The Correlation Between Conformations and Activities of ${\omega}$-Pyridylalkenoic Acids (${\omega}$-피리딜 알켄산의 형태와 활성간의 상관관계)

  • Molecular mechanics and conformation search methods were carried oyt to investigate the relationship between conformations and thromboxane synthetase ingibitory activities of om ega-pyridylalkenoic acids. The initial geometries of ${\omega}$-pyridylalkenoic acids and heme part of cytochrome P-450 were obtained from MM+ geometry optimization. The bond lenths and angles were not varied by step during the conformation searching. Stable conformers of some ${\omega}$-pyridylalkenoic acids were obtained by comformational search method. The distances were 8.5~10.8 ${\AA}$- between N atom at 3-position of pyridine ring and C atom at carboxylic group of stable ${\omega}$-pyridylalkenoic acids. The conformations of ${\omega}$-pyridylalkenoic acids and heme part complex were determined by same method. In theses structures, benzene ring and ethylene group in ${\omega}$-pyridylalkenoic acids are making the structure more rigid and increase inhbitory activity. The electron donating groups in C atom shich is connected to pyridine ring also increase activity.

  • In conclusion, a series of N-Cbz-.alpha.-amono-glutarimides (1a-f), combining common structures such as N-CO-C-N and cyclic imide in a single molecule, were prepared from the (R)- or (S)-N-Cbz-glutamic acid and evaluated for their anticonvulsant activities in MES and PTZ tests in order to develope new and broad spectrum anticonvulsant. In this study, N-Cbz-.alpha.-aminoglutarimides (1) except ac and af, showed significant anticonvulsant activity in both MES and PTZ tests enough to be recommended as promising new anticonvulsant drug candidates. Now we are continuing to investigate further anticonvulsant test (quantification)for these compounds and synthesize their analogues in order to develop more active anticonvulsant and define the structure activity relationship more precisely.

  • Further bioactivity-directed fractionation of the ethanol extract of the seeds of Annona cherimolia has led to the isolation of two mono-tetrahydrofuran acetogenins, xylomaticin (1) and gonionenin (2). The structures of these compounds were characterized on the basis of chemical and spectroscopic data. Compounds 1 and 2 have a relative stereochemistry relationship of threo/trans/threo across the mono-tetrahydrofuran ring with its two flanking hydroxyls. Compounds 1 and 2 are known, but are first isolated from this plant. In brine shrimp lethality test (BST), 1 and 2 exhibited cytotoxic activity.

Inhibitory Activity of Flavonoids from Prunus davidiana and Other Flavonoids on Total ROS and Hydroxyl Radical Generation

  • Since reactive oxygen species (ROS) and hydroxyl radicals ($^-OH$) play an important role in the pathogenesis of many human degenerative diseases, much attention has focused on the development of safe and effective antioxidants. Preliminary experiments have revealed that the methanol (MeOH) extract of the stem of Prunus davidiana exerts inhibitory/scavenging activities on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals, total ROS and peroxynitrites ($ONOO^-$). In the present study, the antioxidant activities of this MeOH extract and the organic solvent-soluble fractions, dichloromethane (CH$_2$Cl$_2$), ethyl acetate (EtOAc), and n-butanol (n-BuOH), and the water layer of P. davidiana stem were evaluated for the potential to inhibit $^-OH$ and total ROS generation in kidney homogenates using 2',7'-dichlorodihydrofluorescein diacetate (DCHF-DA), and for the potential to scavenge authentic $ONOO^-$. We also evaluated the inhibitory activity of seven flavonoids isolated from P. davidiana stem, kaempferol, kaempferol 7-Ο-$\beta$-D-glucoside, (+)-catechin, dihydrokaempferol, hesperetin 5-Ο-$\beta$-D-glucoside, naringenin and its 7-Ο-$\beta$-D-glucoside, on the total ROS, $^-OH$ and $ONOO^-$ systems. For the further elucidation of the structure-inhibitory activity relationship of flavonoids on total ROS and 'OH generation, we measured the antioxidant activity of sixteen flavonoids available, including three active flavonoids isolated from P. davidiana, on the total ROS and 'OH systems. We found that the inhibitory activity on total ROS generation increases in strength with more numerous hydroxyl groups on their structures. Also, the presence of an ortho-hydroxyl group, whether on the Aring or S-ring, and a 3-hydroxyl group on the C-ring increased the inhibitory activity on both total ROS and $^-OH$ generation.

New dammarane-type triterpenoids from the leaves of Panax notoginseng and their protein tyrosine phosphatase 1B inhibitory activity

  • Background: Panax notoginseng has been used as a general tonic agent to invigorate human body for millennia in China and continued to be used until present. Methods: Some chromatographic methods were performed to isolate pure triterpenoids, and their structures were determined by nuclear magnetic resonance (NMR) experiments. Anti-diabetes activities of isolated compounds were evaluated through their inhibitory activity of protein tyrosine phosphatase 1B (PTP1B) enzyme. Results and Conclusion: Three new dammarane-type triterpenoids, notoginsenoside-LX (1), notoginsenoside-LY (2), and notoginsenoside-FZ (3) together with eighteen known compounds were isolated from the Panax notoginseng leaves. The structure-activity relationship of the compounds with dammaranetype triterpenoids and their PTP1B inhibitory activity were also reported. Results showed that compounds 2, 15, 20, and 21 can significantly inhibit the enzyme activity of PTP1B in a dose-dependent manner, with inhibitory concentration 50 ($IC_{50}$) values of $29.08{\mu}M$, $21.27{\mu}M$, $28.12{\mu}M$, and $26.59{\mu}M$, respectively. The results suggested that Panax notoginseng leaves might have potential as a new therapeutic agent for the treatment of diabetes.

