소음 스트레스가 면역반응에 미치는 영향에 관한 실험적 연구 (Effect of Sound Stress on Immune Response)

  • This study was undertaken to assess the effect of sound stress on humoral and cellular immune responses to thymus-dependent and independent antigens in mice. After mice were exposed to 4 hr daily sound stessors(83㏈) for 4 days before or after immunization, the primary and / or secondary immune response to sheep red blood cells(SRBC), polyvinylpyrroridone(PVP) or picry1 chloride(TNCB) were assayed. When mice were exposed to sound stressor before or after immunization, delayed-type hypersensitivity reaction and contact sensitivity to TNCB was remarkably depressed compared with those of the unstressed control mice. However, the primary and secondary hemagglutinin response of the stresed mice to SRBC showed a pronounced increase compared with that of the unstressed mice, In contrast to antibody response to SRBC, the primary antibody response of the stressed mic to PVP was almost not detected. surprisingly, the secondary antibody response to PVP of the mice receiving the secondary sound stress was markedly increased when the immune-depressed mice received the secondary immunization with PVP at 46 days after the primary immunization. The susceptibility of mice to intraven-oulsy infected Candida albicans was not changed by the sound stress.

소음인(少陰人) 소합향원(蘇合香元)이 Stress 생쥐의 뇌(腦) Serotonin 함량(含量)에 미치는 영향(影響) (A Study on Effect of Soeuminsohabhyangwon on the Brain Serotonin contents of Stressed Mice)

  • This study was aimed to evaluate the anti-stress effect of Soeuminsohabhyangwon on the mice in Cold and Swimming stress.In order to investigate the anti-stress effect of Soeuminsohabhyangwon in Cold and Swimming stressed mice, the serotonin contents were measured by HPLC method in various part of mouse brain The following results were observed. 1. In Cerebral Cortex of Frontal Lobe, the serotonin content was decreased in the Control group as compared with Normal group and the serotonin content was increased in the SHW group as compared with Control group. 2. In hypothalamus, the serotonin content was decreased in the Control group as compared with Normal group and the serotonin content was increased with statistical significance in the SHW group as compared with Control group. 3. In corpus striatum, the serotonin content was decreased in the Control group as compared with Normal group and the serotonin content was increased with statistical significance in the SHW group as compared with Control group. 4. In hippocampus, the serotonin content was decreased in the Control group as compared with Normal group and the serotonin content was increased with statistical significance in the SHW group as compared with control group.Base on the above results, it may be concluded that Soeuminsohabhyangwon are effective to reduce stress.

두중(杜仲)의 수치(修治)에 따른 실험적(實驗的) 연구(硏究) (EXPERIMENTAL STUDIES OF EUCOMMIAE CORTEX ACCORDING TO PROCESSING)

  • In order to estimate the effects of Eucommiae Cortex, which is one of the most important herb medicines used for invigorating the liver and kidney and strengthening ones and muscles, fetus-soothing etc., this experiment was conducted the quantitative analysis of geniposidic acid and geniposide by HPLC, and the analgesic effects of mice, the lipid metabolism of rats, the catecholamine concentration in the brain and the plsma of rats stressed by immobilization. The results obtained are as follows. 1. The contents of geniposidic acid and geniposide were 22.3mg/g, 5.4mg/g in Eucommiae Cortex without processing, and 119.8mg, 10.4mg/g in Eucommiae Cortex fried with salts. 2. In analgesic effects of mice, there were all significant in Eucommiae Cortex without processing and Eucommiae Cortex fried with salts. 3. In the effects of lipid metabolism of rats, there were not significant on the change of organ weight and the level of serum total lipid and total cholesterol, triglyceride, phospholipid, HDL-cholesterol. But, there were significant on the level of free fatty acid. 4. In tile effects of the catecholamine concentration in the brain and the plsma of rats stressed by immobilization, there were all significant in Eucommiae Cortex without processing and Eucommiae Cortex fried with salts, especially in case of norepinephrine. From the results mentioned above, there were significant on the analgesic effects of mice, and the catecholamine concentration in the brain and the plsma of rats stressed by immobilization. And there were different from Eucommiae Cortex without processing and Eucommiae Cortex fried with salts in the contents of geniposidic acid and geniposide. These results are relative to invigorating the liver and kidney and strengthening ones and muscles, and fetus-soothing, therefore when we use Eucommiae Cortex, it is more desirable to use Eucommiae Cortex with processing.

