The Relationship among Sun-Screening Agent Use, Bone Health Promotion Behavior and Bone Mineral Density of Female College Students (여대생의 자외선 차단제 사용과 골 건강증진행위 및 골밀도와의 관계)

  • Purpose: The purpose of this study is to investigate relationships among sun-screening agents use, bone health promotion behavior, and bone mineral density. Methods: The participants of this study were 105 female college students. The data were collected using a questionnaire about sun-screening agent use and bone health promotion behavior. Bone mineral density was measured with Ultrasound Bone Densitometer/Osteo Pro Series. Data were analyzed with the SPSS/Win 18.0 program. Results: The mean Z-score in female college students was $-1.04{\pm}1.26$, and the T-score was $-1.16{\pm}1.22$, 21.39% of the participants have osteopenia. Bone mineral density showed a significant correlation with weight. Bone mineral density showed no significant correlation between sun-screening agent use, and bone health promotion behavior. There was a significant correlation between protection power of sun-screening agents and the protection by clothes. Conclusion: According to the results of this study, we need to develop an intervention program for bone health promotion of female college students.

  • UV-screening agents have some adverse effects that raise consumers' concern. The organic agents often cause irritation and may penetrate into human body while the inorganic agents raise aesthetic issues because they often turn opaque. Organic agents with high molecular weights and nano-sized inorganic agents have been developed respectively to minimize transdermal intrusion into human body and suppress turning opaque. Recently, we reported preparation of powdery UV-screening agents made of polysilsesquioxane, an organic-inorganic hybrid material. Powders would not penetrate into epidermis and organic-inorganic hybrid nature would suppress the opaqueness problem. In this study, we continued our research on this powdery polysilsesquioxane UV-screening agent, SESQUV, regarding its chemical composition, its synergic UV-screening effects when mixed with other organic agents, and its applicability in practical formulation. Results showed SESQUV was promising UV-screening agents useful in sunscreen formulation.

  • Diazepam (DZ) is one of the most frequently prescribed drugs as an antianxiety agent, muscle relaxant, and anticovulsant and sometimes causes intoxication due to accidental overdose, misuse or abuse. Screening or confirmation methods for DZ and NDZ in plasma are very important for clinical and toxicological studies and in forensic cases. GC/MS assay with SPE was developed for the determination of diazepam and its metabolite, nordiazepam in human plasma. Diazepam in plasma was extracted by a rapid and sensitive procedure based on C18 bonded-phase extraction. (omitted)

  • Sunscreen is divided into the organic agent of UV absorption and inorganic agent of reflection. These are evaluated by sun protection factor (SPF) in-vivo test requiring high cost and time, while in-vitro tests are adopted commonly because of short test time, easy result collection. Generally, test method of SPF use SPF 290a by UV-vis spectrometer. The evaluate by SPF 290 has low reproducibility. Although analysis using UV-vis spectrometer has high reproducibility, it is hard to separated results of transmission, adsorption, and reflection. In this study, suggested method of elipsometer has some merit such as high reproducibility, easy separation of transmission/adsorption/re- flection, analysis using various incident angle. We tested the validity of elipsometer for SPF measurement, using commercially available sun-block (SPF 50).

  • Recently, we developed a new anti-osteoporotic agent, DW-1350, which not only inhibited osteoclast formation but also induced osteoblast differentiation through the in vitro randomized screening studies. We identified inhibitory activities of DW-1350 for each step of osteoclast differentiation, fusion and pit formation process in co-culture system with mouse bone marrow and primary osteoblasts.   • An attempt was made to develop a proteinchip for screening of MITF (microphthalmia transcription factor) inhibitor. Binding of MITF to E-box causes transcription of several pigmenting genes including tyrosinase gene. We investigated binding of MITF and its DNA binding site (E-box) using a protein-DNA chip with various detection methods including flurorescence (Cyt3). SPR (surface plasmon resonance) and SPRi (surface plasmon resonance imaging). A fusion protein (MITF-Maltose Binding Protein) was attached on the glass plate by chemical modification. An inhibitory synthetic DNA oligomer, artificially designed based on the E-box sequence, inhibited the binding of MITF and E-box. These results showed the potentials of flurorescence-based MITF protein chip as a microarray for high throughput screening (HTS) system of depigmenting agents.

