Butyrate-induced differentiation of PC12 cells to chromaffin cells involves cell adhesion and induction of extracellular proteins and cell adhesion proteins

  • PC12 cells were differentiated into the cells of chromaffin phenotype by butyrate treatment. Cells were aggregated and formed tight cell adhesion. To investigate the molecular change in this differentiation, we examined expression levels of cell adhesion proteins and extracellular proteins during butyrate induced-differentiation of PC12 cells. Integrin ${\beta}1$, integrin ${\alpha}7$, E cadherin, VCAM, collagen-I, fibronectin, desmoglein and connexin were increased during differentiation. The levels of clusterin and secreted clusterin were also increased. These increased levels of cell adhesion proteins and extracellular proteins appear to induce cell aggregation and tight cell adhesion. The levels of p21, p27 and p16 were increased probably because of differentiation-related growth arrest during differentiation. Prolonged incubation of butyrate up to 1 day was required for differentiation. Signal transduction pathways for this differentiatiom could not be identified since various inhibitors had no effect. The results showed that butyrateinduced differentiation of PC12 cells to chromaffin cells involves tight cell adhesion and induction of extracellular proteins and cell adhesion proteins.

  • Purecell, hydroxyethyl-cellulose-2-hydroxypropyltrimethyl ammonium chloride ether, was developed as a thickener and evaluated its functions for stability and safety when it was added in the cosmetic composition. Compared with a commercially available thickener Natrasol, Purecell have a basic chemical backbone of hydroxyethylcellulose which has been widely used in the cosmetic industry. pH and viscosity of Purecell was lower than those of Natrasol, however, a similar pH value to was shown when the Purecell was added into a shampoo or cream. The Purecell was stable to temperature and light. When 0.5% of the Purcell was added to a shampoo having cationic surfactant, a separation occurred due to the same cationic surfactant of the Purecell and shampoo. However, the shampoo and cream made with Purecell improved a smoothness and adaptability of hair. It also showed a negative response for Patch test of human, indicating the Purecell was safe for use.

Enhanced biological effects of Phe140Asn, a novel human granulocyte colony-stimulating factor mutant, on HL60 cells

  • Granulocyte colony-stimulating factor (G-CSF) is a cytokine secreted by stromal cells and plays a role in the differentiation of bone marrow stem cells and proliferation of neutrophils. Therefore, G-CSF is widely used to reduce the risk of serious infection in immunocompromised patients; however, its use in such patients is limited because of its non-persistent biological activity. We created an N-linked glycosylated form of this cytokine, hG-CSF (Phe140Asn), to assess its biological activity in the promyelocyte cell line HL60. Enhanced biological effects were identified by analyzing the JAK2/STAT3/survivin pathway in HL60 cells. In addition, mutant hG-CSF (Phe140Asn) was observed to have enhanced chemoattractant effects and improved differentiation efficiency in HL60 cells. These results suggest that the addition of N-linked glycosylation was successful in improving the biological activity of hG-CSF. Furthermore, the mutated product appears to be a feasible therapy for patients with neutropenia.

Do Trunk Muscles Affect the Lumbar Interbody Fusion Rate? : Correlation of Trunk Muscle Cross Sectional Area and Fusion Rates after Posterior Lumbar Interbody Fusion Using Stand-Alone Cage

  • Objective : Although trunk muscles in the lumbar spine preserve spinal stability and motility, little is known about the relationship between trunk muscles and spinal fusion rate. The aim of the present study is to evaluate the correlation between trunk muscles cross sectional area (MCSA) and fusion rate after posterior lumbar interbody fusion (PLIF) using stand-alone cages. Methods : A total of 89 adult patients with degenerative lumbar disease who were performed PLIF using stand-alone cages at L4-5 were included in this study. The cross-sectional area of the psoas major (PS), erector spinae (ES), and multifidus (MF) muscles were quantitatively evaluated by preoperative lumbar magnetic resonance imaging at the L3-4, L4-5, and L5-S1 segments, and bone union was evaluated by dynamic lumbar X-rays. Results : Of the 89 patients, 68 had bone union and 21 did not. The MCSAs at all segments in both groups were significantly different (p<0.05) for the PS muscle, those at L3-4 and L4-5 segments between groups were significantly different (p=0.048, 0.021) for the ES and MF muscles. In the multivariate analysis, differences in the PS MCSA at the L4-5 and L5-S1 segments remained significant (p=0.048, 0.043 and odds ratio=1.098, 1.169). In comparison analysis between male and female patients, most MCSAs of male patients were larger than female's. Fusion rates of male patients (80.7%) were higher than female's (68.8%), too. Conclusion : For PLIF surgery, PS muscle function appears to be an important factor for bone union and preventing back muscle injury is essential for better fusion rate.

