The Effects of Environmental Conditions and Chemical Treatments on Seed Germination in Astilboides tabularis (Hemsl.) Engl.

  • Tabulis [Astilboides tabularis (Hemsl.) Engl.] has the potential for becoming a useful horticultural crop. This study was conducted to classify the seed dormancy types and to identify the germination conditions that improve the horticultural usefulness of this plant. We found that A. tabularis (Hemsl.) Engl. produced undifferentiated embryos and had a low germination rate of under 50%. In addition, water submersion led to moisture absorption. Therefore, A. tabularis seeds were designated as morphologically dormant. The germination rate was highest (86.3%) at $30^{\circ}C$ under light conditions. Higher temperatures generally led to an acceleration of the germination process, regardless of light condition. To improve the germination rate, seeds were submerged in various concentrations of growth regulators, such as $GA_3$ and kinetin, as well as minerals such as $KNO_3$ and KCl. An analysis of germination characteristics at $30^{\circ}C$ under light conditions revealed that treatment with 200 and $500mg{\cdot}L^{-1}GA_3$ led to an excellent germination rate of 97.0%. Treatment with $100mg{\cdot}L^{-1}GA_3$ led to a better germination rate than the control as well. However, treatment with kinetin, $KNO_3$, and KCl had no effect on germination, regardless of concentration.

순산소 고율포기시스템 및 침지식 MBR융합공정에서 Biofouling 제어 및 원인물질 규명에 관한 연구 (The Study of Biofouling Control and Cause Material in Hybrid Process of Pure Oxygen and Submerged Membrane Bio-reactor)

  • Membrane bio-reactor (MBR) has several advantages over the conventional activated sludge process, including a high biomass, low sludge production, and better permeate quality. Therefore, the MBR have gained popularity for municipal and industrial wastewater treatment. However the MBR usually were used for sewage and low streng th wastewater treatment because of membrane fouling problem and limitation of oxygen transfer into biomass. In this study, the hybrid process combining MBR and pure oxygen was tested for high strength organic wastewater treatment in the COD loading range from 2 to $10kgCOD/m^3{\cdot}day$. The hybrid process, membrane coupled pure oxygen high compact reactor (MPHCR), had been operated for one year and operation parameters, the effect of COD loading, MLSS concentration and the location of membrane module were studied for membrane fouling characteristic. Also membrane resistance test and the component of foulant was analyzed to investigate what is specific foulant in the MBR.

초음파침지처리에 의한 목재방부제의 주입성 (Penetrating Performance of Wood-preservatives by Ultrasonic Steeping)

  • 본 실험에서는 초음파의 특성을 이용하여 목재를 방부처리하고 그 주입효과에 대해 검토하고자 하였다. 유용성 방부제인 NCU (나프텐산 구리)와 수용성 방부제 AAC (알킬암모늄 화합물)에 소나무 및 잣나무 시편을 침지한 후 40 kHz, 400 W 출력의 초음파를 발진시켰다. 초음파처리 이후 시편의 흡수율 및 침윤도는 시간 경과에 따라 지속적으로 증가하여 120시간 경과 후 AAC처리 잣나무, 소나무 모두에 대해 약 95%의 흡수율을 얻을 수 있었으며, 소나무 및 잣나무 각 시편 두께의 95, 83%에 해당하는 NCU의 침윤도를 얻을 수 있었다. 초음파 처리후의 시편을 SEM을 이용해 관찰하였으며, 초음파 발생에 의해 생성된 고온, 고압의 충격파가 목재 내 약액의 주입통로가 되는 벽공구를 파괴하고 초음파의 탈포작용과 목재내 추출물의 배출 등에 기인하여 약액의 주입성능이 향상된 것으로 판단된다. 초음파 처리에 의한 방부제의 주입은 가압방부처리의 적용이 어려운 소재, 단판 등에 적용이 가능할 것으로 판단된다.

