L-Asparaginase (E.C. 3.5.1.1) is an enzyme involved in asparagine hydrolysis and has the potential to effect leukemic cells and various other cancer cells. We identified the L-asparaginase gene (L-ASPG86) in the genus Mesoflavibacter, which consists of a 1,035 bp open reading frame encoding 344 amino acids. Following phylogenetic analysis, the deduced amino acid sequence of L-ASPG86 (L-ASPG86) was grouped as a type I asparaginase with respective homologs in Escherichia coli and Yersinia pseudotuberculosis. The L-ASPG86 gene was cloned into the pET-16b vector to express the respective protein in E. coli BL21 (DE3) cells. Recombinant L-asparaginase (r-L-ASPG86) showed optimum conditions at 37-40℃, pH 9. Moreover, r-L-ASPG86 did not exhibit glutaminase activity. In the metal ions test, its enzymatic activity was highly improved upon addition of 5 mM manganese (3.97-fold) and magnesium (3.35-fold) compared with the untreated control. The specific activity of r-L-ASPG86 was 687.1 units/mg under optimum conditions (37℃, pH 9, and 5 mM MnSO4).

This paper details a testing facility ("NATURAL FIRE FACILITY") that allows closely-controlled experimental testing on full-scale sub-frames while reproducing the spatially transient temperature conditions measured in real fires. Using this test facility, an experimental investigation of six steel sub-frames under a natural fire was carried out at the Department of Civil Engineering of the University of Coimbra. The main objective of these tests was to provide insight into the influence of these connection types on the behaviour of steel sub-structures under fire. The experimental layout is defined by two thermally insulated HEA300 columns and an unprotected IPE300 beam with 5.7 m span, supporting a composite concrete slab. Beam-to-column connections are representative of the most common joint type used on buildings: welded joints and extended, flush and partial depth plate. Finally, the available results are presented and discussed: evolution of the steel temperature; development of displacements and local deformations and failure modes on the joints zone.
Seismic design codes permit the use of Equivalent Static Analysis of buildings considering torsional eccentricity e with dynamic amplification factors on structural eccentricity and some accidental eccentricity. Estimation of e in buildings is not addressed in codes. This paper presents a simple approximate method to estimate e in RC Moment Frame and RC Structural Wall buildings, which required no detailed structural analysis. The method is validated by 3D analysis (using commercial structural analysis software) of a spectrum of building. Results show that dynamic amplification factor should be applied on torsional eccentricity when performing Response Spectrum Analysis also. Also, irregular or mixed modes of oscillation arise in torsionally unsymmetrical buildings owing to poor geometric distribution of mass and stiffness in plan, which is captured by the mass participation ratio. These irregular modes can be avoided in buildings of any plan geometry by limiting the two critical parameters (normalised torsional eccentricity e/B and Natural Period Ratio 𝜏 =T𝜃/T, where B is building lateral dimension, T𝜃 uncoupled torsional natural period and T uncoupled translational natural period). Suggestions are made for new building code provisions.
Finite element (FE) model based structural damage detection (SDD) methods play vital roles in effectively locating and quantifying structural damages. Among these methods, structural model updating should be conducted before SDD to obtain benchmark models of real structures. However, the characteristics of updating parameters are not reasonably considered in existing studies. Inspired by the l∞ norm regularization, a novel anti-sparse representation method is proposed for structural model updating in this study. Based on sensitivity analysis, both frequencies and mode shapes are used to define an objective function at first. Then, by adding l∞ norm penalty, an optimization problem is established for structural model updating. As a result, the optimization problem can be solved by the fast iterative shrinkage thresholding algorithm (FISTA). Moreover, comparative studies with classical regularization strategy, i.e. the l2 norm regularization method, are conducted as well. To intuitively illustrate the effectiveness of the proposed method, a 2-DOF spring-mass model is taken as an example in numerical simulations. The updating results show that the proposed method has a good robustness to measurement noises. Finally, to further verify the applicability of the proposed method, a six-storey aluminum alloy frame is designed and fabricated in laboratory. The added mass on each storey is taken as updating parameter. The updating results provide a good agreement with the true values, which indicates that the proposed method can effectively update the model parameters with a high accuracy.

