An extracellular protease of Salmonella schottmulleri was purified from culture filtrate by using 0-75% ammonium sulfate precipitation, DEAE Sepharose Fast Flow ion exchange chromatography, Ultrogel HA chromatography and Sephacryl S-200 HR molecular sieve chromatography. To measure enzyme activity, synthetic dipeptide substrate (CBZ-arg-arg-AFC) with low molecular weight was employed as substrate. The molecular weight of the purified enzyme was approximately 80 kDa when determined by gel filtration on Sephacryl S-200 HR and 73 kDa when estimated by SDS-PAGE. The isoelectric point was 5.45. The activity of the purified enzyme was inhibited by metal chelating agesnts such as EDTA and 1.10-phenanthroline. The divalent cations, such as Ca$\^$2+/, Zn$\^$2+/, Fe$\^$2+/, Mg$\^$2+/ enhanced its activity. These results suggested that it was a metalloprotease. It had a narrow pH optimum of 6.5-7.5 with a maximum at pH 7.0 and a temperature optimum of 40.deg.C. It was stable at least for 1 week at 40.deg.C and maintained its activity for 24 hours at 50.deg.C, but it was rapidly inactivated at 65.deg.C. Objective: The objective of this study was to compare fenugreek (FG) with alfalfa (Alf) in ruminal fermentation and methane (CH4) production in vitro. Methods: Whole-plant FG harvested at 11- and 15-wk and Alf harvested at early and mid-bloom maturities, alone or as 50:50 mixture of FG and Alf at the respective maturity, were assessed in a series of 48-h in vitro batch culture incubations. Total fermentation gas and methane gas production, dry matter (DM) disappearance, volatile fatty acids, microbial protein and 16S RNA gene copy numbers of total bacteria and methanogens were determined. Results: Compared to early bloom Alf, FG harvested at 11-wk exhibited higher (p<0.05) in vitro DM and neutral detergent fibre disappearance, but this difference was not observed between the mid-bloom Alf and 15-wk FG. Regardless plant maturity, in vitro ruminal fermentation of FG produced less (p<0.001) CH4 either on DM incubated or on DM disappeared basis than that of Alf during 48-h incubation. In vitro ruminal fermentation of FG yielded similar amount of total volatile fatty acids with higher (p<0.05) propionate percentage as compared to fermentation of Alf irrespective of plant maturity. Microbial protein synthesis was greater (p<0.001) with 11-wk FG than early bloom Alf as substrate and 16S RNA gene copies of total bacteria was higher (p<0.01) with 15-wk FG than mid-bloom Alf as substrate. Compared to mid-bloom Alf, 15-wk FG had lower (p<0.05 to 0.001) amount of 16S RNA methanogen gene copies in the whole culture during 48-h incubation. Conclusion: In comparison to Alf, FG emerges as a high quality forage that can not only improve rumen fermentation in vitro, but can also remarkably mitigate CH4 emissions likely due to being rich in saponins.

The deer antler has been used as a major drug in oriental medicine for a long time. Recently, the demand for easy-to-take health functional foods is increasing due to economic development and changes in diet. As part of research on the development of functional materials for antlers, lactic acid fermentation of antler extract was performed. It was intended to develop a functional material with enhanced total polyphenol and flavonoid content and enhanced antioxidant activity. Lactic acid bacteria fermentation was performed by adding 4 types of lactic acid bacteria starter products, B. longum, Lb. Plantarum, Lb. acidophilus and mixture of 8 types of lactic acid bacteria to the antler water extract substrate, respectively. During the fermentation of lactic acid bacteria, the number of proliferation, total polyphenol and total flavonoid content, DPPH radical scavenging and antioxidant activity were quantified and evaluated. As a result of adding these four types of lactic acid bacteria to the antler water extract substrate, the number of lactic acid bacteria measured was 2.04~5.00×107. Meanwhile, a protease (Baciullus amyloliquefaciens culture: Maxazyme NNP DS) was added to the antler extract to decompose the peptide bonds of the contained proteins. Then, these four types of lactic acid bacteria were added and the number of lactic acid bacteria increased to 2.84×107 ~ 2.21×108 as the result of culture. The total polyphenol contents were 4.82~6.26 ㎍/mL in the lactic acid bacteria fermentation extracts, and after the reaction of protease enzyme and lactic fermentation, increased to 14.27~20.58 ㎍/mL. The total flavonoid contents were 1.52~2.21 ㎍/ml in the lactic acid bacteria fermentation extracts, and after the protease reaction and fermentation, increased to 5.59 ~ 8.11 mg/mL. DPPH radical scavenging activities of lactic acid bacteria fermentation extracts was 17.03~22.75%, but after the protease reaction and fermentation, remarkably increased to 32.82~42.90%.
