리를 통하여 얻어진 목질계 바이오매스 가수분해 물을 이용한 바이오 에탄올 생산에 대하여 연구하였다. 초임계수 처리 가수분해물은 바이오에탄올 생산을 위한 배지의 탄소원으로 사용되었다. 농축된 초임계수 처리 가수분해물 (SCW3)을 사용하여 효모를 배양하였을 때, 다른 두 가지 초임계수 처리 가수분해물 (SCW1, SCW2) 을 사용한 경우에 비하여 효모의 성장속도가 늦었다. 그리고 모든 경우에 0.51에서 0.56 (%, w/v)의 바이오 에탄올이 생산되었다. 그래서 농축된 초임계수 처리 가수분해물 (SCW3)을 활성탄과 수산화 칼슘으로 전처리하여 페놀류 독성물질을 제거하였다. 활성탄 전처리가 보다 효과적으로 94.6%의 페놀류 화합물을 제거하였고, 바이오 에탄올도 0.96 (%, w/v) 생산 할 수 있게 하였고, 환원당을 기준으로 한 바이오에탄올 수율도 0.5에 이르렀다.
Many studies on plant extracts have been reported for the treatment of candidiasis caused by Candida albicans, a representative fungal infection. This study demonstrates the synergistic antifungal activity of the combination of Phellodendri Cortex and Magnoliae Cortex, previously reported to have antifungal efficacy. Considering the antifungal efficacy and the separation of the active constituents, berberine and magnolol, hot water extraction and carbon dioxide supercritical extraction were selected for Phellodendri Cortex and Magnoliae Cortex, respectively. A combination of 0.55 g/L hot water extract of Phellodendri Cortex and 0.59 g/L carbon dioxide supercritical extract of Magnoliae Cortex showed synergistic antifungal activity. The synergistic antifungal activity of 160 μM berberine and 100 μM magnolol, which are representative antifungal compounds of Phellodendri Cortex and Magnoliae Cortex, respectively, contributes to the synergistic antifungal effect of their extracts. The additive decrease in cellular ergosterol level and the increased antifungal efficacy by extracellular ergosterol suggest that disruption of the biological function of ergosterol in the cell membrane is not responsible for the synergistic antifungal activity of berberine and magnolol. Synergistic cellular release of chromosomal DNA upon mixing berberine and magnolol indicates that disruption of the cellular structure is responsible for the synergistic antifungal effect of berberine and magnolol.
초임계수 산화는 난분해성 유기화합물의 분해를 위한 공정으로 각광받고 있다. 본 연구에서는 연속반응기에서 온도 $387-500^{\circ}C$, 압력 250 bar의 초임계수 조건하에서(EDTA 분해효율) 체류시간 15.9-88.9초의 범위의 EDTA 분해효율을 측정하였다. 이때 산화제로는 과산화수소($H_2O_2$)를 사용하였다. EDTA의 분해효율은 온도 및 산화제투입량의 증가에 따라 상승하였으며, 반응물 도입유속의 감소 즉, 체류시간의 증가에 따라서 상승하였다. 본 연구결과 온도 $500^{\circ}C$, 압력 250 bar, 산화제 투입량 400%의 조건에서 최대 99.6%의 분해효율을 나타내었다. 분해효율에 미치는 온도의 영향이 산화제 투입량 증가의 영향보다 컸으며, 5,000 mg/L의 EDTA($COD_{Cr}$로서 3,063 mg/L)의 99% 이상 분해효율은 온도 $500^{\circ}C$와 압력 250 bar의 초임계수 산화조건에서 산화제투입량 200% 이상 및 체류시간 40.1초 이상에서 얻을 수 있었다.
Paclitaxel is a taxane diterpene amide, which was first extracted from the stem bark of the western yew, Taxus brevifolia. This natural product has proven to be useful in the treatment of a variety of human neoplastic disorders, including ovarian cancer, breast and lung cancer. Paclitaxel is a highly hydrophobic drug that is poorly soluble in water. It is mainly given by intravenous administration. Therefore, The pharmaceutical formulation of paclitaxel ($Taxol^{(R)}$; Bristol-Myers Squibb) contains 50% $Cremophor^{(R)}$ EL and 50% dehydrated ethanol. However the ethanol/Cremophor EL vehicle required to solubilize paclitaxel in $Taxol^{(R)}$ has a pharmacological and pharmaceutical problems. To overcome these problems, new formulations for paclitaxel that do not require solubilization by $Cremophor^{(R)}$ EL are currently being developed. Therefore this study utilized a supercritical fluid antisolvent (SAS) process for cremophor-free formulation. To select hydrophilic polymers that require solubilization for paclitaxel, we evaluated polymers and the ratio of paclitaxel/polymers. HP-${\beta}$-CD was used as a hydrophilic polymer in the preparation of the paclitaxel solid dispersion. Although solubility of paclitaxel by polymers was increased, physical stability of solution after paclitaxel/polymer powder soluble in saline was unstable. To overcome this problem, we investigated the use of surfactants. At 1/20/40 of paclitaxel/hydrophilic polymer/ surfactant weight ratio, about 10 mg/mL of paclitaxel can be solubilized in this system. Compared with the solubility of paclitaxel in water ($1\;{\mu}g/mL$), the paclitaxel solid dispersion prepared by SAS process increased the solubility of paclitaxel by near 10,000 folds. The physicochemical properties was also evaluated. The particle size distribution, melting point and amophorization and shape of the powder particles were fully characterized by particle size distribution analyzer, DSC, SEM and XRD. In summary, through the SAS process, uniform nano-scale paclitaxel solid dispersion powders were obtained with excellent results compared with $Taxol^{(R)}$ for the physicochemical properties, solubility and pharmacokinetic behavior.