  • 본 논문에서는 문자출력이 가능한 무선호출기에 음성인식 기술을 접목한, 특성화된 한 음성인식 시스템에 대하여 설명하고자 한다. 시스템 동작 과정은, 일단 호출자가 음성인식 서버와 접속하게 되면 서버는 호출자의 자연스런 입력음성을 인식, 그 결과를 문장 형태로 피호출자의 호출기 단말기에 출력시키는 방식으로 되어 있다. 본 시스템에서는 통계적 음성인식 기법을 도입하여, 각 단어를 연속 HMM으로 모델링하였다. 가우시안 혼합 확률밀도함수를 사용하는 각 모델은 전통적인 HMM 학습법들 중의 하나인 Baum-Welch 알고리듬에 의해 학습되고 인식시에는 이들에 비터비 빔 탐색을 적용하여 최선의 결과를 얻도록 한다. MFCC와 파워를 혼용한 26 차원 특징벡터를 각 프레임으로부터 추출하여, 최종적으로, 83 개의 도메인 어휘들 및 무음과 같은 특수어휘들에 대한 모델링을 완성하게 된다. 여기에 구문론적 기능과 의미론적 기능을 함께 수행하는 FSN을 결합시켜 자연발화음성에 대한 연속음성인식 시스템을 구성한다. 본문에서는 이상의 사항들 외에도 음성 데이터베이스, 레이블링 등과 갈이 시스템 성능과 직결되는 시스템의 외적 요소들에 대해 고찰하고, 시스템에 구현되어 있는 다양한 특성들에 대해 밝히며, 실험 결과 및 앞으로의 개선 방향 등에 대해 논의하기로 한다.

Distribution patterns of Monochamus alternatus and M. saltuarius (Coleoptera: Cerambycidae) in Korea

  • Distribution patterns of two pine sawyer species (Monochamus alternatus which is the main vector insect and M. saltuarius which is the potential insect vector of the pine wood nematode) were investigated in Korea. The data were collected at 89 study sites which were chosen to cover the whole region of South Korea. The selected pine trees were killed in early April and left for I year in the pine stands to be egg-laid by the pine sawyers. Emergence of the beetles from the dead pine trees was checked from early April to late July. M. saltuarius was the most abundant in the mid to northern areas of South Korea, whereas M. alternatus in Jeju-do, southernmost island of Korea. Considering temperature distribution patterns in areas where the two species occur, their thermal distribution boundary may be formed around $13.2^{\circ}C$ of annual mean temperature. The hypothesized distribution map of the two Monochamus species under the invasion of pine wilt disease is suggested on the base of thermal distribution of Korean peninsula.

  • The Marfans syndrome is a generalized connective tissue disease involving eye, musculoskeletal system, cardiovascular system, and inherited autosomal dominant with various expression type. The cardiovascular complications such as aortic aneurysm, aortic dissection, aortic regurgitation, mitral regurgitation and aortic dissection which usually occurs in previously normal sized aorta are poor prognostic factors. However, the aortic dissection which developed in patient with Marfan syndrome and aortic aneurysm was rare. We experienced one case of dissecting aneurysm in patient diagnosed as previous aoritc aneurysm, aortic regurgitation, and Marfan syndrome, receiving successful operation.

Performance Comparison of Continuous Reactors for Bioethanol Production Based on Glycerol (글리세롤 기반의 바이오에탄올 생산을 위한 연속생산반응기의 성능 비교)

  • Ethanol production using glycerol as a carbon source was performed by Enterobacter aerogenes immobilized on calcium alginate beads. To improve the ethanol production, the optimal conditions such as loading amount of immobilized cells and glycerol concentration were investigated. The optimal loading amount of immobilized cells and glycerol concentration were 10 mL of calcium alginate bead and 10 g/L, respectively. Consequently, glycerol consumption rate, ethanol concentration and yield were 0.32 g/$L{\cdot}h$, 3.38 g/L and 0.43 g/g on the batch production, respectively. Continuous production of ethanol was successfully achieved using two types of immobilized cell reactors (continuous stirred tank reactor and packed bed reactor) from 10 g/L of glycerol. In the continuous stirred tank reactor, glycerol consumption, ethanol concentration, specific productivity and yield were 9.8 g, 4.67 g/L, 1.17 g/$L{\cdot}h$, 0.48 g/g, respectively. The concentration of produced ethanol was 38-44% higher comparison to batch fermentation, and continuous stirred tank reactor showed better performance than packed bed reactor.

  • The antimicrobial activity of 70% ethanol extracts from Rosa multiflora Thunberg fruit against Helicobacter pylori was examined. The inhibitory activity of Rosa multiflora Thunberg fruit extracts against H. pylori was determined to clear a zone of 14 mm with 70% ethanol extracts. Purification of inhibitory compounds was carried on Sephadex LH-20 and $C_{18}$ cartridge column chromatography using a gradient procedure, with increasing ethanol ($0{\rightarrow}100%$) in $H_2O$. The chemical structure of the purified inhibitory compounds on H. pylori was identified to be protocatechuic acid, chlorogenic acid and quercetin by FAB-MS, NMR and IR spectrum.

  • Diffusion tensor tractorgraphy which is based on line propagation method with brute force approach is implemented and the vector correlation function is proposed in addition to the conventional fractional anisotrophy value as a criterion to select seed points. For the whole tractography, the proposed method used 41 % less seed points than the conventional brute force approach for $FA{\geq}0.3$ and most of the fiber tracks in the outer region of white matter were removed. For the corticospinal tract passing through region of interest, the proposed method has produced similar results with 50% less seed points than conventional one.