생태적 축산폐수 처리 및 재활용 연못시스템의 Pit 메탄발효 (Methane Fermentation of Pit in Pond System for Ecological Treatment and Recycling of Animal Excreta)

  • 유입폐수의 $BOD_5$가 1차연못에서 88%가 제거되고 있어 Pit의 $BOD_5$ 제거효율이 60%에 달할 것으로 예측된다. 메탄발효 Pit의 환경조건으로 용존산소가 없고, 혐기성 및 중성 pH가 유지되어야 하며, 충분한 체류시간이 확보되어야 하고, 온도변화가 적어야 한다. 분석결과 실험 메탄발효 Pit는 이런 조건들을 만족시키고 있어 Pit설계가 적절함을 알 수 있다. 실험결과 메탄발효 Pit의 설계인자로 폐수체류 기간이 2day, 월류유속은 $1.5m^3m^{-2}day^{-1}$가 적합하며, Pit바닥의 수심은 슬러지층의 온도와 밀접한 관계가 있어 3-3.5m 정도가 적합한 것으로 사료된다. Pit 바닥의 슬러지층 온도가 $16^{\circ}C$ 이상으로 유지되어야 메탈발효가 원활히 일어난다. 우리나라 중부지방과 기후조건이 유사한 지역에 위치한 연못시스템 연구에 의하면, 연못바닥의 온도가 메탄박테리아 활동이 거의 정지하는 $14^{\circ}C$ 이하로 내려가는 기간이 약 7개월이 된다. 온대권의 연못시스템은 연간 슬러지 침전량이 분해량보다 많아 어느 정도 슬러지가 쌓이게 된다. 따라서 여분의 30㎝ 수심을 두어 10-20년에 한번 슬러지를 제거하도록 설계한다. 실험 연못시스템이 설치된 장소는 중부지방보다 평균기온이 약 $3-4^{\circ}C$ 높은 지역으로 연못바닥 지하 1.5m에 위치한 Pit의 수온이 14℃이하가 되는 기간이 Fig.5에서 약 6개월이 된다. 실험 메탄발효 Pit는 좁은 면적의 연못에 설치하기 위해 콘크리트구조로 만들었으나, 1차연못의 규모가 크면 토공만으로 Pit설치가 가능하며 비용이 적게 든다. Pit에서 발생한 가스가 연못상층으로 이용하면서 $CO_2$가 해리되어 정제된 메탄을 회수할 수 있다. 메탄발생이 왕성한 기간에 연못상층에서 포집한 가스는 거의 메탄으로만 구성되어 있어 축산폐수를 처리하면서 메탄가스를 회수하여 연료로 사용하는 것이 가능하다. 메탄발효 Pit가 생태적으로 적응하면 초기보다 처리효율이 증가할 것으로 기대되어 지속적인 실험을 수행할 예정이다.

대기노출형 백색부후균 생물막을 이용한 유기물 처리특성 (Characteristics of Organics Treatment Using White-rot Fungus Biofilm of Atmospheric Exposed Type)

  • 점점 더 강화되는 수질기준, 연료 및 에너지비용 증가 등으로 경제성, 소요부지의 최소화, 운전의 용이성, 슬러지 발생의 최소화, 높은 처리효율을 지니는 폐수처리시스템의 개발은 시급한 실정이다. 따라서 본 연구에서는 난분해성물질을 함유한 오 폐수에 대해 대기노출형 백색부후균 생물막을 이용하여 생물막의 침지형태, 체류시간, 재순환비, 모듈회전수에 따른 유기물 처리 특성을 연구하였다. HBC 링 여재에 부착된 백색부후균 생물막을 대기중에 완전히 노출하여 운전시 침지형 포기조건과 거의 비슷한 제거율을 나타냈다. 대기노출형의 최적조건은 HRT 3$\sim$4 hr, 재순환비 6$\sim$10 Q, 모듈회전수 0.5$\sim$2회/min이며, 이때 유기물 제거율은 COD$_{Cr}$ 65.0$\sim$69.9%, NBDCOD 70.4$\sim$72.7%, BOD$_5$ 88.8$\sim$90.1%, SS 84.2$\sim$90.4%를 나타냈다. 또한 본 연구에서 BOD$_5$의 유출수 평균농도는 8.9 mg/L로 중수도 수질기준 BOD$_5$ 10 mg/L이하를 만족하였으나, NBDCOD의 경우 평균농도가 29.6 mg/L로 중수도 수질기준 20 mg/L보다 높은 것으로 나타났다.

