Peroxynitrite Scavenging Activity of Sabohwanin Lipopolysaccharide-Induced Oxidatively-Stressed Mice (Lipopolysaccharide로 산화 스트레스를 유도한 Mouse에서 사보환(四補丸)의 Peroxynitrite 억제 효과)

  • Objectives : Peroxynitrite (ONOO-), superoxide anion radical (?O2-) and nitric oxide (NO) are cytotoxic because they can oxidize several cellular components such as proteins, lipids and DNA. They have been implicated in the aging process, and age-related diseases such as Alzheimer's disease, rheumatoid arthritis, cancer and atherosclerosis. The aim of this study was to investigate Sabohwan's activity for scavenging ONOO- and its precursors. NO and ?02-. Methods : For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4.5-diaminofluorescein (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Results : Sabohwanblocked tert-butylhydroperoxide (t-BHP)-induced cell death in a dose-dependent fashion. It scavenged t-BHP-induced ONOO-, NO and ?O2- in YPEN cells. Sabohwan inhibited the generation of ONOO-, NO and ?O2- in the lipopolysaccharide (LPS)-treated mouse kidney postmitochondria both in vitro and in vivo. The lipid peroxide level increased and glutathione level decreased in the LPS-treated mice, whereas the ones in the Sabohwanadministered group among the LPS-treated mice reversed toward their natural levels. Conclusions : These results suggest that Sabohwanis an effective ONOO-, ?O2- and NO scavenger, and thereby it might have a potential role as a therapy against the aging process and age-related diseases.

Effects of Short-term Thermal Stress on the Mouse Serum Concentrations of Cortisol and Dehydroepiandrosterone Sulphate (단기 고온 스트레스가 마우스 혈청 Cortisol, Dehydroepiandrosterone Sulphate 농도에 미치는 영향)

  • This study was carried out to investigate the effect of short-term thermal stress on the serum concentrations of cortisol and DHEAS in BALB / c male mice. Cortisol and DHEAS concentrations in serum were measured by radioimmunoassay(RIA). We found there were significantly increased in the cortisol levels in 30 min-stressed group(T30) compared with control group(p<0.01), and then declined without significance in 120 min-stressed group (T120) compared with T30. By contrast, DHEAS levels were decreased without significance in both T30 and T120 compared with control group. Though short-term thermal stress, the continuous decline of DHEAS levels were observed. These results show that short-term thermal stress affects the serum levels of cortisol and DHEAS in mice. Furthermore, we found that DHEAS is a stress-related hormone and will be able to utilize as a stress marker.

  • Objective : The study was aimed to evaluate the anti-stress effect of KYT on the cold-stressed Mice. Methods: The experimental animals were stressed in cold room $(4{\pm}0.5^{\circ}C)$ for one hour in a day during two weeks, and administred 131mg/100g KYT extract for two weeks before stress. For two weeks, while the experiment was taking place, we conducted a research about the change of weight and content of norepinephrine, epinephrine, dopamine, cortisol and GOT, GPT in the mice serum. Results : KYT extract significantly inhibited the decrease of body weight induced by stress, compared with the control group. KYT extract had no significant decreasing effect on the change of content of the serum GOT but had significant decreasing effect on the change of content of the serum GPT compared with the control group. KYT extract had significant decreasing effect on the change of content of the serum cortisol compared with the control group. KYT extract had decreasing effect on the change of content of the serum dopamine compared with the control group. KYT extract had no decreasing effect on the change of content of the serum norepinephrine compared with the control group. KYT extract had significant decreasing effect on the change of content of the serum epinephrine compared with the control group. Conclusions : According to the above results, it is concluded that KYT will be useful as a preventive and remedy against stress disease.

  • In the present study, chronic effects of the hot water extracts of walnut seed (Juglans regia L.) (WSE) and Nelumbo nucifera seed (NSE) were investigated in mice exposed to 2 hr of restraint stress each day for 4 weeks. Corticosterone levels in serum were significantly increased in the vehicle-treated stressed group ($25\;{\mu}g/dL$) compared to that in the control group ($13\;{\mu}g/dL$). This stress induced gastric redness and lesions. However, treatment with WSE and/or NSE significantly protected the stomach from this lesion by 50-60% compared to that in the vehicletreated group. In the amygdala, the administration of WSE and/or NSE also reduced the immediate early gene (c-fos) expression by 70-90% vs. the vehicle-treated group. These suggest that WSE and/or NSE may reduce the appearance of symptoms induced by stress and these materials are useful as anti-stress foods, as natural products tend to be relatively safe compared to chemical products.

