This study was conducted to determine the influence of postharvest dipping treatment with aminoethoxyvinylglycine (AVG) on ethylene production and composition of non-cellulosic neutral sugars in cell walls of 'Tsugaru' apple fruits during storage. Fruits were harvested on August 20, soaked in AVG 50 and 75 $mg L^{-1}$ solution for 5 minutes, and stored in cold storage chamber at $0{\pm}1^{\circ}C$ for 60 days. Fruit quality factor, ethylene productions, and cell wall component changes were investigated at 20 days interval. As a result, the fruit firmness and acid content were much higher in AVG treated fruits than those of untreated one during 60 days of cold storage. Ethylene production of AVG treated fruits was reduced to the level of 1/10 compared with untreated one. As to the change of non-cellulosic neutral sugars in the cell walls of 'Tsu- garu' fruits, the major sugar was arabinose and galactose in water, CDTA and $Na_2CO_3$ soluble fractions. The content of arabinose and galactose in untreated fruits increased as the softening of fruits was in progress, but the fruits treated with AVG showed a little change during storage, so it is predicted that these two cell wall compositional sugars were not solubilized by the treatment of AVG. Accordingly, the marketability of 'Tsu- garu' fruits could remarkably increase when soaking the fruits in AVG solution after harvest.
This experimental was undertaken for the purpose to investigate the different chemical composition in developing fruit of Poncirus trifoiata. In the first place, the differences among the content of tannin and qualitative analysis of free sugars and organic acids in the fruit of Poncirus trifoiata fron Kwang-ju are to be investigated and chemical components are analyzed. 1) The differences of the content of tannin are investigated at the four periods of time and gradual decrease in the sample 2 (July 15) take place. 2) In sample 1 (June 20), xylose, mannose and rhamnose are not identified and later, as the fruit age, otehr three sorts of sugars with the former are founded and there is a conspicuous tendency to increase in the content and especially the increase in glucose and rhamnose seems to be related with the decomposition of tannin, while the decrease in the former from sample 2 to sample 4 with the formation of the latter. 3) Tartaric acid, succinic acid and citric acid are appeared and then in sample 1 (June 20( these are not founded and later all of them are identified and gradual increase in the content take place and especially citric acid and succinic acid seems to be increased in the content. These are related with the synthesis of organic materials in plant body.
창포 뿌리를 주근(main mot)과 측근(lateral root)으로 분리하여 열풍건조 분쇄, 분말화한 후 일반성분, 당 및 아미노산등 화학성분을 분석하여 식품학적 특성을 검토하였다. 주근과 측근 건조분말에는 각각 조단백질 12.76%, 9.84%와 조지방 7.43%, 3.42%로 높게 함유하고 있어 에너지공급원과 식물성 고단백질 원료로서의 우수한 영양학적 가치를 가지고 있음을 확인하였다. 유리당으로는 sucrose, stachyose, raffinose, glucose 및 fructose 등으로, 특히 sucrose가 주근과 측근에 각 10.85%와 12.27%로 높게 함유하고 있었다. 창포뿌리 주근의 건조분말은 총아미노산 함량은 426.81 mg%이었고, 특히 arginine 78.91 mg%와 glutamic acid 69.73 mg%로 높은 함량 수준을 보였다. 측근 건조분말의 총아미노산 함량은 470.13 mg%로 주근 보다 다소 높은 함량을 나타내었고, 특히l arginine이 116.91 mg%로 주근의 arginine 함량보다 매우 높은 함량 수준을 보였다. 주된 유리아미노산으로는 asparagine, arginine, glutamic acid, phenylalanine 등의 순으로 높게 함유되어 있고, 특히, 주근 건조분말에는 asparagine이 128.53mg%, 측근 건조분말에는 arginine이 86.10 mg%로 높은 함량 수준을 나타내었다. 필수아미노산 함량은 유리아미노산의 경우는 phenylalanine, threonine, valine, leucine 등이 높게 함유되어 있었으며, 특히, 주근 건조분말에는 threonine이 15.62 mg%, 측근 건조분말에는 phenylalanine 12.67 mg%로 타 필수 아미노산에 비해 매우 높은 함량 수준을 나타내었다.