최근 프린팅 기술은 디스플레이나 연료전지를 포함한 IT 분야에 폭넓게 사용되고 있지만 핵심 부품인 프린터 헤드의 박막을 적층하는 제조공정에서 발생하는 잔류응력 및 열응력으로 인해 기판이 변형되거나 노즐층이 파손되어 잉크가 새거나 원하는 영역으로 토출되지 않는 문제가 발생하고 있다. 따라서 본 논문에서는 보다 견고하고 신뢰할수 있는 구조를 가진 열전사 잉크젯 프린트 헤드 형상을 제안하고자 한다. 기판과 노즐층의 변형을 줄이기 위해 리브, 기둥, 지지벽 및 개별 피드홀과 같은 다양한 형태의 잉크젯 프린트 헤드 구조가 설계되었으며, FEA 해석을 통해 타당성을 검증하였다. 해석 결과 헤드의 최대 응력 및 노즐층 변형이 최소 40~50%로 급격히 감소하였으며 기둥 및 지지벽 형태의 프린터 헤드를 제작하여 노즐층 변형에 의한 균열 및 잉크 누출이 없는 것을 확인하였다. 따라서 제안된 헤드 형상이 정상 방향의 잉크 토출에 기여하며 대면적 프린팅 기술에도 적용될 수 있을 것으로 기대된다.
본 연구는 순환식과 비순환식 수경재배 온실 내 특정 구역에서 잠재적인 병발생 가능성을 추정하기 위하여 미생물 시료의 채취 및 배양을 통하여 집락형성단위수(CFU)를 분석하였다. 시료의 채취는 배지를 기준으로 각 구역별 3cm 간격으로 $25cm^2$의 면적에 대해 4개의 구역을 각 온실별로 3회 반복하여 이루어졌다. 비순환식과 순환식 수경재배 온실에서 암면과 코이어를 배지로 사용하는 각 2개 농가를 조사대상으로 하였다. 진균류의 경우 비순환식 수경재배 온실에서 유의적으로 높은 수준의 집락형성단위수(CFU)가 관찰되었다. 세균류의 경우는 유의적인 차이는 관찰되지 않았지만, 순환식 수경재배 온실이 비교적 낮은 경향을 나타냈다. 위치 별 CFU의 비교에서는 순환식과 비순환식 모두 배액의 발생에 따른 환경조건의 변화가 적은 배지 부분에서 가장 낮은 수준의 CFU가 관찰되었다. 주성분 분석결과 온실 내 배액의 이동 경로 상에서 수집된 시료가 미생물의 개체수 변화에 많이 기여하고 있는 것으로 판단되었다. 순환식 수경재배 온실은 비순환식 수경재배 온실에 비해 미생물 증식억제에 유리한 조건에 있으며, 이는 외부 환경에 비해 비교적 단순한 생태계로 구성된 온실의 조건을 고려해 보았을 때 잠재적인 병 발생 가능성이 상대적으로 낮음을 의미한다.