Effects of the Guibi-tang and Yishingyojae-dan on the Regional Brain Monoamines Contents of Immobilization Stressed Mice (귀비탕(歸脾湯)과 이신교제단(二神交濟丹)이 구속(拘束)스트레스 생쥐의 뇌부위별(腦部位別) Monoamines 함량(含量)에 미치는 영향(影響))

  • Objectives : This study aimed to evaluate the anti-stress effects of Guibi-tang and Yisiungyojae-dan on the contents of monoamines in the regional brain of mice immobilized stress. Methods : The experimental animals were immobilized in stress cylinder(height: 15cm, diameter: 3cm) for 15 minutes, and administered of Guibi-tang(6.0mg/10g) and Yishingyojae-dan(9.3mg/10g) water extracts for 7 days before stress. The monoamines contents were measured by HPLC method in various part(frontal cortex, hypothalamus, corpus striatum and hippocampus) of mice brain. Results: 1. In frontal cortex, the contents of norepinephrine were decreased with statistical significantly in Yishingyojae-dan administered group compared to control group. The contents of serotonin were decreased with statistical significance in all of the administered group compared to control group. 2. In hypothalamus, the contents of norepinephrine were decreased with statistical significantly in Yishingyojae-dan administered group compared to control group. The contents of serotonin were decreased with statistical significance in all of the administered group compared to control group. 3. In corpus striatum, the contents of dopamine were decreased with statistical significantly in Yishingyojae-dan administered group compared to control group. The contents of serotonin were decreased with statistical significance in all of the administered group compared to control group. 4. In hippocampus, the contents of serotonin decreased with statistical significantly in Yishingyojae-dan administered group compared to control group. Conclusions: This study shows that Guibitang and Yishingyojae-don are significantly effective on reducing and preventing stress in mice. In addition, Yishingyojae-dan is more clear effective than Guibi-tang on the monoamines change in the mice brain.

Methods : The experimental animals were forced cold swimming stress for 3 minutes, and administered of Ansymondam-tang(2.2mg/10g) and Gamiondam-tang(0.83mg/10g) extract for 7 days before stress. The monoamines contents were measured by HPLC method in various part(frontal cortex, hypothalamus, corpus striatum and hippocampus) of mice brain. Results: 1. In frontal cortex, the contents of norepinephrine were increased with statistical significantly in all of the administered group compared to control group. The contents of dopamine and serotonin were increased in Gamiondam-tang administered group has a statistical significance. 2. In hypothalamus, the contents of dopamine were increased with statistical significance in all of the administered group compared to control group. 3. In corpus striatum the contents of norepinephrine were decreased and the contents of dopamine were increased with statistical significance in all of the administered group compared to control group. The contents of serotonin were increased in Gamiondam-tang administered group has a statistical significance. 4. In hippocampus the contents of norepinephrine were decreased with statistical significance in Gamiondam-tang administered group compared to control group. The contents of dopamine were increased in Ansymondam-tang administered group has a statistical significance. Conclusion : This study reaches a conclusion that Ansymondam-tang and Gamiondam-tang has significant effects .on reducing and preventing stress in mice. Especially Gamiondam-tang is more effective in the statistical significance than Ansymondam-tang on the monoamines change in the mice brain.

Immunopotentiating Effect of Protein Extract from Dioscorea quinqueloba in Stressed Mice (단풍마 단백질 추출물의 스트레스로 인한 면역력 저하 개선 효과)

  • It is noted that Dioscorea quinqueloba is a medicinal herb that is widely used to treat cardiovascular disease and is assessed as useful to treat other various medical conditions. The immunopotentiating effects of the protein extract (DQP-1) from Dioscorea quinqueloba were thus formally investigated in vivo under incident of cold stress. In this case study, the spleen and thymus weight in mice was shown to have decreased after a measured exposure to cold stress, while the adrenal gland weight in the mice was shown to have increased. The systematic oral administration of DQP-1 significantly recovered the weight loss of the spleen and suppressed the adrenal gland hypertrophy during the association with cold stress. Additionally, the DQP-1 also restored the ascorbic acid level in the adrenal gland reduced after cold stress. The cold stress exposure lowered the percentage of $CD4^+$ and $CD8^+$ cells in the mouse thymus as determined by the flow cytometric analysis, as well as the levels of some serum immunological cytokines(interleukin-2, interleukin-12, and interferon-${\gamma}$) in the studied mice. The resulting identified weakened immunity caused by cold stress was also recovered by a treatment with DQP-1. The DQP-1 significantly suppressed the formation of serum enzymes of alanine aminotransferase, aspartate aminotransferase, and lactate dehydrogenase, which were systematically elevated during the cold stress episode. These results indicate that DQP-1 can improve immunity in mice that are characteristically weakened under stress.