In this study, the possibility and the optimal conditions for treating slurry type piggery wastewater using supercritical water oxidation were tested in the laboratory. The results could be summarized as follows; The slurry type piggery wastewater, which was diluted 50 times, was treated most effectively at the pressure of 300 bar, the temperature of $550^{\circ}C$ and the residence time of 10 minutes. The air saturated water was injected, as an oxidizing agent, and the removal efficiencies of $COD_{Cr}$, T-N, $NH_4^+$-N and T-Pattheoptimal conditions were 92, 40, 59 and 100%, respectively. 현사시나무 분말(60 mesh 통과)을 압력 $220{\pm}10atm$, 온도 $325{\sim}425^{\circ}C$ 범위 내에서 60초 간 초임계수로 처리한 후에 미분해 고형잔사를 얻을 수 있었다. 고형잔사 내 섬유상 물질을 구성하는 주요 당은 글루코오스와 자일로스였으며, 가장 높은 온도인 $425^{\circ}C$에서 초임계수 처리를 하였을 때 글루코오스/자일로스 구성 비율이 가장 높게 측정되었다. 초임계수 반응온도가 높아질수록 고형 잔사를 구성하는 섬유상 물질의 비율은 감소하였으나, 리그닌의 비율은 상대적으로 증가하였다. 고형잔사에 존재하는 리그닌의 H (p-hydroxyphenyl) : G (guaiacyl) : S (syringyl) 비율은 분석형 열분해법으로 측정하였으며, 반응온도에 따른 변화 없이 비교적 일정하게 나타났다. 고형잔사를 구성하는 리그닌의 H : G : S 조성을 현사시나무에서 단리한 milled wood lignin (MWL)과 비교해보면 G 형 단량체의 비율에는 큰 변화가 없었지만, H 형 단량체 비율은 비교적 낮게 측정되었고, 반면 S 형 단량체 비율은 증가하였다. 초임계수 당화과정에 염산촉매를 첨가하면 H 비율의 감소와 S 비율의 증가는 더욱 두드러지게 나타났다. 니트로벤젠 산화법(nitrobenzene oxidation)에 의하면 현사시나무 MWL에서 획득한 vanillin과 syringaldehyde의 수율은 약 265 mg/g MWL으로 측정되었지만, 초임계수 고형성분의 NBO 분석 결과에 의하면 반응온도를 높여 주거나 염산촉매를 첨가하면 NBO 분해산물은 두드러지게 감소하였다. 이러한 결과는 초임계수 반응 조건에서 리그닌의 주요 결합 양식인 $\beta$-O-4 결합이 비교적 쉽게 끊어지는 것으로 해석되며, 따라서 초임계수 반응 후 고형성분에 존재하는 리그닌은 $\beta$-O-4 결합 대신 탄소-탄소 결합에 의한 축합형 페놀고분자로 예측되었다.
비이온성 계면활성제인 Ethoxylated Nonyl Phenol 계열(NP-Series)을 초임계 이산화탄소내 마이크로에멀젼 형성 연구에 적용하였다. 초임계 이산화탄소내 용해도 측정결과, 기존에 잘 알려진 친이산화탄소성 계면활성제가 아님에도 안정적인 용해도를 나타내었고, 물과의 마이크로에멀젼 형성에 있어서도 안정적으로 형성되었다. 마이크로에멀젼 형성을 위한 NP-시리즈 계면활성제의 친이산화탄소성기에 대한 친수성기의 길이 배합을 실험을 통해 NP-4 계면활성제(N=4)로 최적화하였다. 마이크로에멀젼 형성을 분광학적 방법인 UV-Visible 스펙트럼을 측정하여 확인하고, 마이크로에멀젼내 물의 존재성도 확인하였다. 금속표면처리나 도금의 적용을 위해 산성용액과 마이크로에멀젼 형성을 실험한 결과, 이온성 계면활성제는 산성용액과의 반응에서 마이크로에멀젼 형성이 불안정해지는 반면, 비이온성 계면활성제는 안정적으로 형성되었다. 본 연구결과는 친환경적 용매인 이산화탄소의 응용분야를 보다 넓힐 수 있을 것이다.