Screening of $\alpha$-Amylase and $\alpha$-Glucosidase Inhibitor from Nepalese Plant Extracts (100종 네팔 식물 추출물로부터 $\alpha$-Amylase 및 $\alpha$-Glucosidase저해제의 선별)

  • In the course of screening for anti-acidosis and anti-diabetes agent from natural products, the inhibitory activities of Nepales plant extracts against microbial $\alpha$-amylase and $\alpha$-glucosidase were evaluated. Among the 100 different kinds of ethanol extracts, Cedrus deodara (Roxb.) G. Don and Myrica nagi Thunb showed strong inhibition activities against $\alpha$-amylase. The $IC_{50}$ values of C. deodara (Roxb.) G. Don, M. nagi Thunb and acarose, a commercial available anti-diabetes agent, were 44.5, 47.5 and $50.5\;{\mu}g/mL$, respectively. Considering the crude extract of C. deodara (Roxb.) G. Don, and M. nagi Thunb, these extracts have strong potentials as anti-acidosis or anti-diabates agent. In a while, Cleistocalyx operculatus (Roxb.) extract showed a good inhibition activity to $\alpha$-amylase and $\alpha$-glucosidase, even it was recently reported. The selected three extracts did not show any hemolysis activity against human red blood cell up to 1 mg/mL, and the inhibition activities were maintained by heat or acid treatment. Moreover, treatment of HCl (0.01N) for 1 h into C. operculatus (Roxb.) and M. nagi Thunb increased the inhibition activity from 50% to 70%. Our results suggest that C. deodara (Roxb.) G. Don, M. nagi Thunb, and C. operculatus (Roxb.) are potential as anti-acidosis and anti-diabetes agents.

Screening of Myxobacteria Inhibiting the Growth of Collectotrichum acutatum Causing Anthracnose on Pepper (고추탄저균 성장 억제 점액세균의 탐색)

  • As an effort to search new bacterial biocontrol agents against pepper anthracnose, we screened myxobacteria, which might inhibit the growth of Colletotrichum acutatum, the agent of that plant disease. When 93 myxobacterial strains including 59 Myxococcus spp. and 34 Corallococcus spp. were tested against C. acutatum ACYSJ001 on agar plates, 10 strains identified as the genus Myxococcus significantly obstructed the growth of C. acutatum, whereas the majority of strains belonging to the genus Corallococcus did not demonstrate any counteractive effect. Such results have indicated that the strains of the genus Myxococcus have a high potential to play roles of biocontrol agents for control of pepper anthracnose. These also have revealed that the strains of the genus Myxococcus could be used as excellent microbial resources for screening novel antifungal substances.

Draft Genome Analysis of Antimicrobial Streptomyces Isolated from Himalayan Lichen

  • There have been several studies regarding lichen-associated bacteria obtained from diverse environments. Our screening process identified 49 bacterial species in two lichens from the Himalayas: 17 species of Actinobacteria, 19 species of Firmicutes, and 13 species of Proteobacteria. We discovered five types of strong antimicrobial agent-producing bacteria. Although some strains exhibited weak antimicrobial activity, NP088, NP131, NP132, NP134, and NP160 exhibited strong antimicrobial activity against all multidrug-resistant strains. Polyketide synthase (PKS) fingerprinting revealed results for 69 of 148 strains; these had similar genes, such as fatty acid-related PKS, adenylation domain genes, PfaA, and PksD. Although the association between antimicrobial activity and the PKS fingerprinting results is poorly resolved, NP160 had six types of PKS fingerprinting genes, as well as strong antimicrobial activity. Therefore, we sequenced the draft genome of strain NP160, and predicted its secondary metabolism using antiSMASH version 4.2. NP160 had 46 clusters and was predicted to produce similar secondary metabolites with similarities of 5-100%. Although NP160 had 100% similarity with the alkylresorcinol biosynthetic gene cluster, our results showed low similarity with existing members of this biosynthetic gene cluster, and most have not yet been revealed. In conclusion, we expect that lichen-associated bacteria from the Himalayas can produce new secondary metabolites, and we found several secondary metabolite-related biosynthetic gene clusters to support this hypothesis.

Screening of Antibrowning Agents for Minimally Processed Vegetables (최소가공채소류에 적합한 갈변방지제의 선발)

  • Peeled garlic, soybean sprouts, cut onion and cut green pepper were treated by dipping in solutions of different antibrowning agents, and then stored at $10^{\circ}C$. Surface color and polyphenol oxidase activity of produce were measured through the storage. Tested antibrowning agents include ascorbic acid, citric acid and allyl isothiocyanate. The commodities showed different responses to the antibrowning agent solutions; 1% citric acid for peeled garlic, 1% ascorbic acid for soybean sprouts, 2% citric acid for cut onion, and 1% ascorbic acid for cut green pepper showed better retardation in browning than the other treatments for respective product. For peeled garlic, surface browning was concomitant with increase in polyphenol oxidase activity during storage. On the other hand, there was no positive correlation between surface browning and polyphenol oxidase activity for the other stored products.