A Comparison between Francesco Borromini's Architectural and Structural Design - Focusing on the Churches of San Carlo alle Quattro Fontane and Sant'Ivo alla Sapienza -

  • This study aims to contribute to better understanding of the Baroque architect Francesco Borromini, and, as a consequence, of the Baroque period itself. Recognizing that historical studies have focused mainly on the architectural (i.e., formal) aspects of Borromini's work and largely neglected the technical (i.e., structural) issues, and that the relationship between the architectural and structural worlds in Borromini's activity has consequently never been studied in depth, this research set as its objective the study of this relationship, and has developed a comparative analysis of Borromini's attitude toward architectural problems and questions of tectonics. The investigation has been conducted on two meaningful case-studies in Rome: San Carlo alle Quattro Fontane and Sant'Ivo alla Sapienza. The analysis has highlighted a strong dichotomy between Borromini as an architectural designer and Borromini as a structural designer. While Borromini's design is experimental, innovative, and nonconformist, his structures are cautious, tested, and validated by trustworthy tradition. His unprejudiced use of constructive solutions from different historical periods, regions, and cultural areas allows the definition of his approach to tectonics as "constructive eclecticism." The analysis has also highlighted the independence between the architectural and structural aspects of his work, as the two do not interfere with or limit each other. The dichotomy between form and structure in Borromini's work may reflect the "theatrical" tendency of Baroque culture, where what is shown on the face is often different from what is hidden behind.

  • Among four compounds such as oxyresveratrol, resveratrol, morin and afzelechin isolated from the heartwood of Morns bombycis, oxyresveratrol and resveratrol which belong to stilbenoid indicated high antioxidative and antimicrobial activity, respectively. Based on this result, this experiment was carried out to elucidate the relationship between biological activities of stilbenoids and their chemical structures using two isolated and six synthesized ones prepared by Wittig reaction. Antimicrobial activity of 3,5-dihydroxystilbene(pinosylvin) devoid of hydroxyl group of B-ring was the best among the tested stilbenoids. Antimicrobial activities of the stilbenoids showed negative dependency on the number of hydroxyl groups of B-ring, that is, the fewer number of hydroxyl groups of B-ring, the higher antimicrobial activity. On the other hand, antioxidative activities of the stilbenoids indicated a positive relationship with the number of hydroxyl groups of B-ring. In tetrahydroxystilbenoids, antioxidative activities of the compounds possessing ortho-diphenol structure, 2',3,3',5- and 3,3',4',5-tetrahydroxystilbene, were superior to the others. As a result, it was concluded that resveratrol and oxyresveratrol isolated from the heartwood of M. bombycis had antimicrobial and high antioxidative activities, and these activities of stilbenoids were also dependent on the number and position of hydroxyl groups of B-ring.

The Emergence of Competitiveness in Korea-China Ship Distribution Industry

  • Purpose - This paper reviews the changes in the ship export and import structure between Korea and China. It utilizes the comparative advantage trade theory to analyze time-series statistical data from the market share index, revealed comparative advantage index (RCA), and trade specialization index(TSI). Research design, data, and methodology - Based on their economic phases, both Korea and China have similar country characteristics. The purpose of this research is to understand the two country's trade structures to fortify the Korea-Sino economic relationship including verifying what is working and what is not. Results - Based on the analysis, bilateral economic activity to achieve a plus trade stimulus environment should be realized in the long run. Both countries should establish guarantee-free trade negotiations and boundaries instead of various non-tariff barriers. Conclusion - Reviewing the research, a sound competitive relationship can be grown for mutual benefit including export market diversification in the near future. The review of the Korea-Sino ship industry is keenly important and investigative research about it is timely because it is a major industry in each country.

Neuraminidase Inhibitors from Mushroom Microphorus affinis

  • In the course of screening anti-influenza agents from natural products, four neuraminidase inhibitors were isolated from the methanol extract of mushroom Microphorus affinis by purification using solvent partition, silica gel column chromatography, Sephadex LH-20, and semi-preparative HPLC. The chemical structures of these compounds were identified as ${\alpha}-lupeol$, methyl linoleate, methyl palmitate, and methyl oleate by means of spectral data including GC-MS, $^1H-,\;and\;^{13}C-NMR\;with\;IC_{50}$ values of 5.65, 7.07, 7.12, and $7.52\;\mu\textrm{M}$, respectively. They did not inhibit other glycosidases such as glucosidase, mannosidase, and galactosidase, indicating that they were relatively specific inhibitors of neuraminidase. The relationship between the fatty acid structure and inhibitory activity was investigated. The result showed that, in the case of an aliphatic linear hydrocarbon skeleton, at least one carboxyl (presumably any carbonyl) moiety and sixteen carbons were the necessary requirements for potent inhibition, whereas saturated, unsaturated, free, and ester forms did not have any significant effect on the activity.

Community Structures of Arbuscular Mycorrhizal Fungi in Soils and Plant Roots Inhabiting Abandoned Mines of Korea

  • In this study, we collected rhizosphere soils and root samples from a post-mining area and a natural forest area in Jecheon, Korea. We extracted spores of arbuscular mycorrhizal fungi (AMF) from rhizospheres, and then examined the sequences of 18S rDNA genes of the AMF from the collected roots of plants. We compared the AMF communities in the post-mining area and the natural forest area by sequence analysis of the AMF spores from soils and of the AMF clones from roots. Consequently, we confirmed that the structure of AMF communities varied between the post-mining area and the natural forest area and showed significant relationship with heavy metal contents in soils. These results suggest that heavy metal contamination by mining activity significantly affects the AMF community structure.