제비콩(Dolichos lablab L.) 잎 추출물이 쥐의 혈청효소활성에 미치는 영향 (Effect of Jebikong(Dolichos lablab L.) Leaves Extract on Enzyme Activities in Mice Blood)

  • The effect of Dolichos lablab leave extract on enzyme activities in mice blood was investigated in this study to clarify the new useful application of the Plant leaves. There were not significant differences in the enzyme activities in mice blood among treatment fed with the leave extract and non fed control. The feeding treatment with the extract showed a tendency to activity compared to the nun fed control. The activities of glutamate oxaloacetate transaminase, glutamate pyruvate transaminase and lactate dehydrogenase in mice blood at the treatment fed with lead and the leave extract were significantly low compared to their activities at the non fed treatment with the extract, respectively. The choline esterase activity was high at the leave extract feeding. The cadmium dietary treatment showed the same result as the lead treatment. In conclusion, the physiological function of the Jebikong leave was significantly in creased when the mice was stressed by the hear metal intake. Therefore, the plant leave extract would consider the reduction of heart metal effect.

마우스에서 vitamin E의 장.단기적 항스트레스 효과 (Anti-stress Effect during Long and Short-Term of Vitamin E in Mice)

  • 본 연구에서는 일정기간 스트레스를 받을 때 vitamin E 투여가 스트레스를 얼마나 장 단기적으로 방어할 수 있는지 알아보았다. ICR 마우스를 사람의 나이 20대에 해당되는 5개월까지 사육한 후, 8주간의 스트레스를 가하고 사람의 나이 노년기에 해당되는 18개월 될 때까지 사육하여 스트레스로 인한 생체 내의 산화적 손상지표로 혈청과 간의 대사산물의 변화를 조사하였다. 그 결과, 스트레스는 장 단기적으로 모두 혈청의TG, AST 함량을 증가시키고, HDL-cholesterol 함량을 저하시키는 것에 반해, 여기에 vitamin E의 투여는 스트레스를 받은 쥐에서 혈청 TG와 AST의 함량을 감소시키고, HDL-cholesterol의 함량을 증가시키는 효과를 보여주었다. 스트레스는 간조직의 TAS, SOD 함량 감소와 MDA 함량 증가를 야기시켰으며, 스트레스 받은 쥐에 vitamin E의 투여는 TAS, SOD의 함량을 증진시키고, MDA의 함량을 저하시키는데 긍정적 영향을 주었다. 따라서 젊었을 때 한때 받은 스트레스는 노년기의 TG, HDLcholesterol, AST, TAS, SOD 그리고 MDA의 수치 모두에 있어서 부정적인 영향을 미치는 것으로 나타났다. 이러한 결과를 통해서 한때 어느 기간 받은 스트레스는 시간이 지난 노년기까지 혈청과 간조직에 부정적 영향을 주며 vitamin E는 이를 어느 정도 보호하는데 긍정적 인 효과를 나타낸다는 것을 알 수 있었다.

실험적 마우스 모델에서 예측 불가능한 스트레스가 황체형성호르몬 수용체의 발현과 생식기능에 미치는 영향에 관한 연구 (The Effects of Unpredictable Stress on the LHR Expression and Reproductive Functions in Mouse Models)

  • 본 연구의 목적은 예측 불가능한 만성적인 스트레스가 생식기능과 황체형성호르몬 수용체의 발현에 미치는 영향을 알아보는 것이다. 9 주령 암컷 C57BL/6 마우스를 무작위로 선택하여 대조군과 스트레스군의 두 집단으로 분류하였다. 스트레스군은 35일 동안 하루에 두 번씩 12가지의 서로 다른 스트레스를 무작위로 선택하여 스트레스를 주었다. 대조군에 비하여 스트레스군에서 불안과 관련된 행동들이 유의성 있게 증가하였으며(P < 0.05), 스트레스를 받는 동안의 증체율 또한 유의성 있게 감소하였다(P < 0.01). 그리고 평균 산자수도 대조군에 비하여 스트레스 군에서 유의하게 감소함을 관찰 하였다(P < 0.01). 조직학적인 검사에서 일차, 이차 및 초기 성숙 난포의 비율이 대조군에 비해 스트레스 군에서 유의하게 감소한 반면(P < 0.05) 폐쇄 난포의 비율은 유의하게 증가하였다(P < 0.01). 면역조직화학적 검사를 통해 과립막세포와 황체세포의 황체형성호르몬 수용체 발현을 관찰한 결과, 대조군에 비해 스트레스군에서 그 발현이 감소하였고, 웨스턴 블롯을 통해 난소 내 황체형성호르몬 수용체의 단백질 양을 측정한 결과 또한 대조군에 비하여 스트레스군에서 유의하게 감소하였다(P < 0.05). 본 연구를 통해 난소의 황체형성호르몬 수용체는 예측 불가능한 스트레스에 의해 영향을 받으며, 변화된 황체형성호르몬 수용체가 난자의 난포 발육 불량과 생식기능의 이상에 영향을 미친다는 것을 실험적으로 증명하였다.