Sea-buckthorn fruits (Hippophae rhamnoides L.) are used in Mongolia as traditional medicine due to the health-benefits associated with its bioactive components. The purpose of this study was to investigate the chemical composition of Mongolian sea-buckthorn fruits. In terms of proximate composition, crude fat content was the highest, whereas its crude ash content was the lowest. In organic acid contents, malic acid content ($6.30{\pm}0.005$ mg/g) was the highest. Free sugars were composed of sucrose, xylose, glucose and mannitol. Vitamin C and carotenoid content were 320 mg/100 g and 715.25 mg/100 g, respectively. The major fatty acids were palmitic (C16:0), palmitoleic (C16:1), caprylic (C8:0) and linoleic (C18:2) acid.
DagA, a ${\beta}$-agarase, was produced by cultivating a recombinant Streptomyces lividans in a glucose medium or a mixed-sugar medium simulating microalgae hydrolysate. The optimum composition of the glucose medium was identified as 25 g/l glucose, 10 g/l yeast extract, and $5g/l\;MgCl_2{\cdot}6H_2O$. With this, a DagA activity of 7.26 U/ml could be obtained. When a mixed-sugar medium containing 25 g/l of sugars was used, a DagA activity of 4.81 U/ml was obtained with very low substrate utilization efficiency owing to the catabolic repression of glucose against the other sugars. When glucose and galactose were removed from the medium, an unexpectedly high DagA activity of about 8.7 U/ml was obtained, even though a smaller amount of sugars was used. It is recommended for better substrate utilization and process economics that glucose and galactose be eliminated from the medium, by being consumed by some other useful applications, before the production of DagA.
This study was carried out to investigate the compositions of free sugars, lipids, and nonvolatile organic acids in parts of omija. The major components of free sugars in each part of omija were fructose and glucose. The contents of those were similar in fruits and endocarps, however, the content of glucose was 1.5 times as much as that of fructose in seeds. The content of lipids in endocarps was 2.4 times as much as that in seeds, and major composition of lipids was neutral lipid. In the contents of nonvolatile organic acids, the content of citric acid that content was 61 to 68% depend on each part of sample was highest among other components, and that of malic acid being 25 to 30% was followed. The contents of nonvolatile organic acids of water extract were 74.5, 55.9, and 69.2% as high as those of original sample in fruits, endocarps, and seeds, respectively. The content of oxalic acid in seeds was lower than that of it in original sample.

Objective: A study was conducted to determine the chemical composition of barley and co-products from barley, corn, and wheat produced in South-East Asia or Australia, and to test the hypothesis that production area or production methods can impact the chemical composition of wheat co-products. Methods: Samples included seven barley grains, two malt barley rootlets, one corn gluten feed, one corn gluten meal, one corn bran, eight wheat brans, one wheat mill mix, and four wheat pollards. All samples were analyzed for dry matter, gross energy, nitrogen, amino acids (AA), acid hydrolyzed ether extract, ash, minerals, starch, and insoluble dietary fiber and soluble dietary fiber. Malt barley rootlets and wheat co-products were also analyzed for sugars. Results: Chemical composition of barley, malt barley rootlets, and corn co-products were in general similar across countries. Wheat pollard had greater (p<0.05) concentrations of tryptophan, magnesium, and potassium compared with wheat bran, whereas wheat bran had greater (p<0.05) concentration of copper than wheat pollard. There were no differences in chemical composition between wheat bran produced in Australia and wheat bran produced in Thailand. Conclusion: Intact barley contains more starch, but fewer AA, than grain co-products. There were only few differences in the composition of wheat bran and wheat pollard, indicating that the two ingredients are similar, but with different names. However, corn gluten meal contains more protein and less fiber than corn bran.
Mono- and oligosaccharides are derivatized with p-aminobenzoic ethyl ester (ABEE), strongly absorbs UV light at 254 nm, in the presence of sodium cyanoborohydride. C18-bonded silica column is used for the separation of sugar-ABEE derivatives in an isocratic mode. RP-HPLC conditions are optimized by using ternary mixture as mobile phase and $45^{\circ}C$ as a column temperature. Sugar-ABEE derivatives are separated well within a short run time (ca. 25 min) by reverse-phase partition chromatographic mode. The ($1{\rightarrow}6$) linkage type of dihexose-ABEE derivatives has shorter retention time than ($1{\rightarrow}4$)-linkage type. After acid hydrolysis of glycoproteins with 2M trifluoroacetic acid, monosaccharide composition and contents are determined. This procedure is very useful for the simultaneous analysis of neutral and amino sugars in a single chromatographic step using RP-HPLC without reacetylation of deacetylated amino sugars, which are produced by acid hydrolysis.
