Chemical Doping of $TiO_2$ with Nitrogen and Fluorine and Its Support Effect on Catalytic Activity of CO Oxidation

  • The effect of substrate on catalytic activity of CO oxidation with transition metal Platinum nanoparticles on doped and undoped TiO2 was investigated. Titanium dioxide was doped chemically with non-metal anions including nitrogen and fluorine. Undoped TiO2 was synthesized via simple conventional sol-gel route. Thin films of titania were developed by spin coating technique and the characterization techniques SEM, XRD, UV-Vis Absorption Spectroscopy and XPS were carried out to examine the morphology of films, crystal phase, crystallites, optical properties and elemental composition respectively. XPS analysis from doped TiO2 confirmed that the nitrogen site were interstitial whereas fluorine was doped into TiO2 lattice substitutionally. Catalytic activity systems of Pt/doped-TiO2 and Pt/undoped-TiO2 were fabricated to reveal the strong metal-support interaction effect during catalytic activity of CO oxidation reactions. By arc plasma deposition technique, platinum nanoparticles with mean size of 2.7 nm were deposited on the thin films of doped and undoped titanium dioxide. The CO oxidation was performed with 40 Torr CO and 100 Torr O2 with 620 Torr He carrier gas. Turn over frequency was observed two to three folds enhancement in case of Pt/doped TiO2 as compared to Pt/TiO2. The electronic excitation and the oxygen vacancies that were formed with the doping process were the plausible reasons for the enhancement of catalytic activity.

Hydrothermal Synthesis of $TiO_2$ Nanowire Array for Osteoblast Adhesion

  • Osteoblast is one of cells related with osseointegration and many research have conducted the adhesion of osteoblast onto the surface of implant. In the osseointegration, biocompatibility of the implant and cell adhesion to the surface are important factors. The researches related to cell adhesion have a direction from micro-scaled surface roughness to nano-scaled surface roughness with advancing nanotechnology. A cell reacts and sense to stimuli from extracellular matrix (ECM) and topography of the ECM [1]. Thus, for better osseointegration, we should provide an environment similar to ECM. In this study, we synthesize TiO2 nanowires using hydrothermal reaction because TiO2 provides inertness to titanium on its surface and enables it used as an implant material for the orthopedic treatment such as fixation of the bone fracture [2]. Ti substrate is immersed into NaOH aqueous solution. The solution are heated at $140{\sim}200^{\circ}C$ for various time (10~720 minutes). After heat treatment, we take out the sample and immerse it into HCl aqueous solution for 1 hour. The acid treated sample is heated again at $500^{\circ}C$ for 3 hours [3]. Then, we culture osteoblast on the TiO2 nanowires. For investigating cell adhesion onto nanostructured surface, we conduct several tests such as MTT assay, ALP (Alkaline phosphatase) activity assay, measuring calcium expression, and so on. These preliminary results of the cell culture on the nanowires are foundation for investigating cell-material interaction especially with nanostructure interaction.

Comparisons of microhabitat use of Schlegel's Japanese gecko (Gekko japonicus) among three populations and four land cover types

  • Background: The effective use of habitats is essential for the successful adaptation of a species to the local environment. Although habitats exhibit a hierarchical structure, including macro-, meso-, and microhabitats, the relationships among habitats of differing hierarchy have not been well studied. In this study, we studied the quantitative measures of microhabitat use of Gekko japonicus from three field populations in Japan: one at Tsushima Island, one at Nishi Park, Fukuoka, and one at Ohori Park, Fukuoka. We investigated whether land cover type, a higher hierarchical habitat component, was associated with quantitative microhabitat use, a lower hierarchical component, in these populations. Results: The substrate temperature where we located geckos (SubT) and the distance from the ground to the gecko (Height) were significantly different among the three populations. In particular, SubT on Tsushima Island was lower than it was in the other two populations. Irradiance at gecko location and Height were significantly different among the land cover types. In particular, Height in evergreen needleleaf forest was significantly lower than that in deciduous broadleaf forest. Furthermore, significant interactions between population and land cover type were observed for the SubT and Height variables. Conclusions: The quantitative measures of microhabitat use of G. japonicus varied with population and land cover type, which exhibited significant interaction effects on microhabitat use variables. These results suggest that higher hierarchical habitat components can affect the quantitative measures of lower hierarchical microhabitat use in nocturnal geckos.

