Acute and Subacute Toxicity of New Woohwangchungsimwon in Rats (랫드에서 신우황청심원의 급성 및 아급성독성시험)

  • The acute and subacute toxicity of New Woohwangchungsimwon(NWCH) which was used l-muscone as substitutive material of musk were investigated in S.D. rats. In intraperitoneal acute toxicity test. rats(Sprague-Dawley, SPF) were injected intraperitoneally with dosages of 0, 540, 750, 1,070, 1.500 and 3,000 mg/kg. Body weights were significantly decreased at 540 mg/kg dose group in both sexes and abnormal autopsy findings were founded in both sexes at all treated groups. Intraperitoneal $LD_{50}$ of NWCH was 812.3 mg/kg in male and 872.3 mg/kg in female rats. In the subacute toxicity study, NWCH was administrated orally to both sexes of rats for 4 weeks as several doses(0, 320, 800 and 2, 000 mg/kg). There were neither dead animals nor significant changes of body weights during the experimental period. In addition, no differences were found between control and treated groups in clinical signs, urinalysis, hematology, serum biochemical analysis, and other findings. Above data strongly suggest that no observed adverse effect level of NWCH might be over 2,000 mg/kg/day in this study.

  • Single and 4 weeks oral administration of New wonbangwoohwangchungsimwon (NSCH) which was used l-muscone as substitutive material qf musk, to beagle dogs of both sexes were per-formed to investigate both acute and subacute toxicity. Beagle dogs (3 males and 3 females) in acute experiments were administered orally with single dose of 2,000 mg/kg and groups of 9 male and 9 female beagle dogs in subacute experiments were given daily different dosage of NSCH, 160 mg/kg/day (low dosage group), 400 mg/kg/day (middle dosage group), 1,000 mg/kg/day (high dosage group) once a day for 4 weeks by oral route according to the Established Regulation of Korea Food and Drug Ad-ministration (l996. 4. 16). $LD_{50}$ value for beagle dogs was more than 2,000 mg/kg per oral for both male and females. In animals administered with NSCH, there were neither dead animals nor significant changes of body weights. In addition, no differences were found between control and treated groups in clinical sign, urinalysis, eye examination, hematology, serum chemistry, organ weight and other fingings. No histological lesions were observed in both control and treatment groups. Above data strongly suggset that NSCH in beagle dogs is considered to be safe.

Effects of Trunk Position Sense through Visual Cue Deprivation Balance Training in Subacute Stroke (앉은 자세에서의 시각을 차단한 균형훈련이 아급성기 뇌졸중 환자의 체간 위치감각에 미치는 영향)

  • PURPOSE: The purpose of this study is to investigate effects of trunk position sense through visual cue deprivation balance training in subacute stroke patients. METHODS: The subjects were randomly allocated to two groups: experimental(n=10) and control(n=10). Both groups performed balance training on sitting for 30minute after measurements. Trunk position sense test was assessed using the David back concept to determine trunk repositioning error for four movement(flexion, extension, affected side lateral flexion, non-affected side lateral flexion). Measurements on each test were assessed prior to the balance training and then immediately following the balance training. RESULTS: In comparison of the difference of the trunk position sense between groups, the experimental group decreased significantly in trunk repositioning error of flexion, extension and affected side lateral flexion than control group(p<.05). CONCLUSION: The trunk position sense of the experimental group showed more improvement after the balance training program compared to the control group, Therefore, these results suggest that visual cue deprivation training is considered an effective exercise method for individuals with subacute stroke.

Subacute Toxicity Study on Palmultang(Bawu-Tang) in SD Rats (SD 랫트를 이용하여 팔물탕(八物湯)의 4주 반복투여 독성에 관한 연구)

  • Objectives : Palmultang(Bawu-tang) has been traditionally prescribed as a restorative, The present study was undertaken to determine the possible toxic effects of Palmultang on SD rats. Methods : In this study, we investigated the subacute toxicity of water-extracted Palmultang(Bawu-tang) on SD rats. Twenty rats were orally adiministered Palmultang for 28 days at a dose of 0 mg/kg(control group) or 1500 mg/kg(treated group), respectively. Results : All of subjects was survived. No significant difference in abnormal clinical signs, related to hematological values, serum biochemical values, water and feed intake, coagulation time, autopsy analysis, organ weight, tissue microscopically, funduscopy, urine intake and urinalysis, was detected. Conclusions : Compared with the control group, we could not find any subacute toxic alteration in treated group (1500 mg/kg) for 28 days. This result suggests that Palmultang, a herbal medicine prescription, is a safe prescription to body.