Spray dried $Cr_2O_3$ powder having an agglomerated structure of particles was twice treated into a plasma flame to increase its apparent density. The powder subjected to the first densification treatment did not show the entirely melted state keeping inner particle hollows, and it was fully melted after the second processing only. The powder size as a result of the second treatment decreased, and the apparent density as well as flowability were increased due to melting and surface smoothing effects. But a part of particles after the second densified treatment showed the hollow structure, especially those which were above $30{\mu}m$ in size. This densification behavior of the powder has been qualitatively discussed in terms of the thermal conductivity and inner gas pressure within aggregates exposed to the plasma flame.
In this study, the fiber orientation distribution (FOD) is investigated using both micro-CT (computerized tomography) and image analysis of physically cut specimens prepared from Polyvinyl Alcohol (PVA) fiber reinforced cemented Toyoura sand. The micro-CT images of the fiber reinforced cemented sand specimens were visualized in horizontal and vertical sections. Scans were obtained using a frame rate of two frames and an exposure time of 500 milliseconds. The number of images was set to optimize and typically resulted in approximately 3000 images. Then, the angles of the fibers for horizontal sections and in vertical section were calculated using the VGStudio MAX software. The number of fibers intersecting horizontal and vertical sections are counted using these images. A similar approach was used for physically cut specimens. The variation of results of fiber orientation between micro-CT scans and visual count were approximately 4-8%. The micro-CT scans were able to precisely investigate the fiber orientation distribution of fibers in these samples. The results show that 85-90% of the PVA fibers are oriented between ±30° of horizontal, and approximately 95% of fibers have an orientation that lies within ±45° of the horizontal plane. Finally, a comparison of experimental results with the generalized fiber orientation distribution function 𝜌(θ) is presented for isotropic and anisotropic distribution in fiber reinforced cemented Toyoura sand specimens. Experimentally, it can be seen that the average ratio of the number of fibers intersecting the finite area on a vertical plane to number of fibers intersecting the finite area on a horizontal plane (NVtot/NHtot) cut through a sample varies from 2.08 to 2.12 (an average ratio of 2.10 is obtained in this study). Based up on the analytical predictions, it can be seen that the average NVtot/NHtot ratio varies from 2.13 to 2.17 for varying n values (an average ratio of 2.15).
Background: Refractory ceramic fibers (RCFs) are a suspected carcinogen but have been widely used as insulations. Depending on the temperature, RCFs can transform into crystalline SiO2, which is a carcinogen that can be present in the air during bulk RCF handling. This study analyzed the physicochemical and morphological characteristics of RCFs at high temperatures and determined the exposure levels during the semiconductor scrubber maintenance. Methods: Sampling was conducted at a company that manufactures semiconductor scrubbers using RCFs as insulation. Bulk RCF samples were collected both before and after exposure to a scrubber temperature of 700℃. Airborne RCFs were collected during scrubber maintenance, and their characteristics were analyzed using microscopes. Results: The components of bulk RCFs were SiO2 and Al2O3, having an amorphous structure. Airborne RCFs were morphologically different from bulk RCFs in size, which could negatively affect maintenance workers' health. 58% of airborne RCFs correspond to the size of thoracic and respirable fibers. RCFs did not crystallize at high temperatures. The exposure caused by airborne RCFs during the scrubber frame assembly and insulation replacement was higher than the occupational exposure limit. Conclusion: Workers conducting insulation replacement are likely exposed to airborne RCFs above safe exposure limits. As RCFs are suspected carcinogens, this exposure should be minimized through prevention and precautionary procedures.

Tunnels may undergo a larger or a smaller response compared with the free-field soil. In the pseudo-static procedure, the response of the tunnel is most often characterized by a curve that relates the racking ratio (R) with the flexibility ratio (F), where R represents the ratio of the tunnel response with respect to the free-field vibration and F is the relative stiffness of the tunnel and the surrounding soil. A set of analytical and empirical curves that do not account for the depth and the aspect ratio of the tunnel are typically used in practice. In this study, a series of dynamic analyses are conducted to develop a set of F-Rm relations for use in a frame analysis method. Rm is defined as an adjusted R where the rocking mode of deformation is removed and only the racking deformation is extracted. The numerical model is validated against centrifuge test recordings. The influence of aspect ratio, buried depth of tunnel on results is investigated. The results show that Rm increases with the increase of the buried depth and the aspect ratio. The widely used F-R relations are highlighted to be different compared with the obtained results in this study. Therefore, the updated F-Rm relations with proposed equations are recommended to be used in practice design. The rocking response decreases with either the decrease of the difference of stiffness between surrounding soil and tunnel or the larger aspect ratio of the tunnel section.

Recently, the government is reinforcing safety management at construction sites to prevent safety accidents in construction works, and the safety management plan of workers is an important situation. Meanwhile, modular construction is expected to be gradually expanded to middle and high-rise buildings, but active measures to ensure worker safety are insufficient. This study is a preliminary study of the development of a checklist for preventive worker safety management. The purpose of this study is to derive a checklist classification system for the safety management of workers in the field of modular construction by preceding studies, case analysis, and expert advisory opinions. The classification system consists of large categories of factory manufacturing, transportation, and on-site construction, and the sub-system consists of six sub-classes: foundation work, frame work, modular frame installation work, finishing work, and facility work. Among them, the sub-classification of modular frame installation work consists of 12 unit works, centering on module lifting and assembly module work, which are the main construction processes. And the classification system reflects the three main management factors and contents for defined safety management. It is expected that the research results of this study can contribute to efficient safety management at the modular construction site.

This paper describes the study on a method for improving the structural durability of bolted joints applied to a composite bogie frame. In this study, three bolted joints with and without inserts and screw threads were selected for determining the effect of the inserts, using experiment and analysis. The structural performances of the proposed bolted joints were compared and evaluated using the test method prescribed by the ASTM D5961 standard. The results revealed that the bolted joint having an insert shape without the screw thread offered improved durability for application to a composite bogie frame. Furthermore, the structural integrity of the frame comprising the bolted joints was evaluated using finite element analysis according to the JIS E 4207 standard. The Tasi-Wu and Von-Mises failure criteria were used for determining the failure of the composite structure and bolted joints, respectively. A sub-modeling technique was introduced for investigating the performance of the bolted joints in greater detail. The analysis results demonstrated that the Tasi-Wu failure index of the composite structure near the bolted joints was reduced by approximately one-half after applying an insert without the screw thread. This implies that the structural durability of the bolted joints of a composite bogie frame could be improved by using a metal insert without the screw thread.