Pt Deposits on Bi-Modified Pt Electrodes of Nanoparticle and Disk: A Contrasting Behavior of Formic Acid Oxidation

  • This work presents a contrasting behavior of formic acid oxidation (FAO) on the Pt and Bi deposits on different Pt substrates. Using irreversible adsorption method, Bi and Pt were sequentially deposited on Pt electrodes of nanoparticle (Pt NP) and disk (Pt disk). The deposited layers of Bi and Pt on the Pt substrates were characterized with X-ray photoelectron spectroscopy, transmission microscopy and scanning tunneling microscopy. The electrochemical behaviors and FAO enhancements of Pt NP and Pt disk with deposited Bi only (i.e., Bi/Pt NP and Bi/Pt disk), were similar to each other. However, additional deposition of Pt on Bi/Pt NP and Bi/Pt disk (i.e., Pt/Bi/Pt NP and Pt/Bi/Pt disk) changed the electrochemical behavior and FAO activity in different ways depending on the shapes of the Pt substrates. With Pt/Bi/Pt NP, the hydrogen adsorption was suppressed and the surface oxidation of Pt was enhanced; while with Pt/Bi/Pt disk, the opposite behavior was observed. This difference was interpreted as a stronger interaction between the deposited Bi and Pt on Pt NP than that on Pt disk. The FAO performance on Pt/Bi/Pt NP is much better than that on Pt/Bi/Pt disk, most likely due to the difference in the interaction between the deposited Pt and Bi depending on the shapes of Pt substrates. In designing FAO electrochemical catalysts using Pt and Bi, the shape of a Pt substrate was concluded to be critically considered.

  • Erm 단백질은 23S rRNA의 특정 아데닌 잔기 $N_6$ 위치에 methylation을 일으켜 임상적으로 중요하게 사용되는 macrolide-lincosamide-streptogramin B계 항생제에 내성을 유발시킨다. 최근 ErmC'에서 N-말단 catalytic domain과 C-말단 substrate binding domain를 연결하는 오목한 홈 형성부위에 존재하는 잘 보존된 아미노산 잔기가 기질과 상호작용하는 것으로 제안되었다. 우리는 ErmSF에서 두 domain의 연결 부위의 오목한 홈에 위치하여 기질과의 상호작용이 예상되며 또한 Erm 단백질들 사이에서 매우 높게 보존되어있는 237번 아르지닌 잔기를 치환하여 그 기능을 in vivo, in vitro상에서 검색하여 분석하였다. R237A 변이 단백질을 발현하는 세균은 야생형 단백질을 발현하는 세균과 비교하여 in vivo 상에서는 차이를 나타내지 않았으나 순수분리 한 후 in vitro에서의 효소 활성은 야생형에 비하여 51%만을 나타내어 그 잔기가 기질 부착 기능을 수행하고 있다고 제안할 수 있었다.

Purification and Characterization of Acidic Chitinases from Gizzards of Broiler (Gallus gallus L.)