The Effect of Stress During Pregnancy on the Skin Barrier of Mice Four Weeks After Birth (임신 중 만성스트레스가 생후 4주 유서(幼鼠)의 피부장벽에 미치는 영향)

  • Background and Objectives: The skin barrier protects skin against a harmful environment. Its function includes an antimicrobial barrier as well as a physical barrier. Stress is harmful to the skin barrier and there are many studies on this, but there are few about the effect of stress during pregnancy on the skin barrier of offspring. The aim of this study was to investigate the effect of stress during pregnancy on the skin barrier of offspring by examination with the naked eye, cortisol, weight, TEWL and histologic tests. Materials and Methods: Male hairless mice ten weeks old were coupled with females for three days. After twelve days the females were divided into two groups. We stressed one group and not the other group. The offspring from the non-stressed (N group) and stressed (S group) grew up until four weeks. The S group was exposed to chronic mild stress using the chronic stress model protocol modified method of Wilner and Towell for a week. We made examinations with the naked eye, assessment of cortisol, weight, TEWL, and histologic test to contrast the S group with the N group. Statistical analysis was performed by using Mann-Whitney and Friedman test. Statistical significance was achieved if the probability was less than 5% (p<0.05). Results: 1. By making an examination with the naked eye, S group showed papules, creases and dryness on their skin, but N showed no change compared with normal skin. 2. There was no statistically significant difference between S group and N group in cortisol (p>0.05). 3. There was no statistically significant difference between S group and N group in weight. N group had less weight than S group (p<0.05). 4. There was no statistically significant difference between S group and N group in TEWL (p>0.05). 5. There was no statistically significant difference between S group and N group in TEWL recovery (p>0.05). 6. In TEWL and TEWL recovery, there was statistically significant difference by time between S group and N group (p<0.005). Histologically, S group showed a thicker epidermis and epidermal crest than N group. Conclusions: We found that stress during pregnancy has negative effects on the skin barrier of offspring. Though there were no statistically significant differences between S group and N group in TEWL recovery, we found evidence that the recovery of skin barrier function is interrupted by stress during pregnancy by examination with both the naked eye and histology.

Effect of Tremella fuciformis Berk on Anti-stress activities during Long-Term and Short-Term in Mice (마우스에서 흰목이버섯 (Tremella fuciformis Berk)의 장.단기적 항스트레스 효과)

  • The objective of this study was to evaluate the protective effect of aqueous extracts from Tremella fuciformis Berk(Tf AE) against stress during long-term and short-term in ICR mice. All the animals were randomly divided into two groups which had been bred for 5 months that were treated by immobilization stress for 8 weeks (total 7 months breeding, equivalent to human beings aged 20) with or without Tf AE, and one out of two groups was continuously bred until they become 18 months old (equivalent to human beings aged 60) without Tf AE. Afterwards, the changes of serum and hepatic metabolites were investigated on the basis of the index of stress-related in vivo oxidative damage. As a result, it was found that stress increases serum triglyceride (TG) and aspartate aminotransferase (AST) and decreases serum HDL-cholesterol in the long-term (total 18 months breeding) and short-term (total 7 months breeding). In addition, stress concerned the decrease of total antioxidant status (TAS) and superoxide dismutase (SOD) as well as the increase of malondialdehyde (MDA) in liver. On the other hand, Tf AE-fed groups reversed all these biochemical indices. These results suggest that stress in one's youth causes negative results in TG, HDL-cholesterol, LDL-cholesterol, AST, TAS, SOD and MDA measured in one's senescent. The administration of Tf AE in the stressed mice decreases serum TG and AST that are increased by stress, and exerts influence on the increase of serum HDL-cholesterol. Also Tf AE recovered the values of liver TAS, SOD and MDA in the stressed mice. In conclusion, Tf AE represented protective effect in the stressed mice to some degree.

Effects of Sumsu (Bufonis venenum) Pharmacopuncture Treatment on Depression in Mice

  • Objectives: The main objective of this study was to evaluate the anti-depressant effects of pharmacopuncture using sumsu (Bufonis venenum). Methods: Animals were divided into three groups (control, sham, and experimental), with eight mice per group. The sham and the experimental groups were exposed to 2 hours of immobilization stress daily for 14 days. They were also injected with normal saline (sham) or subjected to pharmacopuncture with sumsu at the acupoints HT7, SP6, and GV20 (experimental). The depression or anxiety-like behaviors of the mice in each group were evaluated 1 day after treatment. Results: There was no difference in locomotor activity between the groups during the open-field test; i.e., all groups had normal motor function. However, the open-field and the forced-swimming tests revealed that depression and anxiety-like behaviors were decreased significantly in the group treated with sumsu pharmacopuncture. Conclusion: Sumsu pharmacopuncture attenuated depressive or anxiety-like behavior in mice stressed with chronic immobilization. These results suggest that sumsu pharmacopuncture has therapeutic potential for treating neuropsychiatric disorders such as anxiety or depression disorder.