유동층 비중차 분리기를 이용하여 혼합 폐 이온교환수지에서 폐 양이온수지와 폐 음이온수지로 분리하였으며, 원소분석과 열분석으로 분리가 잘 되었음을 확인하였다. 비중차 분리를 통해 얻은 수백 미크론의 구형 알갱이 형태의 폐 양이온수지와 폐 음이온수지 각각을 볼밀로써 24시간 습식 분쇄하여 적정 입도 이하의 입자가 균일하게 분산된 고농도 슬러리를 얻은 다음, 희석하여 $COD_{cr}$, 농도 25,000ppm의 시료를 준비하였다. 실험실 규모의 소형 초임계수산화(Super Critical Water Oxidation, SCWO) 장치(반응기 용량 : 220 mL)로써 화력발전소에서 발생한 폐 양이온수지와 폐 음이온수지에 대해 최적의 분해조건을 확립한 다음, 파일럿플랜트(반응기 용량 : 24 L)로 scale-up 실험을 수행하였다. 우선 화력발전소의 폐수지로서 파일럿테스트를 실시한 다음, 원자력발전소의 폐수지로써 파일럿테스트를 수행하여 최적의 분해조건을 설정하였다. 실험실 규모의 소형 SCWO 장치와 파일럿플랜트의 설계 및 운전에서 얻은 경험을 바탕으로 원자력발전소 내에 설치 가능한 규모로 상용설비(처리용량 : 150kg/h)를 설계 중에 있다.

Defatted bovine liver (DBL) is a potential source of protein and minerals. Supercritical carbon dioxide ($SC-CO_2$) and a traditional organic solvent method were used to remove lipid from bovine liver, and the quality characteristics of a control bovine liver (CBL), bovine liver defatted by $SC-CO_2$ ($DBLSC-CO_2$) at different pressures, and bovine liver defatted by organic solvent (DBL-OS) were compared. The $DBLSC-CO_2$ samples had significantly higher (p<0.05) protein, amino acid, carbohydrate, and fiber contents than CBL and DBL-OS. There was a higher yield of lipid from CBL when using $SC-CO_2$ than the organic solvent method. SDS-PAGE analysis demonstrated that the CBL and $DBLSC-CO_2$ had protein bands of a similar intensity and area, whereas DBL-OS appeared extremely poor bands or no bands due to the degradation of proteins, particularly in the 50 to 75 kDa and 20 to 25 kDa molecular weight ranges. In addition, $DBLSC-CO_2$ was shown to have superior functional properties in terms of total soluble content, water and oil absorption, and foaming and emulsification properties. Therefore, $SC-CO_2$ treatment offers a nutritionally and environmentally friendly approach for the removal of lipid from high protein food sources. In addition, $SC-CO_2$ may be a better substitute of traditional organic solvent extraction for producing more stable and high quality foods with high-protein, fat-free, and low calorie contents.

With the recent rapid improvement in the standards of life and westernization of dietary lifestyles, the consumption of high-calorie diets such as high-fat and high-protein red meat and instant foods has increased, while less vegetables containing dietary fiber are consumed. In addition to that, stress, erroneous dietary behaviors, and contaminated environments are linked to the risk of developing ulcerative colitis, which is on the rise. Another cause of ulcerative colitis is that involve laxative abuse, including repeated, frequent use of laxatives, and include such conditions as deteriorated bowel function, irritable bowel syndrome, diarrhea, intestinal inflammation, etc. The present study aimed to investigate the comparative evaluation of pharmacological efficacy between sulfasalazine alone and combination with herbal medicine on dextran sodium sulfate (DSS)-induced UC in mice. Balb/c mice received 5% DSS in drinking water for 7 days to induce colitis. Animals were divided into five groups (n = 9): group I-normal group, group II-DSS control group, group III-DSS + sulfasalazine (30 mg/kg), group IV-DSS + sulfasalazine (60 mg/kg), group V-DSS + sulfasalazine (30 mg/kg) + Radish Extract mixture (30 mg /kg) (SRE). DSS-treated mice developed symptoms similar to those of human UC, such as severe bloody diarrhea and weight loss. SRE supplementation, as well as sulfasalazine, suppressed colonic length and mucosal inflammatory infiltration. In addition, SRE treatment significantly reduced the expression of pro-inflammatory signaling molecules through suppression both mitogen-activated protein kinases (MAPK) and nuclear factor-kappa B (NF-κB) signaling pathways, and prevented the apoptosis of colon. Moreover, SRE administration significantly led to the up-regulation of antioxidant enzyme including SOD and Catalase. This is the first report that Radish extract mixture combined with sulfasalazine protects against experimental UC via the inhibition of both inflammation and apoptosis, very similar to the standard-of-care sulfasalazine.