  • Acidic chitinases from the gizzards of a broiler were purified to homogeneity, using precipitation with $(NH_{4})_{2}SO_{4}$, ion exchanger chromatography, gel filtration, chromatofocusing and hydrophobic interaction chromatography. The enzymes, GAC1 and GAC2, were purified 180- and 194- folds with a recovery of 4.9% and 2.7%, respectively. The molecular mass of GAC1 and GAC2 were 48.2 kDa and 57.8 kDa, respectively. Chromatofocusing resulted in a pI of 3.1 for both enzymes. The purified enzymes were endochitinases that were devoid of ${\beta}-N-acetylglucosaminidase$ and lysozyme activity. Kinetic studies using $[^3H]chitin$ indicate that GAC1 has a $K_m$ and $V_{max}$ of 1.97 mg/ml and 185 mg/mg protein/h, respectively. The GAC2 has a $K_m$ and $V_{max}$ of 0.42 mg/ml and 92.3 mg/mg protein/h, respectively at optimal pH and temperature (pH 5.0 and $60^{\circ}C$). When the pentamer and hexamer of N-acetylglucosamine (GlcNAc) were used as a substrate, the major product by GAC1 was the dimer of GlcNAc with a differential accumulation of the monomer and trimer, depending upon the substrate. However, the GAC2 produced the dimer and trimer in an equal quantity, regardless of the substrate used. The first 9 $NH_2-terminal$ amino acid residues of the purified gizzard chitinase GAC1 and GAC2 shared a 100% homology. The first 25 $NH_2-terminal$ amino acid residues of GAC1 also shared 55-60% homology with animal chitinases and some animal proteins, such as whey protein and oviduct-specific proteins. However, little homology was found with either microbial and plant chitinases, or egg white lysozyme.

새로운 O,O-dialkyl-1-phenoxyacetoxy-1-methylphosphonate 유도체들의 반응성과 제초활성에 관한 2D-QSAR 및 HQSAR 분석 (2D-QSAR and HQSAR Analysis on the Herbicidal Activity and Reactivity of New O,O-dialkyl-1-phenoxy-acetoxy-1-methylphosphonate Analogues)

  • 일련의 새로운 O,O-dialkyl-1-phenoxyacetoxy-1-methylphosphonate (S) 유도체들의 반응성과 치환기가 변화함에 따른 오이(Cucumus Sativa)씨에 대한 발아전 제초활성과의 관계를 2D-QSAR 및 HQSAR 방법으로 검토하였다. 통계적으로 HQSAR 모델이 2D-QSAR 모델보다 양호하였으며 기질분자(S)와 PDH 효소중 $BH^+$ 이온(I) 사이의 경계분자궤도(FMO) 상호작용은 친전자성 반응이 우세하였다. 치환기의 효과로부터 기질분자 (S)내 $R_2$-치환기는 carbonyl 산소원자에 대한 친전자성 반응을, 그리고 phenyl 고리상 X,Y-치환기는 carbonyl 탄소원자에 대한 친핵성 반응에 기여하였으며 $R_2$-치환기보다 X,Y-치환기의 영향이 더 컸다. 2D-QSAR모델 (I 및 II)과 HQSAR 모델의 기여도로부터 X,Y-치환기의 길이가 길수록 제초활성이 증가하는 경향이었으며 적정한 ${\epsilon}LUMO$ 에너지($({\epsilon}LUMO)_{opt.}$=-0.479 e.v.)가 제초활성에 중요한 요소이었다. 그러므로 PDH 효소의 저해활성으로 인한 제초활성은 친핵성반응으로 진행될 것으로 예상되었다. 2D-QSAR 및 HQSAR 두 모델로부터 제초활성에 기여하는 기질분자(S)의 구조 특이성과 요소들을 새로운 제초제 설계에 적용할 수 있음을 시사하였다.

R.F. 스퍼터링법에 의한 상변화형 광디스크의 $(ZnS)_{1-x}-(SiO_2)_x$ 보호막 제조시 기판 바이어스전압의 영향 (The Effects of Substrate Bias Voltage on the Formation of $(ZnS)_{1-x}-(SiO_2)_x$ Protective Films in Phase Change Optical Disk by R.F. Sputtering Method.)