생물계면활성제 생성균주, Pseudomonas aeruginosa F722를 이용하여 다양한 배양조건과 배지조성에서 생물계면활성제 생산성을 검토하였다. 질소원과 탄소원을 검토하기 전에는 P. aeruginosa F722의 생물계면활성제 생산량은 0.78 g/l이었다. 하지만, 질소원과 탄소원을 검토한 후에는 생물계면활성제 생산량이 2배 증가한 1.66g/l이었다. 무기질소원으로 $_NH4$Cl 또는 $NaNO_2$를 첨가하였을 때 생물계면활성제 활성에 효과적이었으며 유기질소원으로는 yeast extract 또는 tryptone을 첨가하였을 때 생물계면활성제 활성이 높았다. 이중 무기 질소원으로 0.05% $NH_4$Cl , 유기 질소원으로 0.1% yeast extract를 질소원으로 첨가하였을 때 가장 최적이었다. 탄소원으로 소수성 기질(n-alkane) 또는 친수성 기질(glucose, glycol)을 첨가하여 생물계면활성제 생산량을 조사하였는데 소수성 기질보다는 친수성 기질인 3.0% glucose를 첨가하였을 때 생물계면활성제 생산량이 높았다. 이때의 탄소원/질소원 비율은 17~20이었다. P. aeruginosa F722는 배양조건 3.0% glucose, 0.05% $NH_4$Cl, 0.1% yeast extract, $35^{\circ}C$, pH 7.0, C/N ratio 20, 5 days에서 생물계면활성제 생산량은 1.66g/l이였다. 질소원이 결핍 후 탄소원을 첨가하여 배양하였을 때가 질소원과 탄소원을 함께 첨가하여 배양했을 때보다 생물계면활성제 생산속도 및 균체 생장속도가 높았다. 최적 배양조건하에서 얻어진 배양액의 표면장력은 30mN/m이었다.
큰느타리버섯 수확 후 배지를 이용한 유기퇴비를 제조하여 퇴비 원료로서의 이용가능성에 대해 실험을 진행하였다. 퇴비의 발아능력을 검증하기 위해 오이, 무, 상추, 배추를 파종한 후 발아율과 발아지수를 측정하였으며 미강과 미생물제의 첨가비율이 높을수록 높은 발아율을 나타내는 것을 확인할 수 있었다. 오이와 무의 발아지수는 전체적으로 높았으며, 상추의 경우 SMS 100%와 미강 5% 첨가구만 발아지수 70을 넘었고 배추는 미생물제 5% 첨가구에서 가장 높은 발아지수를 나타내었다. 또한 상추를 이용하여 SMS 상토 제조비율과 생육특성을 조사하기 위해 생육시험용 상토 제조비율은 일반상토 100%와 SMS 10%, SMS 20%, SMS 30%, SMS 50%, SMS 70%, SMS 100%로 제조하여 상추묘를 이식하였다. 먼저 상토재료의 이화학성 분석을 시행하였으며 SMS 첨가량이 높을수록 질소전량이 높아지는 것을 확인할 수 있었다. 하지만 상추의 SMS 처리구와 일반상토에서의 생육정도를 비교하였을 때, 중량은 SMS 10% 43.9 g, 일반상토 46.4 g로 비슷하였으며 엽록소 함량도 SMS 10% 32.8, 일반상토 SMS 10% 32.8로 같았고 나머지도 비슷한 수치를 나타내었으나 총장에서 SMS 10% 14.6 cm, 일반상토 16.8 cm로 일반상토가 더 큰 것으로 확인되었다.