늙은 호박의 부위별 이화학적 특성 연구로서 호박의 유리당, 유기산, 아미노산, 카로데노이드 및 펙틴물 질의 함량과 특성을 조사하였다. 호박의 중량에 대해 과육부위가 84%로 가장 높은 함유율을 나타내었고, 껍질이 10%, 내부 섬유상물질이 3.5%를 차지하였다. 호박의 유리당은 fructose, glucose, sucrose로 구성되어 있었으나 과육부는 fructose와 glucose가 전체 유리당의 87%를 차지한 반면 내부 섬유상물질은 sucrose의 함량이 높았고, 총 유리당 함량은 과육부가 섬유상부위 보다 3배 높았다. 유리아미노산의 경우 과육부는 serine이 102.0mg%로 전체 유리아미노산의 45%, 섬유상부위는 aspartic acid가 241.3mg%로 70%를 차지하였으며, 총 아미노산은 호박의 부위에 관계없이 aspartic, glu-tamic acid의 순으로 함량이 높았다. 유기산은 malic, cit-ric, succinic, fumaric acid로 구성 되어 있었으나 malic acid가 과육의 경우 85.2mg%, 섬유상 부위의 경우 226.5mg%로서 전체 유기산의 63~70%를 나타내었다 총 카로테노이드의 함량은 내부 섬유상물질이 65.3mg%로 호박 총 카로테노이드의 87%를 차지하였고, 과육, 껍질은 각각 6.6, 3.3mg%였다. 호박의 가용성 펙틴 함량은 수용성 펙틴이 총가용성 펙틴의 58%를 차지하였고, 섬유상부위가 과육부에 비해 그들의 총 함량이 높았다. 가용성 펙틴의 분자량 분포를 조사한 결과 과육부의 펙틴이 섬유상에 비해 다소 분자량이 높은 것으로 나타났다. 호박펙틴의 중성당은 rhamnose, arabinose, galactose, glucose로 분리되었고, 이중 과육은 galactose가 45%, 섬유상은 glucose가 36.5%로 가장 높은 값을 나타내었다.
This experiment was carried out to elucidate the sugar composition of polysaccharides and the structural features of water-soluble polysaccharides(WSP) isolated from Korean oak mistletoe, Loranthus yadoriki Sieb. The 48-hours ball-milled meals of extractive-free dried mistletoe sawdusts were extracted with distilled water for $24hrs{\times}2$ at room temperature. The extracts poured into 95% ethyl alcohol to precipitate. The separated precipitate of WSP, in form of yellowish white powder by lyophilization, was fractionated into four subfractions of WSP-1, WSP-2, WSP-3 and WSP-4 by anion exchange chromatography on DEAE-cellulose column. The sugar composition of WSPs was analyzed by GLC in form of their glycitol acetates, and the structure of polysaccharides in Fractions WSP-1 and WSP-2 was determined by FT-IR and GC-MS after methylation through and acetylation. The sugars of WSPs from Korean oak mistletoe, Loranthus yadoriki, are majorly arabinose and galactose in stem, galactose in leaves very high in content and showed difference in composition and monomeric units between stems and leaves. D-galactose, D-glucose and L-arabinose are the simple sugars consisting of polysaccharides in WSP-1. ($1{\rightarrow}3$)-Linked galactan is the bakcbone with side chain of ($1{\rightarrow}5$)- -L-arabinofuranosyl residues and ($1{\rightarrow}6$)- -D-galactopyranosyl residues, and ($1{\rightarrow}4$)-linked glucan also presents. ($1{\rightarrow}4$)-Linked rhamnogalacturonan and ($1{\rightarrow}4$)- and ($1{\rightarrow}3$)-linked galactan present in WSP-2.