  • 상변화형 광디스크의 보호막으로 사용되는 $ZnS-SiO_2$ 유전체막을 RF magnetron 스퍼트링방법에 의하여 제조하는 경우에 기판 바이어스전압의 영향을 조사하기 위하여, 알곤가스 분위기에서 ZnS(80mol%)-$SiO_2$(20mol%)타겟을 사용하여 Si Wafer와 Corning flass 위에 박막을 증착시켰다. 본 실험에서는 여러 실험 변수를 효과적으로 조절하면서 실험의 양을 줄이고 도시의 산포를 동시에 만족시키는 최적조건으로 타겟 RF 출력 200W, 기판 RF 출력 20W, 아르곤 압력 5mTorr과 증착시간 20분을 얻을 수 있었으며, 신뢰구간 95%에서 확인실험을 수행하였다. 증착된 박막의 열적 저항성을 측정하기 위해 $300^{\circ}C$$600^{\circ}C$에서 열처리시험을 수행하였고, Spectroscopic Ellipsometry 측정을 통한 광학적 데이터를 바탕으로 Bruggeman EMA(Effective Medium Approximation)방법을 이용하여 기공(void)분률을 측정하였다. 본 연구결과에 의하면 특성치 굴절률에 대하여 기판 바이어스인자와 증착시간 사이에는 서로 교호작용이 강하게 존재함을 확인할 수 있었다. TEM분석과 XRD 분석 결과에 의하면 기판 바이어스를 가한 최적조건에서 증착된 미세조직은 기존의 바이어스를 가하지 않을 조건에서 증착시킨 박막보다 미세한 구조를 가지며, 또한 과도한 바이어스전압은 결정구조의 조대화를 야기시켰다. 그리고 적절한 바이어스전압은 박막의 밀도를 증가시키며, 기공분률을 약 3.7%정도 감소시킴을 확인할 수 있었다.

Comparison of Optical Properties of Ga-doped and Ag-doped ZnO Nanowire Measured at Low Temperature

  • Pristine ZnO, 3 wt.% Ga-doped (3GZO) and 3 wt.% Ag-doped (3SZO) ZnO nanowires (NWs) were grown using the hot-walled pulse laser deposition (HW-PLD) technique. The doping of Ga and Ag in ZnO NWs was observed by analyzing the optical and chemical properties. We optimized the synthesis conditions, including processing temperature, time, gas flow, and distance between target and substrate for the growth of pristine and doped ZnO NWs. The diameter and length of pristine and doped ZnO NWs were controlled under 200 nm and several ${\mu}m$, respectively. Low temperature photoluminescence (PL) was performed to observe the optical property of doped NWs. We clearly observed the shift of the near band edge (NBE) emission by using low temperature PL. In the case of 3GZO and 3SZO NWs, the center photon energy of the NBE emissions shifted to low energy direction using the Burstein Moss effect. A strong donor-bound exciton peak was found in 3 GZO NWs, while an acceptor-bound exciton peak was found in 3SZO NWs. X-ray photoelectron spectroscopy (XPS) also indicated that the shift of binding energy was mainly attributed to the interaction between the metal ion and ZnO NWs.

Principles of Physiology of Lipid Digestion

  • The processing of dietary lipids can be distinguished in several sequential steps, including their emulsification, hydrolysis and micellization, before they are absorbed by the enterocytes. Emulsification of lipids starts in the stomach and is mediated by physical forces and favoured by the partial lipolysis of the dietary lipids due to the activity of gastric lipase. The process of lipid digestion continues in the duodenum where pancreatic triacylglycerol lipase (PTL) releases 50 to 70% of dietary fatty acids. Bile salts at low concentrations stimulate PTL activity, but higher concentrations inhibit PTL activity. Pancreatic triacylglycerol lipase activity is regulated by colipase, that interacts with bile salts and PTL and can release bile salt mediated PTL inhibition. Without colipase, PTL is unable to hydrolyse fatty acids from dietary triacylglycerols, resulting in fat malabsorption with severe consequences on bioavailability of dietary lipids and fat-soluble vitamins. Furthermore, carboxyl ester lipase, a pancreatic enzyme that is bile salt-stimulated and displays wide substrate reactivities, is involved in lipid digestion. The products of lipolysis are removed from the water-oil interface by incorporation into mixed micelles that are formed spontaneously by the interaction of bile salts. Monoacylglycerols and phospholipids enhance the ability of bile salts to form mixed micelles. Formation of mixed micelles is necessary to move the non-polar lipids across the unstirred water layer adjacent to the mucosal cells, thereby facilitating absorption.