Apoptosis of Human Hepatocarcinoma (HepG2) and Neuroblastoma (SK-N-SH) Cells Induced by Polysaccharides-Peptide Complexes Produced by Submerged Mycelial Culture of an Entomopathogenic Fungus Cordyceps sphecocephala

  • Three different polysaccharide-peptide complexes (PPC, named as Fr-I, Fr-II, and Fr-III) were produced by submerged mycelial culture of an entomopathogenic fungus Cordyceps sphecocephala, and their anticancer activities were investigated in human hepatocarcinoma (HepG2) and neuroblastoma (SK-N-SH) cells. The highest inhibitory effects of PPC on both HepG2 and SK-N-SH cells were achieved with Fr-I, whereas Fr-III with low molecular mass showed lower inhibition effects. Interestingly, the inhibitory effects of the three fractions were increased after protease digestion, suggesting that the inhibitory effects resulted mainly from the carbohydrate moiety, at least in the case of Fr-II and Fr-III, of PPC. The results of DNA fragmentation in PPC-induced apoptotic cells were confirmed by both DNA ladder assay and comet assay. Our investigation also showed that PPC-induced apoptosis of both cancer cells was associated with intracellular events including DNA fragmentation, activation of caspase-3, and modulation of Bcl-2 and Bax. We conclude that PPC has potential as a novel therapeutic agent for the treatment of both HepG2 and SK-N-SH cancer cells without any cytotoxicity against normal cells.

Immediate loading하에서 치근형 임프란트 주위 변연골 흡수에 대한 연구 (A STUDY OF MARGINAL BONE RESORPTION AROUND IMPLANTS AFTER IMMEDIATE LOADING)

  • Alveolar bone changes after immediate loading on implants up to one year were observed by means of standard intraoral X-ray measurement which were taken at 3 month intervals. At the same time, bone density changes were observed according to digital subtraction method which is a becoming a more and more promising diagnostic tool for implants. Following results were obtained ; 1. There was no significant difference in the amount of alveolar bone loss implant type, sex and implant diameter, but there was difference according to case selection. In fully bone anchored prostheses cases, bone loss was $1.16{\pm}0.15m$ whereas, in partial edentulous cases, it was $1.84{\pm}0.08mm$. 2. Alveolar bone loss after immediate loading showed a higher degree of bone loss than after submerged loading in the initial three months. But there were no significant difference at the 12th month. 3. According to the one year bone density change observation at the alveolar bone surrounding the implant, significant change was observed vertically, whereas no significant change could observed horizontally. According to the above mentioned results, we can conclude that immediate loading of implants results in a higher degree of alveolar bone loss in one year than submerged loading. But since alveolar bone loss rate decreases to a reasonable rate after the initial 3 months of rapid bone loss, immediate loading of implants seems to be an acceptable treatment modality for patients with good bone conditions. Fully bone anchored cases showed an favorable outcome, but partial edentulous cases showed more bony resorption. So this cases considered in case selections. Bone density changes observation in the study was performed for only one year therefore a more longitudinal observation may be studied.

새로 개발한 조월견방부제 ″쉬세요(Swi-Se-Yo)″의 효과에 관하여 (Study on the Effect of a New Antiseptic Preparation ″Swi-Se-Yo″ on the Carry-Over Cocoons in Silk Reeling Process)