양신탕(養神湯)의 항(抗)스트레스 효과(效果)에 대(對)한 실험적(實驗的) 연구(硏究) (The Experimental study on the Anti-Stress effect of Yangsintang(YST))

  • Objective: The study aimed to evaluate the anti-stress effect of YST on the cold-stressed Mice. Method: The experimental animals were stressed in cold room ($4{\pm}0.5^{\circ}C$) for 1 hour in a day during 2weeks, and administerd 115mg/100g YST extract for 2weeks before stress. For 2weeks, we conducted a research about the change of weight and content of norepinephrine, epinephrine, dopamine, cortisol and GOT, GPT in the mice serum. Results: YST significantly inhibited the decrease of body weight induced by stress, compared with the control group. YST had no significant decreasing effect on the change of content of the serum GOT But decreased the content of the serum GPT. YST decreased the content of the serum cortisol. YST decreased the content of the serum dopamine a little and decreased the content of the serum epinephrine significantly. Conclusion: According to the above results, it is concluded that YST will be useful as a remedy against stress disease.

단기 속박스트레스가 마우스 혈청 Cortisol, Dehydroepiandrosterone Sulphate 농도에 미치는 영향 (Effects of Short-term Immobilization Stress on the Mouse Serum Concentrations of Cortisol and Dehydroepiandrosterone Sulphate)

  • 단기 속박스트레스가 혈청 cortisol과 DHEAS 농도에 미치는 영향을 알아보고자 단기 속박스트레스 (IS)를 받은 BALB / c 숫컷 생쥐의 혈청 cortisol과 DHEAS 농도를 방사선면역측정 (RIA)을 이용하여 분석하였다. 그 결과 cortisol의 경우 30분 IS를 받은 그룹 (Ⅰ-30N)에서 대조군에 비해 유의적인 증가를 보였으며 (p<0.01), 120분 부과 (Ⅰ-120N) 시에도 대조군에 비해 유의적인 증가를 보였다. 가시오갈피 (SG) 처리군에서도 30분 IS를 받은 그룹 (Ⅰ-30T)에서 대조군에 비해 유의적 증가를 보였고 (p<0.01), 120분 부과(Ⅰ-120T) 시에도 대조군에 비해 유의적으로 증가하는 경향을 나타내었다. SG 처리군과 비처리군 모두에서 cortisol의 계속적인 증가를 관찰할 수 있었다. 30분 부과 그룹에서 SG 비처리군 (Ⅰ-30N)에 비해 SG 처리군 (Ⅰ-30T)에서 cortisol 농도의 유의적인 감소를 나타내었고, 120분 부과 그룹에서도 SG 비처리군 (Ⅰ-120 N)에 비해 SG 처리군 (Ⅰ-120T)에서 cortisol 농도의 유의적인 감소를 나타내었다. 이에 반해, DHEAS의 경우 30분 속박 (Ⅰ-30N)시 대조군에 비해 유의성 없는 감소 경향을 보였으나, 120분 속박 (Ⅰ-120N)시 대조군에 비해 유의적인 감소를 나타내었다 (p<0.01). SG 처리군에서도 30분 속박 (Ⅰ-30T) 시 대조군에 비해 유의성 없는 감소 경향을 보였으나, 120분 속박 (Ⅰ-120T)시 대조군에 비해 유의적인 감소를 나타내었다 (p<0.01). 그러나 30분, 120분 부과그룹 모두에서 SG 처리에 대한 유의적 차이는 없었다. 따라서 본 연구를 통하여 단기 속박이라는 국한된 스트레스에도 불구하고 DHEAS가 지속적으로 감소됨으로써 단기 속박스트레스가 혈청 cortisol과 DHEAS에 영향을 미치고 있음을 알 수 있었으며, 이를 통하여 DHEAS가 스트레스 관련 호르몬임을 확인할 수 있었다.

돌외 에탄올 추출물 엑스의 만성 스트레스-유도 불안작용에 대한 개선작용 (Ameliorating Effects of Herbal Ethanol Extract from Gynostemma pentaphyllum on Chronic Stress-Induced Anxiety in Mice)

  • The effects of herbal ethanol extract from Gynostemma pentaphyllum (GP extract) on chronic stress-induced anxiety in mice were investigated. The animals were treated with GP extracts (50 and 100 mg/kg/day, p.o.) for 21 days before exposure to electric footshock (EF; duration and interval 10 sec for 3 min, 2 mA) for chronic stress once a day. The ambulatory locomotor activity was reduced by chronic EF stress and it was recovered by 12.9-15.1% in GP extract-treated groups. The grip strength was also significantly decreased by chronic EF stress, however, the EF-stressed groups treated with GP extract increased grip strength from 13.9% to 56.8% compared to EF-stressed groups. In addition, the serum levels of corticosterone were significantly elevated by chronic EF stress to 197% of the control levels, which was reduced to 73.1% by treatment with GP extract (100 mg/kg). In contrast, the brain levels of dopamine and serotonin were reduced to 67.6% and 63.1% by chronic EF stress, which was recovered to 90% of the control levels by treatment with GP extract. These results indicate that GP extract shows the ameliorating effects on chronic EF stress-induced anxiety in mice and it can be developed as the promising anti-anxiety agent.