  • Kikuchi disease(subacute necrotizing lymphadenitis), first reported by Kikuchi and Fujimoto in 1972, is a benign self-limiting illness characterized by fever, neutropenia and cervical lymphadenopathy and develops predominantly in young women, especially in Asia. The cause of Kikuchi disease is unknown, but postinfectious(virus or bacteria) hyperimmune reaction has been suggested. Few pediatric cases have been reported. We experienced a case of Kikuchi disease associated with Yersinia pseudotuberculosis infection in 12-year-old girl. After she was admitted with cervical lymphadenopathy and headache, followed by sustained fever with leukopenia. All symptoms and signs did not seem to be responded to antimicobial treatment. Open biopsy of cervical lymph node was performed and showed findings consistent with subacute necrotizing lymphadenitis. Serologic studies were all negative except for Y. pseudotuberculosis. Fever subsided and lymphadenopathy improved after administration of oral steroid for 1 week.

  • Subacute necrotizing lymphadenitis was first reported by Kikuchi and Fujimoto in 1972. Young females no more than 30 years of age are mainly affected. It usually manifests as fever and cervical lymphadenopathy. We experienced one case of subacute necrotizing lymphadenitis with hepatic complication in an 11-year-old boy. Symptoms presented were URI signs, diarrhea, headache, and weight loss along with fever and cervical lymphadenopathy. Elevated serum AST/ALT levels were also noted up to 682/1560 (IU/L) and were normalized within one month. We performed aspiration biopsy of the liver twice (at admission and 5 months thereafter). The hepatic histopathologic findings were nonspecific.

Effects of Repetitive Transcranial Magnetic Stimulation on Motor Recovery in Lower Extremities of Subacute Stage Incomplete Spinal Cord Injury Patients: A Randomized Controlled Trial

  • The aim of this study was to investigate whether repetitive transcranial magnetic stimulation (rTMS) can improve motor recovery in the lower extremities of the patients with subacute stage spinal cord injury (SCI). This study was conducted with 19 subjects diagnosed with paraplegia because of SCI. The experimental group included 10 subjects who underwent active rTMS, and the control group included 9 subjects who underwent sham rTMS. The SCI patients in the experimental group underwent conventional rehabilitation therapy, and active rTMS was applied daily to the hotspot of the lesional hemisphere. The SCI patients in the control group underwent sham rTMS and conventional rehabilitation therapy. The participants in both the groups received therapy five days per week for six weeks. Latency, amplitude, and velocity were assessed before and after the six-week therapy period. A significant difference in post-treatment gains for the latency and velocity was observed between the experimental and control groups (p < 0.05). However, no significant differences in the amplitude were observed between the two groups (p > 0.05). The results of this study indicate that rTMS may be beneficial in improving motor recovery in the lower extremities of subacute stage SCI patients.

Hepatotoxicity of Uhwangchungsimwon: Acute and Subacute Studies in Rats (우황청심원의 간독성시험 : 랫드에서의 급성 및 아급성시험)

  • Uhwangchungsimwon(UC) is widely used as a herbal medicine on various circulatory disorders in Korea. The objective of this investigation was to characterize the acute, subacute hepatotoxic potency of orally administered UC in rats. In the acute and short-term studies, male rats of 150~170g were gavaged with 0, 7.5 g/kg once daily for up to 1, 5, 10 consecutive days. No differences in body weight, serum enzyme activities, absolute and relative liver weight and histopathological examination on liver between control and UC-fed groups were found. In the subacute study, UC was administered orally to both sexes of rats for 30 days(0, 1.875, 3.75 or 7.5 g/kg/day). There were no-doserelated hepatotoxic signs of general symptoms, body weight gain, water consumption and serum biochemical analysis. Slight decreases of food consumption observed at 3.75 and 7.5 g/kg groups of both sexes were due to be full of UC fed. Gross necropsy and histopathology revealed no evidence of hepatotoxicity related to UC. Our data indicate that hepatotoxicity was not caused by administration of UC up to 7.5 g/kg/dayfor 30 days in rats.