본 연구는 고형배지경 양액재배에서 화학 양액 대체가능성을 검토하기 위하여 여과액비, 농축액비에 양액 및 부산물의 혼합처리가 고추의 생육과 수량에 미치는 영향을 검토하였다. 처리는 질소함량을 기준으로 여과액비, 농축액비에 부산물과 양액을 혼합하는 처리구를 두어 전기 전도도와 pH를 조정하여 고추 양액재배를 실시한 결과를 요약하면 다음과 같다. 1. 여과, 농축액비에는 다량 및 미량원소를 함유하고 있으며 부유물질(SS)이 낮아 수경재배시 관배수의 막힘문제 없이 활용이 가능하였다. 또한 여과, 농축액비는 인산, 칼슘, 마그네슘 함량이 낮고 칼륨이 높은 양분불균형를 나타내었다. 2. 양액재배에서 고추의 합계수량은 여과액비 단독시 용구에서 원예연 표준양액재배 대비 총수량 59%를 나타내었으며 농축액비의 경우 양액 대비 14%에 불과하였다. 퇴비단여과액비+부산물, 농축액비+부산물 처리구의 수량은 대조구 대비 각각 60, 54%를 나타내어 여과액비 단독시 용구와 비슷한 수량을 나타내어 부산물 첨가 효과가 나타나지 않았다. 3. 여과액비와 농축액비에 양액을 50%: 50% 비율로 혼합 처리구의 수량은 표준양액과 대등한 수량을 나타내었다. 결론적으로 여과액비, 농축액비 50%에 양액을 50% 첨가하면 고추 양액 재배에 활용이 가능 할 것으로 판단된다.
본 실험은 양액의 종류와 배지의 종류, 그리고 양액의 농도가 20일 무의 생육과 몇가지 품질에 미치는 영향을 알아보고자 실시하였다. 20일 무의 양액재배시 적절한 양액의 구명을 위하여 기존의 Cooper, Hoagland, Yamazaki양액의 비교실험을 실시한 결과, YamaBaki양액이 엽장, 엽수, 지상부와 지하부 생체중 등 전반적으로 Hoagland와 Cooper 양액보다 좋은 결과를 나타내었다. 근형지수는 모든 처리구에서 약 0.66으로 비교적 낮은 수치를 보였다. 14가지 조합의 배지를 이용하여 고형배지경을 실시하였다. 그 결과 모래 배지가 가장 균일하고도 좋은 생육의 20일 무를 배출하였고, 피트모스는 반대의 생육을 보였다. 근형지수의 경우 전반적으로 고형배지경이 순수수경재배보다 높은 수치를 나타내었다. Yamazaki 양액의 적정농도 수준을 구명을 위하여 모래를 배지로 하여 20일 무의 생육과 품질을 비교 조사한 결과, 1.0mS/cm 이상의 농도에서는 전반적으로 20일 무의 생육이 좋았으며, 특히 1.5mS/cm가 가장 좋은 생육을 보였다. 비타민 C의 함량과 티오시아네이트 함량은 1.0mS/cm이상의 처리구에서는 현저한 차이가 없었으나, 0.5mS/cm는 상당히 높은 수치를 보였다. 또한 무기물 함량의 경우는 모든 처리구에서 통계적 유의성이 없었으나, 칼륨이 1.5mS/cm에서 비교적 높은 함량을 나타내었다.
본 연구는 절화용 국화의 양액재배를 위한 코코넛 배지의 이용 가능성을 검토하기 위해 수행되었다. 본 연구에서 코코넛 배지는 중과피에서 유래된 dust와 fiber 그리고 가공된 chip을 이용하여 조성하였다. 시험구의 처리는 코코넛 단용배지 (dust 100%), dust와 fiber 그리고 dust와 chip을 각각 70:30의 부피비로 혼합한 혼용배지, 그리고 비교를 위해 펄라이트를 대조구로 하였다. 배지별 함수율은 펄라이트배지에서 75.8%로 가장 낮았고 코코넛 더스트 단용배지에서 93.1%로 가장 높게 나타났다. 배액의 pH는 코코넛 단용 및 혼용배지에서 5.8 ~ 6.5사이였으나 펄라이트배지에서 6.7 ~ 7.3의 범위로 높게 나타났다. 배액의 EC는 코코넛 단용배지에서 가장 높고 펄라이트배지에서 가장 낮은 것으로 나타났다. 국화의 초장, 엽면적, 그리고 건물량은 펄라이트배지와 코코넛 단용배지와 비교하여 코코넛 혼용배지에서 우수한 것으로 나타났다. 배지처리별 개화소요일수는 차이가 없었다.

제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