  • In silk reeling process the carry-over cocoons must be submerged in the reeling baths filled with reeling water and left until reopening the operation. Under the detention the carry-over cocoons are apt to decay without any antiseptic treatment. Thus an useful antiseptic for the cocoons is urgently needed, and various antseptic agents have been tested for their applicability to the process. However, such an useful agent has not been developed yet. Formalin has been the only chemical used for antisepticizing carry-over cocoons, although it has many defects as the antiseptic for the cocoons. In these circumstances, recently we newly prepared an antiseptic useful for preventing the carryover cocoons from decaying. We named the new antiseptic preparation "Swi-Se-Yo." The Korean term "Swi-Se-Yo" literally means "please take a rest". Through a series of experiments with Swi-Se-Yo we obtained the following results: 1) Swi-Se-Yo, in 0.05% aqueous solution, exerted a good antiseptic effect on the boiled Cocoons submerged in the reeling baths and the effect lasted for 45 hours. The duration of the effect is about two times longer than that of Formalin. 2) The percentage of cocoon reel ability of the carry-over cocoons treated with Swi-Se-Yo was 6% higher than that of Formalin and was equal to that of flowing cold water. 3) The percentage of raw silk yield of the carry-over cocoons treated with Swi-Se-Yo was almost equal to that of Formalin and to that of flowing cold water. 4) The quality of raw silk of the carry-over cocoons treated with Swi-Se-Yo is the same as that of flowing cold water. Besides the above favourable results, Swi-Se-Yo has many advantages as an antiseptic. Chemically it is very stable. Its antimicrobial action is very strong and the spectrum is very broad. It can be available in water-soluble powder and in small bulk. And it is not harmful to human and domestic animals. Considering these profitable properties of Swi-Se-Yo, it will have a good reputation as a carry-over cocoon antiseptic. (The chemical composition and manufacturing method of Swi-Se-Yo will be published in the near future.)

Pilot Scale Plant의 황 충진 MBR을 이용한 고효율의 질소제거 공법 개발 (Development of High-rate Nitrogen Removal Process Using Submerged MBR Packed with Granular Sulfur of Pilot Scale Plant)

  • In this study, a process combined biofiltration with sulfur-utilizing autotrophic denitrification and membrane separation was proposed to examine the efficiency of nitrogen removal. As an experimental device, hollow-fiber module was installed in the center of reactor to generate the flux forward sulfur layer in the cylinder packed with granular sulfur. In addition, a simple module was installed in activated sludge aeration tank which inside and outside of sulfur-using denitrification module was covered with microfilter and the module was considered as an alternative of clarifier. The experiment for developing new MBR process was carried out for three years totally. As the results of first two-year experiment, successful nitrogen removal performance was revealed with lab-scale test and pliot scale plant using artificial wastewater and actual plating wastewater. In this year, pilot scale test using actual domestic wastewater was performed to prove field applicability. As the results, high-rate nitrogen removal performance was confirmed with about 0.19 kg ${NO_3}^--N/m^3$ day of rate. Also significant fouling and pressure increase were not found during the experiment. And, the production ratio of sulfate and the consumption ratio of alkalinity showed a slightly higher value about 311 mg ${SO_4}^{2-}/L$ and 369 mg $CaCO_3$/L, respectively. In conclusion, the developed MBR process can be utilized as an alternative for retrofiting existing wastewater plants as well as new construction of advanced sewage wastewater treatment plants, with cost-effective merit.

지베렐린 생산에 관한 연구 (Studies on the Production of Gibberellic acid)

  • 본 실험에서 G. fujkuroi I-892를 우수 균주로 선정한 후 자외선과 NTC를 처리하여, 지베렐린 생산성이 30%이상 증가된 변이주 G. fujkuroi G-471을 분리하였다. (178.6mg/$\ell$) 이 변이주에 적당한 최적 배지조성과 발효조건을 조사한 결과 saccharose 10%, ammoniun tartarate 50mM malt extract 1.0% KH$_2$PO$_4$, 0.5% MgSO$_4$0.5%, FeSO$_4$0.0002%,, 미량원소 0.002%(v/v)으로 나타났으며, 초기pH5.0. 통기량 1vvm 교반속도 400rpm의 발효조건에서 253mg/$\ell$가 생산되어 기존의 배양조건에서 보다 30%정도 생산성이 증가되었다. 발효액의 초기pH는 지베렐린 생산에 상당한 영향을 미쳤으며, 초기pH를 4.0으로 조절한 경우가 가장 좋은 생산성을 나타냈다.
